Screening of Hepatoprotective Substances from Higher Fungi by Primary Cultured Rat Hepatocytes intoxicated with Carbon Tetrachloride ($CCl_4$로 독성을 유발시킨 초대배양 간세포를 이용한 고등균류로부터 간세포 보호물질의 검색)

  • Hepatoprotective effects of polysaccharides extracted from liquid cultured mycelia were screened by measuring the glutamic pyruvate transaminase activity of the primary cultured rat hepatocytes intoxicated with carbon tetrachloride. Sixty among 75 isolates of higher fungi showed to be hepatoprotective effect, and these were 13 of Ganoderma lucidum, 5 of Lentinus edodes, 1 of Pleurotus ostereatus, 4 of Coriolus versicolor, 2 of Lyophyllum spp., 7 of Grifora frondosa, 3 of Agaricus spp., 14 of Schizophyllum commune and 11 of Cordyceps spp.. Especially, 10 isolates, Ganoderma lucidum IY003 and IY009, Lentinus edodes IY103, Lyophyllum sp. IY402, Agaricus sp. IY701 and IY703, Schizophyllum commune IY804, IY810 and IY818, Cordyceps sp. IY902, were indicated below 80% of glutamic pyruvate transaminase activity.

Comparative analysis of useful β-glucan and polyphenol in the fruiting bodies of Ganoderma spp. (영지버섯 균주별 자실체의 베타글루칸과 폴리페놀 함량 비교 분석)

  • This study was carried out to compare the medicinal effects of various fruiting body of Ganoderma species and Cordyceps militaris, Phelinus linteus extracts. ${\beta}$-glucan and polyphenol are useful ingredient in mushrooms and they were known to have antioxidant activity. We analyzed ${\beta}$-glucan and polyphenol contents of fruiting body of Ganoderma spp., Cordyceps militaris, and Phellinus linteus. Most Ganoderma spp. exhibited ${\beta}$-glucan contents of 15 to 20%. Cordyceps militalis showed the highest ${\beta}$-glucan level of 25%. Interestingly, eight strains of Ganoderma spp. was analyzed to have higher contents of ${\beta}$-glucan than Phelinus linteus. Polyphenol contents was measured after extraction with different solvents. (D.W., 70% EtOH, 80% MeOH) The level of polyphenol in ASI 7020 strain was at maximum in the water extraction and ASI 7086 showed the highest level in the 70% EtOH extraction. The amounts of polyphenol in strain ASI 7113 was at maximum in the 80% MeOH extraction.

Phylogenetic Study of Ganoderma spp. Based on the DNA Sequences in ITS II Region (ITS II 영역의 DNA 염기서열 분석에 의한 불로초(Ganoderma)속의 계통분류학적 고찰)

  • The internal transcribed spacer II regions (ITS II) of the ribosomal DNA gene repeat from Ganoderma spp. were amplified using polymerase chain reaction (PCR) and sequenced. Sequences from 9 species including Ganoderma lucidum, G. tsugae, G. pfeifferi, G. resinaceum, G. australe-applanatum, G. oregonense, G. neo-japonicum, G. applanatum and Inonotus xeranticus as an out-group were compared. The spacer regions of them were $247{\sim}257$ nucleotides in length and contained partial sequences of 5.8S and 25S gene. The reciprocal homologies of each ITS II sequence of the species were in the range of $70{\sim}100%$ except outgroup species, I. xeranticus. According to the analysis of ITS II sequences, Ganoderma spp. constructed 5 clusters. Ganoderma lucidum isolates were to be divided into two groups. One group was consisted of isolates from South Korea. The other group comprised isolates from UK. G. lucidum isolates belonging to the group I were closely related with G. tsugae. These results suggested that G. lucidum from Korea should be G. tsugae, otherwise G. tsugae was to be synonym of G. lucidum.

Classification of Isolates of Ganoderma lucidum, Schizophyllum commune and Cordyceps spp. by Electrophoretic Patterns of Isozymes (Isozyme patterns 차이에 의한 영지, 치마버섯 및 동충하초의 계통분류)

  • To classify fungal species employed for pharmacological effects, mycelial proteins of six isolates of Ganoderma lucidum, five isolates of Schizophyllum commune and five isolates of Cordyceps spp. were separated on polyacrylamide gel to compare them by esterase, acid phosphatase, leucine aminopeptidase and peroxidase patterns. Similarity of isozyme patterns among the isolates of G. lucidum IY003, IY004, IY005 and IY008 was indicated over 70%, but that among the isolates of G. lucidum IY009, IY010 and others was indicated from 48% to 9%. Highest similarity of isozymes of S. commune was observed to be between IY803 and IY805, and similarity between these two isolates was 57%. Similarity among other isolates was shown to be from 40% to 56%. Isozyme patterns of Cordyceps spp. were comparatively different, even though they were originated from the same kind of insect as their isolate. Similarity between Cordyceps spp. IY901 and IY904, which was isolated from moths, was 67% and that of IY905 and IY909, which was originated from the larvae, was 42%. Similarity among other isolates was shown to be from 12% to 67%.

Comparative analysis of amino acid contents of the fruiting bodies in Ganoderma spp. (영지버섯 균주별 자실체의 아미노산함량 비교 분석)

  • Ingredient content for the general composition of essential amino acids and non-essential amino acids were analyzed by the fruiting bodies of Ganoderma sp. The results obtained are as followes. The ASI 7004 showed that high concentrations than other strains in essential amino acids(His, Ile, Leu, Phe, Thr, Val) and the non-essential amino acids(Ala, Ser, Gly). In addition, The ASI 7002 showed that high concentrations than other strains in essential amino acids(Lys) and the non-essential amino acids(Asp, Glu). ASI 7022 showed that high concentrations than other strains in essential amino acids(Met) and the non-essential amino acids(Pro). In General, contents of Amino acid was higher than Phellinus.

Comparisons of growth characteristics, biological activities, nutritional contents, and sugar contents of Ganoderma spp. strains (영지 균주별 생육특성, 생리활성, 영양성분 및 당 성분 함량 비교)

  • This study was carried out to compare the growth characteristics, biological activities, β-glucan contents, sugar contents, and amino acid contents of 14 strains of Ganoderma spp. Among the 14 strains of Ganoderma spp., KMCC02960 (G. meredithae) and KMCC02932 (G. tropicum) showed excellent growth characteristics such as those with respect to the size and yield of fruiting bodies. The highest 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity was observed in KMCC02932 (G. tropicum). The nitrite scavenging activities of KMCC02824 (G. lucidum) and KMCC02852 (G. neo-japonicum) were higher than those of the other strains. The total polyphenol contents of the extracts from KMCC02824 (G. lucidum) and KMCC02852 (G. neo-japonicum) were higher than those of the other strains. KMCC03018 (G. lingzhi) showed the highest β-glucan content of 33.4%. In an analysis of the 4 types of monosaccharides, 2 types of disaccharides, and 4 types of sugar alcohols, only KMCC02996 (G. weberianum) and KMCC03018 (G. lingzhi) were commonly detected out of the 14 strains of Ganoderma spp. Eighteen amino acids, including eight essential amino acids, were identified: the highest total amino acids and total essential amino acids were found in KMCC02932 (G. tropicum), which had the highest levels of tyrosine and phenylalanine. Although the contents of amino acids differed by strain, cysteine, tyrosine, and phenylalanine were the most abundant amino acids in the analyzed extracts.

Selection of Newly Isolated Mushroom Strains for Tolerance and Biosorption of Zinc In Vitro

  • Nine newly isolated mushroom strains were tested to assess both their zinc tolerance and potential for zinc removal from an aqueous solution. Four strains of ectomycorrhizal fungi, namely Clavariadelphus truncatus (T 192), Rhizopogon roseolus (T 21), Lepista nuda (T 373), and Tricholoma equestre (T 174), along with five strains of white rot fungi, Lenzites betulina (S 2), Trametes hirsuta (T 587), Ganoderma spp. (T 99), Polyporus arcularius (T 438), and Ganoderma carnosum (M 88), were investigated using zinc-amended solid and liquid media. Their biosorption properties were also determined. The colony diameter and dry weight were used as tolerance indices for fungal growth. C. truncatus and T. equestre were not strongly inhibited at the highest concentrations of (225 mg/l) zinc in solid media. The most tolerant four strains with solid media, C. truncatus, G carnosum, T. hirsuta, and T. equestre, were then chosen for tolerance tests in liquid media. An ectomycorrhizal strain, C. truncatus, was also detected as the most tolerant strain in liquid media. However, the metal-tolerant strains demonstrated weak activity in the biosorption studies. In contrast, the highest biosorption activity was presented by a more sensitive strain, G. carnosum. In addition, seven different biosorbent types from G. carnosum (M 88) were compared for their Zn (II) biosorption in batch experiments.

Effect of Buprofezin Concentration on the Formation and Reversion of Protoplast of Ganoderma spp. and Coriolus versicolor (Buprofezin이 Ganoderma및 Coriolus 속균(屬菌)의 원형질체(原形質體) 나출(裸出) 및 재생(再生)에 미치는 영향(影響))

  • Effects of buprofezin, a chitin synthesis inhibitor of insects, on mycelial growth, protoplast formation and reversion of Coriolus versicolor, Ganoderma applanatum and G. lucidum were investigated. The mycelial growth of C. versicolor, G, applanatum and G, lucidum was severely inhibited by buprofezin treatment, and the inhibition rate severely as the concentration of the buprofezin increased. Aerial mycelia and oidia formation of the mushrooms were increased by buprofezin treatment, but mycelial morphology was not changed. The rate of protoplast formation and reversion of G, applanatum was greatly increased by the treatment of the buprofezin, while that of G. lucidium was decreased. The rate of protoplast formation of C. versicolor was also increased when buprofezin was added to the medium, but the rate of protoplast reversion was not affected by the treatment, From the results obtained in this experiment, we found that the rate of protoplast. formation and reversion was increased by the treatment of the buprofezin in the mushrooms such as C, versicolor and G, applanatum, whose mycelial growth was fast on the medium, while that of G. lucidum, whose mycelial growth was relatively slow, was decreased by the treatment.

Comparative analysis of ganoderic acid A, F, and H contents in the fruiting bodies of Ganoderma spp. (영지버섯 유전자원의 가노데릭산 A, F, H의 함량 분석)

  • This study was carried out for examining the amount of ganoderic acid A, F, H from fruiting body extracts of the various Ganoderma species. The preserved Ganoderma species are extracted by using three kinds of solvent. Among them, five strains which have the large amount of ganoderic acid A, F, H were selected. First of all, Strains with the highest amount of ganoderic acid A were sorted out acorrding to solvent as follows. ASI 7013 has the highest amounts with 0.988 mg/g from D.W. extracts. And ASI 7023, 7059, 7026, 7060 were in order of content. In EtOH extracts, ASI 7037 has the highest amounts of ganoderic acid A with 0.940 mg/g. And ASI 7021, 7034, 7026, 7038 were in order of content. In MeOH extracts, ASI 7162 has the highest amounts with 0.833 mg/g. And ASI 7091, 7032, 7056, 7125 were in order of content. secondly, Strains with the highest amount of ganoderic acid F were sorted out acorrding to solvent as follows. ASI 7011 has the highest amounts with 0.722 mg/g from D.W. extracts. And ASI 7113, 7011, 7104 were in order of content. In EtoH extracts, ASI 7037 has the highest amounts with 0.811 mg/g. And ASI 7007, 7027, 7012, 7014 were in order of content. In MeoH extracts, ASI 7162 has the highest amounts with 0.833 mg/g. And ASI 7021, 7034, 7026, 7125 were in order of content. Finally, Strains with the highest amount of ganoderic acid H were sorted out acorrding to solvent as follows. ASI 7013 has the highest amounts with 0.985 mg/g in D.W. extracts. And ASI 7059, 7135, 7001, 7141 were in order of content. In EtoH extracts, ASI 7016 has the highest amounts with 2.842 mg/g. And ASI 7089, 7159, 7007, 7002 were in order of content. In MeoH extracts, ASI 7003 has the highest amounts with 6.969 mg/g. And ASI 7034, 7169, 7159, 7103 were in order of content.

Historical Record of Mushroom Research and Industry in Korea

  • Two kinds of mushrooms, Gumji (金芝; Ganoderma) and Soji, were described in old book of Samguksagi (History of the three kingdoms; B.C 57~A.D 668; written by Bu Sik Kim in 1145) in Korea-dynasty. Many kinds of mushrooms were also described in more than 17 kinds of old books during Chosun-dynasty (1392~1910) in Korea. Nowadays, mushroom cultivation has been increased through out the world last decade years. Production of mushrooms has also been increased 10-20% and many varieties have been cultivated. Similar trends were also observed in Korea. Approximately two hundred commercial strains of 37 species in mushrooms were developed and distributed to cultivators. Somatic hybrid variety of oyster mushroom 'Wonhyeong-neutari' were developed by protoplast fusion, and distributed to grower in 1989. The fruiting body yield index of somatic hybrids of Pleurotus ranged between 27 and 155 compared to parental values of 100 and 138. In addition, more diverse mushroom varieties such as Phellinus baumi, Auricularia spp., Pleurotus ferulae, Hericium erinaceus, Hypsizigus marmoreus, Grifola frondosa, Agrocybe aegerita and Pleurotus cornucopiae have been attempted to cultivate in small scale cultivation. Production of mushrooms as food was 190,111 metric tons valued at 800 billion Korean Won (one trillion won if include mushroom factory products; 1dollar = 1,040 Won) in 2011. Major cultivated species are Pleurotus ostreatus, Pleurotus eryngii, Flammulina velutipes, Lentinula edodes, Agaricus bisporus, and Ganoderma lucidum, which cover 90% of total production. Since mushroom export was initiated from 1960 to 1980, the export and import of mushrooms have been increased in Korea. Technology developed for liquid spawn production and automatic cultivation systems lead to the reduction of the production cost resulting in the increasement of mushroom export. However some species were imported because of high production cost for these mushrooms requiring the effective cultivation methods. Developing of effective post-harvest system will be also directly related to mushroom export. In academic area, RDA scientists have been conducting mushroom genome projects. One of the main results is the whole genome sequencing of Flammulina velutipes for molecular breeding. An electrophoretic karyotype of of F. velutipes was obtained using CHEF with 7 chromosomes, with a total genome size of approximately 26.7 Mb. The mususcript of the genome of F. velutipes was published in PLOS ONE this year. For medicinal mushrooms, we have been conducting the genome research on Cordyceps and its related species for developing functional foods using this mushroom. In 2013, Korea Food and Drug Administraion (KFDA) approved Cordyceps mushroom for its value as an immune booster.
