Objectives : Oxidative DNA damage is a known risk factor of lung cancer. The glutathione peroxidase (GPX) antioxidant enzyme that reduces hydrogen peroxide and lipid peroxides plays a significant role in protecting cells from the oxidative stress induced by reactive oxygen species. The aim of this case-control study was to investigate effects of oxidative stress and genetic polymorphisms of the GPX1 genes and the interaction between them in the carcinogenesis of lung cancer. Methods : Two hundreds patients with lung cancer and 200 age- and sex-matched controls were enrolled in this study. Every subject was asked to complete a questionnaire concerning their smoking habits and their environmental exposure to PAHs. The genotypes of the GPX1 and 8-oxoguanine glycosylase 1 (hOGG1) genes were examined and the concentrations of urinary hydroxypyrene (1-OHP), 2-naphthol and 8-hydroxydeoxyguanosine (8-OH-dG) were measured. Results : Cigarette smoking was a significant risk factor for lung cancer. The levels of urinary 8-OH-dG were higher in the patients (p<0.001), whereas the urinary 1-OHP and 2-naphthol levels were higher in the controls. The GPX1 codon 198 polymorphism was associated with an increased risk of lung cancer. Individuals carrying the Pro/Leu or Leu/Leu genotype of GPX1 were at a higher risk for lung cancer (adjusted OR=2.29). In addition, these individuals were shown to have high urinary 8-OH-dG concentrations compared to the individuals with the GPX1 Pro/Pro genotype. On the other hand, the polymorphism of the hOGG1 gene did not affect the lung cancer risk and the oxidative DNA damage. Conclusions : These results lead to a conclusion that individuals with the GPX1 Pro/Leu or Leu/Leu genotype would be more susceptible to the lung cancer induced by oxidative stress than those individuals with the Pro/Pro genotype.

A subtracted cDNA library of a marine scuticociliate, Philasterides dicentrarchii, in response to 17β-estradiol exposure was constructed using suppression subtractive hybridization (SSH). As a result of SSH, 275 clones were isolated, and among them, only glutathione peroxidase (GPX) gene was isolated as an antioxidative enzyme responding to 17β-estradiol. The semi-quantitative reverse-transcriptase polymerase chain reaction (RT-PCR) analysis revealed that the transcription of GPX gene of P. dicentrarchii was clearly increased by exposure to 17β-estradiol. The GPX transcription was also clearly increased by exposure to xenoestrogens such as bisphenol A (BPA) and genistein.

We investigated high light-induced alterations in antioxidant enzymes by exposing green vegetative cells of the alga Haematococcus pluvialis to excess irradiance to induce the production of astaxanthin, a carotenoid pigment. Total activity of catalase decreased approximately 70% after high light exposure, whereas glutathione peroxidase (GPX) activity was slightly enhanced. Total activity of superoxide dismutase and ascorbate peroxidase (APX) also slightly decreased. Overall, we did not observe dramatically elevated levels of antioxidant isozymes, although APXn, GPX2, and GPX3 isozyme increased slightly. ${H_2}{O_2}$ content increased about sixfold after high light exposure, demonstrating severe cellular oxidative stress, whereas lipid peroxidation was notably reduced. Concomitantly, astaxanthin accumulation increased about sevenfold. This result suggests that probably massively accumulated astaxanthin may be one of the antioxidant protector against high light stress.
The purpose of this study was to investigate the effect of endurance training and Rooibos-tea treatment during 12 week on lipid peroxidation(MDA), oxidative DNA damage(8-hydroxyguanine), and antioxidant enzymes(SOD, GPX) in human. The subjects were divided into three groups; Train+Rooi, Train, and Rooi groups. The Train+Rooi and Rooi group took 3 g of Rooibos-tea for 12 weeks. Blood samples were taken from antecubital vein at before training, after 6week, and after 12 week training. Data were analyzed by two-way ANOVA with repeated measures using the SPSS/PC+. The results are summarized as follows: MDA and 8-hydroxyguanine concentration were no significantly differences between group(p>.05). SOD and GPX concentration were significantly increased in Train+Rooi, Rooi group than Train group(p<.05). This results suggested that effects of Rooibos-tea treatment has associated with improve scavenger of antioxidant.
The present study evaluated the effects of different anesthesia techniques on oxidative stress in beagle dogs. Ten dogs were randomly assigned to either total intramuscular anesthesia with medetomidine-tiletamine/zolazepam (MTZ) combination (group T, 40 ${\mu}g/kg$ medetomidine and 2 mg/kg tiletamine/zolazepam) or volatile anesthesia with isoflurane (group I, 2% isoflurane and 100% oxygen). Heart rate, respiratory rate, and rectal temperature for vital signs and the concentration of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) for oxidative stress were measured. SOD activity decreased significantly from baseline anesthesia in both groups ($p$ < 0.05). CAT and GPx activities were also decreased significantly after anesthesia between both groups ($p$ < 0.05). CAT activity decreased significantly from baseline after anesthesia in both groups, but activities of group I were significantly higher compared with group T after anesthesia ($p$ < 0.05). GPx activity in group T decreased significantly from baseline after anesthesia, but activities of group I were significantly higher compare with that of group T 1 hour after the conclusion of anesthesia ($p$ < 0.05). In conclusion, general anesthesia seems to induce oxidative stress, and volatile anesthesia with isoflurane attenuates oxidative injuries in beagle dogs.

In order to investigate the cellular peroxidative damage due to heated oil intake and the preventive effect of vitamin E on it rats were fed heated corn oil with acid value of 4.02 at the level of 10 Cal% and three different levels of vitamin E that were 0, 40 and 200 mg/kg diet. Control group was fed fresh corn oil and 40mg/kg diet of vitamin E. After ech feeding period of 0, 3 and 6 weeks, liver superoxide dismutase (SOD), glutathione peroxidase (GPX) activities and microsomal content of vitamin E and lipid peroxide (LPO) were measured as well as cellular morphology was examined. SOD activities and LPO contents were higher, while GPX activities and vitamin e contents were lower in heated oil groups than control group. Electromicroscopic observation revealed the loss of inner mitochondrial membrane and cristae and irregular arrangement of nuclear membrane and chromatin in heated oil groups. As dietary vitamin e level was increased, SOD activity and LPO content were decreased, but GPX activity and vitamin E content in the liver increased and cellular peroxidative damage reduced progressively. This phenomena was more remarkable in 6 weeks of feeding than 3 weeks.
Four-d-old broiler chicks were randomly assigned to 3 groups with 9 replicates (8 birds/cage) under high ambient temperature; birds fed a basal diet (CON), a basal diet supplemented with 0.25% of probiotic complex (LPB, 1 × 106 Lactobacillus plantarum, 1 × 106 Bacillus subtilis, and 1 × 106 Saccharomyces cerevisiae) and 0.5% probiotic complex (HPB). Immediately after 28-d feeding trial, 6 birds having average body weight per group were sacrificed for evaluating the effects of probiotics on antioxidant parameters in the serum, intestine, and liver of birds. As results, serum biochemical parameters of nitrogen components including total protein, albumin, urea nitrogen, and glutathione were unaffected by dietary probiotics. In addition, serum superoxide dismutase (SOD), glutathione peroxidase (GPX), and glutathione S-transferase (GST) activities, and lipid peroxidation (MDA) were not changed by dietary probiotic supplement in birds. In the intestinal mucosa, SOD activity in the HPB group significantly (P<0.05) increased compared with that in the CON and the LPB groups. Lipid peroxidation in the HPB group significantly (P<0.05) decreased compared with that in the CON group. However, there was no statistical difference in GPX, and GST activities in the intestinal mucosa among treatment groups. In the liver, the activities of SOD, GPX, and GST, and the level of MDA were unaffected by probiotic supplement. In conclusion, 0.5% of probiotics significantly increased SOD activity and decreased lipid peroxidation in the intestinal mucosa, suggesting that probiotic complex could be potential to improve the small intestinal antioxidant capacity of bird under high ambient temperature conditions.
The purpose of this study was to investigate the effect of taurine on sperm characteristics and gene expressions(bax and Gpx) in fresh boar semen during in vitro storage. The motility of spermatozoa in Modena, Modana plus taurine 25 mM, Modana plus taurine 50 mM, Modana plus taurine 75 mM and Modana plus taurine 100 mM were 63.1%, 65.1%, 65.3%, 82.5% and 80.8%, respectively.
