Application of Modified Stokes Expression to Model the Behavior of Expanded Beds with Feed Streams Containing E. coli Homogenates

  • To model the behavior of expanded beds with aqueous feed streams containing different amounts of glycerol, we previously developed a modified Stokes expression that correlates the terminal settling velocity of a particle of a solution with the properties of the particle (particle diameter and density) and the solution (density and viscosity). Two empirical parameters, the effective diameter of the poly-disperse resins for protein adsorption and an exponent of non-unity for $(\rho_{P}-\rho)/\mu$ term, are introduced in the modified Stokes expression. We applied the same type of the modified Stokes expression in combination with the Richardson-Zaki correlation to the published results [1], and found that the expansions of the beds with feed streams containing different amounts of E. coli homogenates can also be successfully described.

Development of Tools for Measurement of Inner Shell Deformation of HANARO Reactor

  • It was estimated by an analysis method thai the inner shell of HANARO reactor will be deformed due to pressure, loads, creep and growth during reactor operation. To confirm the analysis validity and safe operation of reactor, we developed tools to remotely measure the straightness of the inner shell located 12m below the pool top. The performance and the accuracy of the measurement tools have been verified through tests using a dummy inner shell and steel straight edge. The accuracy of the measurement shows very good results with a maximum error of 0.06mm by steel straight edge. The technical experiences described in this paper will be a good reference not only for the operation and maintenance of HANARO but also for the next performance of the measurement in the future.

  • The HANARO reactor has a vertical hole for a cold neutron source (CNS) in the heavy-water reflector tank, i.e., the CNS hole, which was considerably deformed during its welding to the horizontal cold neutron (CN) beam tube. This paper presents an investigation of the form of the CNS hole for the optimal design of the a vacuum chamber for the CNS. In addition, the installation method of the vacuum chamber into the CNS hole for minimizing the water thickness between the vacuum chamber and the nose of the CN beam tube is proposed.

Kinetic Study of pH Effects on Biological Hydrogen Production by a Mixed Culture

  • The effect of pH on anaerobic hydrogen production was investigated under various pH conditions ranging from pH 3 to 10. When the modified Gompertz equation was applied to the statistical analysis of the experimental data, the hydrogen production potential and specific hydrogen production rate at pH 5 were 1,182 ml and 112.5 ml/g biomass-h, respectively. In this experiment, the maximum theoretical hydrogen conversion ratio was 22.56%. The Haldane equation model was used to find the optimum pH for hydrogen production and the maximum specific hydrogen production rate. The optimum pH predicted by this model is 5.5 and the maximum specific hydrogen production rate is 119.6 ml/g VSS-h. These data fit well with the experimented data($r^2=0.98$).

Hemolytic Activity of Culture Supernatant of Xenorhabdus nematophilus, a Symbiotic Bacterium of Entomopathogenic Nematodes

  • Lysis of erythrocytes isolated from human, rabbit, and mouse blood samples was investigated with the culture supernatant of Xenorhabdus nematophilus in a primary form. Prior to use, the culture supernatant of the bacteria was concentrated and the concentrate was dialyzed against Tris-HCl buffer (10 mM, pH 8.1) by ultrafiltration using PM-5 membrane with a molecular weight cut-off of 5,000. At $30^{\circ}C$, the supernatant exhibited no lytic activity towards three types of erythrocytes. However, at $4^{\circ}C$, the supernatant showed selective lytic activity towards rabbit erythrocytes within 90 min. yet did not lyze human or mouse erythrocytes. Microscopic examination clearly revealed that most of the rabbit erythrocytes had been fumed into ghost forms.

Oxidation of Dibenzothiophene Catalyzed by Surfactant-Hemoprotein Complexes in Anhydrous Nonpolar Organic Solvents

  • In anhydrous organic solvents, the complexes formed between AOT (dodecylbenzene sulfuric acid sodium salt) and hemoproteins, such as hemoglobin, myoglobin, or cytochrome c, displayed remarkably higher activity than the hemoprotein powders to oxidize dibenzothiophene, a model compound of organic sulfurs contained in fossil fuels. In slightly hydrophobic organic solvents, such as ethyl acetate and butyl acetate, dibenzothiophene was completely oxidized catalytically by the cytochrome c-AOT complex with cumene hydroperoxide (${\alpha},{\alpha}-dimethylbenzyl$ hydroperoxide) as an oxidant. In highly hydrophobic organic solvents, such as decane and hexadecane, however, the activity of the cytochrome c-AOT complex decreased, presumably due to the aggregation of the hemoprotein-AOT complex in these solvents.

  • Entomopathogenic nematodes are used for insect control. Herein, an extracellular protease was partially purified from a culture supernatant of Xenorhabdus nematophilus, a symbiotic bacterium of an entomopathogenic nematode, Steinernema glaseri: using precipitation with $80\%$ v/v isopropyl alcohol followed by gel permeation chromatography with a packed Sephacryl S-300 HR media. The partially purified protease exhibited maximal activity at pH 7 in the presence of 1 mM $CaCl_2$. The protease was identified as a metallo-protease based on the inhibition of its activity by the metal chelating agent, EDTA.

  • This study is a seismic analysia of absorber rod in KMRR Reactivity Control Mechanism. The model being studied i8 two coaxial tubes (control absorber rod and flow tube) immersed in the water and partially coupled (overlap) by water Hap. The hydrodynamic mass effects by the water in each surrounding conditions are considered in the model. The natural frequencies, stresses and displacements of the system due to Safe Shutdown Earthquake are computed in the cases of in-phase modes and out-of-phase modes of two coaxial tubes. The results show that maximum stresses are well below the allowable limit and maximum displacements at the ends of both tubes in out-of-phase modes are so huck that the tubes contact each other in the overlap zone.

  • 그 동안 IT 아웃소싱에 관한 많은 연구성과에도 불구하고 아웃소싱 계약체결에 있어서 실무적으로 꼭 필요한 아웃소싱의 적정가격에 관한 연구는 매우 미진한 형편이다. 특히 아웃소싱 의뢰업체 입장에서의 적정가격결정 모형이나 사례를 찾아보기는 더욱 어려운 것이 현실이다. 아웃소싱 가격의 적정성은 공급업체 만이 아니 라 의뢰업체의 입장에서도 아웃소싱의 성패를 좌우하는 중요한 요건이다. 본 연구는 TCO 개념을 이용하여, IT 아웃소싱 의뢰업체의 입장에서, 적정 가격을 산정하는 모형을 실제 적용사례를 통해 제시한다. 본 연구는 아웃소싱을 하지 않을 경우의 총소요비용과 아웃소싱을 할 경우의 총소요비용을 비교하여 아웃소싱의 효과를 측정하고 계약가격의 적정성을 판단할 수 있도록 하고자 하였다. 본 연구를 통하여 제시한 TCO분석방법 및 결과는 아웃소싱을 준비하거나 계획중인 업체에게 자체적으로 아웃소싱 계약가격을 산출하고, 아웃소싱을 통한 비용절감항목을 판단하며, 계약가격을 비교할 수 있는 자료로 사용되어 지기를 기대한다.

Transcription Factor for Gene Function Analysis in Maize (옥수수 유전자 기능 분석을 위한 전사인자의 이해)

  • Transcription factors are essential for the regulation of gene expression in plant. They are binding to either enhancer or promoter region of DNA adjacent to the gene and are related to basal transcription regulation, differential enhancement of transcription, development, response to intercellular signals or environment, and cell cycle control. The mechanism in controlling gene expression of transcription can be understood through the assessment of the complete sequence for the maize genome. It is possible that the maize genome encodes 4,000 or more transcription factors because it has undergone whole duplication in the past. Previously, several transcription factors of maize have been characterized. In this review article, the transcription factors were selected using Pfam database, including many family members in comparison with other family and listed as follows: ABI3/VP1, AP2/EREBP, ARF, ARID, AS2, AUX/IAA, BES1, bHLH, bZIP, C2C2-CO-like, C2C2-Dof, C2C2-GATA, C2C2-YABBY, C2H2, E2F/DP, FHA, GARP-ARR-B, GeBP, GRAS, HMG, HSF, MADS, MYB, MYB-related, NAC, PHD, and WRKY family. For analyzing motifs, each amino acid sequence has been aligned with ClustalW and the conserved sequence was shown by sequence logo. This review article will contribute to further study of molecular biological analysis and breeding using the transcription factor of maize as a strategy for selecting target gene.
