Postprandial hypoglycemic effect of mulberry leaf in Goto-Kakizaki rats and counterpart control Wistar rats

  • Postprandial hypoglycemic effect of mulberry leaf (Morus alba L.) was compared in two animal models: Goto-Kakizaki (GK) rats, a spontaneous non-obese animal model for type II diabetes, and their counterpart control Wistar rats. First, the effect of a single oral administration of mulberry leaf aqueous extract (MLE) on postprandial glucose responses was determined using maltose or glucose as substrate. With maltose-loading, MLE reduced peak responses of blood glucose significantly in both GK and Wistar rats (P < 0.05), supporting the inhibition of $\alpha$-glucosidase by MLE in the small intestine. With glucose-loading, MLE also significantly reduced blood glucose concentrations, measured at 30 min, in both animal models (P < 0.01), proposing the inhibition of glucose transport by MLE. Next, dried mulberry leaf powder (MLP) was administered for 8 weeks by inclusion in the diet. By MLP administration, fasting blood glucose was significantly reduced at weeks 4 and 5 (P < 0.05), but then returned to values that were similar to those of the control at the end of experimental period in GK rats. Insulin, HOMA-IR, C-reactive protein, and triglycerides tended to be decreased by MLP treatment in GK rats. All other biochemical parameters were not changed by MLP administration in GK rats. Collectively, these findings support that MLE has significant postprandial hypoglycemic effect in both non-obese diabetic and healthy animals, which may be beneficial as food supplement to manage postprandial blood glucose. Inhibitions of glucose transport as well as $\alpha$-glucosidase in the small intestine were suggested as possible mechanisms related with the postprandial hypoglycemic effect of MLE.

The Combined Effect of Gamma Knife Irradiation and p53 Gene Transfection in Human Malignant Glioma Cell Lines

  • Objective: The purpose of this study is to elucidate in vitro responses to combined gamma knife irradiation and p53 gene transfection on human malignant glioma cell lines. Methods: Two malignant human glioma cell lines, U87MG (p53-wild type) and U373MG (p53-mutant) were transfected with an adenoviral vector containing p53 (MOI of 50) before and after applying 20Gy of gamma irradiation. Various assessments were performed, including, cell viability by MTT assay; apoptosis by annexin assay; and cell cycle by flow cytometry, for the seven groups: mock, p53 only, gamma knife (GK) only, GK after LacZ, LacZ after GK, GK after p53, p53 after GK. Results: Cell survival decreased especially, in the subgroup transfected with p53 after gamma irradiation. Apoptosis tended to increase in p53 transfected U373 MG after gamma irradiation (apoptotic rate, 38.9%). The G2-M phase cell cycle arrest markedly increased by transfecting with p53, 48 hours after gamma knife irradiation in U373 MG (G2-M phase, 90.8%). Conclusion: These results suggest that the in vitro effects of combined gamma knife irradiation and p53 gene transfection is an augmentation of apoptosis and G2-M phase cell cycle arrest, which are more exaggerated in U373 MG with p53 transfection after gamma knife irradiation.

Improvement of Thunderstorm Detection Method Using GK2A/AMI, RADAR, Lightning, and Numerical Model Data

  • To detect thunderstorms occurring in Korea, National Meteorological Satellite Center (NMSC) also introduced the rapid-development thunderstorm (RDT) algorithm developed by EUMETSAT. At NMCS, the H-RDT (HR) based on the Himawari-8 satellite and the K-RDT (KR) which combines the GK2A convection initiation output with the RDT were developed. In this study, we optimized the KR (KU) to improve the detection level of thunderstorms occurring in Korea. For this, we used all available data, such as GK2A/AMI, RADAR, lightning, and numerical model data from the recent two years (2019-2020). The machine learning of logistic regression and stepwise variable selection was used to optimize the KU algorithms. For considering the developing stages and duration time of thunderstorms, and data availability of GK2A/AMI, a total of 72 types of detection algorithms were developed. The level of detection of the KR, HR, and KU was evaluated qualitatively and quantitatively using lightning and RADAR data. Visual inspection using the lightning and RADAR data showed that all three algorithms detect thunderstorms that occurred in Korea well. However, the level of detection differs according to the lightning frequency and day/night, and the higher the frequency of lightning, the higher the detection level is. And the level of detection is generally higher at night than day. The quantitative verification of KU using lightning (RADAR) data showed that POD and FAR are 0.70 (0.34) and 0.57 (0.04), respectively. The verification results showed that the detection level of KU is slightly better than that of KR and HR.

Cloning and Characterization of GL-7-ACA Acylase Gene from Pseudomonas sp. GK16

  • The gene coding for glutaryl-7-aminocephalosporanic acid (GL-7-ACA) acylase was cloned from Pseudomonas sp. GK16 and some of its characteristics were analyzed. The complete nucleotide sequence revealed that the putative open reading frame is 2160 bases long and encodes 720 amino acids. By SDS-PAGE three proteins, approximately corresponding to 70, 54 and 16 kDa of molecular weight, were detected in E. coli cells carrying pGAP18. The largest protein should be a precursor which is not processed yet, while the other two proteins must be derived from the precursor by the proteolytic processing.

  • A gene (GenBank AF096282) coding for a $\alpha$-glucosidase (TcaAG, EC 3.2.1.20) from Thermus caldophilus GK24 was expressed in Saccharomyces cerevisiae, a generally recognized as safe (GRAS) host. The thermostable $\alpha$-glucosidase was produced inside of the GRAS host at 0.04 unit/mg-dry cell by the constitutively expressing ADH1 promoter and at 1.2 unit/mg-dry cell by the inductively expressing GALl0 promoter, respectively. No $\alpha$-glucosidase activities were found in the medium when the MF-alpha signal sequence from S. cerevisiae or $\alpha$-amylase signal sequence from Aspergillus oryzae were fused before the $\alpha$-glucosidase gene for the secretion.

  • A measurement of indoor microorganism using Reuter Centrifugal Air Sampler(RCS) was undertaken during October 1991-February 1999 and 6-Stage Cascade Air Sampler was undertaken during May 2001-June 2001 in Seoul. The results were as follows : 1.   • Hyperacute liver failure from mushroom intoxication in children is rare and has a low survival rate. We report a case of hyperacute liver failure from mushroom intoxication in a 29-month-old boy. The patient ingested a mushroom about three days prior to presentation. He was admitted to the hospital with vomiting, abdominal pain, seizures, and hematemesis. During the hospitalization the patient developed hepatic encephalopathy (stage IV-a), and a coagulopathy. He recovered fully with specific medication, Penicillin GK and N-acetylcysteine.

An H.323 Gatekeeper Architecture Providing Handoff function (핸드오프를 지원하는 H.323 게이트키퍼 구조)

  • H.323 proposes to use Mobile IP and H.323 ad hoc conference signaling to provide Handoff function to an H.323 mobile terminal. But the H.323 ad hoc conference signaling has a drawback. It requires an H.323 terminal to do a complex conference signaling which takes a longer signaling time. In this paper, we propose an GK architecture that provides Handoff function effectively using 3$^{rd}$ P'||'||'||'||'||'&'||'||'||'||'||'R(Third party initiated Pause and Rerouting) signaling which are done through H.245 logical channel. The GK which is implementing 3$^{rd}$ P'||'||'||'||'||'&'||'||'||'||'||'R signaling only requires an H.323 endpoint to do the H.323 basic signaling in reestablishing media channel and gives the faster Handoff signaling. To do this, our GK has derived H.245 control channel using tunneling f3r all H.323 calls including the fast connect calls which enable terminals communicate each other if they doesn't have H.245 control channel .municate each other if they doesn't have H.245 control channel .

Production and Properties of a Bacillus subtilis Mannanase from Recombinant Lactobacillus paracasei (재조합 Lactobacillus paracasei로부터 Bacillus subtilis의 Mannanase 생산과 효소특성)

  • A gene coding for mannanase (manA) from Bacillus subtilis was introduced into a shuttle vector pGK12 between Escherichia coli, B. subtilis and Lactobacillus paracasei. As a result of transferring the resultant plasmid, designated pGK12M3, into three different strains, the manA gene was found to be expressed in L. paracasei as well as in B. subtilis and E. coli. In a 4 L fermentor culture, the production of mannanase by recombinant L. paracasei (pGK12M3) reached a maximum level of 5.4 units/ml in an MRS medium with a fixed pH 6.5. Based on the zymogram of mannanase, it is assumed that mannanase produced by recombinant L. paracasei is not maintained stably with proteolytic degradation. The optimal temperature and thermostability of mannanase produced by recombinant L. paracasei were also found to be different from those of enzymes produced by B. subtilis.

Sensitivity Analysis for CAS500-4 Atmospheric Correction Using Simulated Images and Suggestion of the Use of Geostationary Satellite-based Atmospheric Parameters (모의영상을 이용한 농림위성 대기보정의 주요 파라미터 민감도 분석 및 타위성 산출물 활용 가능성 제시)

  • As part of the next-generation Compact Advanced Satellite 500 (CAS500) project, CAS500-4 is scheduled to be launched in 2025 focusing on the remote sensing of agriculture and forestry. To obtain quantitative information on vegetation from satellite images, it is necessary to acquire surface reflectance through atmospheric correction. Thus, it is essential to develop an atmospheric correction method suitable for CAS500-4. Since the absorption and scattering characteristics in the atmosphere vary depending on the wavelength, it is needed to analyze the sensitivity of atmospheric correction parameters such as aerosol optical depth (AOD) and water vapor (WV) considering the wavelengths of CAS500-4. In addition, as CAS500-4 has only five channels (blue, green, red, red edge, and near-infrared), making it difficult to directly calculate key parameters for atmospheric correction, external parameter data should be used. Therefore, thisstudy performed a sensitivity analysis of the key parameters (AOD, WV, and O3) using the simulated images based on Sentinel-2 satellite data, which has similar wavelength specifications to CAS500-4, and examined the possibility of using the products of GEO-KOMPSAT-2A (GK2A) as atmospheric parameters. The sensitivity analysisshowed that AOD wasthe most important parameter with greater sensitivity in visible channels than in the near-infrared region. In particular, since AOD change of 20% causes about a 100% error rate in the blue channel surface reflectance in forests, a highly reliable AOD is needed to obtain accurate surface reflectance. The atmospherically corrected surface reflectance based on the GK2A AOD and WV was compared with the Sentinel-2 L2A reflectance data through the separability index of the known land cover pixels. The result showed that two corrected surface reflectance had similar Seperability index (SI) values, the atmospheric corrected surface reflectance based on the GK2A AOD showed higher SI than the Sentinel-2 L2A reflectance data in short-wavelength channels. Thus, it is judged that the parameters provided by GK2A can be fully utilized for atmospheric correction of the CAS500-4. The research findings will provide a basis for atmospheric correction of the CAS500-4 in the future.
