Discrimination of the geographical origin of commercial sesame oils using fatty acids composition combined with linear discriminant analysis (지방산 조성과 선형판별분석을 활용한 유통판매 참기름의 원산지 판별)

  • In this study, the fatty acid (FA) composition of commercial sesame oils (n = 62) was investigated using gas chromatography with flame ionization detector (GC-FID). Multivariate statistical techniques, including principal component analysis (PCA) and linear discriminant analysis (LDA), were applied to the chromatographic data of the FAs to discriminate the geographical origin of sesame oils. A statistically significant difference was observed in the content of C16:0, C18:0, C18:1, and C18:2 between domestic and imported sesame oils. A satisfactory recovery rate of 82.8-100.2 % was achieved for C16:0, C18:0, C18:1, C18:2, and C18:3. The correlation of C16:0, C18:1, and C18:2 in domestic sesame oils showed opposite trends compared to imported oils. The PCA plot demonstrated that sesame oils were clustered in distinct groups according to their origin. LDA was used to predict sesame oil samples in one of the two groups. C16:0 (Wilks λ = 0.361) and C18:1 (Wilks λ = 0.637) demonstrated the highest discriminant power for classifying the origin of the samples. The correct prediction rates were 88.9 % and 100 % for the domestic and imported samples, respectively. Further, 60 of the 62 sesame oil samples (96.8 %) were correctly classified, indicating that this approach can be used as a valuable tool to predict and classify the geographical origin of sesame oils.

Determination of Preservatives in Raw Materials of Functional Foods by HPLC-PDA and GC-FID (HPLC 및 GC를 이용한 건강기능식품 원료 중 보존료 함유량 조사)

  • Preservatives, as food additives, are occasionally intrinsic to natural raw materials or sometimes generated during the fermentation process as reported in many research articles. Preservative compounds in raw food materials may persist in the final food product, which is not supposed to include such preservative compounds. In this study, we validated an analytical method for preservative compounds in raw materials of functional foods. Quantification of benzoic acid and sorbic acid was determined using HPLC-PDA analysis after distillation, whereas propionic acid was quantified with GC-FID. A significant set of validation data (accuracy, precision, linearity, recovery, etc) was acquired. A total of 212 samples were collected for analysis of naturally occurred preservatives, and preservatives were detected in 85 samples. Most of the detected samples showed less 10 mg/kg of preservatives. The results of this study provide fundamental data on naturally occurring preservatives in raw materials of functional foods. Moreover, building up a database of naturally occurring preservatives could solve problems in the current scientific data.

  • Ethanolysis of squid viscera oil with immobilized lipase was investigated for reducing the free fatty acid contents and enhancing the function of the oil by stepwise addition method of Shinmada[1]. Tendency of oil variation during Ethanolysis showed increased content of diglyceride, monoglyceride and fatty acid ethyl ester with reduced free fatty acid contents. The oil composition was analyzed using GC-FID and compared before and after ethanolysis. Structural analysis of the lipid was performed by HPLC-UV spectrophotometer during ethanolysis. The transformed oil was thought to has suitable properties for functional oil production.

  • In the quantitative analysis of oxygenated exhaust emissions (unburned methanol, formal- dehyde) from methanol fueled vehicles, the oxygen contained in oxygenated exhaust gases lowers the FID (Flame Ionization Detector) response factor of conventional THC analyzer and leads to erroneous HC reading. For correct measurement of various HCs including oxygenated HCs emitted from FFV(Flexible Fuel Vehicle), first of all, the measurement technique of real HC emissions should be established. GC and HPLC-DNPH measuring methods specified by the EPA are used in this paper to analyze unburned methanol and formaldehyde components in the exhaust emissions. In emission test of FFV, unburned methanol and formaldehyde are emitted mostly during cold transient period, and it is shown that formaldehyde emission level is proportional to engine displacements. In view of the HC emission level, vehicle using M85 has 40% advantage over gasoline-fueled vehicle in OMHCE and has a good potential of a low emission vehicle.

  • UV photolysis process is little known in parts of air pollution treatment, so there are not many applications in field. Therefore we have to do more experiment and study application possibility for treatment of VOCs(Volatile organic compounds). To solve these problems, we have been studying for simultaneous application of this technology. It has shown that concentration of TCE and B.T.X., diameter of reactor and wavelength of lamp have effected on decomposition efficiency. Analysis of TCE and B.T.X. concentration was carried out by GC-FID. A cylinderical reactor consisting of a quartz tube and a centrally located lamp(${\psi}25mm$) was used. The length and diameter of reactor were 1800mm, 75mm. It has shown that the generated ozone concentration goes up 250ppm when using 64watt ozone lamp. When using Photolysis process only, the rates of fractional conversion of each material are TCE 79%, Benzene 65%, Toluene 68%, Xylene 76%. This phenomenon can be rationalized in terms of the different bond energy that indicates how easily VOCs species can be decomposed.

  • A plasticizer is a substance which is added to a material to improve its processability, flexibility and stratchability. Phthalates and adipates are the most frequently used plasticizers of poly(vinyl chloride) (PVC). However, they can migtate into food from PVC food packaging, and some of them are especially suspected as endocrine disruptors. In this study, Simultaneous analysis of 13 phthalates and 9 adipates were carried out by dual-column gas chromatography system equipped wi two FID detectors for rapid confirmation and quantification. The Proposed method was validated with > 0.993 of linearity in the ranges of 10-500 mg/l, < $3.5\%$ RSD of reproducability in 10 inter-days sample preparations, and > $98.1\%$ of recoveries for all the plasticizers. DEHA was detected in all the 3 PVC wraps at levels of 176.9-198.5mg/g. Among the 51 samples of PVC gaskets, the targeted plasticizers were detected in 41 samples. Of these plasticizer detected samples,40 contained DIDP at the levels of 157.3-374.7 mg/g and one contained DMP at the levels of 165.6 mg/g. Also, some plasticizers were detected in other packaging materials such as PET, PP, PE, Pulp. But it might be attributed to contamination in manufacturing.

  • The purpose of this study was to investigate the seasonal variation of PAHs and to estimate their source characteristics in Taegu area. To do this, four sampling sites were selected to represent an industrial, a traffic, a traffic & residential, and a residential area in Taegu. Total of 72 samples had been collected from January, 1999 to September, 1999 on glass micro fiber filters by high volume air sampler. The PAHs in the total suspended particulate were extracted by a soxhlet process with dichloromethane and analyzed by GC/MSD, GC/FID. A statistical analysis was performed for the PAHs data set using a principal component analysis to derive important factor inherent in the interactions among the variables. The specific conclusions of this research are: 1) There was a significant seasonal and local variation in the atmospheric concentration of PAHs. The seasonal variation is winter>spring>Fall>summer, and the local variation is industrial>traffic>graffic & residential>residential area. 2) To evaluate the correlation between a measured PAHs and other affecting factors such as air pollutant concentration and meterological data, statistical analysis was performed. PAHs have negative correlation with temperature (r=-0.593, p<0.05), radiation(r=-0.535, p<0.05), and O3(r=-0.719, p<0.05), but have positive correlation with NO(r=0.615, p<0.05) 3)Finally, multivariate analysis was performed for the PAHs dat set to identify and to estimate the source contributions of PAHs. According to results of statistical analysis, it could be identifies as three factors such as vehicular/gasoline, vehicular/diesel, and combustion in Taegu area.

Development of Analytical Method for Propylene Glycol in Foods (식품 중 프로필렌글리콜의 분석법 개발)

  • Standardized method based on extraction, filtration, and gas chromatography (GC) was developed far propylene glycol analysis to set hygienic norm of safety measure for foods under governmental control. Various columns were tested fur propylene glycol analysis by GC with flame ionization detector. Known amount of propylene glycol was spiked into wheat flour dough and analyzed by developed method. Results showed 101.60% recovery rate for propylene glycol with HP-5 column. Reproducibility test of standards recorded 0.30 for standard variation and 0,42% for relative variation. Using analytical method established, contents of propylene glycol in more than hundred different foods were monitored. Propylene glycol was detected in most foods, indicating propylene glycol is not only commonly added during food preparation, but also is contained naturally in food.

  • Comparisons of enzymatic method, gas chromatographic method, and the Iatroscan method for the determination of cholesterol in each parts of Korean squid were undertaken. The Iatroscan method was the most suitable procedure for the rapid and simple determination of net cholesterol concentration in parts of Korean squid. 5$\alpha$-cholestane is used as a good internal standard. The cholesterol contents in body, leg, viscera, eye, skin, and liver part of Korean squid, Todarodes pacificus STEEN STRUP by Iatroscan method were 178.9, 321.4, 168.9, 159.5, 608.8 and 634.2 mg%, respectively.
