Sakarya is famous for cucurbit productions in Turkey and cucurbits can grow as big as 560 kg of weight per fruit in its agricultural areas. There is no or limited information about contaminant levels and profiles of the agricultural fields in Sakarya. The purpose of this study is to investigate the levels of polycyclic aromatic hydrocarbons (PAHs) (naphthalene, phenanthrene, pyrene, and fluoranthene) and heavy metal (As, Cd, Cu, Cr, Ni, Pb, Zn) concentrations of the selected fields. Total 33 soil samples were collected from 12 counties of Sakarya where both cucurbits have been produced and organochlorine pesticides have been applied to the fields for more than 30 y during the historical plantation periods. Heavy metal and PAH contents in the soil samples were measured by an Inductively Coupled Plasma Emission Spectroscopy and a Gas Chromatography-Mass Spectrometry. The highest phenanthrene, pyrene, and fluoranthene concentrations were measured as 63.50 ng/g, 134.34 ng/g, 140.0 ng/g, respectively in the soil samples from Geyve County. Cu, Ni, and Cr concentrations were measured as 108.2 mg/kg, 219.9 mg/kg, and 173.1 mg/kg, respectively in Geyve's samples which were also the highest and 2-7 times more than the limit values given in the Turkish Soil Pollution Control Regulation. Precautions need to be taken for Sakarya's agricultural fields which are an important milestone of Turkey's cucurbit and fruit productions since the contaminants can be accumulated in the fruits and edible parts of the plants.

The purpose of this study was to investigate and evaluate the safety of the grains, nut products, beans and oilseeds being sold in Gyeonggi province by analyzing mycotoxins. A multi-mycotoxins analysis method based on LC-MS/MS was validated and applied for the determination of eight mycotoxins, including aflatoxins ($B_1$, $B_2$, $G_1$ and $G_2$), fumonisins ($B_1$, $B_2$), zearalenone and ochratoxcin A in 134 samples. The limit of detection (LOD) and limit of quantitation (LOQ) for the eight mycotoxins ranged from 0.14 to $8.25{\mu}g/kg$ and from 1.08 to $7.21{\mu}g/kg$, respectively. Recovery rates of mycotoxins were determined in the range of 61.1 to 97.5% with RSD of 1.0~14.5% (n=3). Fumonisin $B_1$, $B_2$, zearalenone, and ochratoxin A were detected in 22 samples, indicating that 27% of grains, 12.5% of beans and 11.8% of oilseeds were contaminated. Fumonisin and zearalenone were detected simultaneously in 2 adlays and 3 sorghums. Fumonisin $B_1$ and $B_2$ were detected simultaneously in most samples whereas fumonisin $B_1$ was detected in 1 adlay, 1 millet and 1 sesame sample. The average detected amount of fumonisin was $49.3{\mu}g/kg$ and $10.1{\mu}g/kg$ for grains and oilseeds, respectively. The average detected amount of zearalenone was $1.9{\mu}g/kg$ and $1.5{\mu}g/kg$ for grains and beans, respectively. In addition, the average amount of ochratoxin A was $0.08{\mu}g/kg$ for grains. The calculated exposure amounts of fumonisin, zeralenone and ochratoxin A for grains, beans and oilseeds were below the PMTDI/PTWI.

Baculovirus was originally isolated from the alfalfa looper and contains a 134-kbp genome with 154 open reading frames (ORF). The major capsid protein VP39 together with some minor proteins forms the nucleocapsid ($21nm{\times}260nm$) that encloses the DNA with p6.9 protein. They are double-stranded, circular, supercoiled DNA molecules in a rod-shaped capsid. Wild-type baculoviruses exhibit both lytic and occluded life cycles that develop independently throughout the three phases of virus replication. Recombinant baculoviruses can transfer their vectors and express their recombinant proteins in a wide range of mammalian cell types. Especially, inclusion of a dominant selectable marker in these baculoviral vectors can express diverse recombinant genes in many cells. Baculoviral vectors were reconstructed with cytomegalovirus (CMV) promoter,uroplakin II promoter, polyhedron promoter, vesicular stomatitis virus G (VSVG), enhanced green fluorescent protein (EGFP), protein transduction domain (PTD) gene and so on. These reconstructed vectors were infected into various cell and cell lines. We performed transfection and expression of these recombinant vectors comparison with other control vectors. From this study, we knew that transfection and expression of these recombinant vectors have higher efficacy than any control vector. Distribution of benthic macrofauna was surveyed in the subtidal area and on the sand flat of Sindu-ri coast during July 2002. A total of 134 species, 3,511 individuals and 388g biomass of macrobenthos were sampled inclusively from the study sites. 109 species with a mean density of $1,298ind./m^2$ and biomass of $69.4g/m^2$ were collected from 10 stations established in the subtidal area. Polychaetes were represented as a dominant faunal group of the subtidal area in both species richness and density with values of 54 species and $813ind./m^2$. Cluster analysis based on the faunal composition showed that the subtidal area could be divided into four stational groups according to sedimentary characteristics: Cirrophorus armatus - Lumbrineris longifolia association on the mud sediment with pebbles near aqua-farms; Aonides oxycephala association on the rocky bed; Scoloplos armiger - Grandifoxus cuspis association on the sand-dominated sediment along the lowest low water; a association with higher evenness on the sand-dominated sediment in the depth. Although having characteristics of high value in species richness, density and species diversity, the macrobenthic association around the farming area was dominated by the opportunistic species such as Lumbrineris longifolia and Tharyx sp. Natural community represented by S. armiger and G. cuspis was developed in most sand-dominated areas. 50 species were sampled with a mean density of $2,443ind./m^2$ and biomass of $381.3g/m^2$ from eight+ stations along two transects on Sindu-ri sand flat. Mollusks were represented as a dominant faunal group of the sand flat in species richness, density and biomass with values of 20 species, $1,345ind./m^2$ and $350.4g/m^2$, respectively. Umbonium thomasi, Veneridae sp., Mandibulophoxus mai, Armandia lanceolata, Eohaustorius spinigerus, Urothoe convexa were dominant species and these species accounted for over 83% of total individuals. There were three distinct zones of macrobenthos on Sindu-ri sand flat according to the extent of exposure time. The upper zone was dominated by Scopimera globosa, the middle zone was characterized by M. mai, and the lower zone was dominated by E. spinigerus. Sindu-ri coastal area was considered as a healthy habitat for macrobenthos, except for around the farming area. It was considered that Sindu-ri coastal area including the sand flat connected to the sand dune of natural monument was a valuable habitat as a protected site in having no pollution source and no artificial structure.
Purpose: The serologic diagnosis of rotaviral infections is not commonly used in clinical practice, but is used in seroepidemiologic studies. In this study, the usefulness of Escherichia coli-expressed recombinant VP6 proteins of group A rotavirus in the serodiagnosis of rotavirus infections by ELISA was evaluated. Methods: The recombinant VP6 proteins of group A rotavirus expressed in E. coli Rosetta II strain were purified and identified. One hundred sera from 22 children (4 healthy neonates, 13 healthy children, and 5 immunocompromised children) who had serial sera samples prior to and after rotavirus infections were provided by the Gyeongsang National University Hospital, a member of the National Biobank of Korea. IgG, IgA, and IgM antibodies against rVP6 were analyzed by ELISA in all of the patients and Western blot analysis in 4 neonates. Results: ELISA tests using rVP6 proteins of group A rotavirus as antigen revealed that IgG, IgA, and IgM antibodies increased after rotaviral infections in most neonates and healthy children. IgG antibodies also increased after rotaviral infections in most immunocompromised children without an adequate increase in IgM or IgA antibodies. Western blot analysis in four neonates revealed very early IgM antibody responses, even in the sera with low optical densities in ELISA tests. Conclusion: Our study showed that ELISA using rVP6 as an antigen is a valid diagnostic tool for seroepidemiologic studies of rotavirus infections and Western blot analysis is a sensitive test in detecting IgG, IgA, and and IgM antibodies in patients with rotavirus infections.
This study was conducted to determine the effects of different copper (inorganic and organic) and energy (tallow and glycerol) sources on growth performance, nutrient digestibility, gas emission, diarrhea incidence, and fecal copper concentration in growing pigs by using a 2${\times}$2 factorial design. In this trial, 96 pigs (63 d of age) were employed, with an average initial weight of 28.36${\pm}$1.14 kg. The dietary treatments were i) basal diet with 134 ppm copper (Korea recommendation) as $CuSO_4$+tallow; ii) basal diet with 134 ppm Cu as $CuSO_4$+glycerol; iii) basal diet with 134 ppm copper as CuMet+tallow; and iv) basal diet with 134 ppm copper as CuMet+ glycerol. Throughout the entire experimental period, no differences were noted among treatment groups with regard to the magnitude of improvement in ADG (average daily gain), ADFI (average daily feed intake) and G/F (gain:feed) ratios. The nitrogen (N) digestibility of pigs fed on diets containing organic copper was improved as compared with that observed in pigs fed on diets containing inorganic copper (p<0.05). An interaction of copper${\times}$energy was observed in the context of both nitrogen (p<0.05) and energy (p<0.01) digestibility. Ammonia emissions were significantly lower in the organic copper-added treatment groups than in the inorganic copperadded treatment groups (p<0.05). Mercaptan and hydrogen sulfide emissions were reduced via the addition of glycerol (p<0.05). No significant effects of copper or energy source, or their interaction, were observed in reference to diarrhea appearance and incidence throughout the entirety of the experimental period. The copper concentration in the feces was significantly lower in the organic copper source treatment group than was observed in the inorganic copper source treatment group (p<0.05). The results of this experiment show that organic copper substituted for inorganic copper in the diet results in a decreased fecal copper excretion, but exerts no effect on performance. The different energy (tallow and glycerol) sources interact with different copper sources and thus influence nutrient digestibility. Glycerol supplementation may reduce the concentrations of odorous sulfuric compounds with different Cu sources.
This study was conducted by surveying 616 male workers living in Ulsan City regarding their health status based on lifestyles such as alcohol consumption, smoking and exercising as well as physical measurements and biochemical tests. The average height, weight and BMI(body mass index, $kg/m^2$) of the subjects was 170.9cm, 70.2kg and 24.2, respectively. The rate of drinking was $80.9\%$ and the rate of smoking was $53.4\%$. Seventy four percent of subjects responded that they exercise regularly. The results of the blood biochemical tests revealed that the average hemoglobin concentration was 14.7g/dl, and the levels of GPT(glutamic pyruvic transaminase) and GOT (glutamic oxaloacetic transaminase) were 32.74unit/l, 26.99 unit/l, respectively. The average hemoglobin concentration for the subjects aged in the 50s was 14.39g/dl, which was significantly lower than those in the 20s(14.81g/dl), 30s(14.69g/dl) and 40s(14.73g/dl). The blood glucose level and the cholesterol level also increased with age. Also investigated was the blood pressure of the subjects increased with age,. and there was a significant increase(p < 0.05) for the subjects in the 50s compared to those in the 20s. The frequency of alcoholic beverages was significantly correlated with systolic/ diastolic blood pressure(p < 0.05) and $\gamma-GTP(gamma\;glutamyl\; transpeptidase)$(p<0.01). The duration of smoking showed a negative correlation(p < 0.05) with the hemoglobin and positive correlations with diastolic blood pressure, cholesterol and $\gamma-GTP(p<0.01)$. The study shows that blood pressure, blood glucose level, cholesterol level, GOT, GPT and $\gammaGTP$ level, increase with age, which indicates higher possibility of degenerative diseases, calling for nutritional education in terms of advisable lifestyles regarding eating habits, alcohol consumption, smoking and regular exercise.
A novel technology for homogeneous deposition of zeolite particles on a porous support was developed so that those particles played a seeding role for the growth of zeolite crystals. After the particles were dispersed in water, the aqueous solution was 134 through the bore of a porous tubular support. By keeping the other side of the support in a vacuum, the aqueous solution passed through the pores of the support, leading the particles to be homogeneously deposited on the support. The amount of the deposited particles was investigated by changing the following operating parameters: a particle concentration in the solution, a time for deposition, and the feeding rate of the solution. The amount of the deposited particles increased from 0.0019 g to 0.0208 g as the concentration of the particles was changed from 0.01 wt% to 0.3 wt%, while the feeding rate and the deposition time were kept to 100 mL/min and 4 min, respectively. As the deposition time was varied from 1 min to 4 min, the deposition amount increased from 0.0004g to 0.0019g at the typical condition of the rest parameters. Also, it was observed that the deposited weight increased from 0.0029 g to 0.01 g as the feeding rate increased from 100 mL/min to 300 mL/min. However, the total permeance of water and ethanol decreased through the zeolite membrane as the deposited weight increased.
The aim of the study was to obtain the extracts of various native wild mushrooms and select the useful resources though biological activity evaluation. The anti-oxidant potential, nitrite scavenging activity, and -glucan content of wild mushrooms collected from Eumseong and Bonghwa in Korea were investigated. Based on the results of this study, Ganoderma lingzhi (OK1362) showed the highest DPPH radical scavenging activity (73.2%), ferric reducing anti-oxidant power (0.134), reducing power (0.155), nitrite scavenging activity (53.6%), total polyphenol content (28.9 mg GAE/g), flavonoid content (10.0 mg QE/g), and -glucan content (25.2%) when compared to other wild mushrooms sampled in this study. In addition, it was confirmed that Perenniporia fraxinea (OK1360), Amanita sp. (OK1398), and Russula sp. (OK1406) had relatively high anti-oxidant and nitrite scavenging potentials. In conclusion, our results can provide fundamental data for extracting beneficial compounds from wild mushrooms.
Lactobacillus salivarius KLW001, a species of lactic acid bacteria (LAB), was isolated from a weaning piglet in a swine farm, South Korea, to develop an antimicrobial probiotic strain for piglets. Herein, we report the draft genome sequence of the strain. The genome contains 2,326,706 bp with a G+C content of 33.0% in 166 contigs (${\geq}500bp$). From the genome, we found out 4 genes related to antibiotic resistance, 36 genes for phages, 3 genes for bile hydrolysis, and 27 CRISPR spacers.
