청폐사간탕(淸肺瀉肝湯)이 $CCl_4$로 유발(誘發)된 휜쥐의 간손상(肝損傷)에 마치는 영향(影響) (The Effects of Chungpyesagan-tang on the Recovery of Liver Function in Rat Injured by $CCl_4$)

  • 1. Objectives The present study was carried out to investigate the effects of Chungpyesagan-tang on the $CCI_4-induced$ Liver Damage in Rats. 2. Methods Sprague-Dawley rats were devided into 5 experimental groups : Normal, $NS+CCI_4(Solid$ extract of $CCI_4$ injection group after Normal Saline feed), $CP+CCI_4(Solid$ extract of $CCI_4$ injection group after Chungpyesagantang feed), $CCI_4+NS(Nomal$ Saline feed group after $CCI_4$ injection), $CCI_4+CP(Solid$ extract of Chungpyesagantang feed group after $CCI_4$ injection). Biochemical assays for serum enzyme activities such as AST, ALT, ALP, BUN, Creatinine, Uric Acid, Total Protein, Albumin, Total Cholesterol, Triglyceride, Glucose, and mRNA Revelation of Cytochrome p450 and activities such as LPO(Lipid Peroxidation), GSH(Glutathione), GST(Glutathione-S- Transferase), Glutathione Reductase, Glutathione Peroxidase, SOD(Superoxide Dismutase), Catalase, Hydroxyproline, and ${\beta}-Glucuronidase$ were performed. 3. Results 1) $CP+CCI_4$ showed significantly lower relation of Cytochrome p450 than $NS+CCI_4$. 2) As to LPO Hydroxyproline, $CCI_4+CP$ showed significantly lower activity than $CCI_4+NS$. 3) As to GSH GST Glutathione Peroxidase Catalase, $CP+CCI_4$ showed higher activity than $NS+CCI_4$, $CCI_4+CP$ showed significantly higher activity than $CCI_4+NS$. 4) As to Glutathione Reductase SOD, $CCI_4+CP$ showed significantly higher activity than $CCI_4+NS$. 5) As to ${\beta}-Glucuronidase$, $CP+CCI_4$ showed signficantly lower activity than $NS+CCI_4$. 4. Conclusions Chungpyesagantang has the recovering effects on the $CCI_4-induced$ Liver Damage.

살균제 carbendazim이 랫드 간 해독체계에 미치는 영향 (The Effect of Fungicide Carbendazim on Hepatic detoxication systems of rat)

  • Carbendazim의 랫드 간 해독체계에 대한 영향을 검색하기 위하여 간장독성의 지표 효소인 혈장 ALT(alanine aminotransferase) 및 AST(aspartate aminotransferase) 활성, 간에 대한 독성, 해독 및 대사에 대한 영향을 구명하기 위하여 간 GSH(glutathione), GST(glutathione-S-transferase), cytochrome P450 및 cytochrome P450 reductase 활성을 375, 750, 1,500 mg/kg 약량에서 측정한 결과 혈장 ALT 및 AST 활성이 120분 후에 약간의 증가가 있었으나 특이한 독성증상은 관찰되지 않았다. GSH는 고농도와 중농도 120분에서 20%의 함량증가가 있었고, GST는 120분까지 $36{\sim}50%$ 정도의 활성저해가 있었으나 240분에는 활성이 회복되었다. Cytochrome P450함량은 60분까지 $25{\sim}50%$까지 함량이 저하되었으나 120분에 70%이상 회복되었고, 240분 저농도에서는 거의 회복되었으며, cytochrome P450 reductase도 120분까지 $25{\sim}50%$ 활성저해가 있었으나, 240분에는 무처리 군의 활성과 유사하게 회복되어 대사관련 효소에 대한 손상은 크지 않을 것으로 판단되었다. 이상의 결과에서 benzimidazole 계 살균제인 carbendazim은 생체내 해독 및 대사관련 체계에 영향이 적은 농약으로 급성적인 중독은 일으키지 않을 것으로 판단되었다.

양파(Allium cepa) 추출물의 간보호 및 항산화 효과 (The Hepatotprotective and Antioxidative Effects of Onion (Allium cepa) Extracts in Rat Hepatocyte Primary Culture)

  • 본 연구의 목적은 양파추출물의 간보호 및 항산화 효과를 조사하기 위함이다. 간 손상을 유발시키는 t-BHP (1.5 mM) 존재하에 간세포를 0, 0.01, 0.05, 0.1 및 0.3 mg/ml의 다양한 농도의 양파추출물로 1시간 동안 일차배양하였다. t-BHP는 GOT와 LDH 활성 및 TBARS 농도를 증가시켰으며 MTT값은 감소시켰다. 0.05 mg/ml 농도의 양파추출물 첨가는 t-BHP에 의해 증가된 GOT 및 LDH 활성을 감소시켰으며 0.1 mg/ml 농도의 양파추출물은 t-BHP에 의해 감소된 MTT 값을 증가시켰다. 또한 0.01 mg/ml 농도의 양파추출물 첨가는 t-BHP에 의해 증가된 TBARS 농도를 감소시켜 양파추출물이 t-BHP에 의해 유발된 간손상과 지질과산화를 억제시켰다. t-BHP 처리는 간세포의 catalase, GSH-Px 및 GSH-Rd 활성을 현저히 감소시켰다. 그러나 0.1 mg/ml 농도의 양파추출물 첨가는 t-BHP에 의해 감소된 catalase GSH-Px 및 GSH-Rd 활성을 증가시켰으며 특히 catalase 활성은 t-BHP 무첨가군 수준까지 증가시켰다. 또한 hydroxyl radical을 생성하는 Fenton 시약의 존재하에 plasmid DNA를 양파추출물과 함께 배양한 결과 양파추출물은 농도 의존적으로 hydroxyl radical에 의해 유도된 single-strand 절단을 억제하였다. 이상과 같이 간세포 일차배양에서 양파추출물은 t-BHP에 의해 유발된 간독성, 간세포 생존율 감소, 지질과산화를 농도 의존적으로 억제시켰고 또한 t-BHP에 의해 억제된 GSH-Px, GSH-Rd 및 catalase의 활성을 증가시켰다. 이와 같이 양파추출물의 간보호 및 항산화 효과는 항산화 효소 특히 catalase의 활성 증가와 hydroxyl radical에 의해 유도된 산화억제 및 이에 따른 지질과산화 억제에 기인하는 것으로 사료된다.

Active-Site Mutants of Human Glutathione S-Transferase P1-1: Effects of the Mutations on Substrate Specificity and Inhibition Characteristics

  • In order to gain further insight on the relationship between structure and function of glutathione S-transferase (GST), the six active-site mutants, R13T, K44T, Q51A, Q64A, S65A, and D98A, of human GST P1-1 were expressed in Escherichia coli and purified to electrophoretic homogeneity by affinity chromatography on immobilized GSH. The active-site mutants showed marked differences in substrate specificity. The substitution of Gln51 with threonine resulted in a drastic decrease in the specific activities to <10% of the wild-type value. The substitution of Arg13 with threonine resulted in more decreased specific activity toward cumene hydroperoxide and in the $I_{50}$ values of S-(2,4-dinitrophenyl) glutathione and benanstatin A. These results suggest that the substitution of Arg13 with threonine changes the conformation of the active site to increase the affinity for the product or electrophilic substrate. Lys44 seems to be in the vicinity of the H-site of hGST P1-1 or may contribute to some extents to the electrophile binding.

천궁 물추출물이 간암예방효소계에 미치는 영향 (Effect of Cnidii Rhizoma Water Extract on Chemopreventive Enzymes for Hepatocarcinoma)

  • Cnidii Rhizoma water extract (CRW) was tested for liver cancer chemopreventive potential by measuring the inhibition of phase I enzyme and benzo[a]pyrene-DNA adduct formation and induction of phase II detoxification enzymes. There was 17.0% inhibition in the activity of cytochrome P450 1A1 enzyme with the treatment of 150 mg/ml CRW. At concentration of 30 mg/ml CRW, the binding of $[^3H]B[a]P$ metablites to DNA of NCTC-clone 1469 cell was inhibited by 33.3%. CRW was potent inducer of quinone reductase (QR) and glutathione S-transferase (GST) activities in cultured murine hepatoma Hepalc1c7 cells. However, hepatic glutathione (GSH) level was not influenced by CRW. These findings suggest that CRW has chemopreventive potential of liver cancer by inhibiting cytochrome P450 1A1 activity and benzo[a]pyrene-DNA adduct formation and inducing QR and GST activities.

Thermal Effects on Bioaccumulation and Antioxidant Enzyme Response in the Liver and Gills of Tilapia Oreochromis niloticus Exposed to Arsenic

  • The effects of waterborne arsenic (As) exposure on bioaccumulation and antioxidant defenses were examined in the liver and gills of tilapia, Oreochromis niloticus, under thermal stress. Tilapia were exposed to different As concentrations (0, 200, and $400{\mu}g\;L^{-1}$) at three water temperatures (20, 25, and $30^{\circ}C$) for 20 days. After As exposure, higher levels of As accumulation were observed in the gills compared with the liver in elevated water temperatures. In terms of the antioxidant response, glutathione (GSH) levels and the activities of glutathione reductase (GR), glutathione peroxidase (GPx), and glutathione S-transferase (GST) significantly decreased in the liver and gills of tilapia exposed to As for 20 days, regardless of the As concentration (200 and $400{\mu}g\;L^{-1}$) or water temperature. These enzyme activities exhibited greater fluctuations in the liver and gills of tilapia after As exposure in water warmer than $20^{\circ}C$. The present findings suggest that the simultaneous stress of temperature change and As exposure can accelerate As accumulation and alter the antioxidant enzymes activities of tilapia.

Cancer Chemopreventive Potential of Scenedesmus spp. Cultured in Medium Containing Bioreacted Swine Urine

  • Scenedesmus spp. were cultured for 51 days in newly developed medium, KEP I (Kim and Ecopeace: initials of corresponding author and environmental company) made with Bacterio-Mineral-Water (3%, v/v) that had been bio-reacted with swine urine medium to 10% (v/v) Bold's Basal medium, and investigated for cancer chemopreventive potential by measuring the induction of quinone reductase (QR), glutathione S-transferase (GST), and reduced glutathione (GSH), and inhibition of cytochrome P450 (CYP) 1A1 activity. The activitives of QR and GST of Scenedesmus spp. cultured in KEP I medium were increased by 3.0-fold and 1.5-fold, respectively. However, Scenedesmus spp. cultured in control medium (CT) increased the activitives of QR and GST by 1.8-fold and 1.3-fold, respectively. Scenedesmus spp. in KEP I medium strongly inhibited CYP 1Al activity. These results show that Scenedesmus spp. in KEP I medium has cancer chemopreventive potential and may be a candidate for further development as a chemopreventive agent.

Effect of Taurine Supplementation on Hepatic Lipid Peroxide Metabolism in Streptozotocin-induced Diabetic Rats

  • The purpose of this study was to examine the effect of taurine supplementation time on the activity of the enzymes metabolizing lipid peroxide in the liver of streptozotocin(STZ)-induced diabetic rats, Sprague-Dawley male rats were fed the purified diet for 7 weeks. They were supplemented with or without 1% taurine in drinking water before or after STZ injection or during all experimental procedure. In comparison to diabetic group without taurine, glucose-6-phosphatase(G6Pase) activity was decreased in diabetic group supplemented with taurine before STZ injection, and it was increased in diabetic group supplemented with taurine after STZ injection, but the difference was not significant. Glutathione S-transferase(GST) activity was significantly increased by the injection of STZ. However, the GST activities of diabetic groups exposed to taurine after STZ injection or during all experimental procedure were significantly decreased. Glutathione peroxidase(GPx) activities was significantly decreased by STZ injection. However, only in diabetic group supplemented with taurine before STZ injection, GPx activities was not decreased by the STZ injection. These results suggest that taurine supplementation may change the activities of GSH-related enzymes metabolizing lipid peroxide in the liver of streptozotocin(STZ)-induced diabetic rats and that may be helpful for the prevention of diabetic complication.

An evaluation of heat on protein oxidation of soy protein isolate or soy protein isolate mixed with soybean oil in vitro and its consequences on redox status of broilers at early age

  • Objective: The objective of this study was to evaluate effects of heat treatment and soybean oil inclusion on protein oxidation of soy protein isolate (SPI) and of oxidized protein on redox status of broilers at an early age. Methods: SPI mixed with soybean oil (SPIO) heated at $100^{\circ}C$ for 8 h was used to evaluate protein oxidation of SPI. A total of two hundred and sixteen 1-day-old Arbor Acres chicks were divided into 3 groups with 6 replicates of 12 birds, receiving basal diet (CON), heat-oxidized SPI diet (HSPI) or mixture of SPI and 2% soybean oil diet (HSPIO) for 21 d, respectively. Results: Increased protein carbonyl, decreased protein sulfhydryl of SPI were observed as heating time increased in all treatments (p<0.05). Addition of 2% soybean oil increased protein carbonyl of SPI at 8 h heating (p<0.05). Dietary HSPI and HSPIO decreased the average daily gain of broilers as compared with the CON (p<0.05). Broilers fed HSPI and HSPIO exhibited decreased glutathione (GSH) in serum, catalase activity and total sulfhydryl in liver and increased malondialdehyde (MDA) and protein carbonyl in serum, advanced oxidation protein products (AOPPs) in liver and protein carbonyl in jejunal mucosa as compared with that of the CON (p<0.05). Additionally, broilers receiving HSPIO showed decreased glutathione peroxidase activity (GSH-Px) in serum, GSH and hydroxyl radical scavenging capacity in liver, GSH-Px activity in duodenal mucosa, GSH-Px activity and superoxide anion radical scavenging capacity in jejunal mucosa and increased AOPPs in serum, MDA and protein carbonyl in liver, MDA and AOPPs in jejunal mucosa (p<0.05). Conclusion: Protein oxidation of SPI can be induced by heat and soybean oil and oxidized protein resulted in redox imbalance in broilers at an early age.

Pu-erh Tea Powder Preventive Effects on Cisplatin-Induced Liver Oxidative Damage in Wistar Rats

  • Background: Chemotherapy is one of the major means for control of malignancies, with cisplatin (CDDP) as one of the main agents, widely used for the treatment of various malignant solid tumors. However, prevention of hepatotoxicity from cisplatin is one of the urgent issues in cancer chemotherapy. In this study, we aimed to investigate the effects of pu-erh tea on hepatotoxicity through body weight and tissue antioxidant parameters like, liver coefficient, serum alanine aminotransferase (ALT), serum aspartate aminotransferase (AST), superoxide dismutase (SOD), glutathione peroxidase (GSH-PX), malondialdehyde(MDA) and glutathione (GSH) levels, and light microscopic evaluation by histological findings. Materials and Methods: The rats were randomly divided into five groups: Control (n=10), cisplatin (3 mg/kg p.i., n=10), cisplatin+pu-erh (0.32 g/kg/day i.g., n=10), cisplatin+pu-erh (0.8 g/kg/day i.g., n=10) and cisplatin+pu-erh (1.6 g/kg/day i.g., n=10). Pu-erh tea powder was administrated for 31 consecutive days. The rats were sacrificed at the end on the second day after a single dose of cisplatin treatment for measuring indices. Results: Pu-erh tea powder exhibited a protective effect by decreasing MDA and GSH and increasing the SOD and GSH-PX levels and GSH-PX/MDA ratio in camparison with the control group. Besides, pu-erh tea was also able to alleviate the pathological damage to some extent. Conclusion: Pu-erh tea powder is protective against cisplatin-induced liver oxidative damages, especially at the medium dosage (0.8 g/kg/d).
