Characterization of Mucilage Produced from the Solid-state Fermentation of Soybean Grit by Bacillus firmus

  • Mucilage containing ${\gamma}-polyglutamic$ acid (PGA) was efficiently generated by the solid-state fermentation (SSF) of soybean grit by Bacillus firmus NA-1. B. firmus NA-1 was shown to be a glutamate-dependent strain for PGA production. The SSF of soybean grit was optimized in order to produce mucilage with a fortification of 5% glutamate, resulting in higher levels of mucilage production (6.14%) and a higher consistency index ($1.1\;Pa\;sec^n$). The sticky mucilage was comprised of 38% PGA, 7% levan, and some biopolymers. With regard to the viscoelastic properties of the mucilage solution, the viscous modulus (G") obtained from soybean grit fortified with 5% glutamate was approximately 64 times higher titan that of the mucilage solution obtained without glutamate. Although the addition of glutamate in the SSF of soybean grit influenced the rate of PGA production, the molecular weight of PGA remained unaltered, and was detected in a range between 1,400-1,440 kDa.

Potential Detection of Irradiated Dried Agricultural Products by Viscosity Measurement (점도측정법을 이용한 방사선 조사 건조농산품의 검지 가능성)

  • Dried vegetables, white ginseng and spices, which were exposed to gamma and electron beam irradiation, were used in a detection study by measuring their starch content and viscosity change. The samples tested showed different levels of starch content(15.64~60.86%), which was not directly proportional to the viscosity of the samples. The correlation coefficients between irradiation dose and viscosity change were lower in the samples, such as cabbage, carrot, clean vegetable(chunggyungchae), garlic, mushroom, green onion, and red pepper, while some higher coefficients were found in ginger(R2=0.9271), white ginseng (R2=0.6223) and onion (R2=0.7909). Thus, dried ginger and white ginseng were selected to be used for a detection of irradiated samples using specific parameters(threshold values). Specific parameter for the nonirradiated ginger and ginseng were 13.31 and 13.93, respectively. On the other hand, gamma and electron beam irradiated samples at 2.5 kGy, the lowest dose for a commercial purpose, showed decreased values, 11.92 and 11.15 in ginger, and moreover 4.40 and 5.10 in ginseng. It is expected that a proportional decrease in a specific parameter with the absorbed doses will be a potentially useful index for detecting whether starchy foods have been irradiated or not.

Performance Analysis of Economic VaR Estimation using Risk Neutral Probability Distributions

  • Traditional value at risk(S-VaR) has a difficulity in predicting the future risk of financial asset prices since S-VaR is a backward looking measure based on the historical data of the underlying asset prices. In order to resolve the deficiency of S-VaR, an economic value at risk(E-VaR) using the risk neutral probability distributions is suggested since E-VaR is a forward looking measure based on the option price data. In this study E-VaR is estimated by assuming the generalized gamma distribution(GGD) as risk neutral density function which is implied in the option. The estimated E-VaR with GGD was compared with E-VaR estimates under the Black-Scholes model, two-lognormal mixture distribution, generalized extreme value distribution and S-VaR estimates under the normal distribution and GARCH(1, 1) model, respectively. The option market data of the KOSPI 200 index are used in order to compare the performances of the above VaR estimates. The results of the empirical analysis show that GGD seems to have a tendency to estimate VaR conservatively; however, GGD is superior to other models in the overall sense.

  • We present a theoretical analysis of Gaussian beam propagation in nonlinear Kerr media by using aberration-free approximation and Huygens-Fresnel diffraction integral and obtain a simple analytic formular for Z-scan characteristics. Z-scan experiments are carried out on amorphous $As_2S_3$ thin film and compared with the theory developed, showing good agreement. The sign and the value of ${\gamma}$ have been measured at 633 nm to be $+8.65{\times}10^{-6}\textrm{cm}^2/W$. We also measured the far-field intensity profiles, which confirm again self-focusing effect.

  • This study is concerned with assessment of diethylnitrosamine(DEN) induced liver cell carcinogenesis by measurement of changes preceding the development of neoplasms. Therefore, it was undertaken to investigate the changes of liver-specific enzyme activities in rats (Sprague-Dawley) by ad libitum feeding of DEN. And also, as another objective index in urine, the level of urinary biopterin was measured by high performance liquid chromatography(HPLC) method. The results were as follows ; 1. Minor behavioral change, brittleness of hair and decreased amount of water and diet intake were observed in rats 7 weeks after DEN administration. 2. The body and liver weights were significantly(p<0.05) decreased in rats 11 weeks after DEN administration. 3. The ratio of liver weight to body weight was rather stable and not significantly decreased in the all treatment groups. 4. The liver specific enzyme activities(AST, ALT,$\gamma$-GTP) were significantly(p<0.05) increased in all treatment groups compared to control group. 5. Compared to normal level, urine biopterin level was significantly (p<0.05) increased in all treatment groups(p<0.05). In conclusion, this result confirmed that the DEN was one of the potent hepatocarcinogens. In experimental model of rats exposed to DEN, the results indicated that values of liver specific enzyme activities(AST, ALT, $\gamma$-GTP) and urine biopterin level could be useful complementary tumor indices.

Spiral Waves and Shocks in Discs around Black Holes: Low Compressibility and High Compressibility Models

  • Some authors have concluded that spiral structures and shocks do not develop if an adiabatic index $\gamma$ > 1.16 is adopted in accretion disc modelling, whilst others have claimed that they obtained well defined spirals and shocks adopting a $\gamma$ = 1.2 and a $M_2/ M_1$ = 1 stellar mass ratio. In our opinion, it should be possible to develop spiral structures for low compressibility gas accretion discs if the primary component is a black hole. We considered a primary black hole of 8M0 and a small secondary component of 0.5M$\bigodot$ to favour spiral structures formations and possible spiral shocks via gas compression due to a strong gravitational attraction. We performed two 3D SPH simulations and two 2D SPH simulations and characterized a low compressibility model and a high compressibility model for each couple of simulations. 2D models reveal spiral structures existence. Moreover, spiral shocks are also evident in high compressibility 2D model at the outer disc edge. We believe that we could develop even well defined spiral shocks considering a more massive primary component.

Comparative Effect of Gamma Irradiation and Phosphine Fumiagion on Color and Organoleptic Quality of White Ginseng (감마선과 Phosphine 처리가 백삼의 색도 및 관능적 품질에 미치는 영향)

  • Comparative effects of phosphine fumigation and gamma irradiation were evaluated on the qualities of white ginseng prepacked in a commercial laminated film in terms of water absorption, Hunter color parameters(L, a, b) and organoleptic properties(overall color/appearance, flavor) during 6 months of storage at ambient(20oC, 70% RH) and accelerated(40oC, 90% RH) conditions. Moisture content of packed white ginseng(initial 8.44%) linearly increased with storage period, thereby reaching to about 10% (r=0.9966) in ambient and more than 15%(r=0.9886) in accelerated conditions, respectively, at the 6th month of storage. The storage at severe conditions resulted in remarkable changes in Hunter's color values, decrease in whiteness(L) and increase in redness(a) and yellowness(b), while both treatments, phosphine and irradiation less than 5kGy, did not cause any significant changes in color parameters. Phosphine fumigation was shown detrimental to overall flavor of white ginseng(p<0.01), however or ganoleptic qualities of stored samples were effectively maintained by irradiation less than 5kGy(p< 0.05). As the index on the guality of white ginseng stored at severe conditions its organoleptic quality was highly correlated with moisture content(r= 0.9777) and Hunter color values(over 0.9), suggesting the critical values of 15.5% moisture and 72.56, 7.15, 21.45 in Hunter's L, a, b values, respectively.

A Correlation between Urinary Methylhippuric Acid and Hematologic Finding and Urinalysis Index due to Xylene Exposure

  • This study was performed to investigate the urinary methyl hippuric acid (UMHA) excretion among aircraft maintenance workers exposed to xylene by examination of blood test, urine test and biochemical tests. Study subjects (301) were divided into 2 groups; non-exposed group (n=123, who were not exposed to xylene), and exposed group (n=178, who were exposed to xylene). The average of blood level and MHA level was normal. The ${\gamma}$-GTP level was 35.5 IU/L (45.7%) in the exposed group and 27.9 IU/L (44.7%) in the non-exposed (control) group. The proportion of total cholesterol (TG) level higher than 150 mg/dl were 34.3% in the exposed group and 21.1% in the non-exposed group. These results were showed statistically significant difference (${\chi}^2$ = 6.10, P<0.05). Other items showed no statistically significant differences. The results of urine tests were no statistically significant differences in protein, occult blood and glucose level.

Effect of ${\gamma}$-Ray Irradiation on Surface Oxidation of Ultra High Molecular Weight Polyethylene/Zirconia Composite Prepared by in situ Ziegler-Natta Polymerization

  • Novel ultra-high molecular weight polyethylene (UHMWPE)/zirconia composites were previously prepared by the in situ polymerization of ethylene using a Ti-based Ziegler-Natta catalyst supported on to the surface of zirconia, as a bearing material for artificial joints. Tribological tests revealed that a uniform dispersion of zirconia in UHMWPE markedly increased the wear resistance. The effects of zirconia content on the oxidation behavior of the ${\gamma}$-ray-treated UHMWPE/zirconia composite surfaces were examined. The oxidation index that estimates the oxidation degree as the content of total carbonyl compounds was monitored using Fourier transform infrared spectroscopy-attenuated total reflectance. The changes in the surface composition due to the oxidation were confirmed by electron spectroscopy for chemical analysis. The extent of oxidation decreased with increasing zirconia content, which was attributed to the increased crystallinity as well as the decreased polymer portion of the UHMWPE/zirconia composites.

Partial safety factors for retaining walls and slopes: A reliability based approach

  • Uncertainties in design variables and design equations have a significant impact on the safety of geotechnical structures like retaining walls and slopes. This paper presents a possible framework for obtaining the partial safety factors based on reliability approach for different random variables affecting the stability of a reinforced concrete cantilever retaining wall and a slope under static loading conditions. Reliability analysis is carried out by Mean First Order Second Moment Method, Point Estimate Method, Monte Carlo Simulation and Response Surface Methodology. A target reliability index ${\beta}$ = 3 is set and partial safety factors for each random variable are calculated based on different coefficient of variations of the random variables. The study shows that although deterministic analysis reveals a safety factor greater than 1.5 which is considered to be safe in conventional approach, reliability analysis indicates quite high failure probability due to variation of soil properties. The results also reveal that a higher factor of safety is required for internal friction angle ${\varphi}$, while almost negligible values of safety factors are required for soil unit weight ${\gamma}$ in case of cantilever retaining wall and soil unit weight ${\gamma}$ and cohesion c in case of slope. Importance of partial safety factors is shown by analyzing two simple geotechnical structures. However, it can be applied for any complex system to achieve economization.
