Effect of Ganoderma lucidum Mycelial Fractions on the Functional Recovery of Primary Cultured Hepatocytes. (영지 균사체 분획이 일차배양 간세포 기능회복에 미치는 영향)

  • The cultured mycelia of Ganoderma lucidum were extracted and the extract was separated into six fractions by organic solvent fractionation. The antihepatotoxic activity of all the fractions was evaluated by measuring activities of glutamic pyruvic transaminase (GPT) and glutamic oxaloacetic transaminase (GOT). Among the fractions tested, the high-polarity fractions such as aqueous and n-butanol fractions significantly reduced activities of GPT and GOT in CCl4- and galactosamine-intoxicated rat primary hepatocytes. When intracellular synthetic activities were measured by pulsing the rate primary cultured hepatocytes with [3H]-uridine and [3H]-leucine, activities of DNA, RNA and protein. When direct toxicities of the fractions were measured against human hepatoma(SK-Hep-1), the non-polarity fractions such as n-butanol and ethyl acetate fractions showed potent direct cytotoxicities even at the concentration of 1 $\mu\textrm{g}$/ml. These data showed that Ganoderma lucidum has hepatoprotective and hepatotoxic recovery principles in its mycelia.

  • The effect of Ganoderma lucidum (Polyporaceae) extract on initiation and promotion of liver preneoplastic lesions in chemical carcinogenesis were examined in male rats. Male Fisher 344 rats, 8 weeks old, were injected i.p. with diethylnitro-samine(DENA` 200 mg/kg) as an initiator and were fed on diet containing 0.02%-acetaminofluorene (2-AAF) as a promoter for 2 weeks (group 1, 2` 2-4 week) and were fed on basal diet (group 3 and 4). The rats in test groups(group 1 and 3) were fed on diet containing 0.1% of Ganoderma lucidum extract for 6 weeks (2-8 week).

  • In order to investigate the effects of Ganoderma lucidum on the fermentation of media by Kluyveromyces fragilis KCCM 35458, with 0%, 0.1%, 0.5%, and 1.0% (w/v) of Ganoderma lucidum extract were added to skin milk and fermented in the suspension culture at $30^{\circ}C$ for 72 hours. The pH, acidity, $CO_2$ evolution, alcohol production, number of yeast cells and lactose content were investigated. The results are summarized as follows; 1. As fermentation time advanced by the pH was decreased and the acidity was increased in the media of 1.0% Gnaoderma lucidum extract as fermentation time advanced. 2. $CO_2$ evolution was increased in the all of Genoderma lucidum extract and significantly effect showed in 1.0% extract medium. 3. During 72 hours culturing, alcohol was detected in all Gandoderma lucidum extract media, 1.0% Ganoderma lucidum extract medium showed extremly high in alcohol and shortened fermentation time. 4. The number of yeast was increased more at logarithmic phase and the growth of yeast was more rapid in all Ganoderma lucidum extract especially 0.5%, 1.0% media samples than control. 5. As fermentation time goes by, the lactose content of media were decreased more rapidly in 1.0% Ganoderma lucidum extract medium than control.

  • A submerged cultivation of Ganoderma lucidum was carried out in an air-lift fermenter system, and the effects of different pH processes on extracellular branched ${\beta}$-1,3-glucan(EPS) production and mycelial growth(MDW) were investigated. The controlled pH process improved the production of branched ${\beta}$-1,3-glucan and biomass in comparison to the uncontrolled pH process. However, the maximum production of branched ${\beta}$-1,3-glucan were obtained by the bi-staged pH process. From these results, we confirmed that the bi-staged pH process was the most effective for improving the production of branched ${\beta}$-1,3-glucan from submerged culture of G. lucidum.

Phylogenetic Study of Ganoderma spp. Based on the DNA Sequences in ITS II Region (ITS II 영역의 DNA 염기서열 분석에 의한 불로초(Ganoderma)속의 계통분류학적 고찰)

  • The internal transcribed spacer II regions (ITS II) of the ribosomal DNA gene repeat from Ganoderma spp. were amplified using polymerase chain reaction (PCR) and sequenced. Sequences from 9 species including Ganoderma lucidum, G. tsugae, G. pfeifferi, G. resinaceum, G. australe-applanatum, G. oregonense, G. neo-japonicum, G. applanatum and Inonotus xeranticus as an out-group were compared. The spacer regions of them were $247{\sim}257$ nucleotides in length and contained partial sequences of 5.8S and 25S gene. The reciprocal homologies of each ITS II sequence of the species were in the range of $70{\sim}100%$ except outgroup species, I. xeranticus. According to the analysis of ITS II sequences, Ganoderma spp. constructed 5 clusters. Ganoderma lucidum isolates were to be divided into two groups. One group was consisted of isolates from South Korea. The other group comprised isolates from UK. G. lucidum isolates belonging to the group I were closely related with G. tsugae. These results suggested that G. lucidum from Korea should be G. tsugae, otherwise G. tsugae was to be synonym of G. lucidum.

Effects of Antioxidative stress, Antimutagenicity and Cytotoxicity of Cancer cells in Fractional Extracts from Ganoderma lucidum Karst (영지버섯(Ganoderma lucidum Krast)의 순차 분획물의 산화적 스트레스 억제효과와 항돌연변이 및 암세포 증식 억제 효과)

  • This study was performed to determine the antioxidative effect of the hexane, dichloromethane, ethylacetate, butanol and water fractions of Ganoderma lucidum extracts on the inhibition of malondialdehyde(MDA) and bovine serum albumin(BSA) conjugation reaction, the inhibition of lipid peroxidation and the scavenging effect on 1,1-diphenyl-2-picryl-hydrazyl(DPPH) radical, the antimutagenic capacity as measured by the Ames test and the inhibitory effect on cancer cell. Ganoderma lucidum is believed to have possible antioxidative capacities, although the results have varied according to the assay method. The most effective antioxidative capacity was inhibition of lipid peroxidation. Among the five fractions, water fraction showed strong inhibition rates on MDA & BSA conjugation reaction, and ethylacetate fractions showed the most effective inhibition rate on lipid peroxidation and scavenging effect on DPPH radical. The indirect and direct antimutagenic effects of ethanol extracts of Ganoderma lucidum were examined by Ames test using Salmonella typhimurium TA98 and TA100. Among the samples, the water fraction did not have any antimutagenic effect. The inhibition rates on mutagenicity in the presence of 2.5 mg/plate were nearly $100\%$ for Salmonella typhimurium TA98 and TA100 except the hexane fraction of the direct mutagenicity mediated by 2-Nitrofluorene in Salmonella typimurium TA98($64.69\%$). Under the 2.5 mg/plate concentration, the inhibitory effects of hexane and dichloromethane fraction were superior to that of the other fractions on the direct mutagenicity for Salmonella typhimurium TA100 and indirect mutagenicity for Salmonella typhimurium TA98 and TA100. The inhibitory effect of Ganoderma lucidum extracts on cell proliferation in HeLa and MCF-7 was investigated by U test. The dichloromethane fraction showed highly antiproliferative effect in HeLa and MCF-7($IC_{50}$: 0.122 mg/mL, 0.272 mg/mL, respectively) cells while the water faction had a weak inhibitory effect($IC_{50}$: 0.691 mg/mL, 10.919 mg/mL respectively). These results suggest that Ganoderma lucidum may have antioxidative, antimutagenic and anticancer capacities and may be a candidate of the prevention and dietetic treatment of chronic diseases and the development of antioxidative, antimutagenic and anticancer functional food.

Effects of Panax ginseng and Ganoderma lucidum Extract on the Growth of Lactic Acid Bacteria (인삼 및 영지추출물이 유산균 생육에 미치는 영향)

  • This study was carried out to Investigate the effects of Panax ginseng, Ganoderma lucidum extract and crude polysaccharide of G. lucidum on the growth of lactic acid bacteria. p. ginseng extract contained 60.7% carbohydrate and 27.5% protein, whereas G. lucidum contained 35.9% carbohydrate and 46.3% protein. The total sugar and protein content of crude polysaccharide of G. lucidum were 47.2% and 15.2%, respectively. Two amino acids(hg, Trp) were detected in p. ginseng extract and 11 amino acids (hg, Trp, Ua, Lys, Ser, etc.) in C. lucidum extract. By the addition of p. ginseng, 5. lucidum extract and crude polysaccharide, the cia. p. ginseng was more effective on the growth of 1. casei an: G. lucidum was more effective on that of S. thermophilus. The effect of free amino acids on the growth of tactic acid bacteria was also examined. Arginine and lysine stimulated the growth of L. casei, whereas Lysine, serine, arginine, and glutamic acid stimulated the growth of 5. thermophilus.

The Protective Effects of Ganoderma lucidum on the DNA Damage and Mutagenesis (DNA손상 및 돌연변이에 대한 명지버섯의 방어효능)

  • Ganoderma lucidum is commonly known as medically potent mushroom, which has been widely used in China and other oriental countries for the treatment of various diseases, including cancer. In this report, we investigated the anti-oxidant and protective effect of Ganodema lucidum extract (GLE) against the DNA damage induced by free radical and U.V. In the assay of cell growth inhibition, the inhibitory cell growth rate induced by hydroxyl radical was dose-dependently decreased by GLE. This results support that GLE has a detoxifying activity against cytotoxicity of hydroxyl radical in E. coli cell. GLE also protected ColE1 plasmid DNA damage in the concentration of 200$\mu\textrm{g}$ per reaction on the DNA fragmentation assay. The nuclear tailing by hydrogen peroxide in single cell gel electrophoresis(SCGE) was decreased by GLE in the concentration of 50$\mu\textrm{g}$/ml. These data indicate that Ganoderma lucidum has an anti-oxidative activity to hydrogen peroxide. The mutation rate after irradiation of U.V. was reduced by 50$\mu\textrm{g}$/ml GLE and total number of Rif (Rifampicin) resistant mutants was decreased in a concentration dependent manner when added the GLE exogenously in a culture media. According to the results, it is likely that GLE has not only an anti-oxidative activity to hydroxyl radical but also an anti-mutagenic activity to U.V. mutagenesis.

Effect of ginseng residue on the growth of Ganoderma lucidum (Ganoderma lucidum의 생육에 미치는 홍삼박의 영향)

  • The ginseng residue, a by-product of ginseng tea manufacture, was used as growth promoting substance in the submerged cultivation of Basidiomycetes for its effective utilization. Ginseng residue contained about 46% of total sugar, 14% of crude protein, 12% of ash, and 0.16% of crude saponin. Among inorganic substances in ginseng residue, amount of Mg, Na, K and Ca were much more than other inorganic substances. All ginsenosides existed in red ginseng residue. Ganoderma lucidum and Pleurotus ostreatus could be excellently cultured in potato dextrose broth. Most effective additional concentration of residue extract for growth of these fungi was shown to be 0.2%, and the contents of crude protein and amino acid in mycelium were increased when 0.5% of residue extract were added to the medium.

Taxonomy of $Ganoderma$ $lucidum$ from Korea Based on rDNA and Partial ${\beta}$-Tubulin Gene Sequence Analysis

  • In the present study, a phylogenetic analysis was undertaken based on the internal transcribed spacer (ITS) rDNA and partial ${\beta}$-tubulin gene sequence of the $Ganoderma$ species. The size of the ITS rDNA regions from different $Ganoderma$ species varied from 625 to 673 bp, and those of the partial ${\beta}$-tubulin gene sequence were 419 bp. Based on the results, a phylogenetic tree was prepared which revealed that Korean $Ganoderma$ $lucidum$ strains belong in a single group along with a $G.$ $lucidum$ strain from Bangladesh.
