Electrochemical Immunosensing of GOx-labeled CRP Antigen on Capture Antibody Monolayer Immobilized on Calixcrown-5 SAMs

  • Insulating effects on Au electrode according to the thickness and density of coated materials are well-known. To do electrochemical immunoassay reproducibly the glod electrode would be coated with self-assembled monolayers and antobodies. To get reproducibility, the antobody monolayer should be packed at highest density so that the amount of immobilized antibody at defined area should be the same. The calix[4]crown-5 SAMs could provide the basis for the antibodies to be immobilized reproducibly and at highest density. But the insulating effect would be highest too. We proved that the compactly packed protein monolayers on SAMs inhibited the electron transfer by block the free shuttling of redox molecules. The inhibition was minimized by inserting redox molecules in between the proteins during immobilization process. In this paper, we demonstrated that the calix[4]crown-5 SAMs would provide the protein monolayers with highest density and new method to minimize the insulating effect by inserted redox molecules in between the compactly packed protein monolayers.

Quantification of Genetically Modified Canola GT73 Using TaqMan Real-Time PCR

  • Event-specific PCR detection methods are the primary trend in genetically modified (GM) plant detection owing to their high specificity based on the flanking sequence of the exogenous integrant. Therefore, this study describes a real-time PCR system for event-specific GM canola GT73, consisting of a set of primers, TaqMan probe, and single target standard plasmid. For the specific detection of GT73 canola, the 3'-integration junction sequence between the host plant DNA and the integrated specific border was targeted. To validate the proposed method, test samples of 0, 1, 3, 5, and 10% GT73 canola were quantified. The method was also assayed with 15 different plants, and no amplification signal was observed in a real-time PCR assay with any of the species tested, other than GT73 canola.

하이브리드 로켓 추진 시스템의 혼합 연소실 적용에 따른 Trade-off 평가 (Trade-off Evaluation due to Application of Mixing Chamber for Hybrid Rocket-Propulsion System)

  • The intermediate mixing chamber is one of various methods for improving the regression rate and combustion efficiency of the hybrid rocket. The mixing chamber with its non-combustible material makes the propulsion performance increase, but it leads to a low fuel-loading density in the combustion chamber; therefore, this performance-related trade-off between the mixing chamber and the low fuel-loading density was studied. In this study, the trade-off was conducted by comparing the intermediate-mixing-chamber case with a w/o-mixing-chamber case. The small hybrid-sounding rocket is designed with internal ballistics for comparing the rocket length to the weight. In addition, an external ballistic analysis was conducted for comparing the performances of the w/- and w/o-mixing-chamber cases. As a result, the intermediate-mixing-chamber case shows that the length and the weight were decreased to 12 % and 8 %, respectively; furthermore, when compared with the normal cases, the estimated altitude result of the w/-mixing-chamber case was increased to approximately 75 m.

  • 일반적으로 하이브리드 연소를 모델링 할 경우 고체 연료의 표면 온도를 이용하여 후퇴율을 계산하기 때문에 정확하게 고체연료의 표면온도를 예측하는 것이 필요하다. 따라서 본 연구는 하이브리드 고체 연료에 열전대를 삽입한 후, 연소실험을 통해 연료의 표면 온도를 측정하였고, 본 연구에서의 산화제 유속 범위에서의 고체 연료 표면 온도 변화를 고찰하였다.

Improved Sensitivity of a Glucose Sensor by Encapsulation of Free GOx in Conducting Polymer Micropillar Structure

  • A simple process of fabricating micropillar structure and its influence upon enhancing electrochemical biosensor response were studied in this work. Conducting polymer PEDOT was used as a base material in formulating a composite with PVA. Micro porous PC membrane filter was used as a template for the micropillar of the composite on ITO electrode. This structure could provide plenty of encapsulating space for enzyme species. After dosing enzyme solution into this space, Nafion film tent was cast over the pillar structure to complete the micropillar cavity structure. In this way, the encapsulation of enzyme could be accomplished without any chemical modification. The amount of enzyme species was easily controllable by varying the concentration of the dosing solution. The more amount of enzyme is stored in the sensor, the higher the electrochemical response is produced. One more reason for the sensitivity improvement comes from the large surface area of the micropillar structure. Application of 0.7 V produced the best current response under the condition of pH 7.4. This biosensor showed linear response to the glucose in 0.1~1 mM range with the average sensitivity of $14.06{\mu}A/mMcm^2$. Detection limit was 0.01 mM based on S/N = 3.

Polypyrrole-Glucose Oxidase 효소전극의 배위자 크기에 따른 전기화학적 특성 (Electrochemical Properties of Polypyrrole-Glucose Oxidase Enzyme Electrode with Different Dopants)

  • We synthesized polypyrrole (PPy) by electrolysis of the pyrrole monomer solution containing support electrolyte, KCl and/or p-toluene sulfonic acid sodium salt (p-TS). The electrochemical behavior, was investigated using cyclic voltammetry and AC impedance. In the case of using electrolyte p-TS, the oxidation potential of the PPy was about -02 V vs Ag/AgCl reference electrode, while the potential was about 0 V for using electrolyte KCl. The falloff of the oxidation potential gave a sign of an improvement in the electron hopoing mechanism on the backbone. The AC impedance plot gave a hint of betterment of mass transport. PPy doped with p-TS improved in mass transport or diffusion. That was because the PPy doped with p-TS was more porous than PPy with KCl. We attained an effect of good kinetic parameters, in the case of PP-GOx enzyme electrodes doped with p-TS, which were determined by 58 mmol dm$\^$-3/ for apparent Michaelis constant and by 581 ㎂ for maximum current respectively.

  • 정확한 산화제 유량조절은 하이브리드 로켓의 추력제어에 매우 중요하다. 산화제 유량제어를 위해 스텝모터와 니들밸브를 결합하여 장치하고 Labview 프로그램으로 제어하는 산화제 유량제어 장치를 설계하였다. 하이브리드 로켓 연소실험에 사용한 산화제는 기체산소를 사용하였고, 추진제로는 PolyCarbonate, PolyEthylene PMMA를 사용하였다. 본 연구에서는 초기 추력제어 실험에서 발생한 추력 섭동(Oscillation)을 감쇠시키기 위한 연구로 추력제어 실험에서 공급되는 산화제 배관 유속의 변화를 통해 발생되는 추력 섭동의 원인을 분석하였으며, 추진제 종류에 따라 달라지는 안정적인 제어 조건을 찾기 위한 연구를 수행하였다.

하이브리드 로켓 클러스터링 모델의 동시 점화 및 유량 분배 연구 (A Study on the Simultaneous Ignition and Flow Distribution of Hybrid Rocket Clustering Model)

  • 본 연구는 달착륙선을 위한 하이브리드 추진 시스템의 기초 연구로서 동시 점화 및 산화제 유량 분배를 통한 클러스터링 연구를 수행하였다. 클러스터링 연소 실험을 위해 지상 연소 실험 시스템을 구축하였으며, 산화제와 연료는 각각 기체 산소(gas oxygen)와 고밀도 폴리에틸렌(High Density Poly Ethylene)을 사용하였다. 실험 결과 화약 점화를 사용한 동시 점화 장치는 동일한 시점에 점화가 되었으며, 유량 분배 장치를 통해 비교적 균일하게 산화제 분배가 이루어져 각 연소실 내에서의 압력이 비교적 동일하게 나타난 것을 확인하였다.

A Solution towards Eliminating Transaction Malleability in Bitcoin

  • Bitcoin is a decentralized crypto-currency, which is based on the peer-to-peer network, and was introduced by Satoshi Nakamoto in 2008. Bitcoin transactions are written by using a scripting language. The hash value of a transaction's script is used to identify the transaction over the network. In February 2014, a Bitcoin exchange company, Mt. Gox, claimed that they had lost hundreds of millions US dollars worth of Bitcoins in an attack known as transaction malleability. Although known about since 2011, this was the first known attack that resulted in a company loosing multi-millions of US dollars in Bitcoins. Our reason for writing this paper is to understand Bitcoin transaction malleability and to propose an efficient solution. Our solution is a softfork (i.e., it can be gradually implemented). Towards the end of the paper we present a detailed analysis of our scheme with respect to various transaction malleability-based attack scenarios to show that our simple solution can prevent future incidents involving transaction malleability from occurring. We compare our scheme with existing approaches and present an analysis regarding the computational cost and storage requirements of our proposed solution, which shows the feasibility of our proposed scheme.

Biosensing interfaces based on the dendrimer-underlying layer on gold

  • Structually organized mono- and multilayers were developed on gold for the catalytic and affinity biosensing using hyper-branched dendrimers. For the catalytic biosensing interface, a new approach to construct a multilayered enzyme film on the electrode surface was developed. The film was prepared by layer-by-layer depositions of dendrimers and periodate-oxidized glucose oxidase. The voltammograms obtained from the GOx/dendrimer multilayered electrodes revealed that bioelectrocatalytic response is directly correlated to the number of deposited bilayers. From the analysis of voltammetric and ellipsometric signals, the coverage of active enzyme per layer during the layering steps was estimated, demonstrating the spatially-ordered multilayer formation. As an extension of the study, dendrimers having various degrees of ferrocenyl modification were prepared and used. The resulting electrodes were electrochemically characterized, and the density of ferrocenyl groups, active enzyme coverage, and sensitivity were estimated. For the affinity-sensing surrface, a biosensor system based on avidin-biotin interaction was developed. As the building block of affinity monolayer, G4 dendrimer having partial ferrocenyl-tethered surface groups was prepared and used. And the biotinylated and electroactive dendritic monolayer was used for the affinity-sensing surface interacting with avidin. Electrochemical characterization of the resulting biosensor was conducted using free enzyme in electrolyte in terms of degree of surface coverage with avidin and subsequent surface shielding.
