유방암 세포(EMT6) 이식 마우스에서 녹차폴리페놀 음용이 시스플라틴의 암 조직 성장 억제에 미치는 영향 (Effects of Dietary Tea Polyphenol on Tumor Growth Inhibition by Cisplatin in EMT6 Breast Tumor-bearing Mice)

  • 본 연구는 녹차폴리페놀이 시스플라틴의 항암작용과 신장 독성에 미치는 영향을 유방암 세포(EMT6)와 암세포 이식마우스를 이용하여 in vitro와 in vivo 실험으로 관찰하였다. 배양한 EMT6 세포에서 녹차폴리페놀은 시스플라틴에 의한 세포 독성을 증가시켰다. 마우스에 EMT6 세포를 주사하여 유발된 종양의 크기가 시스플라틴군(CP)보다 시스플라틴+녹차폴리페놀군(CP+GTP)에서 유의하게 작았고, 종양조직 p53와 caspase-3 활성화가 시스플라틴군(CP)보다 시스플라틴+녹차폴리페놀군(CP+GTP)에서 유의하게 높았으며, 신장 GGT와 AP 활성은 시스플라틴군(CP)보다 시스플라틴+녹차폴리페놀군(CP+GTP)에서 유의하게 높았고, 신장 조직학적 소견에서 신세뇨관 확장과 괴사가 시스플라틴군(CP)보다 시스플라틴+녹차폴리페놀군(CP+GTP)에서 유의하게 낮았다. 이상의 결과 녹차폴리페놀은 EMT6 유방암 세포를 이용한 in vitro 및 in vivo 실험에서 시스플라틴의 항암작용을 증강시키면서 신장에 대한 독성 부작용은 감소시키는 효과가 있는 것으로 추측된다. 녹차폴리페놀의 시스플라틴 항암작용 증강과 신장 독성 억제 및 감소 효과는 시스플라틴에 의한 암 치료 시 화학요법제의 보조제로서 이용가치가 있는 것으로 생각되며, 항암화학요법제에 대한 보조제로의 개발을 위해서는 대규모 동물실험을 통한 효과 입증 및 부작용에 대한 실험과 임상연구가 뒷받침되어야 할 것으로 사료된다.

분말녹차 첨가 김치의 품질 특성 (Quality Characteristics of Kimchi Added with Green Tea Powder)

  • 본 연구의 목적은 김치에 분말상태의 녹차를 첨가하고, 김치에 첨가된 녹차가 젖산균의 산생성과 생육, 관능적특성 및 몇 가지 휘발성냄새성분에 미치는 영향을 조사하고자 하는 것이다. 김치 제조원료에 분말녹차를 0.2, 0.4, 0.6, 1.2%(w/w)의 농도로 첨가하였다. pH는 녹차의 첨가농도가 증가함에 따라 비숙성시료와 숙성1일에는 약간 증가하였으나, 숙성2일과 숙성3일에는 보다 현저하게 증가하였다. 산도는 녹차의 첨가농도가 증가함에 따라 비숙성시료와 숙성1일에는 대체적으로 증가하였으나, 숙성2일과 숙성3일에는 대체적으로 감소하였다. 한편 녹차의 첨가로 젖산균수는 대체적으로 큰 변화가 없었다. 전반적인 기호도, 맛, 냄새는 녹차 0.2% 또는 0.4% 첨가시료가 다른 시료보다 관능성이 우수하였고, 조직감은 숙성2일 또는 숙성3일시료의 경우 녹차의 첨가농도가 높을 때 관능성이 저하되었다. 색상은 녹차의 첨가농도가 증가할수록 관능성이 저하되었다. 비숙성시료에 비하여 숙성시료에서 diallyl sulfide와 methyl trisulfide가 새로이 생성되었으며, 일부의 냄새성분 함량은 숙성과정 중에 증가하거나 변화를 보였다.

배양조건이 Bacillus subtilis 융합주의 ${\gamma}-Glutamyltranspeptidase{\;}({\gamma}-GTP)$ 활성에 미치는 영향 (Culture Characteristics on the Activity of ${\gamma}-Glutamyltranspeptidase{\;}({\gamma}-GTP)$ by Bacillus subtilis Fusant)

  • Bacillus subtilis 돌연변이주 중 ${\gamma}-GTP$활성이 높은 SM-2와 SM-10을 융합시켜 획득한 융합주 중 ${\gamma}-GTP$활성이 높은 융합주 FG-21의 배양적 특성을 조사하였다. 융합주 FG-21에 의한 ${\gamma}-GTP$의 생산은 1% glycerol, 1% peptone, 0.1% citric acid, 5 mM $K_2HPO_4$, 1 mM $FeCl_3$, 1 mM $MgCl_2$, 1 mM $NH_4Cl$, pH 7.0의 배지에 의해 $37^{\circ}C$에서 36시간 배양했을 때 621 U/mL으로 최대한 활성을 보였다. 융합주 FG-21이 생산한 biopolymer A가 단백질함량이 38.4%이고 B. subtilis K-1이 생산하는 biopolymer B의 단백질함량은 19.3%로 융합주 FG-21이 생산하는 Biopolymer A보다 함량이 낮았다. 융합주 FG-21의 biopolymer가 모균주의 biopolymer B보다 단백질함량이 높은 것은 상대적으로 높은 ${\gamma}-GTP$활성이 높아서 생성된 PGA가 levan보다 함량이 많기 때문인 것으로 사료된다. 총당함량은 biopolymer A가 58.5%, biopolymer B가 76.5%로 나타났다. 융합주 FG-21과 모균주가 생산하는 biopolymer의 단백질함량과 총당함량비는 융합주 FG-21이 생성한 biopolymer는 38 : 59. 모균주가 생산한 biopolymer B는 19 : 78의 비율이었고 HPLC 분석에 의한 biopolymer A와 B의 fructose함량은 각각 537.7 mg/g biopolymer, 764.4 mg/g biopolymer으로 나타났다. Biopolymer A와 B의 glutamic acid함량은 각각 163.7 mg/g, 94.3 mg/g이었다. Biopolymer A와 B의 fructose와 glutamic acid의 함량비는 각각 78 : 22, 89 : 11의 비로 함유되어 있었다. 따라서 융합주에 의해 생산된 biopolymer는 모균주보다 구성상태가 변화된 biopolymer를 생산한 것으로 판단되어 fusion과 같은 균주개량을 통하여 물질의 구성적, 물질적 변화된 biopolymer를 생산할 수 있을 것이라 사료된다.

Effects of Green Tea Polyphenols and Fructo-oligosaccharides in Semi-purified Diets on Broilers' Performance and Caecal Microflora and Their Metabolites

  • This study was conducted to examine the effects of green tea polyphenols (GTP) and fructo-oligosaccharides (FOS) supplement on performance, counts of caecal microflora and its metabolites production. In female broiler chickens fed on semi-purified diets from 28 to 42 d of age, dietary green tea polyphenols (GTP) and fructo-oligosaccharides (FOS) significantly reduced mortality (p<0.05). Dietary GTP significantly decreased the total count of caecal microflora, each colonic population count and caecal flora metabolites contents when compared to other groups (p<0.05). Dietary FOS did not influence the total count of caecal flora but it selectively increased Bifidobacteri and Eubacteria counts (p<0.05) and decreased the count of other microflora and concentrations of caecal phenols and indole (p<0.0.5). These results suggest that GTP and FOS in semi-purified diets can decrease mortality and change the caecal colonic flora population, but GTP shows antibiotic-like effects of non-selectively decreasing all colonic flora and then metabolites, and FOS acts selectively by increasing profitable microflora and decreasing production of caecal microflora metabolites besides volatile fatty acids.

Ashbua gossypii에서의 리보플라빈 측쇄의 기원 (The Origin of Ribityl Side Chain of Riboflavin in Ashbya gossypii)

  • E. coli에서 ribosyl-HTP(hydroxytriamino pyrimidine)를 GTP로 부터 합성하는 효소 GTP cyclohydrolase II가 발견된 뒤 riboflavin의 ribityl group이 guanine nucleotide의 ribosyl group에서 직접 유래한다는 가설이 제안되었다. 본 연구에서는$(U- ^{14}C)$ guanosine을 media에 첨가하여 배양한 riboflavin overproducer 균주 Ashybya 에서 추출 정제한 riboflavin과 RNA에 각각 Incorporate된 guanosine label의 specific radioactivity를 비교 측정함으 로써 ribity I group 이 guanosine에서 기훤한다는 결과를 얻을 수 있었다. 이는 GTP cyclohydrolase II가 riboflavin 생합성의 초기 단계에서 직접 관여한다는 가설을 지지해 주는 것이다.

Biochemical Characterization of Oligomerization of Escherichia coli GTP Cyclohydrolase I

  • GTP cyclohydrolase I (E.C. 3.5.4.16) is a homodecameric protein that catalyzes the conversion of GTP to 7,8-dihydroneopterin triphosphate (H2NTP), the initial step in the biosynthesis of pteridines. It was proposed that the enzyme complex could be composed of a dimer of two pentamers, or a pentamer of tightly associated dimers; then the active site of the enzyme was located at the interface of three monomers (Nar et al. 1995a, b). Using mutant enzymes that were made by site-directed mutagenesis, we showed that a decamer of GTP cyclohydrolase I should be composed of a pentamer of five dimers, and that the active site is located between dimers, as analyzed by a series of size exclusion chromatography and the reconstitution experiment. We also show that the residues Lys 136, Arg139, and Glu152 are of particular importance for the oligomerization of the enzyme complex from five dimers to a decamer.

Physicochemical Properties and Shelf-Life of Regular-Fat Sausages with Various Levels of Grape Tomato Powder Prepared by Different Drying Methods

  • This study was aimed to investigate the physicochemical properties, texture, and antioxidant, and antimicrobial activities of regular-fat sausages (RFSs) mixed with 0.25 and 0.5% of oven-dried and freeze-dried grape tomato powder (GTP, 150 ㎛) during storage at 4℃. RFSs were made by six treatments that included: control (CTL), REF (sausages with 0.1% ascorbic acid alone), F1GTPSs (F1) and F2GTPSs (F2) (sausages with 0.25% and 0.5% freeze-dried GTP), and O1GTPSs (O1) and O2GTPSs (O2) (sausages with 0.25% and 0.5% GTP oven-dried at 100℃). Sausages with added oven-dried grape tomato powders (OGTPs) showed decreased pH, lightness (L), total plate count (TPC), and thiobarbituric acid-reactive substances (TBARS) compared to the sausages mixed with freeze-dried GTP (FGTPSs), but also had the highest redness (a) and yellowness (b) values among the treatments. With increasing levels of GTP, the hardness and chewiness of the sausages gradually decreased and these were decreased more in the FGTPSs (F) than in the OGTPSs (O). Compared to the FGTPSs, OGTPSs had higher antioxidant and antimicrobial activities, which extend the shelf-life of meat products. Application of OGTP to RFSs resulted in higher lipid antioxidant, antimicrobial activities, improving physicochemical properties and extended the shelf-life.

Effects of Thiamine Pyrophosphate on the Inhibition of Self-splicing of Primary Transcripts of T4 phage Thymidylate Synthase Gene in the Presence of GTP

  • Effects of GTP on the inhibition of self-splicing of primary transcripts of the phage T4 thymidylate synthase gene (td) by thiamine pyrophosphate and its analogs have been investigated. The order of the inhibitory efficiency for compounds tested was as follows: thiamine pyrophosphate > thiamine monophosphate > thiamine. of all compounds examined, thiamine pyrophosphate was the most potent inhibitor, Increasing GTP concentration in splicing reaction tended to overcome the suppressive effects of self-splicing by thiamine pyrophosphate and its analogs. The inhibition by thiamine pyrophosphate was most sensitized to a higher concentration of GTP, It has been speculated that the key structural features in thiamine pyrophosphate and its analogs responsible for the inhibition of splicing may be a thiamine moiety in which the phosphorylation of 2-hydroxylethyl group on 5-position of thiazolium ring rendered further stimulation of inhibition in self-splicing reaction..

The Status of Guanine Nucleotides in Taxol-Stabilized Microtubules Probed by 31P CPMAS NMR Spectroscopy

  • Rapid exchange and hydrolysis of the tubulin-bound guanine nucleotides have been known to govern the dynamics of microtubules. However, the instability and low concentration have made it difficult for the microtubule-bound GTP to be observed directly. In this study, we circumvent these problems by lyophilization and using cross-polarization techniques. $^{31}P$ NMR signals were detected from the tubulin-bound GTP in microtubules for the first time. Analysis of the $^{31}P$ CPMAS NMR spectrum indicates that GTP hydrolysis was delayed by the presence of taxol.

서울시 교직원 당뇨병의 역학적 특성 및 관련위험요인분석 (A Study of the Epidemiological Characteristics and Related Factors in School personnel with Diabetes Mellitus)

  • Purpose: To describe the prevalence, epidemiological characteristics, and related factors of diabetes mellitus in school personnel. Method:5,384 school personnel (2,638 males 2,746 females) received physical examinations at the Seoul School Health Center in 2002. Using the American Diabetes Association criteria, and fasting blood sugar(FBS) was classified as normal at 110>=FBS, borderline 111-125, and the diabetes group 126<=FBS. Related factors of each group were evaluated by gender. Results: In the diabetes group, there were 4.74% males and 0.8% females, and the borderline group was 6.37% males and 2.22% females. The older age group gad a higher distribution in both males and females in the diabetes group. In the diabetes group, the mean and abnormal rate of BMI, systolic hypertension, diastolic hypertension, total cholesterol, GOT, GPT, and GTP were higher than in the normal group. In life style factor, the diabetes group gad a higher distribution of meat eating and smoking more than 20 years than the normal group. The effect of FBS on BMI, BP(blood pressure), total cholesterol, GOT, GPT, and GTP were investigated after controlling for confounding variables. In the borderline and diabetes group, the mean of BMI, BP, total cholesterol, GOT, GPT, and GTP was higher in dose-response effect. In the logistic multiple regression, the related factors of the diabetes group were hypertension and abnormal GTP in males, and abnormal total cholesterol and GTP in females. In the borderline group, the related factors were meat eating, hypertension, and high GOT in males and the older age group, and hypertension, total cholesterol, and abnormal GTP in females. Conclusions: It is possible to manage all related factors of diabetes mellitus except for age. An appropriate program for health promotion is necessary.
