Studies on the Volatile Flavor Components and Biochemical Characterizations of Artemisia princeps and A argyi (강화쑥의 생화학적 특성 및 휘발성 향기성분에 관한 연구)

  • This study has attempted to examine the effect of Artemisia princeps and A. argyi on liver function-related enzymes in rats with $CCl_4$ adminisration. The activities of serum aspartate aminotransferase(AST), alanine aminotransferase(ALT) and alkaline phosphatase(ALP) from A. princeps were decreased by 33, 23 and $19\%$, respectively, compared to control. The activities of AST, ALT and ALP from A. argyi were decreased by 37, 33 and $26\%$, respectively. Total phenol contents were 10.2 mg/mL and 4.7 mg/mL in A. princeps, and A. argyi, respectively. Also, flavonoid contents were $6.1\;mg\%\;and\;3.6\;mg\%$ in A. princeps, and A. ar효i, respectively. Ethanol extract from A. argyi showed higher electron donating ability toward DPPH than A. princeps. A total of 31 volatile components(3 hydrocarbons, 10 terpenes, 5 carbonyls, 8 alcohols and 5 esters) were indentified in A. princeps, and A. argyi. The major volatile components of A. princeps were $\delta$-3-carene($2.2\%$) in terpenes and nerolidol($0.9\%$) in alcohols. The major volatile components of A. argyi were eugenol($1.4\%$) in alcohols and thyl pentadecanoate($1.1\%$) in esters.

The Sensory Properties and Flavor Components of the White Bread Added with Arrowroot juice (칡즙 첨가 식빵의 관능적 특성과 향기성분)

  • The sensory properties and flavor components of the white bread added with arrowroot juice. The lightness of bread crumb decreased significantly as arrowroot juice was added. While the yellowness increased slightly, redness increased remarkably. The hardness, chewiness and adhesiveness of the white bread added with arrowroot juice increased more than those of the control bread, but they had no statistical significance. While the gumminess increased significantly and springiness decreased significantly, the cohesiveness did not indicate significant differences among the comparison groups. In sensory evaluation, the texture, flavor and sweetness did not indicate significant difference among the comparison groups, while the color and overall acceptability indicated significant difference. The optimum concentration of arrowroot juice in the white bread was 25% based on the sensory evaluation scores. The main flavors components of the white bread added with 25% arrowroot juice were compounds translated by arrowroot juice and the compounds formed by amino-carbonyl reaction. The translated flavors were methoxy phenol, ${\beta}-damascenone$, benzylcyanide, and menthofuran. The compounds formed by amino-carbonyl reaction were alkyl pyrazines, pyrroles and furans.

Sample Preparation for Quantitative Determination of Bisphenols in Canned Beverages Coated with Epoxy by RP-HPLC (RP-HPLC 에 의한 에폭시 통조림관 음료 중 비스페놀류의 분석을 위한 시료의 전처리)

  • A suitable method of sample treatments to minimize the analytical interferences was presented in order to determine bisphenols [bisphenol F (BPF), bisphenol A (BPA), bisphenol F diglycidyl ether (BFDGE), and bisphenol A diglycidyl ether (BADGE)] in various canned beverages coated with epoxy resin by the reversephase high performance liquid chromatography (RP-HPLC) equipped with a fluorescence detector and the gas chromatography with mass selective detection. The recovery test of bisphenols was performed using 1, 5, and 10 ${\mu}g/L$ bisphenols spiked beverages with the combined technique of the solid-phase extraction (SPE) and the liquid-phase extraction (LPE). Both BPA and BADGE showed quite adequate resolutions in HPLC-chromatograms. The recoveries of BPA obtained by LPE with diethyl ether were higher than those obtaind with methylene chloride on coffee, shikhye and fruit juice. For cola and tea, the recoveries of BPA obtaind by SPE were higher than those by LPE with diethyl ether. The recoveries of BADGE were less than those of BPA for all beverage samples treated by either SPE or LPE method. In survey of bisphenols for eighteen commercial canned beverage samples, BPA contents of coffee, cola, tea, shikhye, and fruit juice were in the range of $1.3{\sim}11.6,\;0.5{\sim}0.9,\;1.0{\sim}1.3,\;2.4{\sim}7.9$, and $3.0{\sim}3.4\;{\mu}g/L$, respectively, but there was no detection of BPA in beer sample.

Volatile Compounds Analysis of Certified Traditional Doenjang (전통식품 품질인증 된장의 향기성분 분석)

  • The purpose of this study was to provide a basis for the management of traditional Doenjang by analyzing characteristics of volatile compounds in local Doenjang certified as a traditional food. The main compounds in Doenjang were acids, esters, aldehydes, and pyrazines, whereas relatively high intensities of acetic acid, ethyl alcohol, benzaldehyde, ethyl acetate, ethyl 2-methyl butanoate, 2,5-dimethyl pyrazine, and tetramethylpyrazine were detected among identified compounds. The analysis revealed that the composition of basic volatile compounds in Doenjang was similar, but isovaleric acid, 2-methylbenzaldehyde, tetramethylpyrazine, benzaldehyde, ethyl alcohol, ethyl caprylate, furfural and butanoic acid can serve as marker compounds for quality evaluation since they were specifically abundant in only some kinds of Doenjang. As a result, the quality status of Doenjang certified as a traditional food was determined by constructing a database of the volatile compounds, which can be suggested as a quality control method.

Inhibition of an Inducible Nitric Oxide Synthase Expression by a Hexane Extract from Perilla frutescens cv. Chookyoupjaso Mutant Induced by Mutagenesis with Gamma-ray (방사선 유도 돌연변이 약용들깨 핵산 추출물의 Inducible Nitric Oxide Synthase 저해활성)

  • In earlier investigations, seeds of Perilla frutescens(L.) Britt. cv. Chookyoupjaso were irradiated with 200 Gy gamma ray to generate mutagenesis. The aim of this study is to investigate the effects of a hexane extract from Perilla frutescens (L.) Britt. cv. Chookyoupjaso mutant 45 on the actions of anti-inflammatory activity on inducible nitric oxide synthase, and an identification of the major active compound. The hexane extract from P. frutescens exhibited activity of inhibition of a NO production ($IC_{50}$, $295.1{\mu}g\;ml^{-1}$). The hexane extract was further divided into subfractions by silica-gel chromatogarphy. Inhibition of the NO production by various fractions was assayed in LPS-stimulated RAW 264.7 cells. Among the seven fractions, the 5th fraction was the most effective ($IC_{50}$, $19.5{\mu}g\;ml^{-1}$). The 5th fraction suppressed the expression of protein of iNOS in LPS-induced RAW 264.7 cells, and GC/MS analyses showed that isoegomaketone is a major bio-active compound in the 5th fraction. The result indicated that isoegomaketone has a good potential to be developed as an anti-inflammation agent.

Fermentation and Quality Characteristics of Yakju According to Different Rice Varieties (쌀 품종에 따른 약주의 발효 및 품질 특성)

  • This study was performed to improve quality of traditional Yakju and compares quality of Yakju according to different rice cultivars. The pHs of Ilmi Yakju and Hanarum Yakju showed 4.14 and 4.07, respectively and the other Yakju's pHs were ranged from 3.92 to 3.98. The content of total acid of Yakju using Indica rice imported from Thailand was the highest among the samples. The major components of free sugar was glucose and the highest content of total free sugars was found in Indica Yakju. The content of reducing sugars in Yakju using Indica rice was the higher than other samples. The ethanol content of Hanarum Yakju showed higher than those Yakju's and the lowest ethanol content found for the indica Yakju. The volatile compounds from the rice Yakju were identified by GC-MS. Twenty-one volatile compounds were found in rice Yakju. And the major volatile compounds were ethanol, acetic acid, 1-methyl-1-propanol, 2-methyl-butane, 3-methyl-1-butanol, iso-amylalcohol and 1-hexanol from Yakju. As the result of sensory test, higher scores for smell and color were found for the Yakju used Hanarum Yakju. The highest score sweat and acid were found for the Anda rice Yakju in sensory test. The result of sensory evaluation indicated that Hanarum Yakju and Anda Yakju were better than the other samples, and the value of bitterness was no significant in this test.

Effect of Differences between Korean Ministry of Environment Standard and ISO Standard on Determination of Petroleum Hydrocarbon Content in Soil (토양오염공정시험기준과 국제표준간의 분석방법 차이가 유류 정량에 미치는 영향)

  • The main objective of this study is to assess the compatibility between Korean ministry of environment (KME) standard and ISO (KS I ISO) standard for the determination of BTEX and TPH content in soil. We carried out comparison analysis for both methods using CRM and matrix spiked samples. In case of GC-MS analysis for BTEX, we got statistically (significance level: 0.05) the same results from KME standard (ES 07600.1) and ISO standard (KS I ISO 15009). However, it showed statistically (significance level: 0.05) different results when TPH was analyzed by KME standard (ES 07552.1) and ISO standard (KS I ISO 16703). To clarify the reason why both methods produced different results for TPH content, we also did some additional experiments in terms of differences in extraction, clean-up and target hydrocarbon range. Extraction with polar and non-polar compounds mixed solvent (acetone+n-heptane) of KS I ISO 16703 showed higher extraction efficiency than with only non polar solvent (dichloromethane) extraction of ES 07552.1 by about 9%. While column type clean-up of KS I ISO 16703 showed the reduction in TPH content between before and after clean-up, batch type of clean-up of ES 07552.1 did not show any changes in TPH content through clean-up process. The target hydrocarbon range of ES 07552.1 and KS I ISO 16703 is $C_8{\sim}C_{40}$ and $C_{10}{\sim}C_{40}$, respectively. From this point of view, kerosene and JP-8 contaminated soil showed higher RPD (relative producibility deviation) values between results by both method than that of lubricant or diesel contaminated soil. The higher content of hydrocarbon ($C_8{\sim}C_{10}$) in kerosene and JP-8 played an important role in increasing RPD values in addition to the effects caused by different solvents and clean-up method. Consequently, it was concluded that both methods (ES 07552.1 and KS I ISO 16703) were not compatible.

Effects of Nitrogen Fertilization on Leaf Yield and Pyranocurmarine Accumulation in Angelica gigas Nakai

  • Angelica gigas Nakai is one of the most widely used herbal medicines and is known to have many pharmaceutical effects including an anti-oxidant, anti-cancer etc. This study was carried out to investigate an effect of fertilization on leaf yield, production of dry-matter and accumulation of pyranocurmarine compounds such as decursin (DE) and decursinol angelate (DA) in Angelica gigas Nakai. Effect of fertilization was determined from response surface regression equation composing of 2 by 3 factorial arrangement of urea, sodium dihydrogen phosphate and potassium chloride. Yield of leaf in Angelica gigas Nakai significantly increased until 100 days after transplanting. Production of leaf also tended to increase with increasing nitrogen fertilization. Model of regression equation showed that leaf production depended upon nitrogen ($Pr>{\mid}t{\mid}$ : 0.087, 0.256 and 0.079). Also, statistical results between nitrogen application level and production of dry-matter showed significant relationship (p<0.05) and contents of dry-matter was highest in 10 kg 10a-1 treatment on 24 Sep. Active compound isolated and purified from leaf and root of Angelica gigas Nakai was identified as DE and DA by gas chromatograph-mass spectrophotometry (GC-MS). Concentration of DA as prevalent compound in leaf was highest on 20 Aug. but decreased on 24 Sep. Amount of DE and DA accumulated in Angelica gigas Nakai significantly increased with growth stages and nitrogen level. The result of our investigation imply that nitrogen fertilization is important factor for production of leaf and accumulation of pyranocurmarine in Angelica gigas Nakai as a medicinal/food materials.

A Scientific Analysis of Gold Threads Used in Donggungbi-Wonsam(Ceremonial Robe Worn by a Crown Princess, National Folklore Cultural Heritage No.48) (동궁비 원삼에 사용된 금사의 과학적 분석)

  • This study identified material properties through scientific analysis on Jikgeumdan(satin with gold threads) from Donggungbi-Wonsam and the gold threads used in the embroidery. The Donggungbi-Wonsam's base of gold threads were estimated to have used mulberry fiber's Korean paper(Hanji) because non-wood-based fibers were observed. The X-ray spectrometer showed that the Tongsuseulan of Donggungbi-Wonsam was a flat gold thread of pure gold and Jikgeumdan of flat silver thread of its Saekdong and Hansam. High sulfur levels were detected in the flat silver thread, which appeared to have formed silver sulfide by either manufacturing process using sulfur or conservation environment. he dragon insignia's embroidery is also described as two types twisted gold threads; pure gold and alloying-gold and silver. while dragon insignia's border line is decorated with a twisted gold thread of pure gold. In particular, it was investigated that adhesives such as an animal glue, a protein-based compound by gas chromatography mass spectrometry. Additionally, XRF and Raman spectroscopy analysis on the mixture substances between the metal surface and the base paper of gold threads identified talc and quartz in the gold threads and Seokganju(hematite) in the flat silver threads.

A New Perilla Variety 'Bomnaeyeop 1 ho' with Laborsaving Plant Type (노력절약형 엽채소용 잎들깨 신품종 '봄내엽1호')

  • A new perilla cultivar, 'Bomnaeyeop 1 ho' was developed by, Kangwon National University in 2008. Its characteristics were investigated three times from 2006 to 2008 under optimal condition of cultivation. 'Bomnaeyeop 1 ho' can be characterized by dark green leaf color and tall plant height. This cultivar has more number of flower cluster and ear per cluster compare to 'Saeyeupsil' control. From result of GC/MS analysis, main compounds consist of perilla ketone, beta-caryophyllene and myristicin in 'Bomnaeyeop 1 ho' cultivar.
