Docosahexaenoic acid (DHA) rich oil was obtained from blue fin tuna (Thunnus thynnus orientalis) orbital tissue with centrifugation of 12,000 rpm under vaccum $(10^{-1}\;Torr)\;at\;4^{\circ}C$. The effect of DHA rich oil (DHA content; $27.8\%$) on $CCl_4-induced$ acute injury was investigated biochemically and histopathologically. Dosage of DHA rich oil on 24h before $ CCl_4-administration$ prevented significantly the increase of glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GTP) values. No necrosis of hepatocytes was observed in rat livers treated with DHA oil on 24h prior to $CCl_4-administration$. These results suggested that DHA oil controls the accumulation of fat in the liver and prevented the liver injury.
Obesity, a state of having excessive body fat enough to attack one's health, is most blamed for causing chronical degenerative illness such as hypertension, diabetes mellitus as well as arteriosclerosis. According to many studies, the program for the weight control was the most demanded at the worksites. This study was designed to investigate the correlation between the degree of obesity and blood parameters of the employees at the worksites. The results of periodic medical examination for 571 employees(male: 418, female: 153) were analyzed; Age, BMI, systolic blood pressure(SBP), diastolic blood pressure(DBP), hemoglobin(Hb), fasting blood sugar(FBS), total-cholesterol(TC), serum glutamic oxaloacetic transaminase (sGOT), serum glutamic pyruvic transaminase(sGPT) and $gamma$-glutamyl transpeptidase($gamma$-GTP). Pearson correlation coefficient was used to find out the correlation between BMI and each blood parameters. In the case of male, 27.3% of the subjects were over-weight. The subject's age and their blood levels of uric acid, FBS, sGOT and sGPT did not show any significant difference regardless of BMI's, while SBP, DBP, Hb, TC and $gamma$-GTP levels of the overweight group were significantly higher than those of normal group. In the case of female, 9.2% of the subjects were over-weight. This study shows that the implementation of weight control program is required at the worksites
The present study was conducted to investigate the effects of dietary supplemental dry powder of chinese medicine refuse (CMR) on the growth performance and physiological status of broiler chicks. The treatments consisted of corn-soybean meal control diet, CMR 4 and 8% diets. A total of 120 Arbor Acre broiler chicks were randomly allotted to 12 pens ; four pens per treatment and 10 birds per pen. The body weight gain and feed intake did not show ant differences among the treatment groups, but feed conversion was significantly higher (P〈0.05) in CMR 8% group than that of the control group. Hunt L and b did not show ant differences among the treatment groups, but hunt a was darker (P〈0.05) in CMR 4 and 8% group than that of the control group. The blood concentrations of total protein, sugar, glutamic-oxaloacetic-transaminase (GOT), glutamic-pyruvic-transaminase (GPT) Hemogloben were not different among the treatment groups. The pH of thigh muscle were lower in CMR 4 and 8% groups than that of the control group, but crude fat, crude ash of thigh muscle and bone hardness in brolier chicks did not show ant differences among the treatment groups. The saturated fatty acid content of the thigh muscle tende to decrease dietary CMR increased.
Objective : To investigate the effects of Artemisiae Capillaris Fructus on liver injury induced $CCl_4$. Method : Carbon tetrachloride-induced hepatic injury in $CCl_4$,-treated rats caused changes in the glutamic oxaloacetic transaminase(GOT), glutamic pyruvic transaminase(GPT) and alkaline phosphatase(ALP) activities in the serum. Result : The Methanol extract of the Artemisiae Capillaris Fructus was found to protect the rats against such changes significantly. Protective activities were shown to be present in ether-soluble and buthanol-soluble fractions prepared from the above methanol extract. Conclusion : we have examined the detoxyfying effect, one of several medicinal effects of Artemisiae Capillaris Fructus. on liver disorder experimentally induced by $CCl_4$. as an animal model of liver disorder.
This study was conducted to investigate the effects of mugwort extracts on the blood ethanol concentration and liver function in rats. Sprague-Dawley rats were used, the rats administered with 25% ethanol (5g/kg$.$B.W.) were devided into three groups (CON-E ; 25% ethanol administered to the CON-E) according to the administered ethanol concentration and the levels of administered mugwonts. Mugwont roots extracts were administered via the caudal vein. Ethanol concentration was measured at the time of 0, 1, 2 and 3hr by gas chromatography. GOT(Glutamic Oxaloacetic Transaminase) and GPT(Glutamic Pyruvic Transaminase) were measured at the time of 0 and 5hr. Components of each extracts were analyzed by using high performance liquid chromatography. Cell number, GOT and GPT were investigated by using rat hepatocyte culture. Megwort extracts were added at the levels of 1% or 2%. Hepatocyte culture were into five groups according to the addition levels. The results were summarized as follows ; 1. Catechin contents of 8∼10mg/100g and the contents of (-)-epigallocatechin was high in the water extracts. 2. Ethanol degradation efficiency declines in the following order : MDW-E>MOH-E>CON-E. 3. The numbers of rat hepatocytes declines in the following order : 2% MDW-L>1%MDW-L>1%MOH-L>CON-L>2%MOH-L. These results suggest that crude catechin of mugwort extracts may play important roles to degrade ethanol and recover liver function in rats.
Effects of acupuncture at Taekjung and Ungyo($GV_{28}$) bloodletting on serum glutamic oxaloacetic transaminase (GOT) activity, glutamic pyruvic transaminase activity(GPT), total Protein, Albumin, Alkalin Phosphatase(ALP), ${\gamma}$-GT, lactic dehydrogenase (LDH), total cholesterol and triglyceride were determined. 1) In the Ungyo bloodletting group, GOT, GPT, ALP, ${\gamma}$-GT and LDH activity showed a tendency to decrease compared to those of $CCl_4$ only group and Taekjung acupuncture group. 2) Serum albumin content showed a tendency decrease by treatment of $CCl_4$ and to increase by treatment of Ungyo bloodletting. 3) The values of serum total protein, total cholesterol and triglyceride showed no significant difference among all the treatment groups. 4) All the biological values showed no significant difference between the $CCl_4$ only group and Taekjung acupuncture group except the values of albumin. Results from this study indicate that the Ungyo bloodletting showed the possibility of liver function recovery in liver-harmed rats by $CCl_4$.
The effects of Ganoderma Lucidum consumption (GL) on the liver function and lipid metabolic disorder induced by alcohol consuming were studied in rats using GL powder(1%, 2%, 4%) and 10% ethly alcohol. Thirty-five rats(Sprague-Dawley, male) were divided into five groups and fed experimental diets for seven weeks. The concentrations of serum cholesterol, HDL-cholesterol and triglyceride were the highest in the alcohol group, while the levels were apparently reduced by in groups fed GL. The atherogenic indices(AI) also tended to be higher in the alcohol group. Glutamic oxaloacetic transaminase(GOT) activities were significantly lower in body alcohol + 2% GL and alcohol + 4% GL groups, and glutamic pyruvic transaminase(GPT)activities lower only in the alcohol + 2% GL compared with the alcohol group. Alkaline phosphatase(ALP) activities were not significantly different among the groups. The elevated levels of liver cholesterol and triglyceride due to alcohol consumption, were apparently reduced by GL supplementation. On microscopic observation of liver tissues, fat droplets appeared extensively on the liver-lobule in the alcohol group, while fat droplets appeared only on the central vein in the alcohol + 1% GL group. However, in groups administered 2% and 4% GL, fat droplets appeared similar to the alcohol group. The concentrations of fecal bile acid were significantly increased in groups fed GL. These results indicate that GL exerted some beneficial effects on lipid metabolic disorder caused by long-term alcohol consumption. Thus, it seems that GL may be effective in preventing or curing some aspects of alcohol toxicity. More detailed studies are needed in order to determine proper dietary levels of GL for combating alcohol toxicity.
이 연구는 설치류에서 양파추출물의 효능을 평가하기 위하여 실시하였다. 실험동물은 대조군(control)과 양파추출물 투여군(ACE)으로 나누어서 진행하였다. ACE그룹은 대조군에 비해 적혈구(RBC), 헤모글로빈(HGB), 헤마토크릿(Hct) 수준이 약간 증가하는 것을 보였다(p<0.05). 반면, 헤모글로빈, 단핵구, 림프구 및 호중구는 대조군과 비교하였을 경우 유의미한 변화(p<0.05)가 없었다. GOT (glutamic oxaloacetic transaminase) 및 GPT (glutamic pyruvic transaminase) 수준을 분석한 결과 대조군에 비해 ACE그룹이 유의적으로 감소하였다(p<0.05). 혈당, 총단백질, HDL-콜레스테롤은 ACE 그룹에서 약간 높았고, 트리글리세라이드, 총콜레스테롤 수치는 대조군에 비해 ACE그룹에서 더 낮았다(p<0.05). 간 조직과 혈액의 면역과 염증에 관련된 사이토카인(인터루킨 1알파(IL-1α), 인터루킨 1베타(IL-1β), 종양괴사인자알파(TNF-α), 인터루킨 6(IL-6), 인터루킨 10(IL-10), 인터페론감마(INF-γ), 인터루킨 18(IL-18), 인터루킨 2(IL-2), 과립구 대식구 집락자극인자(GM-CSF)) 수준은 모두 정상 범위 내에 있는 것으로 확인되었다. 본 연구결과는 고농축 식이성 양파추출물은 혈액학적 지표와 면역 기능에 독성이 없다는 것을 보여주는 기초데이터로 활용할 수 있을 것이다.
산화된 사료에 첨가한 비타민 E, C 와 glycyrrhizin이 나일 틸라피아의 성장 및 Edwardsiella tarda 감염 시 저항력에 미치는 영향을 알아 보기 위해 thiobarbitric acid (TBA) 값이 $80\~88$ mg/kg 사료만을 단독으로 또는 이 사료 100 g 당 비타민 E (50 mg), C (60 mg)과 glycyrrhizin ($25\~200$ mg) 등을 각기 또는 상호 혼합하여 7 주간 사육하였다. 사육후 성장을 조사한 결과 비타민 E, C 및 glycyrrhizin 등을 각기 또는 혼합한 실험군이 산화된 사료만을 투여한 대조군에 비해 성장율이 좋았으며, 혈청 내의 glutamic oxaloacetic transaminase (GOT)와 glutamic pyruvic transaminase (GPT) 값은 산화된 사료를 투여한 실험군에서만 매우 높게 나타났다. 질병원인 E. tarda를 인위적으로 감염시켰을 때 저항력에 있어서도 glycyrrhizin 25 mg 투여군을 제외한 비타민 E, C 및 glycyrrhizin 등을 투여한 실험군이 산화된 사료만을 투여한 대조군에 비해 생존율이 현저히 높았다. 이상의 결과를 종합해 볼 때 산화된 사료만을 나일 틸라피아에 투여했을 때 성장 및 간의 활성도가 떨어지는 반면, 비타민 E, C 및 glycyrrhizin 등을 섞어서 투여했을때 성장 및 간의 활성도가 증가되었을 뿐만 아니라 E. tarda에 대한 저항력도 증가되었음을 알 수 있었다.
Objectives : Effects of Kyungohkgo($Qi{\acute{o}}ng\;y{\grave{u}}\;g{\grave{a}}o$) on intellectual development and learning ability were investigated growth and intellectual impairment rat with insufficient nutrition diet. Methods : We divided male Sprague-Dawley rats into 4 groups(A, B, C, D). They were normal group(A), growth deficiency rat with insufficient nutrition diet group(B), growth deficiency rat with 0.1% Kyungohkgo($Qi{\acute{o}}ng\;y{\grave{u}}\;g{\grave{a}}o$) group(C) and growth deficiency rat with 0.2% Kyungohkgo($Qi{\acute{o}}ng\;y{\grave{u}}\;g{\grave{a}}o$) group(D). They were administered for 5 weeks. We measured body weight, serum growth hormone, insulin-like growth factor and thyroid stimulating hormone, RBC, concentration of Hb and PCV ratio, total WBC and its composition, the values of plasma glutamic oxaloacetic transaminase(GOT) and glutamic pyruvic transaminase(GPT) activities and morris water maze test in escape distance, escape time and escape speed. Results : 1. Body weight showed a tendency to increase in the Kyungohkgo($Qi{\acute{o}}ng\;y{\grave{u}}\;g{\grave{a}}o$) groups and 0.2% Kyungohkgo($Qi{\acute{o}}ng\;y{\grave{u}}\;g{\grave{a}}o$) group showed significantly different than control groups. 2. Serum growth hormone, insulin-like growth factor and thyroid stimulating hormone showed a tendency to increase in Kyungohkgo($Qi{\acute{o}}ng\;y{\grave{u}}\;g{\grave{a}}o$) groups, however these values showed no significantly different. 3. About the counts of RBC, 0.1% and 0.2% Kyungohkgo($Qi{\acute{o}}ng\;y{\grave{u}}\;g{\grave{a}}o$) groups showed significantly different than control groups. Concentration of Hb was higher than control group in Kyungohkgo($Qi{\acute{o}}ng\;y{\grave{u}}\;g{\grave{a}}o$) groups. And 0.2% Kyungohkgo($Qi{\acute{o}}ng\;y{\grave{u}}\;g{\grave{a}}o$) group showed significantly different than control groups in PCV ratio. 4. The counts of total WBC and its composition showed no significantly different in all treatment groups. 5. The values of plasma glutamic oxaloacetic transaminase(GOT) and glutamic pyruvic transaminase(GPT) activities showed no significantly different in all treatment groups. 6. In the morris water maze test, in escape distance and escape speed, Kyungohkgo($Qi{\acute{o}}ng\;y{\grave{u}}\;g{\grave{a}}o$) groups showed no significantly different than control group. But Kyungohkgo($Qi{\acute{o}}ng\;y{\grave{u}}\;g{\grave{a}}o$) groups showed the increasing tendency. Conclusions : So Kyungohkgo($Qi{\acute{o}}ng\;y{\grave{u}}\;g{\grave{a}}o$) have an effect of promoting growth of rats and might be effect to treat various kinds of growth delay in children.
