Effects of High-Molecular-Weight Glutenin Subunits and Agronomic Traits on Bread Wheat Quality Parameters (밀의 고분자 글루테닌 조성과 농업 형질이 품질 특성에 미치는 영향)

  • Improving flour quality is one of the major targets of wheat breeding programs. This study determined the optimum high-molecular-weight glutenin subunits (HMW-GS) to improve flour quality, and analyzed the correlation between agronomic and quality traits in Korea. A total of 180 wheat varieties, including 55 Korean and 125 foreign cultivars, carrying various Glu-1 alleles, were evaluated for their quality and agronomic traits. Results indicated that Glu-A1b, Glu-B1b, and Glu-D1f were the most prevailing alleles for each Glu-1 locus for Korean wheat cultivars. Korean wheat cultivars recorded shorter days to heading (DTH) and longer days to maturity (DTM) compared to foreign cultivars. In addition, an interaction effect was found between Glu-A1 and Glu-B1 alleles on several quality parameters. The combination of Glu-A1c and Glu-B1i showed a higher protein content, dry gluten content, and higher sodium dodecyl sulfate (SDS) sedimentation value than other Glu-A1×Glu-B1 combinations. Cultivars carrying Glu-A1a or Glu-A1b, Glu-B1i or Glu-B1al, and Glu-D1d for each Glu-1 locus exhibited a longer mixing time and stronger mixing tolerance. The DTM positively correlated with the protein content, gluten index and SDS sedimentation value. However, a negative correlation was observed between DTH and quality traits. Owing to the above results, this study suggests that an increase in the frequency of Glu-B1i or Glu-B1al, Glu-D1d coupled with a short DTH and long DTM could significantly improve wheat quality properties.

  • The hairy root was induced form potato tuber disc by infection of a. rhizogenes. The detection of the agropine and mannopine by paper electrophoresis confirmed that induced hairy root was transformed by A. rhizogenes. The activity of peroxidase was the highest at 5 weeks and isozyme pattern of peroxidase revealed 3 cathodic bands and 2 anodic bands and new C4 band(pI 4.6) was observed at 7 weeks after cultivation in hairy root was isoelectric focusing. To study the effect of Ca2+ on cell wall formation in hairy root, channel blocker of Ca2+ was treated. The activity of $\beta$-glucan synthetase II(GS II) related to cell wall synthesis was inhibited by about 50% in diltiazem and flunarizine treatment than that of control, but stimulated in CaCl2 treatment. Therefore these results showed that Ca2+ might be an effective factor in the cell wall formation. The activity of GS II by NaF treatment was increased by about 30%. This result suggested that the activity GS II is changed through phosphorylation process.

Genomic Selection for Adjacent Genetic Markers of Yorkshire Pigs Using Regularized Regression Approaches

  • This study considers a problem of genomic selection (GS) for adjacent genetic markers of Yorkshire pigs which are typically correlated. The GS has been widely used to efficiently estimate target variables such as molecular breeding values using markers across the entire genome. Recently, GS has been applied to animals as well as plants, especially to pigs. For efficient selection of variables with specific traits in pig breeding, it is required that any such variable selection retains some properties: i) it produces a simple model by identifying insignificant variables; ii) it improves the accuracy of the prediction of future data; and iii) it is feasible to handle high-dimensional data in which the number of variables is larger than the number of observations. In this paper, we applied several variable selection methods including least absolute shrinkage and selection operator (LASSO), fused LASSO and elastic net to data with 47K single nucleotide polymorphisms and litter size for 519 observed sows. Based on experiments, we observed that the fused LASSO outperforms other approaches.

  • In common structural systems, there are some limitations to provide adequate lateral stiffness, high ductility, and architectural openings simultaneously. Consequently, the concept of T-Resisting Frame (TRF) has been introduced to improve the performance of structures. In this study, Configuration of TRF is a Vertical I-shaped Plate Girder (V.P.G) which is placed in the middle of the span and connected to side columns by two Horizontal Plate Girders (H.P.Gs) at each story level. System performance is improved by utilizing rigid connections in link beams (H.P.Gs). Plastic deformation leads to tension field action in H.P.Gs and causes energy dissipation in TRF; therefore, V.P.G. High plastic deformation in web of TRF's members affects the ductility of system. Moreover, in order to prevent shear buckling in web of TRF's members and improve overall performance of the system, appropriate criteria for placement of web stiffeners are presented in this study. In addition, an experimental study is conducted by applying cyclic loading and using finite element models. As a result, hysteresis curves indicate adequate lateral stiffness, stable hysteretic behavior, and high ductility factor of 6.73.

Modeling High Power Semiconductor Device Using Backpropagation Neural Network (역전파 신경망을 이용한 고전력 반도체 소자 모델링)

  • Using a backpropagation neural network (BPNN), a high power semiconductor device was empirically modeled. The device modeled is a n-LDMOSFET and its electrical characteristics were measured with a HP4156A and a Tektronix curve tracer 370A. The drain-source current $(I_{DS})$ was measured over the drain-source voltage $(V_{DS})$ ranging between 1 V to 200 V at each gate-source voltage $(V_{GS}).$ For each $V_{GS},$ the BPNN was trained with 100 training data, and the trained model was tested with another 100 test data not pertaining to the training data. The prediction accuracy of each $V_{GS}$ model was optimized as a function of training factors, including training tolerance, number of hidden neurons, initial weight distribution, and two gradients of activation functions. Predictions from optimized models were highly consistent with actual measurements.

A Study on Oxygen Reduction Reaction of PtM Electrocatalysts Synthesized on Graphene for Proton Exchange Membrane Fuel Cell (고분자전해질연료전지를 위한 그래핀 기반 PtM 촉매들의 산소환원반응성 연구)

  • In this research, we investigate electrical performance and electrochemical properties of graphene supported Pt (Pt/G) and PtM (M = Ni and Y) alloy catalysts (PtM/Gs) that are synthesized by modified polyol method. With the PtM/Gs that are adopted for oxygen reduction reaction (ORR) as cathode of proton exchange membrane fuel cells (PEMFCs), their catalytic activity and ORR performance and electrical performance are estimated and compared with one another. Their particle size, particle distribution and electrochemically active surface (EAS) area are measured by TEM and cyclic voltammetry (CV), respectively. On the other hand, regarding ORR activity and electrical performance of the catalysts, (i) linear sweeping voltammetry by rotating disk electrode and rotating ring-disk electrode and (ii) PEMFC single cell tests are used. The TEM and CV measurements demonstrate particle size and EAS of PtM/Gs are compatible with those of Pt/G. In case of PtNi/G, its half-wave potential, kinetic current density, transferred electron number per oxygen molecule and $H_2O_2$ production % are excellent. Based on data obtained by half-cell test, when PEMFC singlecell tests are carried out, current density measured at 0.6V and maximum power density of the PEMFC single cell employing PtNi/G are better than those employing Pt/G. Conclusively, PtNi/Gs synthesized by modified polyol shows better ORR catalytic activity and PEMFC performance than other catalysts.

Performance and Metabolism of Calves Fed Starter Feed Containing Sugarcane Molasses or Glucose Syrup as a Replacement for Corn

  • The aim of this study was to evaluate the effect of replacing corn grain for sugar cane molasses (MO) or glucose syrup (GS) in the starter concentrate on performance and metabolism of dairy calves. Thirty-six individually housed Holstein male calves were blocked according to weight and date of birth and assigned to one of the starter feed treatments, during an 8 week study: i) starter containing 65% corn with no MO or GS (0MO); ii) starter containing 60% corn and 5% MO (5MO); iii) starter containing 55% corn and 10% MO (10MO); and iv) starter containing 60% corn and 5% GS (5GS). Animals received 4 L of milk replacer daily (20 crude protein, 16 ether extract, 12.5% solids), divided in two meals (0700 and 1700 h). Starter and water were provided ad libitum. Starter intake and fecal score were monitored daily until animals were eight weeks old. Body weight and measurements (withers height, hip width and heart girth) were measured weekly before the morning feeding. From the second week of age, blood samples were collected weekly, 2 h after the morning feeding, for glucose, ${\beta}$-hydroxybutyrate and lactate determination. Ruminal fluid was collected at 4, 6, and 8 weeks of age using an oro-ruminal probe and a suction pump for determination of pH and short-chain fatty acids (SCFA). At the end of the eighth week, animals were harvested to evaluate development of the proximal digestive tract. The composition of the starter did not affect (p>0.05) concentrate intake, weight gain, fecal score, blood parameters, and rumen development. However, treatment 5MO showed higher (p<0.05) total concentration of SCFAs, acetate and propionate than 0MO, and these treatments did not differ from 10MO and 5GS (p>0.05). Thus, it can be concluded that the replacement of corn by 5% or 10% sugar cane molasses or 5% GS on starter concentrate did not impact performance, however it has some positive effects on rumen fermentation which may be beneficial for calves with a developing rumen.

  • Developing effective tools for predicting absorption, distribution, metabolism, excretion properties and toxicity (ADME/T) of new chemical entities in the early stage of drug design is one of the most important tasks in drug discovery and development today. As one of these attempts, support vector machines (SVM) has recently been exploited for the prediction of ADME/T related properties. However, two problems in SVM modeling, i.e. feature selection and parameters setting, are still far from solved. The two problems have been shown to be crucial to the efficiency and accuracy of SVM classification. In particular, the feature selection and optimal SVM parameters setting influence each other, which indicates that they should be dealt with simultaneously. In this account, we present an integrated practical solution, in which genetic-based algorithm (GA) is used for feature selection and grid search (GS) method for parameters optimization. hERG ion-channel inhibitor classification models of ADME/T related properties has been built for assessing and testing the proposed GA-GS-SVM. We generated 6 different models that are 3 different single models and 3 different ensemble models using training set - 1891 compounds and validated with external test set - 175 compounds. We compared single model with ensemble model to solve data imbalance problems. It was able to improve accuracy of prediction to use ensemble model.

A Precise Localization Method for a High Speed Mobile Robot using iGS and Dual Compass (iGS와 듀얼 컴퍼스를 이용한 고속 이동로봇의 정밀 위치 인식기법)

  • This paper proposes a precise localization algorithm for a quickly moving mobile robot. In order to localize a mobile robot with active beacon sensors, a relatively long time is needed, since the distance to the beacon is measured using the flight time of the ultrasonic signal. The measurement time does not cause a high error rate when the mobile robot moves slowly. However, with an increase of the mobile robot's speed, the localization error becomes too high to use for accurate mobile robot navigation. Therefore, in this research into high speed mobile robot operations, instead of using two active beacons for localization an active beacon and dual compass are utilized to localize the mobile robot. This new approach resolves the high localization error caused by the speed of the mobile robot. The performance of the precise localization algorithm was verified by comparing it to the conventional method through real-world experiments.
