  • Essential oils of the musk from wild muskrat (Ondatra zibethicus) were analyzed by gas chromatography/mass spectrometry. Fifty-six chemical components were separated, and 14 volatile compounds (78% of composition of total essential oil) of macrocyclic musk, mainly cyclopentadecanone, 12-hydroxy-14-methyl-oxa-cyclotetradec-6-en-2-one, cycloheptadecanone and 9-cycloheptadecen-1-one, were identified by the comparison of Wiley/NBS Library from the results of GC-MS. However, l-muscone, a compound reported by earlier workers in the musk from muskrat, was not detected using the spike test in this study. Interestingly, cyclohexadecanone, which has the same molecular weight (Mw 238) as l-muscone, was identified. In addition, it is considered that because of possessing much higher amounts of macrocyclic components compared to the original musk, muskrat has higher economic values in perfume and pharmaceutical industries.

  • Mugwort (Artemisia montana), which is a perennial plant mainly distributed throughout Northeast Asian regions, has been used as a preferred source of various foods and traditional medicines in Korea. In particular, as essential oils extracted from mugwort were reported to be biologically active, its steam distillate has been widely used to treat various conditions, such as itching, hemorrhoids, and gynecological inflammation. Therefore, efforts have been devoted to develop effective methods for the collection of bioactive essential oils from mugwort. In this study, five mugwort extracts were obtained using different extraction conditions, namely, 6 % ethanol at room temperature and at 80 ℃, pure ethanol, n-hexane, and an adsorbent resin. To evaluate the five extracts of mugwort, area-under-the-curve values (AUCs), chemical profiles, and major bioactive essential oil contents were investigated using gas chromatography-mass spectrometry (GC-MS). An overall assessment of the volatile components, including essential oils, in the five extracts was conducted using AUCs, and the individual essential oil in each extract was identified. Furthermore, the four major essential oils (1,8-cineole, camphor, borneol, and α-terpineol), which are known to possess anti-microbial and anti-inflammatory activities, were quantified using authentic chemical standards. Based on the evaluation results, pure ethanol was the best extractant out of the five used in this study. This study provides evaluation results for the five different mugwort extracts and would be helpful for developing extraction methods to efficiently collect the bioactive oil components for medical purposes using chemical profiles of the extracts.

  • 본 연구의 목적은 열안정성을 향상시키는 첨가제 (열안정제)를 이용하여 exo-tetrahydrodicyclopentadiene (exo-THDCP, $C_{10}H_{16}$)의 열안정성을 향상시키는 것이다. 실험 중 실시간으로 추출한 시료를 gas chromatography-mass spectrometry (GC-MS)를 이용해 성분 분석하여 첨가제 성능-exo-THDCP의 열분해성-을 확인하고, 반응시간에 따른 exo-THDCP의 분해생성물의 성분 및 조성 변화를 통해 첨가제의 작용 메커니즘을 규명하였다. 열안정제로써 수소공여체를 사용하여 실험한 결과 1,2,3,4-tetrahydroquinoline (THQ), benzyl alcohol (BnOH) 등을 첨가하였을 때 exo-THDCP의 열안정성이 향상되었다. 이러한 수소공여체 물질들은 개시반응이 진행된 exo-THDCP 라디칼에 수소원자(hydrogen radical)를 제공함으로 라디칼의 반응성을 완화시켜 1차 생성물의 활성을 감소시키어 2차생성물질인 $C_{11}$이상 생성물의 발생을 감소시키는 것으로 밝혀졌다.

자기치유성 마이크로캡슐 합성 공정에서의 포름알데히드 잔류량 연구 (A Study on Remaining Formaldehyde Concentration in the Synthesis of Self-Healing Microcapsules)

  • 우레아-포름알데히드(UF) 캡슐막의 마이크로캡슐 합성 공정에 있어서 반응 폐액에 포함된 포름알데히드의 잔류량을 GC-MS로 분석하였다. 포름알데히드의 반응에 영향을 주는 주된 인자로서 pH, 암모늄클로라이드 투입량, 온도의 3가지를 선정하고, 이 인자들이 반응 폐액 중의 포름알데히드 농도에 미치는 영향을 조사하였다. 실험 조건 중 암모늄클로라이드를 0.025g 투입한 경우에는 캡슐막 형성이 안 되거나 캡슐막이 약한 경향이 있어서 기본적으로 이 조건은 마이크로캡슐화에 부적절한 것으로 판단된다. 본 연구의 실험 조건에서 포름알데히드 잔류량이 최소화되는 마이크로캡슐화 조건은 70℃의 온도와 2.5의 pH에서 암모늄클로라이드를 0.050g 투입하는 조건임이 확인되었다. 본 연구의 결과는 보다 안전한 작업환경에서 마이크로캡슐화를 수행하는데 기여할 수 있을 것으로 기대된다.

자외선 열화 전후 건조 옻의 용매별 용출 특성 (Extraction Characteristics of Dried Asian Lacquer by Solvents Before and After UV Degradation)

  • 건조칠기 유물 세척 시 사용되는 다양한 용매을 대상으로 클리닝 효과를 비교하기 위해 실험을 진행하였다. 건조칠과 자외선에 24일 노출시킨 건조칠에 물, 에탄올, 아세톤, 헥산을 가하여 녹아 나오는 물질을 열분해/GC/MS로 검출하였다. 용출되는 양은 아세톤과 에탄올에서 많았으며, 극성이 높은 용매인 물은 저분자량 용출에 비교적 효율이 좋았다. 비극성 용매인 헥산은 용출량이 매우 낮았다. UV에 24일 노출되어 열화가 많이 진행된 건조칠은 노출되기 전 건조칠에 비해 용출되는 물질이 크게 증가하였다. 열화가 되면서 고분자 형태의 우루시올 사슬이 절단되고 산화되는 과정을 거쳐 dicarboxylic acid를 포함한 저분자량의 극성 물질을 생성하기 때문인 것으로 보인다. 이와 같은 용출특성은 칠기유물을 클리닝하는 용매를 선택할 때 유물의 열화 상태와 더불어 중요한 참고자료가 될 것이다.

육류가공식품의 조리방법에 따른 PAHs 발생 특성에 관한 연구 (Characteristics of PAH Occurrence during Meat Cooking)

  • In this study, the concentrations of polycyclic aromatic hydrocarbons (PAHs) were investigated in meat process food and for cooking methods (pan-frying and charcoal fire). The methodology involved liquid-liquid extraction, silica gel cartridge clean-up and determination by gas chromatography/mass spectrometry (GC/MS). The recovery of 17 PAHs spiked into these samples ranged from 66.6 % to 98.0% and the coefficient of variation was less than 10%, but that of dibenz(a,h)anthracene was 16.39%. The mean concentration of total PAHs in processed samples was ND~7.2 ng/g, whereas that pan-fried and charcoal-fired samples were ND~22.1 ng/g and, 12.7~367.8 ng/g, respectively. Therefore, the concentrations of total PAHs in cooked samples were higher than in original samples and charcoal-fired samples had the highest total PAH levels.

  • BACKGROUND: Garlic (Allium sativum) contains polyphenols and sulfur compounds that are recognized as antioxidant, antithrombotic, anticancer, antibacterial, antimicrobial, nematicidal, and insecticidal activity. For this reason, the Environmentally-friendly Agriculture Promotion Act allowed the garlic extract as commercial biopesticide material for crop protection, nine commercial biopesticides containing A. sativum extract have been marketed in Korea. METHODS AND RESULTS: The determination of allylmethyl sulfide (AMS), dimethyl disulfide (DMDS), and dipropyl sulfide (DPS) in biopesticides containing A. sativum extract was developed and validated by gas chromatography (GC) mass spectrometry (MS) with head-space sampler. The developed method was validated, and the limit of quantification (LOQ) and recovery rates of AMS, DMDS, and DPS were 0.08, 0.32, and 0.09 mg/L and 90.3-91.3, 86.2-88.3, and 87.6-89.5%, respectively. Anti-nociceptive and anti-inflammatory activities of the essential oil isolated from Cupressus arizonica Greene fruits

  • Background: Cupressus arizonica Greene is a coniferous tree with great importance in fragrance and pharmaceutical industries. Essential oils from C. arizonica (EC) have shown potential antioxidant, and anti-microbial activities. This study aimed at investigating the anti-nociceptive and anti-inflammatory effects/mechanisms of EC. Methods: The EC was evaluated for anti-nociceptive and anti-inflammatory activities on male Wistar rats using a formalin test and carrageenan-induced paw edema, respectively. Also, we pre-treated some of the animals with naloxone and flumazenil in the formalin test to find out the possible contributions of opioid and benzodiazepine receptors to EC anti-nociceptive effects. Finally, gas chromatography/mass spectrometry (GC/MS) analysis was used to identify the EC's constituents. Results: EC in intraperitoneal doses of 0.5 and 1 g/kg significantly decrease the nociceptive responses in both early and late phases of the formalin test. From a mechanistic point of view, flumazenil administration 20 minutes before the most effective dose of EC (1 g/kg) showed a meaningful reduction in the associated anti-nociceptive responses during the early and late phases of the formalin test. Naloxone also reduced the anti-nociceptive role of EC in the late phase. Furthermore, EC at the doses of 1, 0.5, and 0.25 g/kg significantly reduced paw edema from 0.5 hours after carrageenan injection to 4 hours. GC/MS analysis showed that isolated EC is a monoterpene-rich oil with the major presence of α-pinene (71.92%), myrcene (6.37%), δ-3-carene (4.68%), β-pinene (3.71%), and limonene (3.34%). Conclusions: EC showed potent anti-nociceptive and anti-inflammatory activities with the relative involvement of opioid and benzodiazepine receptors.

Elicitation of Innate Immunity by a Bacterial Volatile 2-Nonanone at Levels below Detection Limit in Tomato Rhizosphere

  • Bacterial volatile compounds (BVCs) exert beneficial effects on plant protection both directly and indirectly. Although BVCs have been detected in vitro, their detection in situ remains challenging. The purpose of this study was to investigate the possibility of BVCs detection under in situ condition and estimate the potentials of in situ BVC to plants at below detection limit. We developed a method for detecting BVCs released by the soil bacteria Bacillus velezensis strain GB03 and Streptomyces griseus strain S4-7 in situ using solid-phase microextraction coupled with gas chromatography-mass spectrometry (SPME-GC-MS). Additionally, we evaluated the BVC detection limit in the rhizosphere and induction of systemic immune response in tomato plants grown in the greenhouse. Two signature BVCs, 2-nonanone and caryolan-1-ol, of GB03 and S4-7 respectively were successfully detected using the soil-vial system. However, these BVCs could not be detected in the rhizosphere pretreated with strains GB03 and S4-7. The detection limit of 2-nonanone in the tomato rhizosphere was 1 µM. Unexpectedly, drench application of 2-nonanone at 10 nM concentration, which is below its detection limit, protected tomato seedlings against Pseudomonas syringae pv. tomato. Our finding highlights that BVCs, including 2-nonanone, released by a soil bacterium are functional even when present at a concentration below the detection limit of SPME-GC-MS.
