Engine oil is a substance used for the lubrication of internal combustion systems. However, in some case, defects in engine systems may contaminate engine oil with fuel. Contaminated engine oil can cause problems in the normal functioning of a vehicle. In this study, we investigate the functional properties of engine oil contaminated with diesel fuel. The test results indicate that the engine oil contaminated with diesel fuel has low flash point, pour point, density, kinematic viscosity and cold cranking simulator value. The contaminated engine oil which has low plash point can cause fire and explosion accident. Furthermore, a four ball test indicates that the contaminated engine oil increases wear scar to poor lubricity. Moreover, we investigate the GC pattern using SIMDIST (simulated distillation) for determination of diesel in engine oil. The SIMDIST analytic result, diesel was detected at earlier retention time than engine oil in chromatogram. Thus the SIMDIST method can define whether engine oil is contaminated by diesel fuel or not. We can use the SIMDIST method for the diagnosis of oil condition instead of analyzing other physical properties that require many analytic instruments, large volume of oil sample and long analysis time.
The purpose of this study was to analyze harmful heavy metals, sulfur dioxide and residual pesticides in 30 kinds of oriental medical materials. This study was carried out on 30 samples of oriental medical materials. The examination's methods fallow the guideline of Food and Drug Administration. Inductively Coupled Plasma Mass Spectrometer (ICP-OES) was used to analyze lead, arsenic, cadmium. Mercury was analyzed by amalgamation method. A modified Monier-Williams method was used to analyze sulfur dioxide. The Gas Chromatography / Electron Capture Detector (GC/ECD) was used to analyze residual pesticides. Most of the results were within normal limits at baseline. but Cadmium was detected 0.523 mg/kg more than 0.3 mg/kg in Cinnamomi ramulus and Endosulfan was detected 11.595 mg/kg more than 0.2 mg/kg in Alismatis rhizoma. We needs to keep the quality of herbal medicines assessing and controling for safety of herbal medicines.
Extractives of garlic (Allium scorodorpasum) and green pepper (Capsicum annuum L.) were used to prevent and exterminate rice weevils which contaminate stored rice. The extractives exhibited great efficacy against Sitophilus oryzae and Tribolium castaneum by annihilating all the weevils contained in a bottle. Furthermore, kraft paper coated by 5% extractives also displayed good effectiveness against the rice weevils. When rice weevils were stored with rice in a sack made of extractives-coated kraft paper, the number of living insects was sharply decreased according to elapsed time. In particular, 1,4-benzoquinone, which is very harmful to human, was detected from rice contaminated by Sitophilus oryzae and Tribolium castaneum through the analysis of GC/MS.
본 연구는 울산소재 석유공장의 건조시설에서 발생하는 가스를 스크러버(Scrubber)로 최종 처리하여 배출되는 저 농도의 유해가스와 산 처리 시설에서 발생되는 고 농도로 발생하는 유해가스를 대상으로 연구하였다. 공정별로 벤젠(Benzene), 톨루엔(Toluene), 자일렌(Xylene) 및 총휘발성유기화합물(TVOCs)을 대상으로 측정하였으며 농도 측정은 TVOCs 측정기와 GC-MSD를 이용하여 SAP 전 후단의 TVOCs와 BTX의 농도를 측정, 분석하여 제거효율을 평가하였고, SAP 반응기는 5 단계로 구성하여 실험을 수행하였다. 슬라이딩 아크 프라즈마(slidind arc plasma) 반응기의 단수별에 따른 TVOCs 농도 변화는 유입 TVOCs 농도의 변화에 따른 처리효율 결과 481 ppm 미만에서 94.83%, 481 ppm 이상일 경우에는 1단에서는 89.07% 2단에서는 약 91% 이상으로 처리되는 것을 확인할 수 있었다. 본 연구의 결과는 석유화학공정 및 제조시설에서 SAP 공정을 통한 VOCs 제거에 대한 연구 및 기술개발의 기초가 되어 VOCs의 보다 안정적인 처리와 다양한 기술개발에 효과가 있을 것으로 기대해 본다.
식물체의 향기성분을 분리하는데 사용되는 몇가지 방법을 비교하기 위해서 headspace(purge & trap)법, simultaneous distillation extraction(SDE)법, steam distillation법, 용매추출법 등으로 분리된 휘발성 성분의 조성차이를 분석하여 각각의 사용된 방법들의 특징을 비교하였다. 휘발성 성분 분석시료로는 Anise(Pimpinella anisum L.)씨앗을 사용하였으며 분리된 휘발성분중 GC/MSD를 이용하여 43종의 성분을 확인하였다. Headspace (purge & trap)법에서는 휘발성이 강한 2-butanol, pentanal, 3-hexen-1-ol 등과 같은 alcohol류 및 aldehyde류 성분들을 주요성분으로 확인할 수 있었으며, 용매추출에서는 비점이 높은 성분들인 myristic acid, oleic acid 등의 acid류 성분들과 ethyl hexadecanoate, ethyl octadecanoate 등과 같은 ester 화합물들이 주요성분으로 확인되었다. SDE법과 steam distillation법에서 추출된 성분의 조성은 유사하였으며 terpene 화합물들이 주요성분으로 확인되었다. Anise의 주성분인 anethole은 SDE법, headspace(purge & trap)법으로 추출하였을때 가장 많이 추출되었다.
In this study, antifungal activity of essential oils of Cymbopogon citratus and Ocimum basilicum and two fungicides Mancozeb and Metalaxyl-Mancozeb in six different concentrations were investigated for controlling three species of Phytophthora, including P. capsici, P. drechsleri and P. melonis on pepper, cucumber and melon under in vitro and greenhouse conditions, respectively. Under the in vitro condition, the median effective concen- tration ($EC_{50}$) values (ppm) of plant essential oils and fungicides were measured. In greenhouse, soil infested with Phytophthora species was treated by adding 50 ml of essential oils and fungicides (100 ppm). Disease severity was determined after 28 days. Among two tested plant essential oils, C. citratus had the lowest $EC_{50}$ values for inhibition of the mycelial growth of P. capsici (31.473), P. melonis (33.097) and P. drechsleri (69.112), respectively. The mean $EC_{50}$ values for Metalaxyl-Mancozeb on these pathogens were 20.87, 20.06 and 17.70, respectively. Chemical analysis of plant essential oils by GC-MS showed that, among 42 compounds identified from C. citratus, two compounds ${\beta}$-geranial (${\alpha}$-citral) (39.16%) and z-citral (30.95%) were the most abundant. Under the greenhouse condition, Metalaxyl-Mancozeb caused the greatest reduction in disease severity, 84.2%, 86.8% and 92.1% on melon, cucumber, and pepper, respectively. The C. citratus essential oil reduced disease severity from 47.4% to 60.5% compared to the untreated control ($p{\leq}0.05$). Essential oils of O. basilicum had the lowest effects on the pathogens under in vitro and greenhouse conditions. These results show that essential oils may contribute to the development of new antifungal agents to protect the crops from Phytophthora diseases.
Because of high population diversity in soil microbial communities, it is difficult to accurately assess the capability of biodegradation of toxicant by microbes in soil and sediment. Identifying biodegradative microorganisms is an important step in designing and analyzing soil bioremediation. To remove non-important noise information, it is necessary to selectively enrich genomes of biodegradative microorganisms fromnon-biodegradative populations. For this purpose, a stable isotope probing (SIP) technique was applied in selectively harvesting the genomes of biphenyl-utilizing bacteria from soil microbial communities. Since many biphenyl-using microorganisms are responsible for aerobic PCB degradation In soil and sediments, biphenyl-utilizing bacteria were chosen as the target organisms. In soil microcosms, 13C-biphenyl was added as a selective carbon source for biphenyl users, According to $13C-CO_2$ analysis by GC-MS, 13C-biphenyl mineralization was detected after a 7-day of incubation. The heavy portion of DNA(13C-DNA) was separated from the light portion of DNA (12C-DNA) using equilibrium density gradient ultracentrifuge. Bacterial community structure in the 13C-DNAsample was analyzed by t-RFLP (terminal restriction fragment length polymorphism) method. The t-RFLP result demonstates that the use of SIP efficiently and selectively enriched the genomes of biphenyl degrading bacteria from non-degradative microbes. Furthermore, the bacterial diversity of biphenyl degrading populations was small enough for environmental genomes tools (metagenomics and DNA microarrays) to be used to detect functional (biphenyl degradation) genes from soil microbial communities, which may provide a significant progress in assessing microbial capability of PCB bioremediation in soil and groundwater.
Seventeen microbial species including 10 fungal taxa, two yeasts and five bacteria, were isolated from freshly prepared orange, guava and banana juices kept in open bottles at room temperature for 7 days. Eight different essential oils, from local herbs, were tested for their antimicrobial activity against these test organisms. The essential oils of Cymbopogon citratus, Ocimum basilicum and Origanum majorana were found to be highly effective against these microorganisms. Aspergillus niger, A. flavus and Saccharomyces cerevisiae, the most prevalent microorganisms in juice, showed the highest resistance against these essential oils. GC-MS analysis showed that while e-citral, a'-myrcene, and z-citral represent the major components (75.1 %) of the essential oil of Cymbopogon citratus; bezynen,l-methyl-4-(2-propenyl), 1,8-cineole and trans-a'-bisabolene were the main components (90.6%) of Ocimum basilicum; whereas 3-cyclohexen-l-0l,4-methyl-l(l-methylethyl)-(CAS), c-terpinene and trans-caryophyllene represent the major components (65.1%) of Origanum majorana. These three essential oils were introduced into juices by two techniques namely, fumigation and direct contact. The former technique showed more fungicidal effect than the latter one against A. flavus, A. niger, and S. cerevisiae. The essential oil of Cymbopogon citratus by comparison to other test oils showed the strongest effect against these fungi with a minimum inhibitory concentration of $1.5\;{\mu}l/ml$ medium and a sublethal concentration of $1.0\;{\mu}l/ml$. The antimicrobial activity of this oil is thermostable at $121^{\circ}C$ for 30 min.
Silicon is one of the key elements for healthy growth and development in rice crops. We analyzed the effect of silicon(Si) on some growth parameters, plant mineral contents, and bioactive gibberellins in three rice cultivars. Silicon was applied at the rates of 0 kg/0.1ha(control), 40 kg/0.1ha, and 80 kg/0.1ha throughout the course of experiment. Plant growth parameters were enhanced by the application of elevated Si, though plant height and culm length were more favorably affected than the respective dry weights. The plant mineral contents analyzed also increased in treatments where Si was applied without potassium, demonstrating that Si application promotes the absorption of these minerals in rice crops. The endogenous gibberellins measured in our study showed that $GA_1$ is the only bioactive GA form present in rice seedlings. The endogenous $GA_1$ and its precursor $GA_{20}$ contents increased after Si application. However, this increase in endogenous $GA_1$ and $GA_{20}$ contents, and plant growth parameters were different according to the rice cultivars. Our results indicate that Si is a beneficial element in rice nutrition and that different cultivars of Oryza sativa show differential responses to Si nutrition in terms of their growth and development.
미생물을 이용한 인삼 재배지 내 잔류 tolclofos-methyl의 효과적 분해를 목적으로, tolclofos-methyl에 대한 분해능을 보이는 미생물을 선발하였다. 선발된 미생물은 16S rDNA 염기서열분석을 통하여 Sphingomonas 속으로 동정되었다. 선발 미생물 Sphingomonas sp. 224는 1/10 농도의 LB 배지에 함유된 20 mg/L 농도의 tolclofos-methyl을 배양 72시간 이내에 95% 이상 분해하는 것으로 확인되었다. 또한 이 미생물이 tolclofos-methyl을 분해하여 얻어지는 산물로 2,6-dichloro-4-methyl phenol이 확인됨에 따라 미생물이 생산하는 가수분해 효소에 의한 분해 경로를 가지는 것으로 추정되었다. Tolclofos-methyl 분해 미생물 Sphingomonas sp. 224를 인삼경작지 토양에 처리하여 이들 토양에 잔류되어 있는 tolclofos-methyl에 대한 분해능을 확인 한 결과, 20 mg/Kg 농도의 토양 잔류 tolclofos-methyl에 대하여 14일 이내에 약 50%의 분해력을 보이는 것으로 확인되었다. 이것은 단일 미생물을 이용한 배지 및 토양 내 tolclofosmethyl의 생분해 효과를 처음으로 확인한 연구 결과이다.
