Triacetin이 탄소복합 필터의 연기성분 흡착능에 미치는 영향

  • This study was conducted to evaluate the effect of triacetin(TA) treatment during manufacturing carbon dual filter for the adsorption of cigarette smoke components by activated carbons. The measurements were carried out by separation of activated carbon from carbon dual filter, and the specific surface area analyzed. The specific surface area of activated carbon from the domestic cigarette filter and from the foreign cigarette filter by degassing at 9$0^{\circ}C$ was 163$\pm$32$m^2$/g, and 16.6$\pm$1.9$m^2$/g, respectively. Those values were very lower than that of degassing at 35$0^{\circ}C$ (Domestic brand: 952$\pm$30$m^2$/g, and Foreign brand: 847$\pm$73$m^2$/g). By comparing the adsorption capacity of acetone and benzene with and without triacetin treated activated carbon, there was a 20% reduction of adsorption capacity by 5% triacetin treatment. Also, from the cilia toxicity test with carbon dual filter treated 0 % TA and 8 % TA, the cilia survival time was 706$\pm$74sec. and 603$\pm$64sec. for 0% TA and 8% TA, respectively. The delivery rate of vapour phase of cigarette smoke, which consists of main components of cilia toxicity, was higher at 8% TA filter than 0 % TA filter. Our results indicate that the treated TA covered the micro-pore of activated carbon, and then reduced specific surface area, finally, decreased the adsorption of vapour phase from cigarette smoke.

Analysis on the Frumkin Adsorption Isotherm of the Over-Potentially Deposited Hydrogen (OPD H) at the Polycrystalline Ni | Alkaline Aqueous Electrolyte Interface Using the Phase-Shift Method

  • The Frumkin adsorption isotherm of the over-potentially deposited hydrogen (OPD H) for the cathodic $H_2$ evolution reaction (HER) at the poly-Ni|0.05M KOH aqueous electrolyte interface has been studied using the phase-shift method. The behavior of the phase shift $(0^{\circ}\leq{\phi}\leq90^{\circ})$ for the optimum intermediate frequency corresponds well to that of the fractional surface coverage $(1\geq{\theta}\geq0)$ at the interface. The phase-shift method, i.e., the Phase-shift profile $(-{\phi}\;vs.\;E)$ for the optimum intermediate frequency, can be used as a new method to estimate the Frumkin adsorption isotherm $(\theta\;vs.\;E)$ of the OPD H for the cathodic HER at the interface. At the poly-Ni|0.05M KOH aqueous electrolyte interface, the rate (r) of change of the standard free energy of the OPD H with $\theta$, the interaction parameter (g) for the Frumkin adsorption isotherm, the equilibrium constant (K) for the OPD H with $\theta$, and the standard free energy $({\Delta}G_{\theta})$ of the OPD H with ${\theta}$ are $24.8kJ mol^{-1},\;10,\;5.9\times10^{-6}{\leq}K{\leq}0.13,\;and\;5.1\leq{\Delta}G_{\theta}\leq29.8kJ\;mol^{-1}$. The electrode kinetic parameters $(r,\;g,\;K,\;{\Delta}G_{\theta})$ depend strongly on ${\theta} (0{\leq}{\theta}{\leq}1)$.

Machanism of Cisplatin-induced Apoptosis and Bojungbangam-tang-mediated Anti-apoptotic Effect on Cell Proliferation in Rat Mesangial Cells (Cisplatin과 보정방암탕에 의한 백서 사구체 혈관사이세포의 세포사멸 기전 연구)

  • Cisplatin is a anti-neoplastic agent which is commonly used for the treatment of solid tumor. Cisplatin activates multiple signal transduction pathways involved in the stress-induced apoptosis in a variety of cell types. Previous study reported that cisplatin induces apoptosis through activation of ERK, p38 and JNK in rat mesangial cells, but apoptotic pathway remain known. The present study investigated the apoptotic pathway for cisplatin-indcued apoptosis in rat mesangial cells. cisplatin-induced apoptosis was associated with activation of caspase-3, caspase-8, caspase-9. Caspase-8 inhibition prevented the activation of both caspase-3 and caspase-9. In addition, cisplatin-induced apoptosis increased the expression of Bax, but not the level of Bcl-2. These change of Bax/bcl-2 ratio caused the release of cytochrome c from mitochondria into cytosol. In previous study, the ethanol extract of Bojungbangam-tang (EBJT) inhibited cisplatin-induced apoptosis in rat mesangial cells through inhibition of ERK and JNK activation. However, EBJT did not increase cell proliferation, because it did not prevent cisplatin-induced G2/M phase arrest. These effect of EBJT may be related to p38 activation. Cisplatin-induced G2/M phase arrest are inhibited by treatment with p38 inhibitor and EBJT in rat mesangial cells. Also, p38 inhibition and EBJT treatment on cisplatin-induced G2/M phase arrest are markedly increased the G0/G1 phase and reduced the sub-G1. In conclusion, anti-apoptotic effet of EBJT did not increases cell proliferation, because EBJT did not reduce p38 activation related to cisplatin-induced G2/M phase arrest.

  • In case while modulation index (M) is more than 0.7, the spectrum of motor voltage and current of a conventional two-phase SRP scheme are not reduced considerably. To solve the problems of a conventional two-phase SRP, this paper proposes a two-phase SRP(DZSRP) with double zero vector mode which zero vector is selected as V(111) in case of M $\geq$ 0.7, and zero vector is selected as V(000) if M < 0.7. For the validity of the proposed method, a 16 bit micro-controller C167 was used and the experiments were conducted with the 1.5kw induction motor under load condition. And the experimental results show that the switching noise spectrum for all the M are spread to a wide band area. Also the switching noise and conducted noise are discussed.

  • Following the prior studs regarding that 1.0g of cotton fabric round be dyed uniformly with a reactive dye in the solvent mixture of $2m\ell$ of water and $23m\ell$ of dichloromethane, silk fabric was dyed with C. I. Reactive Blue 203 in the water/dichloromethane two-phase immiscible solvent media. In order to minimize dye loss due to its hydrolysis, the reactive dyeing was carried out in dichloromethane containing a small amount of water. With only $3m\ell$ of water in $22m\ell$ of dichloromethane, 1.0g of silk fabric could be dyed perfectly. The uptake ratio was increased greatly, compared wish that of normal reactive dyeing in a water medium. It would seem that the one of hydrophobic solvents, dichloromethane, can assist the Even dyeing as it disperses a small amount of dye-dissolved phase and conveys this water phase to the fabric entirely and uniformly.

Volatile Component of Pine Needles from Pinus densiflora S. using Solid Phase Microextraction-Ges Chromatography-Mass Spectrometry

  • The volatile components of Pinus densiflora needles were studied by gas chromatography-mass spectrometry(GC-MS), using seven kinds of solid phase microextraction (SPME) fibers, seven in SPME fibers: 100 ${\mu}m$ PDMS, 65 ${\mu}m$ PDMS/DVB, 65 ${\mu}m$ SF-PDMS/DVB, 85 ${\mu}m$ PA, 75 ${\mu}m$ CAR/PDMS, 65 ${\mu}m$ CW/DVB and 50/30 ${\mu}m$ DVB/CAR/PDMS fibers. A total of 40 components were identified by using the seven different SPME fibers. The identified components were classified, according to their functionalities, as follows: 26 hydro-carbons, 7 alcohols, 4 carbonyl compounds, and 3 esters. The major volatile components of Pinus densiflora needles identified by these SPME fibers were $\alpha$-pinene ($1.7\~21.7\;{\mu}g/g$), $\beta$-myrcene ($2.0\~20.1\;{\mu}g/g$), $\beta$-phel-landrene ($4.6\~22.8\;{\mu}g/g$), $\beta$-caryophyllene ($6.7\~26.0\;{\mu}g/g$) germacrene D ($1.1\~11.9\;{\mu}g/g$). In the comparison of the seven SPME fibers, PDMS appeared to be the most suitable fiber for the analysis of hydrocarbon compounds and CAR/DPMS, PDMS/DVB, CW/VB and DVB/CAR/PDMS are shown to be optimal for analysis of the alcohols and carbonyl compounds.

Comparison Solid Phase Microextraction with Purge & Trap on the GC/MS Analysis of Volatile Organic Compounds in Biota Samples (Solid Phase Microextraction 및 Purge & Trap을 이용한 생물시료 중 휘발성 유기화합물의 GC/MS 분석비교)

  • The analysis of n-butylbenzene and 1,2-dibromo-3-chloropropane (DBCP) as volatile organic compounds in biota samples was performed by gas chromatography/mass spectrometry-selected ion monitoring mode. The target compounds, n-butylbenzene and DBCP, in biota samples were extracted by headspace solid phase microextraction (SPME) with $100{\mu}m$ polydimethyl siloxane (PDMS) fiber and purge & trap method. The extraction recoveries of these compounds obtained by SPME was 85.8% for n-butylbenzene and 92.4% for DBCP, respectively. Each value of method detection limit were $0.15{\mu}g/kg$ and $0.05{\mu}g/kg$, respectively. While in the case of purge & trap method, the extraction recovery was 115.2% for n-butylbenzene, 80.9% for DBCP and method detection limit were $0.04{\mu}g/kg$ and $0.70{\mu}g/kg$, respectively. The extraction yields and detection limits of these compounds obtained by purge & trap were equivalent to those by SPME.

Pharmacodynamics of Antitumor Activity of Paclitaxel in Monolayers and Histocultures of Human NSCLC Cells

  • In this study, we evaluated and compared the pharmacodynamics of paclitaxel (PTX) in human A549 NSCLC cells grown as monolayers or as three-dimensional histocultures. Growth inhibitory effects were determined after incubating cells in drug free medium until 96 hr post drug exposure initiation. Cell cycle arrest and apoptosis were measured by flow cytometry. The growth inhibition induced by PTX was significantly different in monolayers and histocultures, and PTX showed significantly less cytotoxicity in histocultures where large resistant fractions were observed. Moreover, although PIX induced significant $G_{2}/M$ arrest followed by apoptosis in monolayers in a drug concentration-dependant manner, $G_{2}/M$ arrest was not elicited in histocultures. However, apoptotic cells appeared from the $G_{2}/M$ phase in histocultures. In this study, we provide first evidence that PIX in three-dimensional histocultures, does not induce $G_{2}/M$ arrest, but rather that it induces $G_{2}/M$ phase specific apoptosis. Overall, our data demonstrate different pharmacodynamics of PTX in traditional monolayer and three-dimensional histocultures.

Induction of Apoptosis in Human Osteosarcoma Cell Lines(Saos-2) by Single Fraction High Dose Irradiation (고용량 방사선 조사 후 골육종 세포주(Saos-2)의 아포프토시스 발생)

  • A single fraction of 50 Gy extracorporeal irradiation, as a modality of limb-sparing operation, has been used to achieve tumor necrosis in osteosarcoma. Although this modality of radiation therapy preserving the mobility of a joint is commonly practiced, the precise knowledge on the radiobiological response of osteosarcoma cell has remained to be elucidated. We therefore observed whether a single high dose irradiation caused apoptosis in osteosarcoma cells and whether the commitment to apoptosis was associated with cell kinetics. We also investigated radiation dose response along the time course for development of apoptosis following single high dose irradiation. The morphologic change in apoptosis was observed by fluorescence with Hoechst 33258 and the degree and the fraction of cells by flow cytometry. Irradiation of osteosarcoma cells with 10, 30 and 50 Gy resulted in chromatin condensation and apoptotic body formation. The degree of apoptosis in osteosarcoma cells was $29.5{\pm}3.56%$, $39.9{\pm}4.83%$ at 24 and 48 hours after 10 Gy irradiation ; $41.1{\pm}3.93%$, $66.9{\pm}5.21%$ at 24 and 48 hours after 30 Gy irradiation ; and $48.0{\pm}3.69%$, $75.6{\pm}4.65%$ at 24 and 48 hours after 50 Gy irradiation. The fraction of cells in cell-cycle kinetic was $39.2{\pm}4.3%$ in G2/M, $22.1{\pm}4.65%$ in G1 at 24 hours after 10 Gy irradiation ; $51.0{\pm}4.3%$ in G2/M, $20.4{\pm}4.7%$ in G1 at 48 hours after 10 Gy irradiation ; $40.3{\pm}3.9%$ in G2/M, $26.1{\pm}4.7%$ in G1 at 24 hours after 30 Gy irradiation ; $59.2{\pm}3.9%$ in G2/M, $5.9{\pm}5.1%$ in G1 at 48 hours after 30 Gy irradiation ; and $44.3{\pm}4.2%$ in G2/M, $21.1{\pm}3.5%$ in G1 at 24 hours after 50 Gy irradiation. The fraction of cells at 48 hours after 50 Gy irradiation could not be observed because of irradiation induced cell death of most of cells. All values for irradiated cells showed accumulation in G2/M phase and reduction in G1 phase, irrespective of irradiation dose. The results suggest that a single fraction of high dose irradiation with 50 Gy results in accumulation of cells at G2/M phase, leading to apoptosis.

Inhibition of Cell-Cycle Progression in Human Promyelocytic Leukemia HL-60 Cells by MCS-C2, Novel Cyclin-Dependent Kinase Inhibitor

  • To elucidate the action mechanism of MCS-C2, a novel analogue of toyocamycin and sangivamycin, its effect on the expression of cell cycle-related proteins in the human myelocytic leukemia cell line HL-60 was examined using Western blotting and a flow cytometric analysis. MCS-C2, a selective inhibitor of cyclin-dependent kinases, was found to inhibit cell growth in a time- and dose-dependent manner, and inhibits cell cycle progression by inducing the arrest at G1 and G2/M phases, in HL-60 cells. The flow cytometric analysis revealed an appreciable arrest of cells in the G2/M phase of the cell cycle after treatment with MCS-C2. The HL-60 cell population increased gradually from 13% at 0 h, to 28% at 12 h in the G2/M phase, after exposure to $2{\;}\mu\textrm{M}$ MCS-C2. Furthermore, Western blot analysis demonstrated that MCS-C2 induced the cell cycle arrest at G1 phase through the inhibition of pRb phosphorylation. Hypophosphorylated pRb accumulated after treatment with $5{\;}\mu\textrm{M}$ MCS-C2 for 12 h, whereas, the level of hyperphosphorylated pRb was reduced. Thus, treatment of the cell with MCS-C2 suppressed the hyperphosphorylated form of pRb with a commensurate increase in the hypophosphorylated form.
