In recent years, global warming is causing dramatic environmental changes and deterioration, such as hypoxia, leading to reduced survival rate and growth performance of farmed aquatic animals. Hence, understanding systemic immuno-physiological changes in fish under environmental stress might be important to maximize aquaculture production. In this study, we investigated physiological changes in rainbow trout exposed to hypoxic stress by monitoring changes in blood chemistry, leukocyte population, and expression levels of related cytokine genes. Hematological and serological factors were evaluated in blood obtained from rainbow trout sampled at a dissolved level of 4.6 mg O2 L-1 and 2.1 mg O2 L-1. Blood and head kidney tissue obtained at each sampling time point were used to determine erythrocyte size, leukocyte population, and cytokine gene expression. The level of LDH and GPT in fish under progressive hypoxia were significantly increased in plasma. Likewise, the (Granulocyte + Macrophage)/lymphocyte ratio (%) of fish exposed to hypoxia was significantly lower than that in fish in the control group. Such changes might be due to the rapid movement of lymphocytes in fish exposed to acute hypoxia. In this study, significant up-regulation in expression levels of IL-1β and IL-6 gene appeared to be involved in the redistribution of leukocytes in rainbow trout. This is the first study to demonstrate the involvement of cytokines in leukocyte trafficking in fish exposed to hypoxia. It will help us understand systemic physiological changes and mechanisms involved in teleost under hypoxic stress.
This study was conducted to determine the circadian rhythm and the effect of starvation for the health assessment of olive flounder Paralichthys olivaceus, reared under 12:12 LD (light:dark) cycle and fed 1% of their body weight once a day. The blood collection was implemented from 10 fish every 3 hours for a day, and analyzed to serval health parameters of the experimental fish. Plasma cortisol, glucose and total protein concentration reflected diel rhythms with high level of photophase and low level of scotophse despites of glutamic oxalacetic transaminase (GOT) and glutamic pyruvate transaminase (GPT) showing no rhythmicity, while electrolyte (sodium, potassium and chloride ion) concentrations arrived at the peak 18 hour and then slowly down. In the experiment of starvation, the result showed that there were no significant differences of the health parameters between the starved group and the fed. In short, our findings described the existence of circadian cortisol with glucose in flat fish, and indicated that the starvation does not almost affect the health of fish.
The purpose of this paper is to find out whether Du-chung tea and Du-chung tea added with Stevia leaves are suitable for everyday use of diabetics. For this purpose, states of diabetic rabbit, induced by giving alloxan to normal rabbit, were examined in two cases. One was to administer Du-chung tea sweetened by sugar to the above same animal and the other to apply Du-chung tea sweetened by Stevia leaves, instead of sugar, under the equal condition. A study was carried out to determine the effects on blood sugar level, contents of BUN (blood urea nitrogen) and total cholesterol, and GPT (glutamic pyruvate transaminase) activity in both groups. The following results were obtained. 1) Sweetness of Du-chung tea added with 0.3-0.5% Stevia leaves was never to the standard sweetness. 2) Sweetness of Du-chung tea with 2% sugar plus 0.2% Stevia leaves was the nearest to the standard sweetness. 3) Du-chung tea with 0.5% Stevia leaves was recognized as the most effective the next was Du-chung tea no sweetening, and the worst was Du-chung tea with sugar. Addition of 0.2% Stevia leaves without lessening sugar concentration of the above Du-chung tea showed the same result as Du-chung tea with sugar. 4) Du-chung tea with Stevia leaves and Du-chung tea without sweetening had the tendency of reducing the total cholesterol content in serum, while only Du-chung tea with Stevia leaves presented remarked difference after 4, 6 and 10 days, when they were compared with control group. 5) Degree of CPT, activity had in general the similar tendency- to that of total cholesterol content. 6) Taken Du-chung tea with Stevia leaves and Du-chung tea without sweetening, the BUN content showed reducing tendency in small quanity, but, having little defference from control group.
Acetaminophen, which causes acute liver min in humans and animals, has made useful inducer of hepatoxicity for studying hepatopreventive drugs. Injinhotang is known as one of the hepatopreventive drugs. However, its mechanism of recovery of hepatoxicity treated with acetaminophen is poorly understood. this study was performed to observe the antioxidative effect of injinhotang extract and its several combination groups. The results were obtained as follows:1. In the study on free radical scavenging effect in vitro(the suppressing effect on peroxidation of linoleic acid on concentration, the scavenging effect of DPPH radical, inhibitory effect of superoxide in xanthine-xanthine oxidase system and the inhibitory effect on lipid peroxidation reaction by hydroxy radical in H2O2-Fe2+system, injinhotang have more effect than its components groups relatively. 2. In the study on antioxidants system in vivo(the level of serum LPO, the level of hepatic LPO, catalase, GSH, GST), only injnhotang has a significant effect. 3. In the study on hepatotoxicity(GOT, GPT, $\gamma$-GTP, ALP, LDH, b ilirubin), only injinhotang has a significant effect. These results suggest that injinhotang has the protective effect on acetaminophen-induced hepatoxicity. The mechanisms of these are supposed to be involved in antioxidant and three drugs' cooperative synergy effect.
This study was undertaken to find out the bio-effects due to be a radiation fractionated exposure. The experimental animals were divided into the control group and the radiation exposure groups of 20cGy, 40cGy and 80cGy with 220 male Sprague-Dawley rats at 6 weeks old. The radiation exposure groups were fractionated exposed from each 20cGy, 40cGy and 80cGy for every 5 days. The chromosome aberrations, the frequency of SCE, the changes of body weight, hematological values and enzyme activities were investigated for the fractionating exposure times and the time after fractionated exposure. The results were summarized as follows 1. The body weight of the radiation exposure groups were significantly decreased compared with control group according to the increasing fractionated exposure times, and it was the lowest values at the immediately after the end of the fractionating exposed, but it was recovered with the level of control group at 3rd weeks gradually increased 1st week after fractionated exposure. 2. The values of WBC, RBC, Hb and Hct in the radiation exposure groups were significantly decreased than those the control group, but the values of GOT, GPT, ALP, and LDH in the radiation exposure groups were significantly increased than those of the control group. 3. The frequency of chromosomal aberration were increased according to the increasing fractionated exposure dose, and it showed the highest at 5th days after fractionated exposed. The types of chromosomal aberration were occurred such as a numerical abnormality, deletion, break and duplication, it was not recovered immediately and maintained high frequency than the control group. 4. The frequency of SCE were significantly increased according to the increasing fractionated exposure dose in 20cGy, 40cGy and 80cGy groups. But it was recovered the level of control group at 7th days after fractionated exposure. According to the above results, this study could confirm that the frequency of chromosomal aberration and SCE were increased with fractionated exposure dose, the other hand, the changes of body weight, hematological values and enzyme activity values were significantly affected according to the increasing fractionated exposure dose.

This experiment was conducted to evaluate anti-Salmonella enteritidis (anti-SE) bacteriophage as feed additives to prevent Salmonella enteritidis in broilers. The experimental diets were formulated for 2 phases feeding trial, and 3 different levels (0.05, 0.1 and 0.2%) of anti-SE bacteriophage were supplemented in basal diet. The basal diet was regarded as the control treatment. A total of 320 1-d-old male broilers (Ross 308) were allotted by randomized complete block (RCB) design in 8 replicates with 10 chicks per pen. All birds were raised on rice hull bedding in ambient controlled environment and free access to feed and water. There were no significant differences in body weight gain, feed intake and feed conversion ratio (FCR) at terminal period among treatments (p>0.05). Relative weights of liver, spleen, abdominal fat and tissue muscle of breast obtained from each anti-SE bacteriophage treatment were similar to control, with a slightly higher value in anti-SE bacteriophage 0.2%. In addition, a numerical difference of glutamic-oxaloacetic transaminase (GOT), glutamic-pyruvic transaminase (GPT) and LDL cholesterol level was observed in the 0.2% anti-SE bacteriophage application even though blood profiles were not significantly affected by supplemented levels of anti-SE bacteriophage (p>0.05). In the result of a 14 d record after Salmonella enteritidis challenge of 160 birds from 4 previous treatments, mortality was linearly decreased with increasing anti-SE bacteriophage level (p<0.05), and Salmonella enteritidis concentration in the cecum was decreased with increasing levels of anti-SE bacteriophage (p<0.05). Based on the results of this study, it is considered that supplementation of 0.2% anti-SE bacteriophage may not cause any negative effect on growth, meat production, and it reduces mortality after Salmonella enteritidis challenge. These results imply to a possible use of anti-SE bacteriophage as an alternative feed additive instead of antibiotics in broilers diet.
The effect of oral vitamin e (800IU/day) and C (500mg/day) supplementation for 10 days and/or smoking cessation for 5 days on oxidative damage to the red blood cells (RBC) of male smokers (22.2$\pm$0.2 years old) was studied. RBC were tested for their ability to protect against smoking-induced oxidative damage by measuring heme proteins (carboxyhemoglobin, hemoglobin, methemoglobin, oxyhemoglobin), hemolysis and thiobarbiturinc acid reactive substances (TBARS). Plasma levels of vitamin c, A, E, $\beta$-catotene, total cholesterol, glutamic pyruvic transaminase(GPT) and glutamic oxaloacetic transaminase(GOT) were also analyzed. In experiment one, a comparison was made of heme proteins and lipid damage to RBC, plasma antioxidant status (indexed by plasma levels of vitamin C, E, A and $\beta$-carotene) between smokers(n=56) and non-smokers (n=16). No differences were found in plasma antioxidant status, heme protein damage and TBARS concentration of RBC. In experiment two, 46 fasting male smokers from experiment one were divided into 4 groups. The groups were smoking with placebo group(SP, n=14), smoking cessation with vitamins supplementatin group (SV, n=13), smoking cessation with placebo group (NSP, n=9) and smoking cessation with vitamins supplementation group (NSV, n=10). After supplementing antioxidant vitamins, significant increases were seen in plasma vitamins supplementation group (NSV, n=10). After supplementing antioxidant vitamins, significant increases were seen plasma vitamin C (p<0.05) and vitamin E levels (p<0.05). The plasma vitamin E level was highest in the NSV group. Vitmain E and C supplementation provided some protection against heme proteins and lipid damage by lowering methemoglobin, hemolysis and TBARS concentration of RBC. Smoking cessation significantly decreased TBARS of RBC and plasma total cholesterol concentration. Supplementing vitamin E and C with smoking cessation considerably lowered plasma total cholesterol. These results point to a special association among smoking, oxidative damage and plasma antioxidant vitamin status. They indicate that increases in plasma antioxidant status can be detected after the supplementation of vitamin C and E and that smoking cessation had an additional effect on plasma vitamin E level. The present data suggest that improved antioxidant status induced by antioxidant supplementation or smoking cessation may help prevent oxidative damage in smokers.
In order to investigate whether Eisenia bicyclis (EB) as a functional food material improves serum lipid status, supplementation of EB powder or EB extract to the high fat/cholesterol diet was tested in 6-week old male Sprague-Dawley rats for four weeks. In the first experiment, four kinds of seaweed powder -Eisenia bicyclis (EB), Undaria pinnatifida (Up), Laminaria japonica (LJ), and Ecklonia stolonifera (ES) were compared with alginic acid (ALG) as well as control. In the second experiment, EB water extract and EB ethanol extract and EB pill containing EB and LJ powder were compared with the EB powder. Amounts of dietary fiber contained in experimental diets were adjusted to provide 5% of diets. Serum total cholesterol was significantly lower only in rats fed LJ or EB powder, but HDL cholesterol was higher in rats fed UP, LJ, ES or EB powder. Ratios of HDL-/total cholesterol of all seaweed groups including ALG were significantly higher than that of the control group. Serum triglyceride was lower in LJ, EB nd ES powder groups than the control group. In comparison of EB extracts, triglyceride level was decreased in rats fed only EB water extract as compared to control rats, but HDL cholesterol was elevated with both water and ethanol extracts. Serum levels of total cholesterol, triglyceride were decreased and HDL cholesterol was increased by the EB pill. Serum levels of thiobarbituric acid-reactive substances were lower in rats fed ES powder, and EB powder, EB water extract or EB pill, as compared to the control rats. Supplementation of EB powder and EB pill reduced serum level of GOT and GPT, respectively, as compared to the control group. In conclusion, EB and EB pill improve serum lipid status and may be utilized as ingredient of functional foods for the purpose of improving serum lipid profile and inhibiting peroxidation of lipids.
Oxidative stress is caused by an imbalance between the production of reactive oxygen and an ability of a biological system, to readily detoxify the reactive intermediates or easily repair the resulting damage. It has been suggested that developmental alloxan-induced liver damage is mediated through increases in oxidative stress. The anti-diabetic effect and antioxidant activity of Phaleria macrocarpa (PM) fractions were investigated in alloxan-induced diabetic rats. After two weeks administration of PM, the liver antioxidant enzyme and hyperglycemic state were evaluated. The results showed that oral administration of PM treatments reduced blood glucose levels in diabetic rats by oral administration (P < 0.05). Serum glutamic-oxaloacetic transaminase (sGOT) and serum glutamic-pyruvate-transaminase (sGPT) were also diminished by PM supplementation. The superoxide dismutase (SOD), catalase (CAT) and glutathione-peroxidase (GPx) activities, and glutathione (GSH) level in the alloxan-induced diabetic rats were significantly decreased (P < 0.05) compared to those in the normal rats but were restored by PM treatments. PM fractions also repressed the level of malondialdehyde (MDA) in the liver. Glutathione reductase (GR), glutathione-S-transferase (GST) and $\gamma$-glutamylcysteine synthase (GCS) were also reduced in alloxan-induced diabetic rats. PM fractions could restore the GR and GST activities, but the GCS activity was not affected in rat livers. From the results of the present study, the diabetic effect of the butanol fraction of PM against alloxan-induced diabetic rats was concluded to be mediated either by preventing the decline of hepatic antioxidant status or due to its indirect radical scavenging capacity.
This study was carried out to compare the dietary behaviors and the nutrient intake of a middle school and a high school girls in Iksan. The subjects consisted of 132 middle school girls and 122 high school girls in Iksan. The proportion of subjects eating meals irregularly was higher in the high school girls (98.6%) than in the middle school girls(78.7%). The proportion of subjects skipping a meal also was higher in the high school girls(61.5%) than in the middle school girls(53.8%) and most of them (65.8%) skipped breakfast though they thought breakfast was the most important meal of a day (87.4%). The subjects tended to overeat at lunch (38.6%), and at dinner (55.9%). The main reasons of skipping a meal were 1. They had no time (49.3%), and 2. They had poor appetite (24.7%) Mean daily energy intake was 2198.5Kcal in the middle school girls and 2150.5Kcal in the high school girls and mean daily intake of protein, iron, vitamin $B_1$, vitamin $B_2$ and niacin was significantly higher (p<0.05, p<0.01) in the middle school girls than in the high school girls. Mean daily calcium, iron, vitamin $B_1$, vitamin A and C intake of middle school girls were lower than those of RDA and all nutrients intake of high school girls except for phosphorus was lower than those of RDA. The index of nutritional quality (INQ) was under 1.0 for calcium, iron, vitamin A, $B_1$ and C. Mean blood level of cholesterol, triglyceride, Hb, Hct, GOT, GPT were 170.9mg/dl, 78.9mg/dl, 13.1g/dl, 39.5%, 18.8mg/d and 15.7mg/dl, respectively.
