SPME-GC-MS를 이용한 담배와 관련된 향료의 분석 (Analysis of Flavor-related Compounds from Tobacco using SPME-GC-MS)

  • 담배에 포함된 향료 성분을 headspace solid phase microextraction(SPME)를 이용하여 gas chromatography-mass spectrometry(GC-MS)의 selected ion monitoring(SIM)방법으로 분석하였다. 본 연구에서 사용한 담배향과 관련된 성분은 estragole, pulegone, trans-anethole, safrole, piperonal, eugenol, methyleugenol, coumarin, trans-isoeugenol, trans-methylisoeugenol 및 myristicin 등 11종이며 분석결과 모든 담배에서 한가지 또는 그 이상의 담배향 성분을 $0.001-1.3{\mu}g/g$ 검출할 수 있었으며 회수율은 89.1-102.9%로 나타났고 상대표준편차는 2.6-25.2%를 얻었다.

쑥에 함유된 monoterpenes 의 함량과 조성의 계절적 변이 (Seasonal Variation in Concentration and Composition of Monoterpenes from Artemisia princeps var. orientalis)

  • The profile and concentration of monoterpene metabolites in the leaf and stem of Artemisia princeps var. orientalis were quantified, and seasonal variation in monoterpenes of Artemisia plant was investigated. Samples were taken from five sites at the campus of Kyungnam University during maturing season. Monoterpenes in leaf and stem were analyzed using gas chromatography-mass spectrometry (GC-MS). The major constituents of A. princeps var. orientalis in both the leaf and stem were 21 monoterpenes.$ {\alpha}-pinene,\;{\beta}-pinene,\;{\beta}-myrcene$, dl-limonene, naphthalene and unknown monoterpenes with 5.49 and 16.27 of retenstion time were present in high concentrations of compounds identified on the leaf and stem of A. princeps var. orientalis. The cmounts of total monoterpenes of leaf were from two to five times higher than stem and rapidly decreased with the time, while that of stem was constnat except early spring. Most of the high percentage of monoterpenes in the leaf were those with later retention time. These results indicated that monoterpenes yields are considered to be more variable than monoterpene composition in responding to the time in both the leaf and stem.

Quantitative and Comparative Analysis of Urinary Steroid Levels upon Treatment of an Anti-Diabetic Drug, CKD-501 using Gas Chromatography-Mass Spectrometry

  • Urinary steroid levels were investigated in the treatment of CKD-501, a new anti-diabetic drug candidate. CKD-501 was administered orally at the dosage of 1, 2, 4 mg/day for 7 days to normal men (n=18). Urine was collected before, during and after stopping the drug administration and the urinary level of androgen, estrogen, progestin and corticoids were quantified using GC-MS (gas chromatography-mass spectrometry). Only urinary corticosteroid and an androgen, DHEA levels among all the analyzed steroids, have been found to increase progressively, reaching significant levels on the last day of drug treatment and later declined after the drug treatment is withdrawn. Therefore, it was thought that an increase in the urinary corticoid and DHEA levels could be a characteristic of CKD-501, since it prominently acts on the glucose sensitivity and suppresses the triglyceride levels. In conclusion, it was found that CKD-501, an anti-diabetic drug candidate, affects the glucocorticoid and DHEA levels and it plays a crucial role in glucose homeostasis.

Determination of volatile compounds by headspace-solid phase microextraction - gas chromatography / mass spectrometry: Quality evaluation of Fuji apple

  • The volatile components in 'Fuji' apple were effectively determined by a headspace solid-phase microextraction (HS-SPME) combined with gas chromatography-mass spectrometry (GC-MS). A total of 48 volatile components were identified and tentatively characterized based on National Institute of Standards and Technology (NIST) MS spectra library and the Kovats GC retention index I (RI). The harvested Fuji apples were divided into two groups: 1-methylcyclopropene (1-MCP) treated and non-treated (control) samples for finding important indicators between two groups. The major volatile components of both apples were 2-methylbutyl acetate, hexyl acetate, butyl 2-methylbutanoate, hexyl butanoate, hexyl 2-methylbutanoate, hexyl hexanoate and farnesene. No significant differences of these major compounds between 1-MCP treated and non-treated apples were observed during 1 month storage. Interestingly, the amount of off-flavors, including 1-butanol and butyl butanoate, in 1-MCP treated apples decreased over 5 months, and then increased after 7 months. However, non-treated apples did not show significant changes for off-flavors during 7 month storage (p<0.05). The non-treated apples also contained the higher levels of two off-flavors than 1-MCP treated apples. These two compounds, 1-butanol and butyl butanoate, can be used as quality indicators for the quality evaluation of Fuji apple.

Analysis of Cow Hide Glue Binder in Traditional Dancheong by Enzyme-linked Immunosorbent Assay

  • Animal glue has been used as a binder in Dancheong since the Joseon dynasty. Binders play an important role in determining the physical characteristics of a painting layer. The analysis of binders can be used to identify the materials and techniques used in traditional Dancheong. Binders can be investigated using physicochemical component analyses methods such as gas chromatography/mass spectrometry, pyrolysis-gas chromatography/mass spectrometry, and fourier transform infrared spectroscopy, but the detection characteristics vary depending on the degradation properties of the pigment and binder. Therefore, cross-validation using a combination of physicochemical analysis and enzyme immunoassay is used to increase the reliability of the results. In this study, we present an enzyme-linked immunosorbent assay (ELISA) as an example of an enzyme immunoassay as a method for analyzing animal glue, a traditional binder used in Korea. The applicability of ELISA was tested using commercial animal glue, in addition to animal glue produced using a variety of extraction conditions. The animal glue was analyzed in a Noerok-additionally coated-replica sample to evaluate the possibility of analyzing the animal glue in a paint layer mixed with pigment. Based on the results, we performed an assay on the use of animal glue in the Dancheong sample of the temples of the Joseon dynasty, that are estimated to have been built in the 17th century.

SPME를 이용한 수용액중의 휘발성 유기화합물 분석 (Determination of Volatile Organic Compounds (VOCs) in Drinking Water using Solid Phase Microextraction (SPME))

  • 16종의 휘발성 유기화합물을 blank water에 첨가하고 고정상으로 $100{\mu}m$ polydimethyl siloxane을 입힌 solid phase microextraction(SPME) fiber를 사용하는 headspace SPME방법으로 추출하여 gas chromatography/mass spectrometry (GC/MS)로 분석하였다. 이 방법의 평균회수율은 97%, 평균상대표준편차는 4.7%, 그리고 검출한계는 $0.01-0.5{\mu}g/l$를 나타냈다. 즉 SPME 방법을 이용한 수용액 중의 휘발성 유기화합물의 결과는 현재 많이 사용되고 있는 기존의 다른 방법보다 우수하고 편리한 방법으로 나타났다.

기체크로마토그래피/질량분석법을 이용한 생체시료 중 프탈산 에스텔류의 분석법 (Determination of Phthalates in Biota Samples Using Gas Chromatography/Mass Spectrometry)

  • 본 연구는 프탈산 에스텔류에 대하여 생물시료 중에 존재하는 가장 일반적인 9종의 분석을 기체크로마토그래피-질량분석기-선택이온검색법에 의해 수행하였다. 생물시료는 유기용매로 추출하고 Florisil 컬럼을 사용하여 정제하였다. 프탈산 에스텔류는 전처리 과정 중 가장 쉽게 오염되는 물질로 생물시료의 경우 전처리 단계가 수질이나 저질시료보다 복잡해지므로 각 전처리 단계의 오염정도를 비교 분석하였다. 표준물질이 첨가된 생물시료에 본 연구에서 개발된 분석법을 적용하여 구한 회수율은 79 - 117%정도로 나타났다. 또한 프탈산 에스텔류의 정량의 정확도, Florisil 컬럼 용리패턴, 검출한계 등을 조사하였다.

SENSITIVE DETERMINATION OF ELEVEN PHENOLIC ENDOCRINE-DISRUPTING CHEMICALS IN HUMAN URINE USING GAS CHROMATOGRAPHY/MASS SPECTROMETRY -SELECTED ION MONITORING

  • I improved an analytical method for determining trace amounts of eleven phenolic endocrine-disrupting chemicals (11 phenolic EOCs) in human urine. The 11 phenolic EOCs were subjected to hydrolysis and then to solid phase extraction with a XAD-4 column. Alkylphenols, chlorophenols, and bisphenol A in XAD-4 column were eluted with acetonitrile, and the eluate was concentrated under a nitrogen stream, and then tert-butyldimethylsilylation. Separation and determination were done by gas chromatography, using mass spectrometry operating in the selective ion monitoring mode for quantitation. For tert-butyldimethylsily (TBDMS) derivatization the recoveries were $91.2{\sim}125.9%$, the limits of quantitation (LOQ) for the 11 phenolic EOCs in the nanogram-per-milliliter range ($0.025{\sim}1.000\;ng/mL$) were thus achieved by using 1 mL of urine, and the SIM responses were linear with the correlation coefficient varying by $0.9300{\sim}0.9943$. Based on the results for urine samples from unexposed individuals, 4-tert-octylphenol and pentachlorophenol were detected in hydrolysed urine sample. Other alkylphenols, chlorophenols and bisphenol A were not detected.

Seasonal variations in the content and composition of essential oil from Zanthoxylum piperitum

  • Seasonal variations in the profile and concentrations of essential oil in Zanthoxylum piperitum were investigated by gas chromatography-mass spectrometry. Seasonal changes in the percentages of the main constituents of the essential oil of both leaves and fruits from Z. piperitum varied. Variations in essential oil yield and the amount of monoterpenes and sesquiterpenes in leaves and fruits at different developmental stages were significant. The characteristic content of essential oil in leaves was determined mainly due to the content of monoterpenes, and that in fruits was determined largely due to the sesquiterpenes. Twenty-nine compounds in the oil from Z. piperitum leaves were detected; the major compounds were ${\beta}$-phellandrene (26.90%), citronella (15.32%), ${\beta}$-myrcene (3.24%), ${\alpha}$-pinene (2.79%), trans-caryophyllene (2.66%), and fanesyl acetate (2.30%). The highest yield of oil (43.89%) in Z. piperitum leaves was obtained in May but decreased gradually beginning in June. The yield of essential oil from Z. piperitum leaves during early periods was higher than that during later periods and usually decreased from early maturation stages to subsequent stages. However, in contrast to leaves, the oil yield in Z. piperitum fruit increased in June, and oil yield later in the season was higher than that earlier in the season. These results indicate that the essential oil produced from Z. piperitum leaves at the early developmental stages was stored in leaves, and might be transferred to fruit at the final developmental stages.

Screening and identification of bioorganic light-emitting substances from marine macrophytes

  • Organic light-emitting materials in marine macrophytes from various coastal environments were identified. Twentyeight species from the solvent fractions were examined and identified as candidates for bioorganic light-emitting materials using photoluminescence (PL) spectra and gas chromatography-mass spectrometry. We selected 16 solvent fractions from a total of 1,221 prepared from Ishige okamurae, Sargassum confusum, Grateloupia elliptica, Chondracanthus intermedius, Porphyra yezoensis, Meristotheca papulosa, Gelidium amansii, and Scytosiphon lomentaria. The maximum light-emitting PL spectra appeared at various colors, mainly between blue and green, based on chromaticity coordinates, from solvent fractions of M. papulosa, G. amansii, G. elliptica, P. yezoensis, S. lomentaria, I. okamurae, and C. intermedius. These results will contribute to the development of novel organic light-emitting materials.
