Cloning, Over-expression, and Characterization of YjgA, a Novel ppGpp-binding Protein

  • Guanosine-5'-diphosphate 3'-diphosphate (ppGpp) serves as alarmone in bacterial stringent responses. In this study, an affinity column was constructed by immobilizing ppGpp to NHS-Sepharose for isolating ppGpp-binding proteins. A novel ppGpp-binding protein, YjgA, was isolated and characterized by MALDI-TOF MS (matrix-assisted laser desorption ionization-time-of-flight mass spectrometry) coupled with two-dimensional gel electrophoresis. YjgA and truncated forms of YjgA were cloned and over-expressed in BL21 (DE3). The binding affinity of YjgA to ppGpp was determined by equilibrium dialysis. The interaction of YjgA with ppGpp was very specific, considering that the dissociation constant of YjgA with ppGpp was measured as $5.2{\pm}2.0{\mu}M$, while the affinities to GTP and GDP were about 60 and 30 times weaker than ppGpp. Expression of yjgA gene in Escherichia coli K-12 MG1655 was examined by reverse transcription polymerase chain reaction (RT-PCR). RT-PCR results revealed that yjgA was expressed from early to late stationary phase. The yjgA deletion mutant exhibited decreased cell number at stationary phase compared to parent strain and the over-expression of YjgA increased the cell number. These results suggested that YjgA might stimulate cell division under stationary phase. In most prokaryotic genome, about half of the protein candidates are hypothetical, that are expected to be expressed but there is no experimental report on their functions. The approach utilized in this study may serve as an effective mean to probe the functions of hypothetical proteins.

An Enhanced Frequency Synchronization Algorithm for 3GPP LTE FDD/TDD Dual Mode Downlink Receiver (3GPP LTE FDD/TDD 듀얼 모드 하향 링크 수신기를 위한 개선된 주파수 동기 알고리즘)

  • In this paper, we propose a coarse and fine frequency synchronization method which is suitable for the 3GPP(3rd Generation Partnership Project) LTE(Long Term Evolution) FDD(Frequency Division Duplexing) / TDD(Time Division Duplexing) dual mode system. In general, PSS(Primary Synchronization Signal) correlation based estimation method and CP(Cyclic Prefix) correlation based tracking loop are applied for coarse and fine frequency synchronization in 3GPP LTE OFDMA(Orthogonal Frequency Division Multiple Access) system, respectively. However, the conventional coarse frequency synchronization method has performance degradation caused by fading channel and squaring loss. Also, the conventional fine frequency synchronization method cannot guarantee stable operation in TDD mode because of signal power difference between uplink and downlink subframe. Therefore, in this paper, we propose enhanced coarse and fine frequency synchronization methods which can estimate more accurately in multi-path fading channel and high speed channel environments and has stable operation for TDD frame structure, respectively. By computer simulation, we show that the proposed methods outperform the conventional methods, and verify that the proposed frequency synchronization method can guarantee stable operation in 3GPP LTE FDD/TDD dual mode downlink receiver.

Relationship between gross primary production and environmental variables during drought season in South Korea (가뭄 기간 총일차생산량과 환경 변수 간 상관관계 분석)

  • Water stress and environmental drivers are important factors to explain the variance of gross primary production (GPP). Environmental drivers are used to generate GPP in Moderate Resolution Imaging Spectroradiometer (MODIS) algorithm and process-based model. However, MODIS algorithm only consider the vapor pressure deficit (VPD) data while the process-based biogeochemical model also uses limited data to express water stress. We compared the relationship between environmental drivers and GPP from eddy covariance method, MODIS algorithm, and Community Land Model 4 (CLM 4) simulation in normal years and drought years. To consider water stress specifically, we used VPD and evaporative fraction (EF). We evaluated the effects from environmental drivers and EF towards GPP products using the structural equation modeling (SEM) in South Korea. We found that GPP products from MODIS algorithm and model simulation results were not restricted from VPD data if VPD was underestimated. We also found that in the cropland area, irrigation effects can relieve VPD effects to GPP. However, GPP products derived from MODIS and CLM 4 had limitation to explain the irrigation effects to GPP. Overall, these results will enhance the understanding of GPP products derived from MODIS and CLM 4.

  • This paper investigates an advanced receiver for interference mitigation of downlink control channel in the 3GPP Rel-13 standard. There are several features for downlink throughput performance improvement with inter-cell interference management such as network coordination and advanced receivers during Rel-10~Rel-12. These features can be operated always under the assumption that UE perfectly decodes control channels (PCFICH and PDCCH) of serving cell. However, the performance of control channels could be deteriorated in the cell edge region due to inter-cell interference. In this paper, we introduce the advanced receivers and analyze performance for control channel interference mitigation (CCIM) based on 3GPP Rel-13 standard. Additionally, we propose UE behavior depending on network condition.

QoS Analysis of 3GPP Service based on PBMN and DiffServ (PBNM과 DiffServ 적용한 3GPP 서비스의 QoS 해석)

  • In this paper, Policy-based QoS in 3GPP service network management techniques are not applied to the DiffServ technology is applied to the first interpretation. The next PBNM and DiffServ associated technologies by applying QoS performance improvement is verified. In this case that PBNM and DiffServ technology is applied, the amount of voice traffic reduced about 1 msec while best-effort traffic occurs 75 percent of the output link capacity. Also, video traffic which is the same as data traffic showed a decreased $0\sim10^{-4}$ packet loss rate than the case that DiffServ technology is applied. We apply the appropriate policy PBNM and DiffServ QoS mechanisms of the existing set of policies is not affected, just by using the appropriate 3GPP Service QoS level to suit the network operation, management can do that was found. This analytical method based on the University of California at Berkeley through NS-2 DiffServ technology into existing systems and next-generation networks mandated PBNM and DiffServ technology is applied to performance evaluation for the case.

Complexity Reduced CP Length Pre-decision Algorithm for SSS Detection at Initial Cell Searcher of 3GPP LTE Downlink System (3GPP LTE 하향링크 시스템의 초기 셀 탐색기 SSS 검출 시 복잡도 최소화를 위한 CP 길이 선 결정 알고리즘)

  • In 3GPP LTE system downlink, PSS (primary synchronization signal) and SSS (secondary synchronization signal) sequences are used for initial cell search and synchronization. UE (user equipment) detects slot timing, frequency offset, and cell ID by using PSS. After that it should detect frame timing, cell group ID, and CP length by using SSS. But in 3GPP LTE, there are two kinds of CP length, so we should operate FFT twice. In this paper, to minimize SSS detection complexity in cell searcher, we propose a CP length pre-decision algorithm that reduces the arithmetical complexity by half at most, with negligible performance degradation.

A Study on the Verification Scheme of SMS Encoding and Decoding Module (SMS 부호화 복호화 모듈 검증 방법에 대한 연구)

  • This paper proposes a test method for compliance of SMS encoder and decoder modules with 3GPP (3rd Generation Partnership Project) specification on SMS PDU (Protocol Data Unit). The existing tools have focused on providing an SMS gateway and on helping to view and edit a single SMS PDU, which rarely help to resolve the compliance test problem. The proposed compliance test method is based on an automatic generation of SMS PDUs fully compliant with the 3GPP specification by using QuickCheck library written in Haskell. By applying the proposed method to a C-based SMS encoder and decoder in Linux Mobile platform, we have found out several critical bugs such as wrong interpretation of time stamps in BCD format. The automatic SMS PDU generator is reusable in that it only depends on the 3GPP SMS specification. The QuickCheck library is also applicable for testing other network protocol data encoders and decoders, as used in this paper.

  • In security layer in the CDMA2000 1x EV-DO, a standard - C.S0024-a v2.0 is being accomplished under the project of 3GPP2(3rd Generation Partnership Project2). Therefore, a security device is needed to implement the security layer which is defined on the standard document for data transfer security between AT(Access Terminal) and AN(Access Network) on CDMA2000 1x EV-DO environment. This paper realizes the security layer system that can make safe and fast transfer of data between AT and AN. It could be applied to various platform environments by designing and implementing the Security Layer in the CDMA2000 1x EV-DO Security Layer to which applies C.S0024-A v2.0 of 3GPP2.

Effect of Grape Pomace Powder, Mangosteen Peel Powder and Monensin on Nutrient Digestibility, Rumen Fermentation, Nitrogen Balance and Microbial Protein Synthesis in Dairy Steers

  • This study was designed to investigate the effect of grape pomace powder (GPP), mangosteen peel powder (MPP) and monensin on feed intake, nutrients digestibility, microorganisms, rumen fermentation characteristic, microbial protein synthesis and nitrogen balance in dairy steers. Four, rumen fistulated dairy steers with initial body weight (BW) of $220{\pm}15kg$ were randomly assigned according to a $4{\times}4$ Latin square design to receive four treatments. The treatments were as follows: T1 = control, T2 = supplementation with monensin at 33 mg/kg diet, T3 = supplementation with GPP at 2% of dry matter intake, and T4 = supplementation with MPP at 30 g/kg diet. The steers were offered the concentrate diet at 0.2% BW and 3% urea treated rice straw (UTRS) was fed ad libitum. It was found that GPP supplemented group had higher UTRS intake and nutrient digestibility in terms of neutral detergent fiber and acid detergent fiber than those in control group (p<0.05). Ammonia nitrogen ($NH_3-N$) and blood urea-nitrogen concentration were higher in monensin, GPP and MPP supplemented groups (p<0.05). Total volatile fatty acids and propionate in the GPP group were higher than those in the control group (p<0.05) while acetate concentration, and acetate to propionate ratio were decreased (p<0.01) when steers were supplemented with GPP, monensin, and MPP, respectively. Moreover, protozoal populations in GPP, MPP, and monensin supplementation were significantly lower than those in the control group (p<0.05), while cellulolytic bacterial population was significantly higher in the control group (p<0.05). Nitrogen retention, microbial crude protein and efficiency of microbial nitrogen synthesis were found significantly higher in steers that received GPP (p<0.05). Based on this study it could be concluded that the GPP has potential as an alternative feed supplement in concentrate diets which can result in improved rumen fermentation efficiency, digestibility and microbial protein synthesis in steers fed on treated rice straw.
