  • Our understanding of the sensitivities of crop responses to changes in carbon dioxide, temperature, and water is limited, which makes it difficult to fully utilize crop models in assessing the impact of climate change on future agricultural production. Genetic coefficients of CERES-Barley model for major domestic cultivars in South Korea (Olbori at Suwon, Albori at Milyang, Saessalbori at Iksan, and Samdobori at Jinju) were estimated from the observed data for daily weather and field trials for more than 10 years by using GenCalc in DSSAT. Data from 1997-2002 annual crop status report (Rural Development Administration, RDA) were used to validate the crop coefficients. The sitecalibrated CERES-Barley model was used to perform crop growth simulation with the 99 treatments of step change combinations in temperature, precipitation and carbon dioxide concentration with respect to the baseline climate (1981-2010) at four sites. The upper boundary corresponds to the 2071-2100 climate outlook from the RCP 8.5 scenario. The response surface of grain yield showed a distinct pattern of model behavior under the combined change in environmental variables. The simulated grain yield was most sensitive to $CO_2$ concentration, least sensitive to precipitation, and showing a variable response to temperature depending on cultivar. The emulated impacts of response surfaces are expected to facilitate assessment of projected climate impacts on a given cultivar in South Korea.

  • Larvae of Bradysia agrestis, an insect vector that transports plant pathogens, were sampled from geographically isolated regions in Korea to identify their cutaneous fungal and bacterial flora. Sampled areas were chosen within the distribution range of B. agrestis; each site was more than 91 km apart to ensure geographical segregation. We isolated 76 microbial (fungi and bacteria) strains (site 1, 29; site 2, 29; site 3, 18 strains) that were identified on the basis of morphological differences. Species identification was molecularly confirmed by determination of universal fungal internal transcribed spacer and bacterial 16S rRNA gene sequences in comparison to sequences in the EzTaxon database and the NCBI GenBank database, and their phylogenetic relationships were determined. The fungal isolates belonged to 2 phyla, 5 classes, and 7 genera; bacterial species belonged to 23 genera and 32 species. Microbial diversity differed significantly among the geographical groups with respect to Margalef's richness (3.9, 3.6, and 4.5), Menhinick's index (2.65, 2.46, and 3.30), Simpson's index (0.06, 0.12, and 0.01), and Shannon's index (2.50, 2.17, and 2.58). Although the microbial genera distribution or diversity values clearly varied among geographical groups, common genera were identified in all groups, including the fungal genus Cladosporium, and the bacterial genera Bacillus and Rhodococcus. According to classic principles of co-evolutionary relationship, these genera might have a closer association with their host insect vector B. agrestis than other genera identified. Some cutaneous bacterial genera (e.g., Pseudomonas) displaying weak interdependency with insect vectors may be hazardous to agricultural environments via mechanical transmission via B. agrestis. This study provides comprehensive information regarding the cutaneous microflora of B. agrestis, which can help in the control of such pests for crop management.

  • Purpose: Various bone graft materials are being used for periodontal tissue regeneration. Th materials are being developed continuously for ideal clinical effects. Therefore, it is necessary to identify the clinical characteristics of each bone graft material through comparing the various bone graft materials statistically and in doing so, proposing a more efficient bone graft material. In this study, the following results were attained through comparing the clinical effects among the bone graft materials, using the statistical method based on the clinical studies published at the department of periodontology of Yonsei hospital. Materials and Method: 6 selected studies of department of Periodontology at Yonsei University Hospital were based on clinical study of bone grafting in intrabony defects. It was compared the clinical parameters among the 6 clinical studies, using the statistical META analysis. Result: When comparing the probing depth reduction, there was a relatively great amount of decease when using the xenograft, Anorganic Bovine Derived Hydroxapatite Bone Matrix/Cell Binding Peptide(ABM/P-15: PepGen $P-15^{(R)}$) and the autogenous bone and absorbable membrane, d, 1-alctide/glycolide copolymer(GC: $Biomesh^{(R)}$). The allogfrafts showed a relatively low decrease in the probing depth and clinical attachment change. It also showed a slight decrease in the bone probing depth. The allografts showed various results according to different bone graft materials. When comparing the ABM/P-15 and bovine bone $powder(BBP^{(R)})$, ABM/P-15 showed a relatively high clinical attachment level and the bovine bone powder showed a relatively high clinical attachment level. The probing depth change and gingival recession change showed a lower value than the mean value between the two bone graft materials. The synthetic bone showed a relatively high decrease in clinical attachment level and periodontal probing depth change. There was a relatively larger amount of gingival recession when using Bioactive Glass(BG) but a relatively low bone regeneration effect was seen. Conclusion: Good restorative results of the periodontal tissue can be attained by applying the various bone graft materials being used today after identifying the accurate clinical effects.

  • Superoxide dismutases (SODs) constitute the first line of cellular defense against oxidative stress in plants. SODs generally occur in three different forms with Cu/Zn, Fe, or Mn as prosthetic metals. We cloned the full-length cDNA of the Thellungiella halophila Cu/Zn-SOD gene ThCSD using degenerate RT-PCR and rapid amplification of cDNA ends (RACE). Sequence analysis indicated that the ThCSD gene (GenBank accession number EF405867) had an open reading frame of 456 bp. The deduced 152-amino acid polypeptide had a predicted molecular weight of 15.1 kDa, an estimated pI of 5.4, and a putative Cu/Zn-binding site. Recombinant ThCSD protein was expressed in Escherichia coli and assayed for SOD enzymatic activity in a native polyacrylamide gel. The SOD activity of ThCSD was inactivated by potassium cyanide and hydrogen peroxide but not by sodium azide, confirming that ThCSD is a Cu/Zn-SOD. Northern blotting demonstrated that ThCSD is expressed in roots, stems, and leaves. ThCSD mRNA levels increased by about 30-fold when plants were treated with sodium chloride (NaCl), abscisic acid (ABA), and indole-acetic acid (IAA) and by about 50-fold when treated with UVB light. These results indicate that ThCSD is involved in physiological pathways activated by a variety of environmental conditions.

  • The clay and yellow loess have capability to adsorb and precipitate particles. The removal efficiencyes of those flocculents on the dinoflagellate, Cochlodinium polykrikoides, have been studied in laboratory and in field near Tongyong fish farm in September, 1996. The removal efficiencyes in the laboratory experiment was $43\%$ for $2\;g/{\ell}$, $64\%$ for $6g/{\ell}$ and $88\%$ for $10\;g/{\ell}$ in one hour after dispersion. No big difference of removal efficiency was found between the raw and the acid-activated loess. In the field survey, the removal rates ranged from 72 to $80\%$ in 30 min after the dispersion. The effect of loess scattering on water quality was estimated. The concentrations of dissolved inorganic nitrogen (DIN), chemical of gen demand (COD) and chlorophyll a decreased more or less after dispersion, while the concentration of suspended solid (SS) increased. The concentrations of dissolved oxygen (DO) and dissolved inorganic phosphorous (DIP) were kept constant. These results indicated that the dispersion concentration of more than $10g/{\ell}$ has a good removal efficiency of above $80\%$ without big variation of water quality after dispersion of yellow loess.

  • The Tomb of Jang Mui was discovered and investigated during the Japanese colonial era. It has provided key archaeological material for identifying the Daebang Commandery mentioned in historical records as Sariwon in Hwanghae-do Province when some bricks used in the construction of the tomb were found to be stamped with the official title "Prefect of Daebang Commandery." The discovery of the Tomb of Jang Mui served to confirm that the brick chamber tombs found along the Daedonggang River in 1909 were related to the Nangnang Commandery. It can be viewed as a major archaeological breakthrough that settled a debate over the locations of the Daesu River and Daebang Commandery that had been ongoing since the early Joseon Dynasty. Despite its significance for archeology and ancient Korean history, the circumstances of the discovery and investigations of the Tomb of Jang Mui have not been thoroughly examined. Inscribed bricks and roof tiles unearthed during the excavation conducted by Sekino Tadashi in 1912 are currently housed at The University Museum of The University of Tokyo. A number of other bricks excavated from the Tomb of Jang Mui are in the collection of the National Museum of Korea. Main agents of the investigations into these materials have not all been identified. Only some records on the materials collected during the investigations by Nomori Gen and others specify the main investigators. Inscribed bricks from other related tombs are also found in the Tokyo National Museum and at several universities in Japan. It is hoped that a comprehensive report incorporating all these materials can be written. Based on a reinterpretation of its structure conducted by Jung In-seung in 2010, the Tomb of Jang Mui has been dated to 348. This means it was not built during the Daebang Commandery period. The most compelling archaeological evidence regarding the location of the Daebang Commandery and its local capital has been undermined. It is hoped that active discussions will be held on the issue of the Daebang Commandery and its local capital.

  • Species identification of animal tissues in meat products is an important issue to protect the consumer from illegal and/or undesirable adulteration; for economic, religious and health reasons. In this reason, accurate analytical methods are needed for the labeling of meat products with requiring simple and fast procedure. Recently, applications of PCR in food analysis have been increased because of their simplicity, specificity and sensitivity. Therefore, in this study, a multiplex PCR assay was developed for the simultaneous identification of eight species of cow, pig, chicken, duck, goat, sheep, horse and turkey from raw meats. The primers were designed in different regions of mitochondrial 16S RNA after alignment of the available sequences in the GenBank database. Two multiplex primer sets were designed as Set 1 (cow, pig, chicken, duck) and Set 2 (goat, sheep, horse, turkey), respectively. Total 274 samples from cow (n = 55), pig (n = 30), chicken (n=30), and duck (n = 30), goat (n = 40), sheep (n = 33), horse (n = 41), and turkey (n = 15) were tested. The primers generated specific fragments of 94, 192, 279, 477 bp (pig, chicken, cow, duck), 670, 271, 152, 469 bp (goat, sheep, horse, turkey) lengths for eight species, respectively. The animal species specificity was 100% in all eight samples in the multiplex PCR assay. The detection limit of the multiplex PCR assay showed from 100 fg to 1 pg of template DNA from extracted from raw meats. When applying multiplex PCR assays to sample from pork/beef and pork/chicken, beef/chicken tested raw mixed meats and heat-treated ($83^{\circ}C$ for 30min, $100^{\circ}C$ for 20min, and $121^{\circ}C$ for 10min) mixtures, detection limit was 0.1% level beef, pork and pork in beef and chicken in pork and 1.0% level pork in chicken. This study suggest that the developed multiplex PCR assay can be used for rapid and simultaneous species identification of cow, pig, chicken, duck, goat, sheep, horse and turkey from meats.
