Reduction of Body Weight by Capsaicin is Associated with Inhibition of Glycerol-3-Phosphate Dehydrogenase Activity and Stimulation of Uncoupling Protein 2 mRNA Expression in Diet-induced Obese Rats

  • Capsaicin is a pungent component of red pepper, which is widely consumed as food adjuncts. The present study was performed to investigate anti-obesity effects of capsaicin in diet-induced obese rats. Male Sprague-Dawley rats (n=14) were fed with a high-fat diet (Control) or high-fat diet containing 0.016% capsaicin (w/w) (Capsaicin) for 8 weeks. The final body weight and the mass of white adipose tissue were significantly lower in capsaicin supplemented group compared to control. Dietary capsaicin ameliorated lipid profiles with decrease in the plasma concentrations of triglycerides and total cholesterol, and decrease in the levels of total lipids and triglycerides in the liver. Activity of glycerol-3-phosphate dehydrogenase (GPDH), an indicator of triglyceride biosynthesis in white adipose tissue, decreased by 35% in the group supplemented with capsaicin. However, consumption of capsaicin increased the expression of uncoupling protein 2 (UCP2) in white adipose tissue, which is related to energy consumption. Our data suggests that capsaicin may reduce body weight and fat accumulation in high fat diet-induced obese rats. These effects may be mediated, at least partially, by the upregulation of UCP2 gene expression and its ability to inhibit GPDH activity.

Inhibitory Effects of Serotonin Derivatives on Adipogenesis (홍화씨 추출물 유래 세로토닌 유도체의 지방전구세포 분화억제 효능에 대한 연구)

  • N-feruloylserotonin (FS) and N-(pcoumaroyl) serotonin (CS), serotonin derivatives, which have been isolated as major and unique phenolics of safflower seed extract (SSE), are member of hydroxycinnamic acid amides and are implicated in the defense against pathogen infection and insect feeding. In this study, we evaluate inhibitory effects of N-(p-Coumaroyl)serotonin and N-Feruloylserotonin on adipogenesis using oil-red O staining, triglyceride and GPDH activity. we found that while serotonin itself did not suppress differentiation of preadipocytes into adipocytes, N-(p-Coumaroyl)serotonin and N-Feruloylserotonin inhibited the differentiation of preadipocytes into adipocytes in a concentration-dependent manner. In addition, they showed antioxidant effects in DPPH assay. Taken together, these results show that N-feruloylserotonin (FS) and N-(pcoumaroyl) serotonin (CS) suppress differentiation of preadipocytes, suggesting the possibility that these serotonin derivatives can be utilized as an anti-obesity agent.

Comparison between Single and Co-culture of Adipocyte and Muscle Cell Lines in Cell Morphology and Cytosolic Substances (지방과 근육 세포주의 단독 및 공동배양을 통한 세포형태학 및 세포물질 비교 연구)

  • Present study was performed to investigate the effect of single and co-culture of adipocyte and muscle cell lines on cell differentiation. 3T3-L1 (adipocyte) and L6 (muscle) cell lines were single-cultured on the condition of 10% fetal bovine serum (FBS)/Dulbeco's modified eagle's medium (DMEM) for 48 h followed by culture within 5% FBS/DMEM as a growth media. Then, the growth media was replaced by differentiation media composed of 2% FBS/DMEM without additives in single- or co-culture of the 3T3-L1 and the L6 cells to induce differentiation of both cell types. In co-culture system, the 3T3-L1 and the L6 cells were grown in separated places by being seeded on a $0.4{\mu}m$ insert membrane and on the bottom of 6 well plate, respectively. Cell differentiation was measured using morphological investigation and cytosolic analysis of glycerol-3-phosphate dehydrogenase (GPDH; for 3T3-L1) and creatine kinase (CK; for L6). Based on the GPDH results, the presence of L6 cells did not stimulate 3T3-L1 differentiation showing more differentiation of 3T3-L1 cells in the single-culture compared to the co-culture condition. In contrast, 3T3-L1 cells in the co-culture promoted differentiation of L6 cells. Enzymatic analysis supported this result showing that 3T3-L1 cells showed statistically (P<0.05) higher GPDH activity in the single-culture than the co-culture, whereas CK results of L6 cells were vice versa (P<0.05). Overall, present results may indicate that co-culture system is more reliable and precise technique compared to single-culture. Further studies on several co-culture trials including different media conditions, supplementation of differentiating substances, molecular biological analysis, etc. should be required to obtain practical and fundamental mass data.

  • Objective: The purpose of this study is to investigate the effects of Ganoderma lucidum herba pharmacopuncture(GHP) on the adipogenesis in 3T3-L1 preadipocytes. Methods: 3T3- L1 preadipocytes were differentiated with adipogenic reagents by incubating for 2 days in the absence or presence of GHP ranging from 1 and 2%. The effect of GHP on cell proliferation of 3T3-L1 preadipocytes was investigated using MTT assay. The effect of GHP on adipogenesis was examined by Oil red O staining and measuring glycerol-3-phosphate dehydrogenase (GPDH) and intracellular triglyceride (TG) content. Results: Following results were obtained from the preadipocyte proliferation and adipocyte differentiation of 3T3-L1. We observed no effect of GHP on preadipocyte proliferation. GHP inhibited adipogenesis, the activity of GPDH and accumulation of intracellular TG content. Conclusions: These results suggest that GHP inhibit differentiation of preadipocyte.

The Effects of Polymannuronates on Leptin in 3T3-L1 Adipocytes (3T3-L1 지방세포 내의 Leptin에 미치는 Polymannuronates의 영향)

  • This study evaluated polymanuronates on the differentiation of 3T3-L1 adipocytes. Polymannuronates increased glucose utilization and reduced the accumulation of triglycerides in the cells. The differentiation showed the same results as Oil red O staining. Also, the polymannuronates inhibited GPDH activity as a result of the restrained adipogenesis promotion process in 3T3-L1 adipocytes. The addition of the differentiation promotion factor to 3T3-L1 promoted the differentiation of adipocytes and increased the expression of the leptin level. However the addition of polymannuronates inhibited differentiation of adipocytes and the leptin secretion level in cells by checking the leptin protein level in the culture media. As well as this, it also inhibited the transcriptional mechanism and leptin mRNA expression. These results suggest that the addition of polymannuronates improves the physiological function of 3T3-L1 cells by reducing the accumulation of triglyceride and GPDH activity, and the repressing expression of leptin at a molecular level.

Comparison of Diglyceride, Conjugated Linoleic Acid, and Diglyceride-Conjugated Linoleic Acid on Proliferation and Differentiation of 3T3-L1 Preadipocytes

  • Conjugated linoleic acid (CLA) reduces fat deposition in several mammalian species. The proposed mechanisms for this effect are reduced preadipocyte proliferation and differentiation. The objective of this study was to investigate the inhibitory effects of diglyceride (DG), CLA, DG-CLA of proliferation and differentiation of 3T3-L1 preadipocytes. Cell viability was determined using WST-8 analysis and cell differentiation was determined by glycerol-3-phosphate dehydrogenase (GPDH) activity. Lipid accumulation in differentiating 3T3-L1 cells was measured by Oil red O staining. The proliferation of preconfluent 3T3-L1 cells by treatments of DG, CLA, and DG-CLA was reduced in a dose-dependent manner. CLA among them was the most effective in reduction of viable cells with increasing concentrations. Treatments of the DG, CLA, and DG-CLA at the concentration of $100{\cdot}\ddot{I}g/ml$ for 48h significantly inhibited differentiation of 3T3-L1 cells (p<0.05). In addition. cytoplasmic lipid accumulation during differentiation of the 3T3-L1 preadipocytes was also inhibited by treatments of the test solutions. DG-CLA was the most effective in the inhibition of differentiation and lipid accumulation in 3T3-L1 cells. These results indicate that the DG including CLA as fatty acids is more effective for anti-obesity than DG or CLA alone and that consumption of DG-CLA as a dietary oil may give a benefit for controlling overweight in humans.

Effects of Yeoldahanso-tang, a Sasang Constitutional Herbal Prescription for Taeeumin, on the Adipogenesis in 3T3-L1 Cells (태음인(太陰人) 열다한소탕(熱多寒少湯)이 3T3-L1 지방전구세포(脂肪前驅細胞) 분화(分化)에 미치는 영향(影響))

  • Objectives : Although Yeoldahanso-tang (YDHST) has been widely used for treatment of obesity and its related diseases such as hyperinsulinemia and hypertension for Taeeumin, no scientific evidence has reported yet to support its ability to work against these metabolic disorders. Our study was aimed to investigate the anti-obesity effect of YDHST extract on the cellular differentiation of 3T3-L1 preadipocytes into adipocytes. Methods : 3T3-L1 preadipocytes were differentiated into adipocytes by adding insulin, dexamethasone and 3-isobutyl-1-methylxanthine (IBMX) for 8 days in the absence or presence of YDHST extract. Anti-obesity effects of YDHST extract were evaluated by Oil Red O staining, glycerol-3-phosphate dehydrogenase (GPDH) activity, triglyceride contents, and leptin production. Results : YDHST extract remarkably prevented lipid accumulation with no cytotoxicity in the differentiated 3T3-L1 cells. In addition, YDHST extract decreased contents of triglyceride 3T3-L1 adipocytes. Consistently, YDHST extract caused a significant inhibition of GPDH activity and leptin production in a dose-dependent manner. Conclusions : Our findings suggest that Sasang constitutional herbal formula YDHST for Tae-eumin has anti-obesity activity by regulation of the adipogenesis process in vitro. Additional study will be required to further confirm the inhibitory effect on adipocyte differentiation by using in vivo animal model.

The Effect of Crataegi Fructus Pharmacopuncture on Adipocyte Metabolism (산사약침이 지방세포 대사에 미치는 영향)

  • Objectives The purpose of this study is to investigate the effects of Crataegi Fructus Pharmacopuncture(CFP) on the adipogenesis in 3T3-L1 cells, lipolysis in rat epididymal adipocytes and histological changes in porcine adipose tissue. Methods Inhibiton of preadipocyte differentiation and/or stimulation of lipolysis play important roles in reducing obesity. 3T3-L1 preadipocytes were differentiated with adipogenic reagents by incubating for 3days in the absence or presence of CFP ranging from 0.01 to 1mg/mL. The effect of CFP on adipogenesis was examined by measuring GPDH activity and by Oil Red O staining. Mature adipocytes from rat epididymal fat pad was incubated with CFP ranging from 0.01 to 1mg/mL for 3 hrs. The effect of CFP on lipolysis was examined by measuring free glycerol released. Fat tissue from pig skin was injected with CFP ranging from 0.1 to 10mg/mL to examine the effect of CFP on histological changes under light microscopy. Results The following results were obtained from present study on adipogenesis of preadipocytes, lipolysis of adipocytes and histological changes in fat tissue. 1. Crataegi Fructus Pharmacopuncture inhibited adipogenic differentiation at the concentration of 1.0mg/mL. 2. Crataegi Fructus Pharmacopuncture decreased the activity of glycerol-3-phosphate dehydrogenase(GPDH) at the concentration of 0.1mg/mL. 3. Crataegi Fructus Pharmacopuncture ok. lipolysis at the concentration of 0.1mg/ml. 4. Crataegi Fructus Pharmacopuncture ranging 0.1 to 10mg/mL failed to exert lysis of cell membrane in porcine fat tissue. Conclusions These results suggest that Crataegi Fructus Pharmacopuncture at relatively high concentration inhibited adipogenesis and increased lipolysis of adipocytes. However, Crataegi Fructus Pharmacopuncture didn't exert any effect on lysis of cell membrane in fat tissue.

Soluble extract of soybean fermented with Aspergillus oryzae GB107 inhibits fat accumulation in cultured 3T3-L1 adipocytes

  • BACKGROUND/OBJECTIVES: This study was conducted to investigate the effects of fermented soybean (FS) extract on adipocyte differentiation and fat accumulation using cultured 3T3-L1 adipocytes. MATERIALS/METHODS: 3T3-L1 adipocytes were treated with FS and nonfermented soybean (NFS) extract during differentiation for 10 days in vitro. Oil red O staining was performed and glycerol-3-phosphate dehydrogenase (GPDH) activity was measured for analysis of fat accumulation. Expressions of adipogenic genes were measured. RESULTS: Soluble extract of soybean fermented with Aspergillus oryzae GB107 contained higher levels of low-molecular-weight protein than conventional soybean protein did. FS extract ($50{\mu}g/ml$) inhibited adipocyte differentiation and fat accumulation during differentiation of 3T3-L1 preadipocytes for 10 days in vitro. Significantly lower GPDH activity was observed in differentiated adipocytes treated with the FS extract than those treated with NFS extract. Treatment with FS extract resulted in decreased expression levels of leptin, adiponectin, and adipogenin genes, which are associated with adipogenesis. CONCLUSIONS: This report is the first to demonstrate that the water-soluble extract from FS inhibits fat accumulation and lipid storage in 3T3-L1 adipocytes. Thus, the soybean extract fermented with A. oryzae GB107 could be used to control lipid accumulation in adipocytes.

Effects of Vitamins on the Differentiation of Preadipocytes from Hanwoo Cattle Adipose Tissues

  • The experiment was conducted to study the effects of water soluble vitamins and retinoic acid on the differentiation of preadipocyte from omental, subcutaneous, intermuscular and intramuscular adipose tissue of Hanwoo. Differentiation was assessed by the change in enzyme activity, glycerol-3 phosphate dehydrogenase in serum free cell culture system. Preadipocytes treated with biotin ($10{\mu}M$) and pantothenic acid ($100{\mu}M$) were significantly (p<0.05) less differentiated than those from the control in all adipose tissue depots except intramuscular tissue. Although there was no significance, vitamin C was shown to stimulate the adipocyte conversion in omental and subcutaneous, but not in intermuscular and intramuscular adipose tissues. Lower values of GPDH activity in intermuscular preadipocyte were interpreted to be caused by relatively higher amounts of protein. In this experiment vitamin C did not stimulate fat deposition in intramuscular adipose tissue but further experiments are needed on the role of vitamin C in preadipocyte differentiation. When treated with different levels of retinoic acids, differentiation of preadipocytes was significantly (p<0.05) reduced from the level of $0.5{\mu}g/ml$ in omental and intermuscular, from $50{\mu}g/ml$ in subcutaneous, and in intramuscular at $500{\mu}g/ml$, thus showing that intramuscular preadipocytes were least responsive to retinoic acid in differentiation. All-trans retinoic acid appeared to inhibit the differentiation in a dose dependent manner, regardless of adipose tissues type.
