  • A symptoms of Diabetic is finded in many recordes, that is said to Diabetic or dryness or migratory arthralgia or flaccid paralysis of limb or abscess or arthralgia, but Diabetic is most closely symptom. So, study the 3-symptoms of Diabetes mellituscause, thses are polyuria and polyphasia and thirst, and this is similar symptoms of So-gal. So, bibliographic study or pathological approach of symptoms of So-gal, and So-gal in oriental medicine has come to the following conclution. The symptoms of So-gal is polyuria and polyphasia and thirst, is due to Simsinbulgyo, that is mean to breakdown of the normal physiological coordination between the heart and the kidney. A thirst is due to heat-transformation have an effect to the upper energizer and the heart and the lung and the stomach in Simsinbulgyo. A polyuria is due to failure of the kidneys Yang, that is due to failure of Yang of the heart in Simsinbulgyo.

Development of aortic endothelial cells to express CD37 and CD73 isolated from alpha 1,3-galactosyltransferase knock-out and MCP expressing pig (alpha 1,3-galactosyltransferase 기능 제거 및 MCP 발현 형질전환 돼지의 대동맥 혈관내피세포에 CD37/CD73 발현 세포주 개발)

  • Acute vascular rejection has been known as a main barrier occurring in a xenograted tissue of alpha 1,3-galactosyltransferase knock-out (GalT KO) pig into a non-human primate (NHP). Adenosine which is a final metabolite following sequential hydrolysis of nucleotide by ecto-nucleotidases such as CD39 and CD73, act as a regulator of coagulation, and inflammation. Thus xenotransplantation of CD39 and CD73 expressing pig under the GalT KO background could lead to enhanced survival of recipient NHP. We constructed a human CD39 and CD73 expression cassette designed for endothelial cell-specific expression using porcine Icam2 promoter (pIcam2-hCD39/hCD73). We performed isolation of endothelial cells (pAEC) from aorta of 4 week-old GalT KO and membrane cofactor protein expressing pig ($GalT^{-MCP/-MCP}$). We were able to verify that isolated cells were endothelial-like cells using immunofluorescence staining analysis with von Willebrand factor antibody, which is well known as an endothelial maker, and tubal formation assay. To find optimal condition for efficient transfection into pAEC, we performed transfection with GFP expression vector using four programs of nucleofection, M-003, U-023, W-023 and Y-022. We were able find that the program W-023 was optimal for pAEC with regard to viability and transfection efficiency by flow cytometry and fluorescent microscopy analyses. Finally, we were able to obtain $GalT^{-MCP/-MCP}/CD39/CD73$ pAEC expressing CD39 and CD73 at levels of 33.3% and 26.8%, respectively. We suggested that pACE isolated from $GalT^{-MCP/-MCP}$ pig might be provided as a basic resource to understand biochemical and molecular mechanisms of the rejections and as an alternative donor cells to generate $GalT^{-MCP/-MCP}/CD39/CD73$ pig expressing CD39 and CD73 at endothelial cells.

Enzymatic Synthesis of 1, 2-Hexanediol Galactoside by Whole Cells of β-Galactosidase-containing Recombinant Escherichia coli (β-galactosidse 함유하는 재조합 대장균 세포를 이용한 1,2-hexanediol galactoside의 합성)

  • Recently, it has been reported that some preservatives used in cosmetics lead to skin problems. Among the many cosmetic ingredients, 1, 2-hexanediol (HD) is used as both a preservative and humectant. In order to develop safer ingredients, we studied the synthesis of 1, 2-hexanediol galactoside (HD-G) by a transgalactosylation reaction using β-galactosidase (β-gal)-containing recombinant Escherichia coli cells. The transgalactosylation reaction was carried out under high-lactose conditions for 24 hr. After 12 hr had elapsed, a new spot was identified by thin-layer chromatography (TLC) analysis, and we presumptively designated this new spot as HD-G. Then, we carried out the purification of the presumptive HD-G spot from the reaction mixture by using silica gel chromatography, and its mass was measured by electrospray ionization-mass spectrometry. The purified new spot on the chromatograph was identified a sodium adduct ion ([M+Na]+, m/z = 303.1423) of HD-G. In addition, when purified HD-G was hydrolyzed using commercially available E. coli β-gal, it was observed that a galactose molecule was released from HD-G. That is, it was demonstrated that HD-G is a galactoside derivative of HD. Finally, we confirmed that HD-G was enzymatically synthesized by E. coli β -gal as a new molecular entity. In the future, we plan to determine the minimum inhibitory concentrations of HD-G against different bacterial species. The cytotoxicity of HD-G against human skin cells will also be examined. It is expected hopefully that the galactosylation of HD would improve the functionality of HD-G.

Computation of Complete Bouguer Anomalies from Free-air Anomalies in East Sea (동해 지역의 프리에어 이상으로부터 완전부우게 이상의 계산)

  • This paper describes the results of complete Bouguer anomalies computed from the Free-air anomalies that derived from Sandwell and DNSC08 marine gravity models. Complete bouguer corrections consist of three parts: the bouguer correction (Bullard A), the curvature correction (Bullard B) and the terrain correction (Bullard C). These all corrections have been computed over the East Sea on a $1'{\times}1'$elevation data (topography and bathymetry) derived from ETOPO1 global relief model. In addition, a constant topographic (sea-water) density of $2,670kg/m^3$($1,030kg/m^3$) has been used for all correction terms. The distribution of complete bouguer anomalies computed from DNSC08 are -34.390 ~ 267.925 mGal, and those from Sandwell are -32.446 ~ 266.967 mGal in East Sea. The mean and RMSE value of the difference between DNSC08 and Sandwell is $0.036{\pm}2.373\;mGal$. The highest value of complete bouguer anomaly are found around the region of $42{\sim}43^{\circ}N$ and $137{\sim}139^{\circ}E$ (has the lowest bathymetry) in both models. These values show that the gravity distribution of both models, DNSC08 and Sandwell, are very similar. They indicate that satellite-based marine gravity model can be effectively used to analyze the geophysical, geological and geodetic characteristics in East Sea.

발효유의 종균 L. casei YIT 9018의 자연돌연변이주의 발생 빈도 및 그 특성에 관한 연구

  • 유산균을 종균으로 하여 유제품을 만드는 경우에 종균의 취급관리는 매우 중요한 일이다. 종균을 취급하는 과정에서 어떤 원인으로 인하여 종균의 활력 특히 산생성력이 감퇴되어 종균으로 사용할 수 없는 경우가 발생한다. 이러한 경우의 원인을 분석하여 그 예방책을 수립하기 위한 기초 연구로써 발효유의 종균으로 사용하고 있는 L. casei YIT9018의 spontaneous lac 변이주의 발생빈도 및 이러한 Lac 변이주의 특성을 검토하였다. parent colony에서의 Lac의 발생빈도는 MRS 배지에서 400 coloney중에서 1개, 10% 환원 탈지분유에서는 984개중 1개의 배율로 나타났다. 분리된 Lac 변이주중에는 Lac Gal도 검출되었다 대부분의 Lac를 탈지유배지에 배양할 경우 10일이 지나면 우유를 응고시켰으나 Lac Gal는 30일이 지난뒤에 응고시켰다. Lac변이주를 MRS-lactose 배지의 plate에 도말하여 배양하면 4일 이후부터는 revertant가 나타났으나 Lac Gal Cell에서는 인정되지 않았다. 얻어진 Lac 변이주의 plasmid를 분리하여 parent와 비교한 결과 plasmid DNA가 소실되었음을 확인하였다.

Separation and Identification of Galactosylmanno-oligosaccharides from Hydrolyzate of Brown Copra Meal by Trichoderma β-Mannanase

  • Three kinds of oligosaccharides were obtained from the hydrolysate of brown copra meal galactomannan by a purified extracellular ${\beta}$-mannanase from Trichoderma sp. These oligosaccharides were identified as Man-Man, ${Gal^2}{Man_3}(6^2 mono-O-{\alpha}-D-galactopyranosyl-4-O-{\beta}-D-mannotriose)$, and ${Gal^2}{Man_6}(6^2-mono-O-{\alpha}-D-galactopyranosyl-4-O-{\beta}-D-mannohexaose)$, where Gal- and Man-represent ${\alpha}$-1,6-D-galactosidic and ${\beta}$-1,4-mannosidic linkages, respectively. The mode of action of ${\beta}$-mannanase on brown copra meal galactomannan is described on the basis of the structure of these oligosaccharides.

Applications of Tandem Mass Spectrometry in the Structure Determination of Permethylated Sialic Acid-containing Oligosaccharides

  • Sets of sialic acid-containing trisaccharides having different internal and terminal linkages have been synthesized to develop a sensitive method for analysis of the reducing terminal linkage positions. The trisaccharides, sialyl($\alpha$ 2-3)Gal($\beta$ 1-3)GalNAc and sialyl($\alpha$ 2-3)Gal($\beta$ 1-X)GlcNAc where X=3, 4 and 6, were synthesized and examined using electrospray ionization (ESI)-collision induced dissociation (CID) tandem mass spectrometry (MS/MS). The compounds chosen for this study are related to terminal groups likely to be found on polylactosamine-like glycoproteins and glycolipids which occur on the surface of mammalian cells. The purpose of this study is to develop tandem mass spectrometral methods to determine detailed carbohydrate structures on permethylated or partially methylated oligosaccharides for future applications on biologically active glycoconjugates and to exploit a faster method of synthesizing a series of structural isomeric oligosaccharides to be used for further mass spectrometry and instrumental analysis.

Application of Relative Gravity Surveying and Modeling to Sinkhole Detection (싱크홀 탐지를 위한 상대중력측량과 중력모델링 기법의 활용)

  • The purpose of this research was to develop and present methods to detect sinkholes which can exist underneath the surface of the ground. First, we buried a water tank with dimensions $1.8{\times}0.8{\times}0.8m$ at a distance of 1.8 m from the surface. This played the role of the sinkhole. Secondly, we created a square zone with sides 12 meters away from the buried water tank. Within this zone, we measured the gravity at 1-meter intervals using a Scintrex CG5 relative gravimeter with a resolution of 0.001 mGal. Additionally, we performed three-dimensional (3-D) gravity modeling to calculate the theoretical values of the relative gravity around our model sinkhole. The resulting values for the relative gravity around the sinkhole depended on the method used. The measured effect of gravity was 0.036 mGal and the effect calculated using 3-D modeling was 0.024 mGal. Our results suggest that sinkholes that are similar in size to the water tank used in this study can be detected using relative gravity surveys. Smaller sinkholes can be detected by reducing the intervals between the relative gravity measurements.

Development of a Reporter System Monitoring Regulated Intramembrane Proteolysis of the Transmembrane bZIP Transcription Factor ATF6α

  • When endoplasmic reticulum (ER) functions are perturbed, the ER induces several signaling pathways called unfolded protein response to reestablish ER homeostasis through three ER transmembrane proteins: inositol-requiring enzyme 1 (IRE1), PKR-like ER kinase (PERK), and activating transcription factor 6 (ATF6). Although it is important to measure the activity of ATF6 that can indicate the status of the ER, no specific cell-based reporter assay is currently available. Here, we report a new cell-based method for monitoring ER stress based on the cleavage of $ATF6{\alpha}$ by sequential actions of proteases at the Golgi apparatus during ER stress. A new expressing vector was constructed by using fusion gene of GAL4 DNA binding domain (GAL4DBD) and activation domain derived from herpes simplex virus VP16 protein (VP16AD) followed by a human $ATF6{\alpha}$ N-terminal deletion variant. During ER stress, the GAL4DBD-VP16AD(GV)-$hATF6{\alpha}$ deletion variant was cleaved to liberate active transcription activator encompassing GV-$hATF6{\alpha}$ fragment which could translocate into the nucleus. The translocated GV-$hATF6{\alpha}$ fragment strongly induced the expression of firefly luciferase in HeLa Luciferase Reporter cell line containing a stably integrated 5X GAL4 site-luciferase gene. The established double stable reporter cell line HLR-GV-$hATF6{\alpha}$(333) represents an innovative tool to investigate regulated intramembrane proteolysis of $ATF6{\alpha}$. It can substitute active pATF6(N) binding motif-based reporter cell lines.

Protective Effects of Angelica keiskei Extracts Against D-Galactosamine (GalN)-induced Hepatotoxicity in Rats

  • Although the vegetable Angelica keiskei (AK) has widely been utilized for the purpose of general health improvement among Korean population, its functionalities are not very well defined. In this study, we examined the effects of methanol extract of AK in rats on the biochemical changes induced by two hepatotoxins, D-galactosamine (GalN) and carbon tetrachloride ($CCl_4$). AK was orally administered once daily for 7 days to male rats at 200 and 500 mg/kg, before hepatotoxins. Effects of AK were assessed 24 hr later. AK pretreatments at 200 and 500 mg/kg significantly blunted GalN-induced elevation in liver lipid peroxidation, plasma aspartate-transaminase (AST) and alanine-transaminase (ALT) activities. AK also prevented, after 500 mg/kg but not after 200 mg/kg, the GalN-induced elevation in triglyceride, total cholesterol and LDL-cholesterol levels. Differently from against GalN-induced toxicity, AK did further elevate the $CCl_4$-induced rise in AST, ALT and lipid peroxidation. These results suggest that AK, when pre-administered prior to GalN, exerted protective effects against GalN-induced hepatotoxicity, in contrast however, AK exacerbated that induced by $CCl_4$. To explore possible mechanism for the toxicity-potentiating effects of AK on $CCl_4$, the activity of hepatic drug metabolism after AK treatment was assessed. It was observed that AK increased the activity of aniline hydroxaylase, a cytochrome P450 isoenzyme responsible for metabolic activation of $CCl_4$. This finding suggests that hepatoprotective effects of AK are not equally expected depending on hepatotoxins employed.
