Effect of Barrel Temperature and Screw Speed on Characteristics of Extruded Raw Ginseng (배럴온도와 스크루 회전속도에 따른 압출성형 수삼의 특성)

  • The objective of this study was to determine effects of the die temperature(100 and $120^{\circ}C$) and screw speed(200 and 300 rpm) on the characteristics of extruded raw ginseng such as crude saponin, ginsenosides, maltol and the color of powder. Crude saponin content increased after extrusion-cooking. Ginsenoside $Rg_1\;and\;Rg_2$ that contained in red ginseng increased from 0.2275 mg/g to 0.2835 mg/g$(Rg_l)$ and 0.1164 mg/g to 0.2230 mg/g$(Rg_2)$ with the increase in die temperature from 100 to $120^{\circ}C$, which increased with the decrease in screw speed from 300 to 200 rpm. Maltol, specific component in red ginseng was detected in extruded ginseng. Total sugar content was not changed by extrusion process, however reducing sugar decreased with the increase in die temperature from 100 to $120^{\circ}C$. In conclusion extrusion process can be applied to red ginseng manufacturing by controling extrusion process variables such as extrusion temperature and screw speed.

Inhibitory Effects of Rice Bran Water Extract Fermented Lactobacillus plantarum due to cAMP-dependent Phosphorylation of VASP (Ser157) on human Platelet Aggregation

  • In this study, we investigated the effect of rice bran water extract fermented with Lactobacillus plantarum KCCM-12116 (RBLp) on ADP ($20{\mu}M$)-, collagen ($10{\mu}g/mL$)-, and thrombin (0.2 U/mL)-stimulated platelet aggregation. RBLp dose-dependently inhibited ADP-, collagen-, and thrombin-induced platelet aggregation, with $IC_{50}$ values of 501.1, 637.2, and > $2,000{\mu}g/mL$, respectively. The platelet aggregation induced by ADP plus RBLp ($750{\mu}g/mL$) was increased by the adenylate cyclase inhibitor, SQ22536, and the cAMP-dependent protein kinase (A-kinase) inhibitor, Rp-8-Br-cAMPS. Treatment with RBLp increased the phosphorylation of VASP ($Ser^{157}$), an A-kinase substrate, which was also inhibited by SQ22536 and Rp-8-Br-cAMPS. It is thought that the RBLp-induced increases in cAMP contributed to the phosphorylation of VASP ($Ser^{157}$), which in turn resulted in an inhibition of ADP-induced platelet aggregation, thereby indicating that RBLp has an antiplatelet effect via cAMP-dependent phosphorylation of VASP ($Ser^{157}$). Thus, RBLp may have therapeutic potential for the treatment (or prevention) of platelet aggregation-mediated diseases, such as thrombosis, myocardial infarction, atherosclerosis, and ischemic cerebrovascular disease.

Comparison of Characteristics of Ganoderma lucidum According to Geographical Origins(III): Classification between Species of Genus Ganoderma Using Dikaryon-Monokaryon Mating

  • A Monokaryotic strain G8M without clamp connections was isolated from germinated basidiospore that was obtained from cultivated fruit body. Strain G8M was used as a tester isolate for 'dikaryon-monokaryon mating'(di-mon mating) with the strains of Ganoderma lucidum, G6 and G35(Korean wild strains), G3(Taiwan), G4(Canada), G15(America), G. oregonense G24, G. resinaceum G28, G. oerstedii G23, and G. subamboinense G29. Isolate G8M was compatible to Korean strains G6 and G35, but was incompatible to foreign strains G3, G4, or G15. Compatible reactions between strains were readily observed macroscopically. Clear barrage lines formed between incompatible strains. These clear lines were not apparent in compatible di-mon matings. The Korean strains were morphologically distinct; they did not form any chlamydospores, and stopped growth at $35^{\circ}C$. The strains of G. lucidum from Korea may be considered as different species from Taiwan, Canadian and American cultures.

Effects of SNPs in Haptoglobin on Average Daily Gain in Pig (Haptoglobin SNP의 돼지 일당 증체량에 관한 효과)

  • In order to provide information of genetic variants for Haptoglobin (Hp) gene, which may be related to weight traits in pig, a total of 235 animals from National Institute of Animal Science (NIAS) were screened with 3 primers. The primer sequences were selected using the porcine cDNA sequences based on NM_214000, and the exon boundaries were estimated. Genetic variants were observed using direct sequencing analysis, and there were 9 SNPs detected at nucleotide positions 503 (A/G), 509 (A/G), 709 (C/T), 734 (C/A), 742 (G/A), 769 (A/G), 840 (C/T), 876 (C/T) and 882 (C/A). All the SNPs were located in coding regions, and mutations caused amino acid changes at nucleotide positions 503, 509, 734, 742 and 769. Allele frequencies of SNPs were estimated for all segments. The SNPs at nucleotide position 509 (p<0.0001) and 734 (p<0.05) were significantly associated with average daily gain, but no significance was observed with other SNPs. From the results, the identified SNPs may be a useful candidate marker for the porcine weight gain traits.

Cloning and Characterization of Squalene Synthase (SQS) Gene from Ganoderma lucidum

  • This report provides the complete nucleotide sequences of the full-length cDNA encoding squalene synthase (SQS) and its genomic DNA sequence from a triterpene-producing fungus, Ganoderma lucidum. The cDNA of the squalene synthase (SQS) (GenBank Accession Number: DQ494674) was found to contain an open reading frame (ORF) of 1,404 bp encoding a 468-amino-acid polypeptide, whereas the SQS genomic DNA sequence (GenBank Accession Number: DQ494675) consisted of 1,984 bp and contained four exons and three introns. Only one gene copy was present in the G. lucidum genome. The deduced amino acid sequence of Ganoderma lucidum squalene synthase (GI-SQS) exhibited a high homology with other fungal squalene synthase genes and contained six conserved domains. A phylogenetic analysis revealed that G. lucidum SQS belonged to the fungi SQS group, and was more closely related to the SQS of U. maydis than to those of other fungi. A gene expression analysis showed that the expression level was relatively low in mycelia incubated for 12 days, increased after 14 to 20 days of incubation, and reached a relatively high level in the mushroom primordia. Functional complementation of GI-SQS in a SQS-deficient strain of Saccharomyces cerevisiae confirmed that the cloned cDNA encoded a squalene synthase.

Chemical Compositions and Antioxidant Activity of Extract from a Extruded White Ginseng (압출성형 백삼추출물의 화학적 조성 및 항산화 활성)

  • Chemical components and antioxidative activities of white ginseng, red ginseng and extruded white ginseng (EWG) were evaluated. Extrusion condition was 20% moisture content, 100 and $140^{\circ}C$ barrel temperature. The results showed that total sugar and acidic polysaccharide contents of white ginseng powder were increased after extrusion treatment of which EWG at $140^{\circ}C$ barrel temperature had higher value than EWG at $100^{\circ}C$ barrel temperature. Free radical scavenging activity of EWG at $140^{\circ}C$ barrel temperature was 80.2 and 45.6% respectively. The butanol fraction of polyphenolic compound and acidic polysaccharide were $27.2{\pm}0.1\;mg/g$ and $217.6{\pm}0.7\;mg/g$, respectively. The ginsenosides were quantified by HPLC and the yield of ginsenoside-Rg3s and Rg3r were achieved by extrusion process.

Effects of Dietary Bong-ip(Morus alba L.), Gam-chei(Glycyrrhizae glabra), Sol-ip(Pinus densiflora) and Dang-gi(Angelica gigas) on Serum Composition in Rats (뽕잎, 감초, 솔잎 및 당귀분말이 흰쥐의 혈청조성에 미치는 영향)

  • The purpose of this study was to investigate the effect of dietary Morus alba L.(Bong-ip, B), Glycyrrhizae glabra(Gam-chei, C), Pinus densiflora(Sol-lp, S) and Angelica gigas(Dang-gi, D)powder on serum composition in rats(Sprague-Dawley male rats, 100-110g). Serum TG(triglyceride, p<0.01), total cholesterol, glucose, total protein, albumin, GGT$({\gamma}-glutamyl$ transferase, p<0.05) were significantly increased D group than that of nomal and other groups, but UA(uric acid, p<0.05) was significantly decreased, and C group(p<0.05) was significantly increased. but C group of urine(p<0.05) was significantly decreased. Also, B and S groups(p<0.05) of BUN(blood urea nitrogen), S group(p<0.05) of ALP(alkaline phosphokinase, Band C(p<0.05) of CPK(creatinine phosphokinsae, p<0.05) were significantly increased. B, S and C groups were better than D group for lipid metabolism, and pretection to liver. Also, B and C groups of glucose were same as normal diet, so Morus alba L. was good food for lipid metabolism and hypoglycemic effect.

The Microwave Dielectric Properties of BMCT Ceramics (BMCT 세라믹스의 마이크로파 유전특성)

  • Ba(Mgl-xCox)TaO3[BMCT] ceramics were prepared by the conventional mixed oxide method. The ceramics were sintered at the temperature of 1525~$1625^{\circ}C$ for 5hr. in air. The crystal structure of BMCT ceramics was investigated by the XRD. The microstructure of the specimens were observed by SEM. The Microwave dielectric properties of BMCT specimens were investigated as a function of composition and sintering temperature. All BMCT ceramics sintered over 1575$^{\circ}C$ were showed a polycrystalline complex perovskite structure. The density of BMCT (90/10) specimen sintered at $1575^{\circ}C$ was 7.75g/㎤. As the Co contents decreased, the ordering parameter of B-site in BMCT increased. In the case of the BMCT(90/10) ceramics sintered at $1575^{\circ}C$ for 5 hours, dielectric constant, quality factor and temperature coefficient of resonant frequency for microwave dielectrics application were a good value o( 25, 17, 845 at 10㎓ and +2.4 ppm/${\circ}$, respectively.

Effect of Cordycepin on the Expression of the Inflammatory Cytokines TNF-alpha, IL-6, and IL-17A in C57BL/6 Mice

  • Culture supernatants of splenocytes from C57BL/6 mice were exposed to 0.3, 1.0, and 3.0 ${\mu}g/ml$ cordycepin plus 3.0 ${\mu}g/ml$ lipopolysaccharide (LPS) to investigate the effects of cordycepin (3'-deoxyadenosine) on the production of inflammatory cytokines. Coadministration of 3.0 ${\mu}g/ml$ cordycepin with LPS in cultured murine spleen cells significantly diminished expression of the inflammatory cytokines tumor necrosis factor-${\alpha}$ and interleukin-6 (IL-6) in a time-dependent manner. Expression of the inflammatory cytokine IL-17A was substantially downregulated in a timeand concentration-dependent manner at all cordycepin concentrations. These findings suggest that cordycepin downregulates the immediate hypersensitivity reaction stimulated by LPS.

Production of ${\gamma}$-Linolenic Acid by Mortierella isabellina IFO 8183 (Mortierella isabellina IFO 8183에 의한 ${\gamma}$-Linolenic Acid 생산)

  • To produce ${\gamma}-linolenic$ acid by a mold, cultural conditions of Mortierella isabellina IFO 8183 were investigated. It was found that the increase of initial pH resulted in the decrease of the ${\gamma}-Linolenic$ acid content and the increase of the C/N ratio of medium resulted in the increase of the lipid content. Addition of sodium acetate into the medium resulted in the increased of cell yield, lipid yield, ${\gamma}-Linolenic$ acid content and ${\gamma}-Linolenic$ acid productivity. Under the optimum coditions(glucose, $NH_4NO_3$, C/N ratio of 40, pH 6.0, $30^{\circ}C$ and 0.5% of sodium acetate), the following results were obtained: cell yield, 0.347(g dry biomass/g glucose; lipid yield, 0.18(g lipid/g glucose); lipid content, 0.52(g lipid/g dry biomass); ${\gamma}-linolenic$ acid content, 60(mg ${\gamma}-linolenic$ acid/g lipid); maximum ${\gamma}-linolenic$ acid concentration, 347mg/l after incubation of 8 days.
