Flavonoid Content and Antioxidant Activities of Gardenia jasminoides Ellis Seed Extracts Produced by Ethyl Acetate, Distilled Water and Methanol (치자(Gardenia jasminoides Ellis) 씨 에틸 아세테이트, 물 및 메탄올 추출물의 플라보노이드 함량 및 항산화 활성)

  • The purpose of this study was to measure the bioactivity and antioxidant activity of seed from Gardenia jasminoides Ellis fructus (GJE). We determined tannin content ($1.517{\pm}0.003mg\;CE/g$, mg of catechin equivalents). The extraction yields of the GJE seed solvent were distilled water (DW) 36.61%, 70% methanol 30.10% and ethyl acetate (EA) 20.40%. The antioxidant activities of the extracts were significantly increased (0.2, 0.4, 0.6 mg/mL). Flavonoid contents (mg QE/g, mg of quercetin equivalents) were observed in the order of 70% methanol (0.799), DW (0.565) and EA (0.117). Antioxidant activities (DPPH, OH, ferrous ion-chelating capacity) were similar to the flavonoid content of each solvent. But ABTS scavenging activity and SOD like ability of DW extract were higher than 70% methanol extract. Comparing the yield and the antioxidant activity of each solvent of GJE seed, it seems to be preferable to use DW and 70% methanol solvent for extraction. As a result of this experiment, it has high antioxidant activity and physiological activity, which is expected to be highly valuable as a functional food and a natural antioxidant.

Reversible Hepatic Toxic Effect of Crocin Dyes in Rats

  • Gardenia jasminodes has been medically used for anti-inflammation, sedation and anti-diarrhea; The extract of this plant has been traditionally used as food colorant and referred as crocin dyes. In the present study, the possible hepatic toxicity of this dye has been evaluated on the basis of its alteration on the marker enzymes, namely, glutamic-oxaloacetic transaminase, glutamic-pyruvic transaminase, alkaline phosphatase, lactate dehydrogenase and gamma-glutamyltransferase. Crocin dyes did not affect hepatic function when they were orally administered to rats in a daily dose of 50 mg/kg for 8 days, but could induce acute hepatic discoloration. A high dose of 100 mg/kg for 2 weeks could induce both hepatic damage and black pigmentation, but a lower dose of 10 mg/kg for 40 days did not The induced black pigmentation and the acute hepatic damage were completely reversible. In conclusion, the crocin dyes have a very low hepatic toxicity in rats, even in high experimental dosages which could hardly happen in human practice. It is therefore suggested that the crocin dyes are safe for coloring foods.

Effects of Gardeniae Jasminoides on RANKL-induced Osteoclastogenesis and Bone Resorption (치자 추출물이 RANKL 유도 파골세포 형성 및 골 흡수에 미치는 영향)

  • Objectives: This study was performed to investigate the effects of Gardenia jasminoides extract (GJ) on osteoclast differentiation and bone resorption in vitro. Methods: To investigate the effect of GJ on osteoclast differentiation, the mouse leukemic myeloid cell line RAW 264.7 was stimulated by RANKL (receptor activator of nuclear factor kB ligand). Osteoclast differentiation was measured by counting TRAP (+) MNC in the presence of RANKL. To elucidate the mechanism of the inhibitory effect of GJ on osteoclast differentiation, gene expression of TRAP, Cathepsin K, MMP-9, NFATc1, c-Fos, MITF, DC-STAMP, CTR, OC-STAMP and Atp6v0d2 was measured using reverse transcription-PCR (RT-PCR). Bone resorption was measured using the bone pit formation assay. Results: GJ decreased the number of TRAP (+) MNCs in the presence of RANKL. GJ inhibited the expression of cathepsin K, MMP-9, TRAP, MITF, NFATc1, c-Fos, iNON, OC-STAMP, Atp6v0d2, and DC-STAMP in the osteoclast, and inhibited bone pit formation in vitro. Conclusions: The results suggest that GJ has inhibitory effects on bone resorption resulting from inhibition of osteoclast differentiation and gene expression.

Lactic Acid Bacteria Increases Hypolipidemic Effect of Crocin Isolated from Fructus of Gardenia jasminoides

  • The fructus of Gardenia jasminoides Ellis (GF) has been widely used as a natural colorant in Asian countries, and also as a Chinese traditional medicine for its homeostatic, antiphlogistic, analgesic, and antipyretic effects. In the present study, its main component, crocin, was fermented with lactic acid bacteria (LAB) and their antihyperlipidemic activity was measured. The GF extract, fermented GF (F-GF), crocin, and fermented crocin (F-crocin) significantly inhibited the increase of serum triglyceride (TG) level in corn oil feeding-induced triglyceridemic mice, as well as that of serum TG and total and LDL cholesterol levels in Triton WR-1339-induced hyperlipidemic mice. These agents also showed hypolipidemic activity in hyperlipidemic mice induced by high fat diet for 5 weeks. F-GF and F-crocin exhibited more potent hyperlipidemic effects than GF and crocin. The results suggest that the hypolipidemic effect of GF and crocin can be synergistically activated by LAB, and that F-GF and F-crocin may improve hyperlipidemia in clinic, compared with GF and crocin.

Pharmacological Screening of Dikamali Resin Extract

  • In the present study, dikamali resin (obtained from the leaf buds and the young shoots of Gardenia gummifera Linn.) was extracted with diethyl ether and the extract was vacuum dried. Qualitative tests confirmed the presence of flavonoids and free phenolic compounds in the extract. The antioxidant property (qualitative) of the extract was performed by TLC method $({\beta}-carotene-linoleate method)$. The $LD_{50}$ of the extract was found to be 2227 mg/kg by Karber's arithmetic method. The extract was screened for analgesic, anti-inflammatory, antipyretic (100, 200 and 400 mg/kg) and anthelmintic (0.1,0.2 and 0.5%w/v) activities by standard methods. The extract exhibited antioxidant property and prevented oxidation of ${\beta}-carotene$. The extract exhibited significant graded dose response for analgesic, anti-inflammatory, antipyretic and anthelmintic activities. The extract caused the death of earthworms in all experimental concentration whereas the standard drug (piperazine) only effected paralysis. The present study proved the claims of dikamali resin mentioned in the Indian system of medicine.

Antibacterial Activity of Soluble Extracts of Gardenia jasminoides against Mycobacteria Other Than Tubercle Bacilli (치자(梔子) 수용성 추출액의 결핵균 외의 기타 Mycobacteria에 대한 항균작용)

  • An attempt was made to verify the antibacterial activities of the soluble extracts of Gardenia jasminoides(SEGJ) against mycobacteria other than tubercle bacilli (MOTT); M. kansasii, M. scrofulaceum, M. simiae, M. szulgai, M.xenopi, M. avium-intracellulare complex, M. gordonae and M. flarescens. In addition, the effect of the SEGJ on the yellow pigmentation and violet pigmentation (crocin reaction) of the colonies of the MOTT grown on7H10-crocin agar plates was observed. The SEGJ gave a growth stimulatory activity against most species of mycobacteria except for M. szulgai at low concentration of the SEGJ in terms of crocin pigment OD=0.02 or less, but exerted an inhibitory activity at high concentration of OD=0.04 or more. The growth-inhibitory activity of the SEGJ was dose-dependent but the yellow pigmentation of the scotochromogens was not affected by the dose of the SEGJ that was growth-inhibitory. Crocin positive reaction was observed only with M. kansasii.

천연염색된 멜란지 얀의 제조와 그의 항균ㆍ소취성

  • Melange yams were prepared by varing the ratio of raw cotton dyed with Indigo, Gardenia and Sappan wood colorants, respectively. The antibacterial activity and deodorization on melange yarns were evaluated for bacteria reduction rate and deodorization rate. The bacterial reduction rate on melange yarns was explored with Staphylococcus aureus, Klebsiella pneumoniae. The melange yarns on the ratio of raw cotton dyed with Sappan wood and Indigo showed high Staphylococcus aureus reduction rate of 99% and 90%, regardless of Al-mordanting treatment, but those with Gadenia hardly showed bacteria reduction rate. The melange yarn showed high Klebsiella pneumoniae reduction rate of 99% for Al-mordanting treatment with Sappan wood. The others were poor. The deodorization of 81% and 98∼99% obtained with melange raw sliver and melange sliver dyed with Indigo, Gardenia and Sappan wood colorants, but dyeing did not significantly affect deodorization rate.

Treatment of produced water in a floating carrier bioreactor

  • Produced water is the largest wastestream of oil and gas exploration. It consists of various organic and inorganic compounds that hinder its beneficial use. This study compared the treatment of produced water in a batch suspended and biofilm activated sludge process. The biofilm carrier material was made from Gardenia Carinata shell. COD, $NH_4{^+}-N$ and $NO_3-N$ removal was monitored in both the suspended (control) and floating carrier bioreactors. The results show a rapid reduction of produced water constituents in the floating carrier bioreactor. COD, $NH_4{^+}-N$ and $NO_3-N$ removal was in the range of 99%, 98% and 97% for the floating carrier bioreactor whereas it was 88%, 84% and 83% for the control bioreactor. The rapid reduction of COD, $NH_4{^+}-N$ and $NO_3-N$ clearly indicate that the floating carrier materials served as an attached growth medium for microorganisms, improved the breakdown of produced water constituents and reduced inhibition of microbial metabolic activities.
