Anti-hyperlipidemic effect of Agaricus brasiliensis fruiting bodies in rats fed with high cholesterol diet (고콜레스테롤 식이의 급여에 의해 고지혈증이 유도된 흰쥐의 신령버섯 자실체의 항고지혈증 효과)

  • In this study, we aimed to investigate the effects of dietary supplementation with fruiting body of Agaricus brasiliensis (AB) mushroom on the lipid profiles of serum and histological patterns of liver of high cholesterol-fed rats. Five-week-old, female Sprague-Dawley albino rats were divided into three groups of 8 rats each, including a normal control-diet (NC) group, a high-cholesterol diet (HC) group, and a group fed high-cholesterol diet supplemented with 5 % powder of Agaricus brasiliensis fruiting bodies (HC+AB). Total serum cholesterol, low density lipoprotein (LDL), and triglyceride (TG) concentrations in the HC+AB group were significantly reduced when compared with those in the HC group. Body weight in the HC+AB group was significantly lower than that in the HC group, whereas no adverse effects were observed on the levels of plasma albumin, creatinine, blood urea nitrogen, uric acid, glucose, and total protein. In the HC+AB group, liver enzyme activities related to liver function, such as GOT and GPT, presented values lower than those in the HC group and were very similar to the ones in the NC group. Excretion of total lipid and cholesterol in feces in the HC+AB group was significantly higher than that in the NC and HC groups, indicating that mushroom feeding inhibits the absorption of lipid cholesterol in the intestine. Liver histopathological analyses revealed that rats fed with HC diet developed fat liver disease, whereas only small amounts of fat were deposited in the livers of the HC+AB group. In conclusion, the results suggest that fruiting body powder of A. brasiliensis provides health benefits to high-cholesterol-fed rats by lowering body weight and the risk of atherogenic lipid profile.

Effect of Fermented Brown Seaweed Waste (FBSW) on Milk Production, Composition and Physiological Responses in Holstein Dairy Cows (발효미역부산물의 첨가가 Holstein 비유우의 비유성적 및 생리반응에 미치는 영향)

  • This study was conducted to determine effects of fermented brown seaweed waste (FBSW) on milk production, composition and physiological responses as functional feed for 60 days in Holstein dairy cows. A total of 24 Holstein dairy cows (average age 49.33 months, average lactation days 175, Reproduction 2.0) were randomly allocated into control(basal diet), 1% FBSW (180g in basal diet) and 2% FBSW (360g in basal diet) groups with 8 replications for 60 days. Daily milk yield and composition (fat, protein, SNF, MUN) were not affected by FBSW supplementation, but Ca level in milk was significantly increased 4.29 mg/dl and 2.91 mg/dl in 1% and 2% groups compared to control group (p<0.05) at the end of the experiment, respectively. The somatic cell count (SCC) in milk was not significant. The plasma $T_4$ level (concentration) were increased in 1% and 2% FBSW compared to control group at the end of the experiment (p<0.05), but between triiodothyronine ($T_3$) and thyroxin ($T_4$) levels were not significant. Concentrations of plasma glucose in control, 1% FBSW and 2% FBSW groups were 64. 37mg/dl, 66.15mg/dl and 73.02 mg/dl and plasma NEFA level was 0.30~0.32 mEq/dl. Concentrations of BUN tended to be higher for FBSW group than control group. Although WBC, RBC, Hb, Hct, T-B, ALP, and GPT levels were not affected by FBSW supplementation, GOT level was significantly decreased in cows fed 1% FBSE diet compared to control group (P<0.05). Therefore we strongly suggest that the 1% FBSW supplementation in basal diet increases the milk yield and Ca level in Holstein dairy cows.

Anti-Obesity Effect of Red Garlic Composites in Rats Fed a High Fat-Cholesterol Diet (고지방-콜레스테롤 식이성 흰쥐에서 홍마늘 복합물의 항비만 효과)

  • Three kinds of dietary composites-R+T, R+F, and R+TF-were combined in green tea (T), dietary fiber (F), and green tea dietary fiber mixture (TF) to red garlic extract (RG), respectively. The effects of their diets on anti-obesity were investigated $in$ $vitro$ and $in$ $vitro$ in obese rats induced high fat-cholesterol. In $in$ $vivo$ rats, the total phenolic content of the R+T and R+TF was 1.9~2.0 times higher, and their total cholesterol adsorption was 9.5~11.5 times higher than that of RG. $In$ $vivo$, male Sprague-Dawley rats were divided into 6 groups (Normal, HFC, HRG, HR+T, HR+F and HR+TF). Afterwards, the diets of the HRG, HR+T, HR+F, and HR+TF groups were supplemented with 1% of RG and its dietary composites (R+T, R+F, and R+TF) for 4 weeks, respectively. The final body weight of the HRG, HR+T, HR+F, and HR+TF groups decreased significantly compared to the group fed high fat-cholesterol (HFC), but the food efficiency ratio was not significantly different from the HFC group. The liver weight of the HFC group doubled compared to the normal group, whereas that of HR+T and HR+TF groups decreased significantly. The weight of visceral and epididymal fat decreased significantly in the groups fed the composites compared to the HFC group. The obesity index of HR+TF group decreased significantly only when compared to the HFC group. The serum lipid profile such as total lipids, cholesterol, triglyceride, LDL- and VLDL-cholesterol, as well as the atherogenic index and cardiac risk factors decreased drastically in all experimental groups compared to the HFC group, and the levels of HR+T, HR+F and HR+TF groups were a similar trend. GPT activity was not significantly different among the groups fed the composites, and it decreased significantly in the HRG group. The content of the lipid peroxide level decreased significantly in the HRG group and in the groups fed the composites, compared to the HFC group. Serum antioxidant activity was the highest in the HR+T group. We suggest that the hypolipidemic and anti-obesity effect of the RG composites, achieved by mixing green tea extract and/or dietary fiber, was due to their total phenolic content and total cholesterol adsorption effect.

Effect of Dietary Supplementation of Alisma canaliculatum (Alismatis Rhizoma) and Viscum album (Mistletoe) on Growth Performance and Immunity in Broiler Chickens (택사와 겨우살이 급여가 육계의 생산성 및 면역성에 미치는 영향)

  • This study was conducted to investigate the effect of dietary supplement of Alisma canaliculatum and Viscum album on the growth performance and immune response of broiler chickens. One hundred and forty Ross broiler chicks (1 day old) were assigned to 4 treatments in 5 replications with 7 birds per replication. The dietary treatments were NC group (without antibiotics), PC group (basal + 50 ppm Oxytetracycline), AC group (basal + Alisma canaliculatum 0.5%), and VA group (basal + Viscum album 0.5%). The results revealed that body weight gain and feed intake were not significantly different among the treatments. Addition of AC to the diet showed significantly higher feed conversion ratios than the VA addition diet and other treatments (p<0.05). The concentration of linolenic acid, ${\alpha}$-linolenic acid and arachidonic acid in the AC and VA treatment were relatively higher than that in the antibiotic treatment, while there was no difference in the concentration of SFA, USFA, and USFA/SFA. The serum IgG concentration in chickens tended to increase in the AC and VA treatment groups compared to NC group. The IL2 concentration in AC and VA groups was significantly higher than in other group (p<0.05). IL6 was not detected, however. GOT, GPT, and T-billirubin in the VA group were significantly higher than in other groups (p<0.05). Thus, the VA additives can result in the damage of liver and kidney. In conclusion, the dietary supplement of a 0.5% of AC and VA showed a similar growth performance and increased immunity response comparable to supplementing a 50 ppm Oxytetracycline. Therefore, it can be said that the AC and VA are possible alternates of antibiotics.

Characteristics of the Health Factors in 45~60 Year Old Korean Women related to Menopausal Stages - Based on 2008~2009 Korean National Health and Nutrition Examination Survey - (2008~2009년 국민건강영양조사를 활용한 45~60세 한국여성의 폐경 여부에 따른 건강인자 특성)

  • We analyzed data from the combined 2008~2009 Korean National Health and Nutrition Examination Survey (KNHANES) to compare the health factors related to menopausal stages in 45~60 year old Korean women. In this study, we classified the subjects into a premenopausal group (n = 439) and a postmenopausal group (n = 683). In the postmenopausal group, age was higher (p < 0.001), monthly income (p < 0.01) and education levels (p < 0.001) were significantly lower than in the premenopausal group. Body fat % and waist circumferences were also higher in the postmenopausal group than in the premenopausal group. The serum glucose (p < 0.05), total cholesterol (p < 0.001), LDL-cholesterol (p < 0.001), triglyceride (p < 0.001), GOT (p < 0.001), GPT (p < 0.001) in the postmenopausal group were higher than in the premenopausal group. The postmenopausal group showed a significantly lower quality of life compared to the premenopausal group (p < 0.01). With regard to dietary quality, nutrient adequacy ratio (NAR) of vitamin A, vitamin $B_1$, vitamin $B_2$ and niacin in the postmenopausal group were significantly lower than in the premenopausal group. The levels of glucose, total cholesterol, LDL-cholesterol, and triglyceride showed a significantly positive correlation with age, waist circumferences, body fat % and BMI. The 45~60 year old Korean women in this study showed high levels of obesity and serum lipids. Also, intakes of the vitamins and minerals of the women did not meet the level of Dietary reference intakes for Koreans. Therefore, nutritional risk may be high in the women, especially in postmenopausal women. In order to prevent the health risk, women's health care including the quality of the meal should be considered.

Effects of Agarum cribrosum on the Improvements in Treating Constipation and Plasma lipid Profiles (구멍쇠 미역의 변비 완화 효과 및 혈액지방에 미치는 영향)

  • The present study was conducted to evaluate the effects of Agarum cribrosum on weight change and defecation states in rats with constipation induced by loperamide. Food intake and body weight both decreased in the 5% Agarum cribrosum and loperamide-treated group (SD5) and 10% Agarum cribrosum and loperamide-treated group (SD10), whereas fecal water contents increased by 1.6 and 2.1-fold in the SD5 and SD10 groups, respectively. The concentrations of total-cholesterol, HDL-cholesterol and triglyceride in the sera of the SD5 and SD10 groups were lower than those in the control (C) group. However, the biochemical parameters, GOT (glutamic oxaloacetic transaminase), GPT (glutamic pyruvic transaminase), and glucose levels, were not affected by the level of Agarum cribrosum. In addition, the concentrations of total-cholesterol and triglyceride in the livers of the SD5 and SD10 groups were also lower than those in the control group. The results of the present study demonstrated that Agarum cribrosum might ameliorate constipation symptoms, and lower lipid concentrations in the blood and liver.

Effect of Enzymatic Hydrolysate of Hamcho (Salicomia herbacea) on Antioxidative Defense System in Rats Fed High Cholesterol Diet (함초(Salicornia herbacea)의 효소적 가수분해물이 고콜레스테롤 식이 흰쥐의 항산화방어계에 미치는 영향)

  • The purpose of the present study was to investigate the effect of enzymatic hydrolysate (extract) of Salicomia herbacea L. (Korean name: Hamcho) on antioxidative defense system in rats fed high cholesterol diet. Rats were divided into six experimental groups which are composed of normal diet group, normal diet group supplemented with 2% Hamcho extract, high cholesterol diet group, high cholesterol diet groups supplemented with 1%, 2% and 4% Hamcho extracts. The activity of serum glutamate oxaloacetae transaminase in rats was not different among all experimental groups, while the activity of glutamate pyruvate transaminase in groups supplemented with Hamcho extract was significantly lower than that of high cholesterol control group. Supplementation of Hamcho extract (SHE) to the high cholesterol fed rats resulted in increased activities of hepatic superoxide dismutase and glutathione peioxidase. However, there was no significant difference in the activity of hepatic catalase among all experimental groups. SHE also resulted in decreased levels of hepatic thiobarbituric acid reactive substances and mitochondrial carbonyl values. Those effects were higher to some extent in 2% and 4% Hamcho extract groups than those of high cholesterol control group. These results suggest that enzymatic hydrolysate of Hamcho may reduce oxidative damage by activation of antioxidative defense system in rats fed high cholesterol diets.

Effect of Deer Antler Drink Supplementation on Plasma Lipid Profiles and Antioxidant Status in Type 2 Diabetic Patients (녹용혼합음료의 섭취가 당뇨환자의 지질양상 및 항산화 영양상태에 미치는 영향)

  • The effect of commercial deer antler drink (provided by Chung-yang Deer Farm) on blood glucose level, plasma lipids and antioxidants state in type 2 diabetic patients were studied. Ten patients with type 2 diabetes participated in the study and consumed 2 pouches (200 mL) of deer antler drink every day for 3 weeks. No significant differences were observed in levels of triglycerides, total cholesterol, low density lipoprotein cholesterol (LDL-C) and high density lipoprotein cholesterol (HDL-C). However, oxidized LDL measured as conjugated dienes decreased in the patients after the trial. Plasma tocopherols and carotenoids levels showed no significant changes. No significant differences were observed in erythrocyte SOD, catalase and GSH-Px in the each group. No significant differences were observed in plasma TRAP. The results would suggest that deer antler drink influences conjugated dienes but long-term intervention trial may be necessary to see further beneficial effect of deer antler drink in diabetic patients.

Modulation of obesity by Gyeongshingangjeehwan18 in ob/ob mice (Ob/Ob 마우스에서 경신강지환(輕身降脂丸)18의 비만조절)

  • Objectives : This study was undertaken to verify the effects of Gyeongshingangjeehwan18 (GGEx18) on obesity using ob/ob male mice. Methods : Eight-week old mice (wild-type C57BL/6J and ob/ob) were used for all experiments. Wild-type C57BL/6J mice were used as lean control and obese ob/ob mice were randomly divided into 5 groups: obese control, GGEx15, GGEx16, GGEx17, and GGEx18. After mice were treated with several kinds of GGEx for 11 weeks, body weight gain, feeding efficiency ratio, plasma lipid and glucose metabolism. Results : 1. Compared with obese controls, GGEx-treated mice had lower body weight gain and feeding efficiency ratio, the magnitudes of which were prominent in GGEx16 and GGEx18. 2. Consistent with their effects on body weight gain, GGEx16 and GGEx18 not only decreased plasma triglycerides levels, but also increased HDL-cholesterol concentration. 3. CT analysis revealed that visceral fat areas were decreased in all treatment groups compared with obese control mice. The decrease in visceral fat area was prominent in GGEx16 and GGEx18, although they were not statistically significant. 4. The size of adipocytes were significantly decreased by GGEx18, whereas the adipocyte number per unit area was significantly increased, suggesting that GGEx18 decreased the number of large adipocytes. Hepatic lipid accumulation was decreased by GGEx16 and GGEx18, and the inhibitory effect was most effective in GGEx18. 5. Plasma GOT and GPT concentrations were significantly lower following GGEx16 and GGEx18 treatment compared with obese controls. Organ weights were not changed by GGEx treatment, indicating GGEx do not show any toxic effects. Conclusions : These results suggest that GGEx may regulate obesity. Of the 4 compositions, GGEx18 seems to be most effective in improving obesity and lipid disorders.

Effect of Yinjinchunggan-tang based Herb Formulae Containing Wasong and Eosungcho on Fibrogenesis (인진청간탕 및 와송 어성초 가미방의 간섬유화억제에 미치는 효과)

  • Objectives : This study was performed to investigate the anti-fibrogenic effect and the effect on cell growth and apoptosis in YJCGT, YJCGT YSO and YJCGT YSCO on thioacetamide-induced rat liver tissue and the immortalized human hepatic cell line LX2. Materials and Methods : LX2 cells were treated with various concentrations (0, 50, 150, 300 ug/ml) of YJCGT, Y+YSO, and Y+YSCO extract for 24, 48 and 72 hours. After the treatment, cell viability was measured by using MTT assay. Caspase inhibitor assay, and cell viability were determined by a colorimetric assay with PMS/MTS solution. Rat liver fibrosis was induced by intraperitoneal thioacetamide injection 150 mg/kg 3 times a week for 5 weeks. After the treatment, body weight, liver & spleen weights, liver function test, the complete blood cell count and the change of portal pressure were studied. After YJCGT, Y+YSO, and Y+YSCO treatment, percentages of collagen in thioacetamide-induced rat liver tissue were measured. Results : The viability of the LX2 cell decreased in a dose- and time-dependent manner. Exposure of LX2 cells to YJCGT, YJCGT+YSO and YJCGT+YSCO induced caspase-3 activation, but co-treatment of YJCGT, YJCGT+YSO and YJCGT+YSCO with the pan-caspase inhibitor Z-VAD-FMK, and the caspase-3 inhibitor Z-DEVE-FMK, blocked apoptosis. There was no difference in rat body weight between the thioacetamide only group and the YJCGT, YJCGT+YSO and YJCGT+YSCO groups. In the YJCGT, YJCGT YSO and YJCGT YSCO groups, the serum level of GPT significantly went down compared with the thioacetamide only group. In the YJCGT, Y+YSO, Y+YSCO groups, white blood cell elevated by thioacetamide injection decreased but RBC, Hgb, and Hct increased. In the Y+YSO group, the portal pressure elevated by thioacetamide injection significantly decreased. In the histological finding, thioacetamide injections caused severe fibrosis, but YJCGT, Y+YSO, and Y+YSCO treatment significantly reduced the amounts of hepatic collagens. Conclusions : YJCGT, Y+YSO, and Y+YSCO inhibit the growth of LX2 cells by inducing apoptosis through caspase activity. YJCGT, Y+YSO, and Y+YSCO have beneficial effects on the treatment of cirrhotic patients as well as patients with chronic hepatitis.
