Storage Stability of the Conversion Pigment from Gardenia jasminoides Yellow Pigment (치자황색소로부터 변환된 색소의 저장안정성)

  • Storage stability of the blue-green pigment, which was converted from Gardenia jasminoides yellow pigment by Staphylococcus epidermides and Gardenia jasminoides yellow pigment, were investigated at various conditions of light, temperature, inorganic ion and pH, The factors that cause the discoloration were light and temperature $(above\;40^{\circ}C)$. The effects of light and temperature on storage stability of blue-green pigment were less than those of Gardenia jasminoides yellow pigment. Also, the effect of light was decreased by using green filter. There were no significant effects of pH and inorganic ion on both pigments.

A Research on the origin and Daily Dose of Gardeniae Fructus in Shanghanlun (상한론(傷寒論)에서 치자(梔子)의 기원(基原)과 1일 복용량)

  • Objective : The purpose of this study is to determine the origin and daily dose of Gardeniae Fructus in Shanghanlun. Methods : Define the origin and daily dose of Gardeniae Fructus by searching Shanhanlun and Herbology Books(本草書). And Compared these with those of current Gardeniae Fructus. Results : According to Shanhanlun, the origin of Gardeniae Fructus is Gardenia jasminoides, but G. jasminoides f. longicarpa must excluded since originally it was used for dyeing. Among the 8 prescriptions containing Gardeniae Fructus, 15 Gardeniae Fructuses for the daliy dose in Chijabekpi-tang(Zhizibopi-tang) is assumed to be 14 Gardeniae Fructuses, as it is found 14 in many other prescriptions same in other prescriptions. The quantity of 14 Gardeniae Fructuses in Shananlun is 3 Ryang(兩), which is equal to 19.5 g in the current measurement. 1 Gardeniae Fructus is equal to 1.4 g. The length of the Gardeniae Fructus in the market which is produced in korea is 22-27 mm, and the diameter is 15-19 mm, whereas in China it is 20-25 mm, and the diameter is 13-18 mm. This meets the standard requirements of the Pharmacopoeia. Conclusions : As a result from above, the origin of Gardeniae Fructus is Gardenia jasminoides, but G. jasminoides f. longicarpa must be excluded. The recommended dose in a day is 14 Gardeniae Fructuses, which is equivalent to 19.5 g, the amount generally used in Shanhanlun.

Pruification of Yellow Color from Gardenia(Gardenin jasminoides Ellis) for Development of Natural Food Color (천연식용색소 개발을 위한 치자 황색색소의 정제도)

  • In order make natural food color from gardenia(Gardenia jasminoides), we investigated optimal conditions of color extraction, in case of water extraction, optimal conditions for color extraction were 7$0^{\circ}C$, 48hrs, pH 7.0 and substrate 10%, respectively. And extracted crude color was purified by activated white clay, were isolated glycoside peak (238nm) and yellow color peak(40nm) from extracted crude color. The pruified color was increased by 27-fold and the yield was 96%.

  • We examined the neuroprotective and neurotrophic effects of genipin fractionated from gardenia (Gardenia jasminoides Ellis) originating from Korea. The neurotrophic effects of the genipin containing fraction was evaluated by microscopically monitoring its potency to induce neurite outgrowth in PC12h cells. The genipin containing fraction from Korean gardenia promoted neurite outgrowth in PC12h cells in this study, similar to previously reported effects by Wako Chemical, Japan. When cells were treated with the genipin containing fraction prior to ${\beta}$-amyloid peptide treatment (active domain of A peptide 25-35 treated), toxicity was significantly diminished (p<0.0l). These results suggest that genipin prepared from Korean gardenia might potentially be used as a precautionary agent in neurodegenerative disease, such as Alzheimer's disease, etc.

  • Anti-Pseudomonas aeruginosa activities for ethanol extract of 34 medicinal plants widely used in the folk medicine were evaluated to screening of anti-Pseudomonas aeruginosa agents. The ethanol extracts of Gardenia jasminoides, Arctium lappa, Citrus unshiu, and Phellodendron amurense showed antimicrobial activities against P. aeruginosa ATCC 27853. The ethanol extracts of Gardenia jasminoides among these medicinal plants showed significant antimicrobial activity against P. aeruginosa. Therefore, we expect that these medicinal plants will be useful for nature antimicrobial agent against P. aeruginosa in future.

Protective Effect of Gardenia jasminoides Against Carbon Tetrachloride-Induced Acute Hepatotoxicity (사염화탄소 유도 급성 간독성 모델에서 치자의 간 보호 효과)

  • Gardenia jasminoides is one of the most widely used herbal preparations for the treatment of liver disorders. This study evaluated the potential beneficial effect of G. jasminoides in a mouse model of carbon tetrachloride ($CCl_4$)-induced liver injury. The mice were treated intraperitoneally with $CCl_4$ (10 ${\mu}l$/kg). They received G. jasminoides (30, 100, 300 mg/kg) 48 h, 24 h and 2 h before and 6 h after administering $CCl_4$. The serum activities of aminotransferase and the hepatic level of malondialdehyde were significantly higher 24 h after the $CCl_4$ treatment, while the concentration of reduced glutathione was lower. These changes were attenuated by G. jasminoides. $CCl_4$ increased the level of circulating tumor necrosis factor-$\alpha$ (TNF-$\alpha$) markedly, which was reduced by G. jasminoides. The levels of hepatic inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) protein expression were markedly higher after the $CCl_4$ treatment. G. jasminoides diminished these alterations. $CCl_4$ increased the level of TNF-$\alpha$, iNOS and COX-2 mRNA expressions, and these increases were attenuated by G. jasminoides. These results suggest that G. jasminoides alleviates $CCl_4$-induced liver injury, and this protection is likely due to the reduced oxidative stress and the downregulation of proinflammatory mediators.

The Dyeing Properties of Cellulose and Protein Fabrics by Yellow Natural Dyes (황색계 천연염료에 의한 셀룰로스, 단백질계 섬유의 염색)

  • In order to analysis on color difference of yellow natural dyes, I have dyed cellulose and protein fabrics. The results of experiment have been analysed by wavelength of maximum absorption, amounts of dye uptake, color difference, Hunter's value and Munsell's value. The results from these analyses are as follows : Bud of pagoda tree, Amur cork, and Curcuma showed greenish yellow color, Gardenia Jasminoides showed reddish yellow color. Barberry root showed reddish yellow color with post-mordanting method on cellulose fabric. Moreover, Dupioni silk was dyed in reddish yellow color by Barberry root and Rhubarb. In addition to Chroma index, Gardenia Jasminoides and Curcuma showed clear color overall. However, dyeing rayon and silk by Barberry root, and dyeing silk by Rhubarb showed clear color. Comparing all the results to actual dyed materials, Bud of pagoda tree had small dye uptake, and both ${\Delta}a$ and ${\Delta}b$ value were short which can't recognized the yellow color easily. Dye uptake of Amur cork and Gardenia Jasminoides was small just like Bud of pagoda tree. However, ${\Delta}b$ value order was Gardenia Jasminoides>Amur cork>Bud of pagoda tree. Therefore, Gardenia Jasminoides recognized reddish yellow because of big value of red color and yellow color. In case of Barberry root and Rhubarb which have larger dye uptake, Baberry root recognized yellow color on rayon only, and couldn't recognized yellow color on bleached cotton fabric, ramie, silk, and dupioni silk. Rhubarb recognized yellow color on rayon with pre-mordanting method only, but recognized silk and dupioni silk as brown like color. Moreover, we could not analyze color by dye uptake, Lab, and H(v/c) for Barberry root and Rhubarb. As a result, I think we need to attach color table for the research paper which handled the color of dyeing materials.

Psychopharmacological Profile of the Water Extract of Gardenia jasminoides and Its Constituents, Genipin and Geniposide, in Mice

  • Gardenia jasminoides (G. jasminoides) is traditionally used to treat insomnia, jaundice, emotional disorders, hepatic disease, and inflammatory disease. Previously, we found that geniposide and the water extract of G. jasminoides increased $Cl^-$ influx in neuroblastoma. Here we examined the sychopharmacological activities of G. jasminoides and its constituents. G. jasminoides extract was orally administered at 100 and 200 mg/kg, and genipin and geniposide were intraperitoneally injected at 2, 10, and 20 mg/kg. G. jasminoides extract (200 mg/kg) significantly decreased total open field activity but increased rearing activity in the center of the open field, suggesting an increase in exploratory activity. Genipin and geniposide did not change open field activity, but geniposide (20 mg/kg) increased rearing activity in the central area. The extract (200 mg/kg) significantly decreased rotarod and wire-balancing activity, but genipin and geniposide did not. No compounds influenced thiopental-induced sleeping or electroshock-induced seizures. The extract (200 mg/kg) significantly increased staying time in the open arms of the elevated plus maze and the entry ratio into the open arms, and geniposide (20 mg/kg) also increased open arm entry. Electroshock stress decreased open arm activity, but the extract and geniposide (20 mg/kg) significantly reversed that effect. This results indicate that G. jasminoides extract and geniposide alleviated anxiety with greater efficacy in stressed animals than normal animals.

Effect of Water Extract of Gardenia jasminoides on the Sensory Quality and Putrefactive Microorganism of Cooked Rice (치자 추출물이 쌀밥의 관능적 특성 및 부패미생물에 미치는 효과)

  • The purpose of this study was to investigate the effect of water extract of Gardenia jasminoides (WEGJ) on the sensory qualities, color value and shelf-life characteristics (pH, total no. of Bacillus and TBA value) of cooked rice. The total color difference(ΔE) was increased when WEGJ was added. The sensory dualities of cooked rice added with WEGJ had no significant difference in color, taste and torture compared with control group. The pH value of cooked rice addeded with 250 ppm and 500 ppm of WEGJ was less declined compared with control group. WEGJ at the concentration of 250 ppm and 500 ppm had an antimicrobial activity and WEGJ at 500ppm showed an antioxidant activity in cooked rice.
