Objective: Selenium-independent glutathione peroxidase (GPx5) is specifically expressed in the mammalian epididymis and plays an important role in protecting sperm from reactive oxygen species and lipid peroxidation damage. This study investigates GPx5 expression in the epididymis of Small Tail Han sheep. Methods: GPx5 expression was studied in three age groups: lamb (2 to 3 months), young (8 to 10 months), and adult (18 to 24 months). The epididymis of each age group divided into caput, corpus and cauda, respectively. Analysis the expression quantity of GPx5 in epididymis and testis by real-time fluorescent quantitative polymerase chain reaction and Western blot. Finally, GPx5 protein locating in the epididymis by immunohistochemical. Results: The results demonstrate that in the lamb group, the GPx5 mRNA, but not protein, can be detected. GPx5 mRNA and expressed protein were detected in both the young and adult groups. Moreover, both the mRNA and protein levels of GPx5 were significantly higher in the young group than in other two groups. When the different segments of epididymis were investigated, GPx5 mRNA was expressed in each segment of epididymis regardless of age. Additionally, the mRNA level in the caput was significantly higher than that in corpus and cauda within same age group. The GPx5 protein was in the epithelial cells' cytoplasm. However, GPx5 mRNA and protein were not detected in the testis. Conclusion: These results suggest that GPx5 is mainly expressed in the epididymis of Small Tail Han sheep, and that the expression level of GPx5 is associated with age. Additionally, GPx5 was primarily expressed in the epithelial cells of the caput. Taken together, these studies indicate that GPx5 is expressed in the epididymis in all age grades.
The effect of various stressors such as reductant ascorbic acid, signalling molecules (salicylic acid and methyl jasmonic acid), heavy metals $(NiCl_2,\;and\;MnSO_4)$ and NaCl on the glutathione peroxidase (GPX) activities and isoenzyme expression patterns were investigated in rice seedling roots. Total GPX activity increased according to the increase of ascorbic acid concentration. Prominent enhancement of GPX1 isozyme due to ascorbic acid contributed to the increase of total GPX activity. GPX showed different reactivity toward salicylic acid and methyl jasmonic acid. GPX activity increased at 0.1 mM salicylic acid, and then decreased thereafter. However, GPX increased gradually in a methyl jasmonic acid concentration-dependent manner, and 3 fold increase of GPX activity was found at 1 mM methyl jasmonic acid. Moreover, GPX1 isozyme increased according to the increase of salicylic acid, while GPX1 isozyme decreased according to the increase of methyl jasmonic acid. When metal ions were treated, GPX activity increased considerably according to the increase of $NiCl_2$ concentration, however, GPX activity increased about 2 fold at 0.5 mM $CuSO_4$ and then decreased. Enhancement of GPX1 isozyme contributed to the increase of total GPX activities in $NiCl_2-treated$ and $MnSO_4-treated$ rice seedlings. Total GPX activity increased 1.7 fold in response to 300 mM NaCl. Especially GPX2 isozyme showed gradual increase according to the increase of NaCl concentration.
In the present study, we investigated and compared the cardiopulmonary and oxidative stress effects of dogs undergoing open and thoracoscopic lung lobectomy. Ten healthy dogs, 5-8 years old, weighing 9-12 kg were used. The animals were randomly assigned to one of two groups according to the type of surgical procedure; open (group 1, n=5) or thoracoscopic lung lobectomy (group 2, n=5). Cardiopulmonary parameters, superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) concentrations were measured. There were statistically significant changes in arterial blood gases values in both groups. Total anesthesia and surgical times were significantly shorter in thoracoscopic lobectomy group compared with open surgery group. Increases in plasma SOD and CAT levels, and decreases in GPx levels were observed in both groups after surgery. Significant difference in GPx levels was found when the groups were compared. The GPx level was significantly lower in the thoracoscopic lobectomy group compared with the open surgery group.
The cDNAs encoding glutathione peroxidase (GPx) were cloned and sequenced from the liver of three Chinese carps with different tolerance to hepatotoxic microcystins, phyto-planktivorous silver carp (Hypophthalmichthys molitrix) and bighead carp (Aristichthys nobilis), and herbivorous grass carp (Ctenopharyngodon idellus). Using genome walker method, a 750 bp 5'-flanking region of the silver carp GPx gene was obtained, and several potential regulatory elements were identified in the promoter region of the GPx gene. The silver carp GPx gene was widely expressed in all tissues examined. Despite phylogenetic analysis, assigning this newly described carp GPx to the group of mammalian GPx2, the carp GPx seems more similar to GPx1 from a physiological point of view. The constitutive expression pattern of the three carp liver GPx gene, shows a positive relationship with their tolerance to microcystins.
The treatment of radish cotyledons with a nitric oxide (NO)-releasing substance, sodium nitroprusside (SNP) resulted in an increased adventitious root development in a dose-dependent manner. However, this NO-mediated enhancement effect was reversed when either 0.5 mM EGTA (an extracellular $Ca^{2+}$ chelator) or 0.1 mM $LaCl_3$ (a calcium channel blocker) was applied with $50\;{\mu}M$ SNP. Our results also showed that guaiacol peroxidase (GPX) and syringaldazine peroxidase (SPX) activities, which are known to play a key role in rooting, were more largely increased during adventitious root induction in the cotyledons treated with SNP. However, the treatment of cotyledons with SNP plus $LaCl_3$ inhibited the SNP-induced increases in the activities of both GPX and SPX. Trifluoperazine (TFP), an antagonist of calmodulin (a specific calcium-binding protein), also delayed adventitious root formation and significantly reduced the root length and number of the SNP-treated cotyledons as well as the deactivation of GPX and SPX enzymes. In conclusion, our results suggest that calcium is involved in the NO response leading to induction of adventitious root through a regulation of GPX and SPX.
The present investigation was undertaken to examine the anticancer activity of the methanol extract from Houttuynia cordata on ICR mouse with induced abdominal cancer and L1210 cancer cells. When the methanol extract of Houttuynia cordata (10∼200 $\mu\textrm{g}$/$m\ell$) was administered orally to ICR mouse with abdominal cancer, 47.8% of the best life prolonging effect was obtained. In case of cytotoxicity study (inhibition of cell proliferation) of Houttuynia cordata extract against L1210 cells, $IC_{50}$/ was found to be 62.8 $\mu\textrm{g}$/$m\ell$. In contrast to such considerable toxicity against cancer cell line, the toxicity demonstrated by the identical extract against normal lymphocytes was very meagre as shown to be < 5% compared with 86.5% in case of L1210 cells at the same condition. To get an insight into the reaction mechanism undelying the anticancer activity, $O_2$ion quantity and antioxidant enzyme activities such as superoxide dismiutase (SOD) and glutathione peroxidase (GPx) of L1210 cells in the presence of Houttuynia cordata extract were measured. The increased values of SOD and GPx enzyme activities in addition to the augmented generation of $O_2$ ion in L1210 cells implied that the reactive oxygen species induding $O_2$ion which were presumably induced by Houttuynia cordata extract might have participated in the process of L1210 cells cytotoxicity.

Fumonisin B1 (FB1) structurally resembles sphingolipids and interferes with their metabolism leading to sphingolipid dysregulation. We questioned if FB1 could exacerbate liver or kidney toxicities in glutathione peroxidase 1 (Gpx1) and catalase (Cat) knockout mice. While higher serum levels of thiobarbituric acid reactive substances (TBARS) and sphinganine (Sa) were measured in Gpx1/Cat knockout mice (Gpx1/Cat KO) than wild type mice after 5 days of FB1 treatment, serum levels of alanine aminotransferase (ALT), sphingosine-1 phosphate (So-1-P), and sphinganine-1 phosphate (Sa-1-P) were found to be relatively low. Although Sa was highly elevated in Gpx1/Cat KO mice and wild mice, lower levels of So and Sa were found in both the kidney and liver tissues of Gpx/Cat KO mice than wild type mice after FB1 treatment. Paradoxically, FB1-induced cellular apoptosis and necrosis were hastened under oxidative stress in Gpx1/Cat KO mice.
As the pH of ground pork increased from 5.0 to 7.0, the corresponding development of 2-thiobarbituric acid reactive substance (TBARS) during storage at $4^{\circ}C$ decreased significantly (p<.001). At the 4th day of refrigerated storage, with the increase in pH of ground pork from 5.0 to 7.0, the release of free iron decreased significantly (p<.05) from 1.50 to .99 ppm. The decrease in free iron content of pH 7 pork well explains the decrease in TBARS absorbances. The fact that the addition of 2% ethylenediamine tetraacetic acid (EDTA) to pH 5 ground pork decreased the oxidative rancidity development (p<.001) strongly supported the above finding that the increased free iron content of pH 5ground pork catalyze the oxidation during storage. The activity of glutathione peroxidase (GPx) at the 4th day of refrigerated storage decreased significantly (p<.05) when the pH of ground pork decreased from 7.0 to 5.0. Both the lower free iron content and the higher activity of GPx were proved to be important factors in controlling the oxidative rancidity of high pH ground pork.
Four-d-old broiler chicks were randomly assigned to 3 groups with 9 replicates (8 birds/cage) under high ambient temperature; birds fed a basal diet (CON), a basal diet supplemented with 0.25% of probiotic complex (LPB, 1 × 106 Lactobacillus plantarum, 1 × 106 Bacillus subtilis, and 1 × 106 Saccharomyces cerevisiae) and 0.5% probiotic complex (HPB). Immediately after 28-d feeding trial, 6 birds having average body weight per group were sacrificed for evaluating the effects of probiotics on antioxidant parameters in the serum, intestine, and liver of birds. As results, serum biochemical parameters of nitrogen components including total protein, albumin, urea nitrogen, and glutathione were unaffected by dietary probiotics. In addition, serum superoxide dismutase (SOD), glutathione peroxidase (GPX), and glutathione S-transferase (GST) activities, and lipid peroxidation (MDA) were not changed by dietary probiotic supplement in birds. In the intestinal mucosa, SOD activity in the HPB group significantly (P<0.05) increased compared with that in the CON and the LPB groups. Lipid peroxidation in the HPB group significantly (P<0.05) decreased compared with that in the CON group. However, there was no statistical difference in GPX, and GST activities in the intestinal mucosa among treatment groups. In the liver, the activities of SOD, GPX, and GST, and the level of MDA were unaffected by probiotic supplement. In conclusion, 0.5% of probiotics significantly increased SOD activity and decreased lipid peroxidation in the intestinal mucosa, suggesting that probiotic complex could be potential to improve the small intestinal antioxidant capacity of bird under high ambient temperature conditions.
SD-994 was prepared from methanol extract of Artemisia argyi by stepwise purification of solvent partioning and silica gel chromatography. In the course of this purification, fractions obtained at each step were investigated for their cytotoxicities against L1210 cells. Fractions A~G prepared from chloroform fraction showed considerable cytotoxicities raging 40~90% against L1210 cells. Subfractions I~IX obtained from fraction A exhibited various cytotoxicities and subfraction I (SD-994) was found to be the most effective compound. $IC_{50}$ values of SD-994 were measured to be $0.5{\;}{\mu\textrm{g}}/ml and less than $0.05{\;}{\mu\textrm{g}}/ml against L1210 cells and normal lymphocytes, respectively: When SD-994 was added to L1210 cell as cytotoxic agent, significantly increased amount of superoxide ($O_2^-$) and dramatically augmented activities of superoxide dismutase (SOD), specially MnSOD and glutathione peroxidase (GPx) were observed according to the concentration and incubation time. Whereas, in case of normal lymphocytes under the same condition, cytotoxicities were not apparent and the generation of superoxide ($O_2^-$) or the activity changes of SOD and GPx were insignificant. These results together indicate that the cytotoxic action of SD-994 against L1210 cell may be achieved via necrosis and/or apoptosis induced by reaction oxygen species which could not probably be completely abolished even by drastically increased antioxidant enzymes, SOD and GPx activities.
