The composite membrane for direct methanol fuel cell (DMFC) was developed using $H_3O^+-{\beta}"-Al_2O_3$ powder and perfluorosulfonylfluroride copolymer (Nafion) resin. The perfluorosulfonylfluroride copolymer (Nafion) resin was mixed with $H_3O^+-{\beta}"-Al_2O_3$ powder and it was made to sheet form by hot pressing. The electrodes were prepared with 60 wt% PtRu/C and 60wt% Pt/C catalysts for anode and cathode, respectively. The morphology and the chemical composition of the composite membrane have been investigated by using SEM and EDXA, respectively. The composite membrane and $H_3O^+-{\beta}"-Al_2O_3$ were analyzed by using FT-IR and XRD. The methanol permeability of the composite membranes was also measured by gas chromatography (GC). The performance of the MEA containing the composite membrane (2wt% $H_3O^+-{\beta}"-Al_2O_3$) was higher than that of normal pure Nafion membrane at high operating temperature (e.g. $110^{\circ}C$), due to the homogenous distribution of $H_3O^+-{\beta}"-Al_2O_3$, which decreased the methanol permeability through the membrane and enhanced the water contents in the composite membrane.

Background and Objectives: Human induced pluripotent stem cell (hiPSC)-derived cardiomyocyte (CM) hold great promise as a cellular source of CM for cardiac function restoration in ischemic heart disease. However, the use of animal-derived xenogeneic substances during the biomanufacturing of hiPSC-CM can induce inadvertent immune responses or chronic inflammation, followed by tumorigenicity. In this study, we aimed to reveal the effects of xenogeneic substances on the functional properties and potential immunogenicity of hiPSC-CM during differentiation, demonstrating the quality and safety of hiPSC-based cell therapy. Methods and Results: We successfully generated hiPSC-CM in the presence and absence of xenogeneic substances (xeno-containing (XC) and xeno-free (XF) conditions, respectively), and compared their characteristics, including the contractile functions and glycan profiles. Compared to XC-hiPSC-CM, XF-hiPSC-CM showed early onset of myocyte contractile beating and maturation, with a high expression of cardiac lineage-specific genes (ACTC1, TNNT2, and RYR2) by using MEA and RT-qPCR. We quantified N-glycolylneuraminic acid (Neu5Gc), a xenogeneic sialic acid, in hiPSC-CM using an indirect enzyme-linked immunosorbent assay and liquid chromatography-multiple reaction monitoring-mass spectrometry. Neu5Gc was incorporated into the glycans of hiPSC-CM during xeno-containing differentiation, whereas it was barely detected in XF-hiPSC-CM. Conclusions: To the best of our knowledge, this is the first study to show that the electrophysiological function and glycan profiles of hiPSC-CM can be affected by the presence of xenogeneic substances during their differentiation and maturation. To ensure quality control and safety in hiPSC-based cell therapy, xenogeneic substances should be excluded from the biomanufacturing process.
Identifying the extracellular metabolites of microorganisms in fresh vegetables is industrially useful for assessing the quality of processed foods. Pectobacterium carotovorum subsp. carotovorum (PCC) is a plant pathogenic bacterium that causes soft rot disease in cabbages. This microbial species in plant tissues can emit specific volatile molecules with odors that are characteristic of the host cell tissues and PCC species. In this study, we used headspace solid-phase microextraction followed by gas chromatography coupled with mass spectrometry (HS-SPME-GC-MS) to identify volatile compounds (VCs) in PCC-inoculated cabbage at different storage temperatures. HS-SPME-GC-MS allowed for recognition of extracellular metabolites in PCC-infected cabbages by identifying specific volatile metabolic markers. We identified 4-ethyl-5-methylthiazole and 3-butenyl isothiocyanate as markers of fresh cabbages, whereas 2,3-butanediol and ethyl acetate were identified as markers of soft rot in PCC-infected cabbages. These analytical results demonstrate a suitable approach for establishing non-destructive plant pathogen-diagnosis techniques as alternatives to standard methods, within the framework of developing rapid and efficient analytical techniques for monitoring plant-borne bacterial pathogens. Moreover, our techniques could have promising applications in managing the freshness and quality control of cabbages.
Xylanase has been used extensively in the industrial and agricultural fields. However, the low-yield production of xylanase from native species cannot meet the increasing demand of the market. Therefore, improving the heterologous expression of xylanase through basic gene optimization may help to overcome the shortage. In this study, we synthesized a high-GC-content native sequence of the thermostable xylanase gene xynB from Streptomyces olivaceoviridis A1 and, also designed a slightly AT-biased sequence with codons completely optimized to be favorable to Pichia pastoris. The comparison of the sequences' expression efficiencies in P. pastoris X33 was determined through the detection of single-copy-number integrants, which were quantified using qPCR. Surprisingly, the high GC content did not appear to be detrimental to the heterologous expression of xynB in yeast, whereas the optimized sequence, with its extremely skewed codon usage, exhibited more abundant accumulation of synthesized recombinant proteins in the yeast cell, but an approximately 30% reduction of the secretion level, deduced from the enzymatic activity assay. In this study, we developed a more accurate method for comparing the expression levels of individual yeast transformants. Moreover, our results provide a practical example for further investigation of what constitutes a rational design strategy for a heterologously expressed and secreted protein.

In this study, conditions for extraction of functional component from Nypa fruticans wurmb was optimized. The yield by extraction with 30% ethanol (LE30, 42.12%) was higher than those hot water extraction (LDW 33.32%), 50% ethanol (LE50, 40.12%) and 70% ethanol (LE70, 34.5%). The extract was purified and analyzed by GC MS. The prevailing compounds found in extract were Cyclodecasiloxane-, pentadecanoic acid, -eicosane, undecanal and tridecanoic acid. The presence of saturated and unsaturated fatty acids in ethanolic extract vindicate the use of this plant to treat many diseases in traditional medicine. The total phenolic contents in the LDW, LE30, LE50, LE70 extract were 128±1.65 mg/g, 205±2.3 mg/g, 210±4.23 mg/g and 180±5.6 mg/g, respectively. The DPPH was highest in LE70 extract (1,000 ㎍/mL, 81.14%), ABTS was highest in LE50 extract (1,000 ㎍/mL, 84.14%). The protective effects against oxidative stress in raw 264.7 cell imparted by the LE50 extract was better than those imparted by the other extracts. The findings of the present study suggest that 50% ethanol is best solvent for extraction of Nypa fruticans Wurmb, considering yield, polyphenol content, and antioxidant activities with extraction cost.
Ipfencarbazone은 tetrazolinone계 화합물로 잡초 세포막의 주성분인 장쇄지방산(VLCFAs) 합성을 억제하여 세포 분열을 저해하는 작용을 하는 제초제이다. 본 연구에서는 국내 신규농약 ipfencarbazone에 대한 안전관리를 위해 농산물 중 ipfencarbazone의 잔류분석법을 개발하였다. 농산물 시료(현미, 대두, 고추, 감귤, 감자) 중 ipfencarbazone을 acetone으로 추출한 뒤 n-hexane을 이용해 분배하고 silica SPE 카트리지를 통한 흡착크로마토그래피법을 이용해 정제하였다. 이를 GC-ECD를 이용해 분석해 정량평가를 하였으며, 재확인을 위해 GC/MS로 분석하였다. 분석법의 직선성은 농도 대비 피크면적과의 결정계수($r^2$)가 0.999 이상으로 우수하였으며, 검출한계 및 정량한계는 각각 0.003, 0.01 mg/kg으로 높은 감도를 나타냈다. 농산물 시료 중 모든 처리구에서 평균 회수율은 80.6%-112.3%(RSD 10% 이내)였고, 경인청과의 실험실간 검증결과에서도 95.1-106.1%를 나타냈다. 모든 결과를 종합해볼 때 개발된 분석법은 모두 CODEX 가이드라인(CAC/GL 40, 2003)의 기준이내를 만족하는 값으로 확인되었다. 따라서 개발된 분석법의 낮은 검출한계 및 정량한계, 우수한 직선성, 회수율 시험을 통한 양호한 정밀성 및 재현성 등을 종합해볼 때 이는 농산물 중 ipfencarbazone을 분석하기 위한 공정분석법으로 사용되기에 적합할 것으로 판단된다.
본 연구에서는 poly (3-hydroxyalkanoate) (PHA)의 생산 비용을 줄이기 위해, 토양에서 분리된 균주 Pseudomonas sp. EML8을 이용하여 폐 튀김유(waste frying oil, WFO)를 단일 탄소원으로 하여 균주의 최적 생장 및 PHA의 생합성 조건을 확립하였다. WFO를 단일 탄소원으로 이용하여 Pseudomonas sp. EML8에 의해 생합성된 PHA를 gas chromatography (GC)와 GC mass spectrometry로 분석한 결과, 7.28 mol% 3-hydrxoyhexanoate, 39.04 mol% 3-hydroxyoctanoate, 37.11 mol% 3-hydroxydecanoate 및 16.58 mol% 3-hydoryxdodecanoate의 단량체로 이루어진 medium-chain-length PHA (mcl-PHAWFO)라는 것을 확인하였다. 플라스크로 배양한 결과, Pseudomonas sp. EML8의 최대 건조세포중량(dry cell weight, DCW) 및 mcl-PHAWFO의 최대 생산수율(g/l)은 WFO 20 g/l, (NH4)2SO4 0.5 g/l, pH 7 및 25℃의 조건에서 확인되었다. 이 결과를 바탕으로 3 l 발효기를 이용하여 48시간 배양한 후에 가장 높은 DCW, mcl-PHAWFO의 함량 및 mcl-PHAWFO의 생산수율(3.0 g/l, 62 wt% 및 1.9 g/l)을 얻었다. 대조군인 신선한 튀김유(fresh frying oil, FFO) 20 g/l를 탄소원으로 사용하여 이와 유사한 DCW, mcl-PHAFFO의 함량 및 mcl-PHAFFO의 생산수율(2.7 g/l, 62 wt%, 1.6 g/l)을 확인했다. 겔 투과 크로마토그래피 분석을 통해 mcl-PHAWFO 및 mcl-PHAFFO의 평균 분자량이 165-175 kDa인 것을 확인했으며, 열중량을 분석한 결과, mcl-PHAWFO 및 mcl-PHAFFO는 각각 260 및 274.7℃의 분해온도값을 보여주었다. 결론적으로 본 연구에서는 Pseudomonas sp. ML8과 WFO는 mcl-PHA의 생산을 위한 새로운 균주와 탄소원으로서의 사용 가능성을 확인하였다.
5,8-Epidioxycholest-6-en-3-ol (1), cholesterol (2), glycerol 1-palmitate (3) and glycerol 1,3-dioleate-2-stearate (4) were isolated from the methanol extract of the sea urchin Diadema setosum, which was collected from the Halong sea, Vietnam. Chemical structures were established based on extensive 1D, 2D-NMR, FAB-MS, EI-MS spectroscopic data and GC-MS analysis. The NMR spectral data of compound 1 were reassigned by using HMQC and HMBC. Compound 1 was found to have strong cytotoxic effect against various cancer cell lines, such as KB ($IC_{50}$, 2.0 $\mu\textrm{g}$/mL), FL($IC_{50}$, 3.93 $\mu\textrm{g}$/mL), and Hep-2 ($IC_{50}$, 2.4 $\mu\textrm{g}$/mL) by in vitro assay.
Objectives : In this study, we investigated the combination effects of Cinnamomi cortex Ethanol Extract (CcEE) and hyperthermia in the human AGS gastric cancer cell line. Methods : AGS cells were treated with the indicated concentrations of CcEE (0, 50 or $60{\mu}g/mL$) for 1h prior to hyperthermia. And then incubated for a further 30 min at the indicated temperatures (37, 42 or $43^{\circ}C$) in a humidified incubator containing 5% $CO_2$ or a thermostatically controlled water bath for hyperthermia. The cell viability was measured by MTT assay, Morphology assay and Trypan blue assay. To investigate the possible molecular signaling pathways, the activation of mitogen-activated protein kinase (MAPK) proteins (ERK, p38 and JNK) and expression of various anti-apoptotic proteins such as Caspase-3, Caspase-9, p53, Cyclin D1 and MMP-2 were assessed by Western blot analysis. In addition, Annexin V and 7-amino-actinomycin D (7-AAD) staining was performed to examine the apoptotic mechanism. Results : Combination of CcEE with hyperthermia effectively suppressed the cell viability and changed cellmorphology compared with CcEE or hyperthermia treatment alone. Combined treatment also abated the expression of Caspase-3, Caspase-9, Cyclin D1 and MMP-2. Whereas, the expression level of p53 was up-regulated by co-treatment. Moreover, combination treatment enhanced phosphorylation of ERK, p38 and JNK. In addition, this combination increased anti-cancer effect by inducing cell death through the apoptosis. Conclusions : Taken together, all these findings suggest that the combination treatment with CcEE and hyperthermia may have therapeutic potential as a promising approach to patients with stomach cancer.
Bacillus species predominantly outgrown in a night soil treatment system were isolated and characterized. Cell interactions took place among them and cell population changed under various culture conditions. Maximum removal of $NH_4\;^+-N$ and cell production occurred under the conditions of 30% DO and C/N ratio of 8. Additions of 0.8% peptone and 0.3% yeast extract to a basal medium influenced the growth of isolates and the removal of $NH_4\;^+-N$ in flask culture, and metal ions such as Ca, Fe and Mg also did. During the flask experiment of nitrogen removal under an aerobic condition, active nitrification by the isolates occurred largely in 1 h with the decrease of COD and alkalinity destructed was only 74.6% of theoretical value. From the nitrogen balance, the percentage of nitrogen lost in the flask culture was estimated to be 29.0%. This conversion of ammonia to $N_2$ under an aerobic condition was confirmed by GC analysis. The B3 process using the Bacillus species seemed to have some economic advantage.
