連續的 溫熱曝露가 흰쥐의 血淸 Glutamic Oxaloacetic Transaminase 및 Glutamic Pyruvic Transaminase의 活性에 미치는 影響 (Effect of Prolonged Heat Exposure on Serum Glutamic Oxaloacetic and Glutamic Pyruvic Transaminase Activities of Rats)

  • 成熟한 Sprague-Dawley 系의 雄性흰쥐를 $30\\pm 0.5^\\circ C$에서 240時間 그리고 $33\\pm 0.5^\\circ C$에서 64時間 各各 溫熱曝露시켰으며, 溫熱曝露 期間中 여러 時間區에서 血淸 glutamic oxaloacetic transaminase (GOT)와 血淸 glutamic pyruvic transaminase (GPT)의 活性을 Reitman 및 Frankel (1957)의 方法에 依해서 Coleman-Model 295E Spectrophotometer로서 各各 550m$\\mu$에서 測定하였다. $23\\pm 1^\\circ C$인 對照群에 比較하여 보면, $30\\pm 0.5^\\circ C$ 및 $33\\pm 0.5^\\circ C$에서 溫熱曝露된 흰쥐는 各各 血淸의 GOT 및 GPT의 活性이 有意性있게 上昇되었다. 서로 다른 高溫의 溫熱에 曝露된 흰쥐의 血淸 GOT 活性은 16 및 30時間 區間中에서 또한 64 및 72時間 區間中에서 各各 有意性있게 上昇되었다. 한편 $30\\pm 0.5^\\circ C$에서 72時間後에는 血淸 GOT 活性은 正常群으로 되돌아가는 傾向이 나타났다. 서로 다른 高熱의 溫熱로 曝露된 흰쥐의 血淸GPTdml 活性은 初期인 4時間區에서 有意性있게 上昇되었으며, 이어서 16時間區와 64 및 72時間 區間中에서 各各 上昇되었다. 한편 $30\\pm 0.5^\\circ C$에서는 114時間後에 약간 增加하는 傾向이 나타났다. $30\\pm 0.5^\\circ C$와 $33\\pm 0.5^\\circ C$에서 各各 溫熱曝露된 흰쥐의 酵素活性을 比較하여 보면, $33\\pm 0.5^\\circ C$에 曝露된 흰쥐의 血淸 GOT 및 血淸 GPT의 活性은 各各 16, 30 및 64時間區에서 有意性있게 높은 活性을 나타내었다. 이러한 結果로 미루어 보아 連續的 溫熱曝露는 非正常의 아미노基 轉移代謝를 招來하는 것으로 思料된다.

低氣壓이 흰쥐의 血淸 Glutamic Oxaloacetic Transaminase 및 Lactic Dehydrogenase 活性에 미치는 影響 (Effect of Low Atmospheric Pressure on Serum Glutamic Oxaloacetic Transaminase and Lactic Dehydrogenase Activities of Rats)

  • 成熟한 Sprague-Dawley 系 雄性 흰쥐를 對照群(760 mmHg)과 低氣壓群인 500 mmHg 와 380 mmHg의 兩實驗群으로 나누어 15日間(1日當 1時間) 曝露시켜, 低氣壓이 血淸 glutamic oxaloacetic transaminase(GOT) 및 血淸 lactic dehydrogenase(LDH) 活性에 미치는 影響을 考察하였다. 血淸 GOT 및 LDH 含量은 兩群 共히 對照群에 比하여 顯著한 變化를 招來하였으며, 특히 GOT는 初期에 減少되고, 後期에 恒定持續性을 나타내어 對照群의 값에 近接하는 傾向이 나타났으며, 한편 LDH의 含量은 初期에 增加하고 後期에는 若干의 減少傾向이 나타났다. 一般的으로 이러한 變化는 흰쥐가 低氣壓의 曝露에 一時的인 恒定持續性을 나타내는 것을 보여주며 酵素의 含量의 變化는 低氣壓의 强度와 順化期間에 따라 다르며, 一般的으로 380 mmHg群은 500 mmHg群에 比하여 顯著한 變化相을 가져오는 것으로 思料된다.

  • Eight cattle and ten sheep were administrated various degree of PMA-B and EPN, and S-GOT, S-GPT and serum cholinesterase activity were tested. Serum cholinesterase activity showed no typical tendency, however, S-GOT/S-GPT ratios were decreased by the administration of PMA-B. It was suggested some degree of liver damage by the administration of the chemicals was recognized.

사염화탄소에 의해 유발된 간독성에 대한 상백피 추출물의 간보호효과 (Protective Effects of Extracts of Mori Cortex Radicis on Carbon Tetrachloride-induced Hepatotoxicity)

  • In order to evaluate the protective effects of extracts of Mori cortex radicis on carbon tetrachloride-induced hepatotoxicity, glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, lactic dehydrogenase, malondiadehyde values and glutathione S-transferase activity were measured in ICR mice. The activities of serum aminotransferase and the hepatic content of lipid peroxide after carbon tetrachloride-treatment were markedly increased than normal control but those levels were significantly decreased by the treatment of butanol fraction of Mori cortex radicis mathanol extract. Glutathione S-transferase activity was decreased by carbon tetrachloride than control, but that also inhibited by the treatment of butanol fraction of Mori cortex radicis methanol extract. These results demonstrate a possible hepatoprotective role of extract role of extract of Mori cortex radicis against ${CCl_4}-induced$ hepatoxicity in vivo.

Glutamic-oxaloacetic transaminase 1 regulates adipocyte differentiation by altering nicotinamide adenine dinucleotide phosphate content

  • Objective: This study was performed to examine whether the porcine glutamic-oxaloacetic transaminase 1 (GOT1) gene has important functions in regulating adipocyte differentiation. Methods: Porcine GOT1 knockout and overexpression vectors were constructed and transfected into the mouse adipogenic 3T3-L1 cells. Lipid droplets levels were measured after 8 days of differentiation. The mechanisms through which GOT1 participated in lipid deposition were examined by measuring the expression of malate dehydrogenase 1 (MDH1) and malic enzyme (ME1) and the cellular nicotinamide adenine dinucleotide phosphate (NADPH) content. Results: GOT1 knockout significantly decreased lipid deposition in the 3T3-L1 cells (p<0.01), whereas GOT1 overexpression significantly increased lipid accumulation (p<0.01). At the same time, GOT1 knockout significantly decreased the NADPH content and the expression of MDH1 and ME1 in the 3T3-L1 cells. Overexpression of GOT1 significantly increased the NADPH content and the expression of MDH1 and ME1, suggesting that GOT1 regulated adipocyte differentiation by altering the NADPH content. Conclusion: The results preliminarily revealed the effector mechanisms of GOT1 in regulating adipose differentiation. Thus, a theoretical basis is provided for improving the quality of pork and studies on diseases associated with lipid metabolism.

상엽과 산사엽 추출물이 사염화 탄소로 유발된 흰쥐의 간 손상에 미치는 영향 (Effects of Mori Folium and Crataegus pinnatifida Leave Extracts on $CC1_4$-induced Hepatotoxicity in Rats)

  • This study was performed to investigate the effect of Mori Folium and Crataegus pinnatifida leave extracts on liver damage induced by CC1$_4$ in rats. 30% and 60% methanol extracts of Mori Folium and Crataegus pinnatifida leaves were administered orally at the dose of 10 mg/kg on every day for 6 days and liver damage was induced by intraperitoneal injection of $CCl_4$ (0.4 ml/kg) on 6th day. 30% Methanol extracts of Mori Folium treated group showed significant protective effect on hepatotoxicity with the lowest Glutamic Oxaloacetic Transaminase (GOT) and Glutamic Pyruvic Transaminase (GPT). The low density lipoprotein (LDL) levels of 30% methanol extracts of Mori Folium and Crataegus pinnatifida leave and 60% methanol extracts of Mori Folium treated groups were significantly lower than normal group, respectively. In particular, LDL level of 30% Mori Folium extracts treated group was significantly decreased compared to $CCl_4$ treated group. These results support that Mori Folium and Crataegus pinnatifida leave extracts are hepatoprotective effects against hepatotoxicity induced by $CCl_4$.

  • Effects of forced physical exercise on the CCl4 hepatotoxicity were examined in adult female rats. Rats were treated with CCl4 (2 mmol/kg, ip) and introduced into a cylindrical cage rotating at 9 rpm for 20 min/hr for 6 hr. Eighteen hr following the termination of exercise serum sorbitol dehydrogenase (SDH), glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT) activities and glucose-6-phosphatase activity were determined as parameters for hepatotoxicity. Physical exercise inhibited the $CCI_{4-}$induced increases in SDH, GOT, GPT activity, and glucose-6-phosphatase activity were determined as parameters for hepatotoxicity.

  • The effects of dichloromethane (DCM) on carbon tetrachloride (CT) toxicity were examined in adult male rats. A concomitant treatment of rats with DCM (0.3, 0.6, 1.2 g/kg, po) significantly potentiated the hepatotoxicity of CT (1.0 g/kg, po) as determined by increase in serum GPT (glutamic pyruvic transaminase), GOT (glutamic oxaloacetic transaminase), and SDH (sorbitol dehydrogenase) activity 24 hr following the treatments. Serum LDH (lactate dehydrogenase) activity was increased by either DCM or CT treatment.

성장장애 흰쥐에서 경옥고(瓊玉膏)와 경옥고가록용(瓊玉膏加鹿茸)의 성장효과 비교 연구 (A Comparative Study on Effects of Kyungohkgo and Kyungohkgo Ga Nokyong on Growth in Growth Deficiency Rat with Insufficient Nutrition Diet)

  • Objectives This study was designed to evaluate the difference of between Kyungohkgo and Kyungohkgo Ga Nokyong effects of growth Methods We divided male Spraque-Dawley rats into 6 groups. They were Normal group, Growth deficiency rat with insufficient nutrition diet group, Growth deficiency rat with 0.1% Kyungohkgo group, Growth deficiency rat with 0.2% Kyungohkgo group, Growth deficiency rat with 0.1% Kyungohkgo Ga Nokyong group and Growth deficiency rat with 0.2% Kyungohkgo Ga Nokyong group. They were administered for 5 weeks. We measured body weight, serum growth hormone, insulin-like growth factor and thyroid stimulating hormone, RBC, concentration of Hb and PCV ratio, total WBC and its composition, the values of plasma glutamic oxaloacetic transaminase(GOT) and glutamic pyruvic transaminase(GPT) activities. Results 1. In body weight, Kyungohkgo 0.1%, Kyungohkgo Ga Nokyong 0.1%, 0.2% groups were showed sighnificantly different than control group. But That groups were not showed significantly different than each others. 2. In serum growth hormone, insulin-like growth factor and thyroid stimulating hormone, we obtained the results of tendency to increase in Kyungohkgo Ga Nokyong group, however these values showed no significantly different. 3. In the counts of RBC, Kyungohkgo group and Kyungohkgo Ga Nokyong group were showed significantly different than control group. But these values showed no significantly different. In the concentration of Hb, Kyungohkgo Ga Nokyong 0.2%group was showed significantly different than control group. Kyungohkgo Ga Nokyong 0.2% group and Kyungohkgo 0.2% group were showed significantly different than control group. 4. The counts of total WBC and its composition showed no significantly different in all treatment groups. 5. The values of plasma glutamic oxaloacetic transaminase(GOT) and glutamic pyruvic transaminase(GPT) activities showed no significantly different in all treatment groups. Conclusions So Kyungohkgo Ga Nokyong and Kyungohkgo have an effect of promoting growth of rats. And We know that the effect of Kyungohkgo Ga Nokyong is better than Kyungohkgo.

붕어, 비둘기, 흰쥐의 肝臟의 Glutamic Dehydrogenase 및 Glutamic Transaminase 의 活性에 관한 比較 硏究 (A comparative Study on the Activities of Glutamic Dehydrogenase and Glutamic Transaminase in Livers of the Crucian carp, Pigeon, and Rat)

  • 붕어, 비둘기, 흰쥐의 肝組織內의 蛋白質含量과 蛋白質代謝에 관하여는 GDH, GOT 및 GPT의 活性과 比較活性, 그리고 GDH 同位酵素를 測定하였다. 1. 蛋白質含量은 흰쥐, 비둘기가 各各 22.0$\pm$0.01mg/ml 및 22.0$\pm$0.16으로서 거의 같은 量의 蛋白質含量으로 組成되었으나 붕어의 肝臟組織內 蛋白質含量은 約 60%에 該當하는 13.0$\pm$0.09이었다(p < 0.01). 2. Glutamic transaminase는 흰쥐, 비둘기, 붕어의 순위로 그 活性度가 낮았다. 卽 GPT의 比較活性은 各各 3.77$\pm$0.18 unit/mg, 1.93$\pm$0.01 및 0.71$\pm$0.07 이었으며 GOT는 8.23$\pm$0.06 unit/mg, 3.95$\pm$0.09 및 0.92$\pm$0.01이었다(p < 0.01). 3. GOT/GDT 比는 흰쥐, 비둘기, 붕어에서 各各 0.20$\pm$0.004, 0.22$\pm$0.005 및 0.20$\pm$0.002로서 별 차이 없었다. 4. GDH 比較活性은 비둘기가 가장 높은 35.7$\pm$0.81 unit/mg이고 붕어는 9.6$\pm$0.16, 흰쥐는 20.5$\pm$0.81이었다. 5. Glutamic transaminase 와 GDH 比較活性이 動物의 種類에 따라 병행하였으며 特히 進化過程과 一致되는 것으로 思料된다. 6. GDH 同位酵素의 樣相은 種特異性이 뚜렷하였다. 흰쥐에서는 음극이 遲延性易動度區劃의 活性이 가장 높았으나, 비둘기에서는 兩極性區劃의 活性이 가장 높았다. 特히 魚類에서는 陰極性區劃이 全然 發見되지 않았으나 兩極性에 4個의 區劃을 볼수 있었다.
