This study was conducted to investigate the effect of acorn extracts on the antioxidative enzyme system. Male Sprague-Dawley rats(110$\pm$10g) were fed on containing normal and high fat diets. They were orally administrated (0.02g/100g B.W) of acorn ethylacetate-extract and water-extract at the same time once a day, respectively. Net weight gain and feed efficiency ratio were increased in high fat diet groups and decreased by acorn extracts administration. The effect of acorn extracts on hepatic glutathione peroxidase(GSH-Px), glutathione S-transferase(GST) and catalase activities did not show significancy in normal fat diet groups. GST and catalase activities and lipid peroxidation content(LPO) were significantly increased in high fat diet groups and this increment were decreased by acorn extracts administration. However GSH-Px activity and GSH content were decreased in high fat diet groups but increased by acorn extracts administration. The activities of lactate dehydrogenase(LDH), alkaline phosphatase(ALP) and aminotransferase in serum were significantly increased in high fat diet group but these increment reduced in acorn extracts administration groups. These results indicate that acorn extracts could improve the liver function and prevent the metabolic diseases by hyperlipidemia.
The present study investigated the protective effects of green tea against acute ethanol-induced lipid peroxidation and the change of antioxidative enzyme activities in various regions of rat brain : cortex, cerebellum, striatum and hippocampus. The following parameters were examined : malondialdehyde(MDA) levels and activities of superoxide dismutase(SOD), catalase and glutathione peroxidase(GSH-Px). Male Sprague-Dawley rats were given the experimental containing 1% green tea powder or control diet for 4 weeks, and at the end of feeding diet group received acute ethanol(5g/kg body weight) or equicaloric sucrose solution intragastrically. Green tea powder significantly decreased MDA levels in the striatum compared to control-non alcohol treated group to 1% green tea-non alcohol treated group without altering the antioxidative enzyme activities. Green tea resulted in a significant increase in GSH-Px activities in the hippocampus compared to either control-non alcohol treated group(0.043units/mg protein) or 1% green tea-non alcohol treated group(0.071units/mg protein). In conclusion, these results suggest that moderate consumption of green tea leaves can exert protective effects against ethanol-induced oxidative stress in brain regions, by reducing MDA concentrations in the striatum and enhancing GSH-Px activities in the hippocampus. 사람 기관지상피세포주인 BEAS-2B에 담배연기농축액(CSC)을 처리하여 유도된 1198 세포주는 대조군 세포주인 1799에 비해 현저하게 낮은 glutathione 농도와 낮은 glutamate-cysteine ligase(GCL), glutathione peroxidase(GPx), glucose-6-phosphate dehydrogenase(G6PD), catalase 효소활성을 보였다. 두 세포주를 포도당 존재 하에서 4시간 hypoxia 처리 후 reoxygenation 하면서 시간에 따른 세포의 항산화계 활성을 측정한 결과, 1799 세포주에서는 의미 있는 변화가 관찰되지 않은 반면, 1198 세포주에서는 hypoxia 처리에 의해 glutathione의 농도 및 GSH/GSSG 비와 G6PD 활성이 감소되었고, reoxygenation 기에는 GPx, glutathione reductase(GRd), G6PD, superoxide dismutase 활성이 감소되었다. 그러나 reoxygenation 2시간 이후에는 GRd와 G6PD 활성의 회복이 관찰되었으며, 그 결과 GSH/GSSG 비율이 회복되었다. 이 실험 결과는 CSC가 능력을 현저히 저하시킬 수 있음을 보여준다. Glutathione은 hypoxia-reoxygenation에 의한 산화적 스트레스 하에서 항산화제로서의 역할뿐 아니라, 세포 내 GSH/GSSG 비의 변화를 통해 산화적 스트레스에 대한 항산화계의 적응 반응 여부를 결정하는 중요한 인자로 작용할 것으로 보여진다.
In the present study, to determine whether the antioxidative components of Korean red ginseng protect against radiation damage and the possible relationship among the radioprotective effects and antioxidant actions, the effects of total saponin (200 mg/kg, ip) and lipophilic fraction (200 mg/kg, oral) preferment of mice on the survival ratio, major antioxidant enzymes (SOD, catalase and glutathione peroxidase) activities, glutathione levels and lipid peroxidation in the liver were exiled for 2 weeks after whole ${\gamma}$-body ${\gamma}$-irradiation (6.5 Gy). The 30-day survival ratio increased from 10% to 57% and 40% for mice treated with total saponin and lipophilic fraction, respectively. On day 14 after ${\gamma}$-irradiation, the ginseng total saponin pretreatment produced a slight increase of antioxidant enzymes activities and significantly Increased reduced glutathione (GSH) contents (p<0.05) in the liver compared with non-treated group. Pretreatment with ginseng total saponin significantly deceased GSSG/total GSH ratio (p<0.05) without change of GSSG in the liver and inhibited the radiation-induced incense in the hepatic malondialdehyde levels. (p<0.05) In these results, GSH plays an important role in the liver in several detoxifications and the reduction of lipid peroxides. Thus, it appears that total saponin of red ginseng exerts its radioprotective effect by accelerating the production of endogenous antioxidants, such as glutathione from radiation induced damages and thereby oxygen free radicals.
The effects of Opuntia ficus-indica (OF) administration on the biochemical parameters of function in liver tissue and serum of $CCl_4$ treated rats were investigated. Opuntia ficus-indica (200 mg/kg) was administered into rats intraperitoneally for two weeks. $3.3m\ell$ of $CCl_4$$_4$ (50% $CCl_4$ : Olive oil = 1 : 1) was treated to rats on the 14th day and 15th day and they were operated on 15th day. We examined the antioxidative enzymatic activity by measuring the level of AST (Aspartate aminotransferase), ALT (Alanine aminotransferase), GSH (Glutathione reduced form), GSSG (Glutathione oxidezed form), GPx (GSH-peroxidase), SOD (Superoxide dismutase) and CAT (Catalase) in serum and liver tissue of rats. OFC administered group showed 24.8% of inhibitory effect in AST activity compared to $CCl_4$ -treated abnormal group (CTA). ALT level of OF administered group was decreased by 60.7% to the level of CTA. GSH, GSSG and GPx of OFC administered group were significantly higher than those of CTA group. SOD and CAT in OFC administered group were increased by 28.3% and by 16.9% respectively compared to those of CTA group.
The present study was conducted to investigate the effects of flowers ethanol extract of Elsholtzia splendens (ESE) on the antioxidant defence system in mice with 7,12-dimethylbenz(a)anthracene (DMBA)-induced oxidative stress. The ESE was pre-administered orally to 2 groups of mice at 10 and 50mg/kg body weight (BW) for 5 weeks. Subsequently, mice with pretreatment of ESE received DMBA intragastrically at a dose of 34 mg/kg BW twice a week for 2 weeks. In DMBA alone group, biomarkers of oxidative stress (TBARS value, carbonyl content, and serum 8-OH-dG) were significantly increased. Also, the antioxidant enzymes were down-regulated. ESE significantly restored the TBARS value and carbonyl content at both doses, while a decrease in the elevated serum 8-OH-dG content was observed only at the higher dose. The DMBA-induced decreases in catalase and superoxide dismutase (SOD) activities were restored to nearly control levels by ESE. Glutathione peroxidase (GSH-px) and glutathione reductase (GR) activities, as well as the reduced glutathione (GSH)/oxidized GSH (GSSG) ratio, were significantly affected by ESE at the higher dose. These results suggest that ESE possesses antioxidant activity, which plays a protective role against DMBA-induced oxidative stress.
The aim of this study was to evalute the effect of multivitamin-mineral supplementation during pregnancy on plasma levels of antioxidants, erythrocyte antioxidant enzyme activities, and lipid peroxidation. A controlled, semi-randomized, prospective trial was performed by comparing the supplement group, which received multivitamin-mineral tables once daily for 10 weeks, with the control group. Plasma levels of $\beta$-carotene, tocopherol, coenzyme Q10, ascorbate, folate, zinc, and selenium and malondialdehyde (MDA), as well as the activities of superocxide dismutase(SOD) and glutathione peroxidase(GSH-Px) in erythrocytes were measured initially (20 wk gestation) and at the end of the intervention (34 wk gestation). In the control group, plasma ascorbate and selenium levels decreased and tocopherol levels increased. In the supplement group, a significant increase in plasma $\beta$-carotene(46%), conenzyme Q10 (42%), and zinc (24%) was observed after 10 weeks of supplementation. No changes were observed in the plasma levels of MDA, and erythrocyte GSH-Px activity, while SOD activity increased in both control group and the supplement group during the intervention. These data suggest that multivitamin-mineral supplementation during pregnancy produced moderate increases in plasma $\beta$-carotens, coenzyme Q10, and zinc concentrations but the enhancement of those plasma antioxidants had on direct on the plasma level of MDA, erythrocytes SOD or GSH-Px activities.
To analyze the correlation between the rusty root and the antiokidative activity in ginseng (Panax ginseng C.A.Meyer) roots, the levels of antioxidative activity in various tissues of healthy and rusty roots. The superoxide dismutase activity in rusty roots (126.9 units/mg protein) was approximately 3.5 times higher than that in healthy roots. The catalase activity in rusty roots was approximately 1.6 times higher than that in healthy roots, whereas the peroxidase activity showed a slight low level in msty roots. The 1.1 diphenyl-2-picryl-hydrazyl(DPPH) free radical scavenging activity in rusty roots was approximately 2.0 times higher than that in healthy roots. The total ascorbate content in healthy roots was 166~240 $\mu\textrm{g}$/g fr. wt. depending on the tissues. Interestingly, the oxidized dehydroascorbate (DHA) content occupied more than 80% in total ascorbate content. The total ascorbate content in rusty roots was a similar level with healthy roots, but the reduced ascorbate content was 3.5~7.5 times higher than that of the healthy roots. The total glutathione content of the epidermis, cortex and stele tissues in 겨sty roots was 7.3, 4.8, 1.2 times higher than the healthy tissues, respectively. The ratio of reduced glutathione (GSH) and oxidized glutathione (GSSG) showed a similar fluctuation of total glutathione content in 겨sty roots. These results indicate that the high antioxidative activity in rusty roots may involve in overcoming the oxidative stress derived from environmental stresses.

1. Objectives This study was carried out to investigate the effects of Gunyuljejo-tang on the $CCl_4$-induced Liver Damage in Rats. 2. Methods Sprague-Dawley rats were devided into 5 experimental groups : Normal, $NS+CCl_4$(Solid extract of $CCl_4$ injection group after Normal Saline feed), $GYJJT+CCl_4$(Solid extract of $CCl_4$ injection group after Gunyuljejo-tang feed), $CCl_4+NS$(Normal Saline feed group after $CCl_4$ injection), $CCl_4+GYJJT$(Solid extract of Gunyuljejo-tang feed group after $CCl_4$ injection). Biochemical assays for serum enzyme activities such as AST, ALT, ALP, BUN, Creatinine, Uric Acid, Total Protein, Albumin, Total Cholesterol, Triglyceride, Glucose, and mRNA Revelation of Cytochrome p450 and activities such as LPO, GSH, GST, Glutathione Reductase, Glutathione Peroxidase, SOD, Catalase, Hydroxyproline, and ${\beta}$-Glucuronidase were performed. 3. Results (1) $GYJJT+CCl_4$ showed lower revelation of Cytochrome p450. (2) $GYJJT+CCl_4$ showed higher GSH activity than $NS+CCl_4$, $CCl_4+GYJJT$ showed higher GSH activity than $CCl_4+NS$ injection significantly. (3) $GYJJT+CCl_4$ showed higher GST activity than $NS+CCl_4$. $CCl_4+GYJJT$ showed higher GST activity than $CCl_4+NS$ significantly. (4) $GYJJT+CCl_4$ showed higher Glutathione Peroxidase activity than $NS+CCl_4$, $CCl_4+GYJJT$ showed higher Glutathione Peroxidase activity than $CCl_4+NS$ significantly. (5) $CCl_4+GYJJT$ showed higher SOD activity than $CCl_4+NS$ significantly. (6) $CCl_4+GYJJT$ showed higher Catalase activity than $CCl_4+NS$ significantly. (7) $GYJJT+CCl_4$ showed lower Hydroxyproline than $NS+CCl_4$ significantly, $CCl_4+GYJJT$ showed higher Hydroxyproline than $CCl_4+NS$ significantly. (8) $GYJJT+CCl_4$ showed higher ${\beta}$-Glucuronidase activity than $NS+CCl_4$, $CCl_4+GYJJT$ showed higher ${\beta}$-Glucuronidase activity than $CCl_4+NS$ significantly. 4. Conclusions Gunyuljejo-tang has the recovering effects on the $CCl_4$-induced Liver Damage significantly.

The present study was conducted to investigate effect of powdered herb of Aster scaber Thunb. (chamchwi) on antioxidant system in ethanol-administrated rats. Four week-old Sprague Dawley male rats which had initial body weights of 97.10$\pm$4.50 g were randomly divided into three groups: control (ethanol treated, vitamin E-deficient group); 5% chamchwi (ethanol-treated, 5% chamchwi powder-supplemented group): 10% chamchwi (ethanol-treated, 10% chamchwi powder-suplemented group). Three groups of rats were suplemented with three experimental diets for 4 weeks and orally administrated 10% ethanol (v/v) daily via drinking water in the last experimental week. Contents of TBARS (thiobarbituric acid reactive substance). glutathione in liver and kidney and serum albumin were determined. The activities of superoxide dismutase (SOD), catalase and glutathione peroxidase (GSH-px) in liver and kidney were also analyzed. Relative weight of liver and spleen to body in chamchwi groups was lower than that in control group (p<0.05). The most remarkable result was that liver TBARS contents in chamchwi groups (5% chamchwi group, 46 $\mu\textrm{g}$ in MDA value; 10% chamchwi group, 35 $\mu\textrm{g}$) were significantly lower (p<0.05) than that in control group (66 $\mu\textrm{g}$). The supplement of chamchwi powder lowered the activity of manganese- superoxide dismutase (Mn-SOD), catalase in liver and GSH-px in kidney. The levels of glutathione in liver and kidney and serum albumin were not significantly different in all experimental groups (p<0.05). These results indicate that powdered herb of Aster scaber decreases lipid peroxidation and acitvity of Mn-SOD increased by alcohol-induced oxidative stress in liver of rats.
