Prognostic Value of Prepro-Gastrin Releasing Peptide in Lung Cancer Patients; NCI-Prospective Study

  • Background: Prior series investigated the expression of prepro-gastrin releasing peptide (prepro-GRP) in the peripheral blood of lung cancer patients. Our aim was to assess any prepro-GRP role as a prognostic factor for small cell lung cancer (SCLC) and NSCLC and correlations with clinical presentation and treatment outcome. Methods: A prospective study was conducted during the time period from the beginning of January 2012 till the end of January 2014. Prepro-GRP expression was analysed using a nested RT-PCR assay in peripheral blood of 62 untreated lung cancer patients attending the National Cancer Institute (NCI), Cairo University, and 30 age and sex matched healthy volunteers. Results: Among the 62 lung cancer cases, there were 24 (38.7%) SCLC, and 38 (61.3%) NSCLC (10 squamous cell carcinomas, 12 adenocarcinomas, 11 large cell carcinomas, 4 undifferentiated carcinomas, and 1 adenosquamous carcinoma). Twenty six patients (41.9%) were prepro-GRP positive. Prepro-GRP expression was higher (58.3%) among SCLC patients compared to NSCLC (squamous cell carcinoma (15.4%), large cell carcinoma (36.4%), and adenocarcinoma (25%)). Mean OS among prepro-GRP negative cases was longer than that among preprogastrin positive cases (17.6 vs 14.9 months). The mean PFS durations among preprogastrin negative versus positive cases were 7.7 vs 4.6 months (p= 0.041). No difference in response to chemotherapy was identified between the groups (p=0.983). Conclusion: Prepro-GRP is suggested to be a useful prognostic marker for lung cancer patients, especially with the fast- growing, bad prognostic SCLC type. More studies should aim at detailed understanding of the mechanisms of prepro-GRP action and its use in monitoring the response to treatment in a larger cohort.

Spatio-Temporal Expression Pattern of Grp 78, a Putative Hoxc8 Downstream Target Gene, During Murine Embryogenesis

  • Grp78, discovered as one of the putative target genes of Hoxc8, is a highly conserved stress protein and functions as a molecular chaperone in the endoplasmic reticulum (ER). In order to see the stage-specific expression pattern of Grp78 during development, mouse embryos from day 7.5 to 17.5 p.c. were isolated, and RT-PCR as well as in situ hybridization was performed. When RT-PCR was performed using Grp78 specific primers, periodic expression pattern was detected. And also a region-specific expression pattern was detected with a strong expression in the trunk part of day 11.5 p.c. embryo, like that of Hoxc8. When in situ hybridization was performed, Grp78 was revealed to be expressed in the endoderm, somite, neuroepithelium cells of neural tube in early embryos. In the case of late embryos, Grp78 expression was detected in the liver, segmental bronchus within cranial lobe of lung, ossification center within the cartilage primordium of rib and vertebra, submandibular gland, as well as metanephros. These expression patterns are very much similar to those of Hoxc8. Since Hoxc8 has been reported to regulate apoptosis during organogenesis, it might be possible that the apoptotic function could have been conveyed through the expression of Grp78, implying that the Grp78 is one of the Hoxc8 downstream target genes.

  • K562 백혈구암 세포의 phorbol 12-myristate 13-acetate(PMA)에 의한 대핵세포로의 분화과정에서 heat shock proteins(HSPs)와 glucose-regulated proteins(GRPs)의 발현을 조사하였다. PMA에 의한 K562 세포의 분화 특징은 세포성장의 억제, 형태학적 변화, gpllIa의 발현 증가, c-myc 발현의 감소 등으로 나타난다. PMA에 의한 대핵세포 분화과정에서, HSP90A, HSP90B 그리고 HSP28 mRNA와 단백질 합성은 현저히 감소하는 반면, GRP78/BiP와 GRP94의 mRNA 합성은 증가하였다. 한편 HSP7OA와 HSP7OB의 mRNA 합성은 감소하였지만, HSP70 단백질의 합성은 변함이 없었다. 이러한 결과는 HSPs와 GRPs가 K562 세포의 증식 또는 대핵세포 분화 과정에서 특이한 역할을 할 것임을 시사하고 있다.

Proteome Analysis of Vernalization-Treated Arabidopsis thaliana by Matrix-Assisted Laser Desorption/Ionization Time-of-Flight Mass Spectrometry

  • In order to gain insight into the molecular changes at the protein level in plants exposed to low temperature for a long period of time (vernalization), proteome analyses of vernalization-treated Arabidopsis thaliana have been carried out by two-dimensional gel electrophoresis followed by matrix-assisted laser desorption/ ionization time-of-flight mass spectrometry. Fourteen proteins including ATP binding/GTP binding/translation elongation factor and glycine-rich RNA-binding protein 7 (GRP7) showed differential expression in vernalization-treated Arabidopsis thaliana. GRP7 showed the most dramatic increase in expression suggesting its involvement in response to vernalization treatment.

바우미 상황버섯 추출물의 항염증 효과에 관한 연구 (Characterization of Anti-inflammation Effect of Aqueous Extracts from Phellinus baumii)

  • 본 연구에서는 바우미 상황버섯의 열수 추출물의 항염증 기능에 관한 연구를 수행하였다. 대식세포주인 RAW264.7을 lipopolyssacharide (LPS)로 활성화 시킨 후, 바우미 상황버섯에서 복수의 조건에서 추출한 추출물을 처리하여 면역 및 염증 발생 경로의 활성도를 측정하여 바우미 상황버섯의 면역증강기능 및 항염증기능을 검증하였다. RAW264.7을 LPS로 처리하였을 때 전형적인 소포체 스트레스를 수반한 염증 반응이 시현되었다. 전형적인 마커인 Grp78, Grp94 및 CHOP의 과발현을 시현하였고, 인터루킨 6 (IL-6)과 염증발현의 표시인자인 nitric oxide(NO) 발현을 증진한 것으로 나타났다. 위와 같이 처리된 세포에 바우미 상황버섯의 추출물을 처리하였을 때, 전반적으로 소포체 스트레스는 경감된 것으로 나타났으며, 특히 염증유발의 신호인자인 NO의 발생량과 이를 생성하는 효소인 iNOS의 발현량이 현격하게 감소한 것으로 나타났다. 이를 종합적으로 분석하면 바우미 상황버섯의 추출물은 유도된 소포체스트레스를 경감하는 효능을 보임과 동시에 항염증의 기능을 보이는 것으로 판단된다.

The Anti-Inflammatory and Anti-Oxidant Activity of Ethanol Extract from Red Rose Petals

  • Red rose petals are usually disposed but they are an abundant source of phenolics and traditionally used as food supplement and as herbal medicine. Of the Various phenolics, they are known to have anticancer, antioxidant, and anti-inflammatory properties. In this study, we investigated the anti-inflammatory effects of red rose ethanolic extracts (GRP) on lipopolysaccharide (LPS)-activated RAW 264.7 cells. The results demonstrated that pretreatment of GRP (500㎍/mL) significantly reduced NO production by suppressing iNOS protein expression in LPS-stimulated cells. Anti-inflammatory effects by red rose petals were observed in the following. Red rose petals inhibited the translocation of NF-κB from the cytosol to the nucleus via the suppression of IκB-α phosphorylation and also inhibited LPS-stimulated NF-κB transcriptional activity. These findings suggest that red rose petals exert anti-inflammatory actions and help to elucidate the mechanisms underlying the potential therapeutic values of red rose petals. Therefore, red rose petals could be regarded as a potential source of natural anti-inflammatory agents.

붉은 장미꽃잎 에탄올 추출물의 활성 분획물 분석 및 항염증 활성 효과 (The Effects of Anti-Inflammatory Activities and Active Fractions Analysis of Ethanol Extract from Red Rose Petals)

  • 붉은 장미꽃잎 에탄올 추출물은 효과가 풍부한 페놀 성분의 공급원이며 전통적으로 식품 보충제 및 약초로 사용되고 있다. 또한 다양한 식물페놀 성분은 항암, 항산화 및 항염증 특성을 갖는 것으로 알려져 있다. 따라서 본 연구에서는 붉은 장미꽃잎 에탄올추출물(GRP) 유래 활성 분획물 분석 및 lipopolysaccharide(LPS)로 유도된 마우스 대식세포주인 RAW264.7 세포와 마우스 폐포 대식세포주인 MH-S 세포에서 붉은 장미꽃잎 에탄올추출물의 항염증 활성 효과를 조사하였다. 그 결과 GRP(500 ㎍/mL)의 전처리 LPS 자극 세포에서 iNOS 단백질 및 염증성 싸이토카인 mRNA 발현을 크게 억제하였다. 빨간 장미꽃잎 에탄올 추출물에 의한 항균활성 효과는 다음과 같이 관찰되었다. 붉은 장미꽃잎은 IκB-α 인산화의 억제를 통해 시토졸에서 핵으로의 NF-κB의 전좌를 억제하고 또한 LPS로 자극된 NF-κB 전사 활성을 억제 하였다. 이러한 연구 결과는 빨간 장미꽃잎 에탄올 추출물이 항염증 작용을 발휘 하고 빨간 장미꽃잎의 잠재적인 치료 가치성 및 기본 메커니즘을 규명 할 수 있었다. 결론적으로 붉은 장미꽃잎 에탄올 추출물이 다양한 활성 성분 함유 및 천연 항염증제의 잠재적 공급원으로 활용 가능성이 있음을 시사 하였다.

Comparisons of Renoprotective Activities between White Ginseng Radix and Rootlet in Spontaneously Hypertensive Rats with Diabetes

  • The renoprotective activities of white ginseng radix and rootlet were compared in spontaneously hypertensive rat (SHR) with diabetes. During oral administration of white ginseng radix (Ginseng Radix Alba, GRA) and white ginseng rootlet (Ginseng Radix Palva, GRP) for four weeks, arterial blood pressure and blood glucose levels were determined at every 10 days. In both GRA- and GRP-treatment groups, arterial blood pressures started to go down after 10 days of administration and maintained throughout the study period. After four weeks administrations of GRA and GRP, diastolic blood pressures were significantly decreased with 17% and 9%, respectively. GRA treatment also decreased blood glucose levels after 10 days of administration when compared with diabetic SHR group. At the end of the experiment, serum creatinine (Scr) and blood urea nitrogen (BUN) were not significantly different between the groups, except 62% higher value of BUN in diabetic SHR group when compared with SHR group. In the diabetic SHR group, the excretion of urinary albumin was increased significantly when compared with SHR. The level of urinary albumin in GRA treated group was markedly reduced when compared with diabetic SHR group $(67.8{\pm}4.7\;vs.\;131.3{\pm}13.5\;mg/24\;h).$ To examine the effects of ginseng radices on an overt diabetic nephropathy, index of kidney hypertrophy and transforming growth $factor-{\beta}1\;(TGF-{\beta}1)$ protein levels were evaluated. The glomerular and tubular cells stained positive for $TGF-{\beta}1$ seemed to be more abundant in diabetic SHR than in those with SHR, and GRA treated rats showed somewhat less $TGF-{\beta}1$ protein in glomerular and tubular cells when compared with diabetic SHR. Our results suggest that GRA might be a useful antihypertensive and antidiabetic agent with renoprotective effect.

경량화 태양광 모듈의 내구성 보완에 관한 연구 (A Study on the Durability Complement of Lightweight Photovoltaic Module)

  • In this study, we fabricated light-weight solar module for various applications such as building integrated photovoltaics (BIPV), vehicles, trains, etc. Ethylene tetra fluoro ethylene (ETFE) film was applied as a material to replace the cover glass, which occupies more than 65% of the weight of the PV module. Glass fiber reinforced plastic (GRP) was applied to the ones with a low durability by replacing the cover glass to ETFE. Moreover, to achieve a high solar power conversion in this study, we applied a shingled design to weight reduced solar modules. The shingled module with GRP shows 183.7 W of solar-to-power conversion, and the output reduction rate after weight load test was 1.14%.

Combined Effects of Multiple Endoplasmic Reticulum Stresses on Cytokine Secretion in Macrophage

  • Cells show various stress signs when they are challenged with severe physiological problems. Majority of such cellular stresses are conveyed to endoplasmic reticulum (ER) and unfolded protein response (UPR) serves as typical defense mechanism against ER stress. This study investigated an interaction between ER stress agents using macropage cell line Raw 264.7. When activated by lipopolysaccharide (LPS), the cell lines showed typical indicators of ER stress. Along with molecular chaperones, the activation process leads to the production of additional inflammatory mediators. Following activation, the macrophage cell line was further treated with TUN and characterized in terms of chaperone expression and cytokine secretion. When treated with TUN, the activated macrophage cell leads to increased secretion of IL-6 although expression of ER stress markers, GRP94 and GRP78 increased. The secretion of cytokines continued until the addition of BFA which inhibits protein targeting from ER to Golgi. However, secretion of cytokines was ceased upon dual treatments with BFA and TG. This result strongly implies that cells may differently deal with various polypeptides depending on the urgency in cellular function under ER stress. Considering IL-6 is one of the most important signal molecules in macrophage, the molecule might be able to circumvent ER stress and UPR to reach its targeting site.
