Partition of Amino Acids Requirement for Maintenance and Growth of Broilers I. Lysine

  • Purified diets containing five graded levels of lysine were fed to a total of 125 growing chicks (25 chicks per treatment) to evaluate lysine requirements for growth and maintenance. A model was developed to separate lysine requirement for maintenance from requirement for growth. Based on weight gain response, the daily lysine requirement for growth was 12.06 mg/g gain and the daily lysine requirement for maintenance was 0.332 times metabolic body size ($W^{0.75}$). Similarly, the lysine requirement for growth was 0.457 mg/mg nitrogen gain and the daily lysine requirement for maintenance was 0.344 times metabolic body size. The plateau of plasma lysine concentration was reached at 354.75 mg intake/day. The total lysine requirement was 414.27 mg/day or 1.0% of the diet, 420.11 mg/day or 1.01% of the diet based on weight gain response and N gain response, respectively. Previous lysine requirements for growing chicks of 1-28 days old were in close agreement with these estimates. As a percentage of protein, lysine requirement was calculated to be 7.3% and the reported lysine content of chick muscle crude protein of 7.46% was closely related.

Maternal and Direct Genetic Parameters for Production Traits and Maternal Correlations among Production and Feed Efficiency Traits in Duroc Pigs

  • Direct and maternal genetic parameters for production traits in 1,642 pigs and maternal genetic correlations among production (1,642 pigs) and feed efficiency (380 boars) traits were estimated in 7 generations of a Duroc population. Traits studied were daily gain (DG), intramuscular fat (IMF), loineye area (LEA), backfat thickness (BF), daily feed intake (FI), feed conversion ratio (FCR) and residual feed intake (RFI). The RFI was calculated as the difference between actual and predicted feed intake. The predicted feed intake was estimated by adjusting the initial test weight, DG and BF. Data for production traits were analyzed using four alternative animal models (including direct, direct+maternal permanent environmental, or direct+maternal genetic+maternal permanent environmental effects). Direct heritability estimates from the model including direct and all maternal effects were $0.41{\pm}0.04$ for DG, $0.27{\pm}0.04$ for IMF, $0.52{\pm}0.06$ for LEA and $0.64{\pm}0.04$ for BF. Estimated maternal heritabilities ranged from $0.04{\pm}0.04$ to $0.15{\pm}0.05$ for production traits. Antagonistic relationships were observed between direct and maternal genetic effects ($r_{am}$) for LEA (-0.21). Maternal genetic correlations of feed efficiency traits with FI ($r_g$ of FI with FCR and RFI were $0.73{\pm}0.06$ and $0.90{\pm}0.05$, respectively) and LEA (rg of LEA with FCR and RFI were $-0.48{\pm}0.05$ to $-0.61{\pm}0.05$, respectively) were favorable. The estimated moderate genetic correlations between direct and maternal genetic effects for IMF and LEA indicated that maternal effects has an important role in these traits, and should be accounted for in the genetic evaluation system.

Ileal Amino Acid Digestibility of Broken Rice Fed to Postweaned Piglets with or without Multicarbohydrase and Phytase Supplementation

  • Most of amino acid (AA) digestibility values for feed ingredients are obtained using pigs cannulated in the distal ileum. The ileal-cannulated pig model uses pigs older than six weeks due to difficulties related to implanting the T-cannula in distal ileum of younger pigs and complications during the post-surgical recovery. However, to properly formulate the diet of weaned pigs, the nutritive value of feed ingredients should be determined with younger pigs. Thus, 25 weaned pigs were used to determine the apparent total tract digestibility (ATTD) of nutrients, energy, and apparent ileal digestibility (AID) and standardized ileal digestibility (SID) ileal AA digestibility of broken rice (BR), with or without multicarbohydrase (MC) and phytase (Phy) supplementation. Piglets were weaned at 23 d of age and individually housed in digestibility cages until 45 d of age. The trial consisted of 7 d of adaptation to the experimental diets and 3 d of excreta (feces and urine) collection. Ileal digesta was collected at slaughter (about 6 weeks of age). A completely randomized experimental design was used to determine the effects of MC and Phy. Reference diets (RD, 5% casein) was replaced by 30% of BR with or without MC, Phy, or MC+Phy. The RD was used to quantify endogenous AA losses. BR with Phy supplied had increased the ATTD of dry matter (p<0.05) and SID of histidine (p = 0.05), arginine, leucine, lysine, valine, alanine, and proline (p<0.05). BR with MC had been increased digestible energy and protein and SID for histidine (p<0.05). There was no interaction between Phy and MC on the BR nutrient digestibilities. Standardized amino acid digestibilities of BR, without enzymes, were lower than those values reported in the literature. The MC and Phy improved the digestibility of some nutrients and energy of BR in post-weaned piglet diets.

Estimation of Gene Effects in Four Bivoltine Silkworm (Bombyx mori L.) Crosses

  • Six generations (P$_1$, P$_2$, F$_1$, BC$_1$, BC$_2$ and F$_2$) of four bivoltine silkworm crosses (SKAU-R-1 ${\times}$ Yakwei, SKAU-R-6 ${\times}$ Yakwei, CSR$_2$ ${\times}$ CSR$_4$ and SH$_{6}$${\times}$ NB$_4$D$_2$) were evaluated in a completely randomized block design with 5 replications for each treatment. The generation mean 2 in respect of 3 metric traits (single cocoon weight, single shell weight, and shell ratio %), were subjected to Cavallis joint scaling test. Additive dominance model was found to be adequate in CSR$_2$${\times}$CSR$_4$ and SH$_{6}$${\times}$ NB$_4$D$_2$ for single cocoon weight and shell weight and SKAU-R-6 ${\times}$ Yakwei for shell ratio(%). Whereas, in rest of the crosses epistasis was evident in the traits under investigation. Magnitude of additive gene effect (d) was greater than dominance(h) in SH$_{6}$${\times}$NB$_4$D$_2$ and SKUA-R-6${\times}$Yakwei for shell ratio (%) and in CSR$_2$${\times}$ CSR$_4$ for shell weight. Thus selection for these traits in early segregating generations of these crosses would be effective for obtaining considerable genetic gain. gain.

Evaluation on Pharmacological Activities of 2,4-Dihydroxybenzaldehyde

  • 4-Hydroxybenzaldehyde, a phenolic compound found in a variety of natural sources, was previously shown to contain anti-inflammatory and related anti-angiogenic and anti-nociceptive activities. The present work was designed to assess some pharmacological activities of 2,4-dihydroxybenzaldehyde (DHD), an analogue of 4-hydroxybenzaldehyde. DHD exhibited a significant inhibition in the chick chorioallantoic membrane (CAM) angiogenesis, and its $IC_{50}$ value was $2.4\;{\mu}g/egg$. DHD also contained in vivo anti-inflammatory activity using acetic acid-induced permeability and carrageenan-induced air pouch models in mice. In the air pouch model, DHD showed significant suppression in exudate volume, number of polymorphonuclear leukocytes and nitrite content. DHD showed an anti-nociceptive activity in the acetic acid-induced writhing test in mice. It also suppressed enhanced production of nitric oxide (NO) and elevated expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophage cells. It was able to slightly decrease the level of reactive oxygen species in the stimulated macrophages. DHD at the used concentrations couldn't modulate the viabilities of RAW264.7 cells. Taken together, like 4-hydroxybenzaldehyde, DHD contains anti-angiogenic, anti-inflammatory and anti-nociceptive activities.

Importance of Leu-5 and Pro-6 in the Inhibitory Activity of the Serratia marcescens Metalloprotease Inhibitor (SmaPI)

  • The Serratia marcescens metalloprotease inhibitor (SmaPI) is a proteinase inhibitor toward Serratia marcescens metalloprotease (SMP). The three-dimensional structure of SmaPI was calculated by computer modeling using the structure complex between SMP and the Erwinia chrysanthemi inhibitor as a template. Based on this model structure, the substitution of the amino acid residues, Ala4, Leu-5, Pro-6, and Thr-7, were located at the hinge region of the N-terminal segment by site-directed mutagenesis. Although the A4R and T7A mutant SmaPIs showed a nearly full inhibitory activity, the inhibitory activity of SmaPI decreased significantly when the Leu-5 was converted to Ala, Gly, Ile, or Val. Surprisingly, the L5I and L5V mutant SmaPIs showed less inhibitory activities than the L5A mutant. From these results, we suggested that the orientations and positions of respective aliphatic groups in the side-chain of position 5 mainly affected the inhibitory activity of SmaPI. The overall side-chain hydrophobicity was only slightly affected. The side-chain of the Leu-5 residue contributed approximately 0.79 kcal/mol out of 8.44 kcal/mol to the binding of SmaPI with SMP The inhibitory activities of P6A and F6G were also severely decreased. The Pro-6 may have a critical role in maintaining the strict conformation of the N-terminal portion that may be important in the inhibitory activity of SmaPI. In conclusion, Leu-5 and Pro-6 have crucial roles in the inhibitory function of SmaPI toward SMP.

Association between Single Nucleotide Polymorphisms of Fatty Acid Synthase and Fat Deposition in the Liver of the Overfed Goose

  • Goose fatty liver is one of the most delicious and popular foods in the world, but there is no reliable genetic marker for the early selection and breeding of geese with good liver-producing potential. In our study, one hundred and twenty-four 78-day-old Landes geese bred in Shunda Landes goose breeding farm, Jiutai, Jilin, China were selected randomly. The fatty livers were sampled each week after overfeeding during a three week period. Polymerase chain reaction-single strand conformation polymorphism and DNA sequencing were used to identify single nucleotide polymorphisms (SNPs) of fatty acid synthase (FAS), which is an important enzyme involved in the synthesis of fat under both physiological and pathological conditions. Least-squares correlation was established between these SNPs and fatty liver weight, abdominal fat weight, and intestinal fat weight of the overfed Landes geese, respectively. The results showed that fatty liver weight of geese with EF and FF genotypes (amplified by primer P1) was significantly higher than that of the EE genotype (p<0.05), and liver weight of CD and DD genotypes (amplified by primer P2) was significantly higher than that of the CC genotype (p<0.05). Different genotype combinations showed different liver weights, and from highest to lowest were ABDD, DDEF, DDFF, DDEE, ABEF, ABFF, AADD, and CDEF. Further analysis of DNA sequencing showed that there were two SNPs within the 5' promoter region the FAS gene. The geese of EF and FF genotypes carried a change of T to C, and the geese of CD and DD genotypes carried a change of A to G. The changes of the bases could potentially influence the binding of some transcription factors to this region as to regulate FAS gene. To our knowledge, this is the first report of SNPs found within the 5' promoter region of the Landes goose FAS gene, and our data will provide an insight for early selection of geese for liver production.

DESIGN OF AN UNMANNED GROUND VEHICLE, TAILGATOR THEORY AND PRACTICE

  • The purpose of this paper is to describe the design and implementation of an unmanned ground vehicle, called the TailGator at CIMAR (Center for Intelligent Machines and Robotics) of the University of Florida. The TailGator is a gas powered, four-wheeled vehicle that was designed for the AUVSI Intelligent Ground Vehicle Competition and has been tested in the contest for 2 years. The vehicle control model and design of the sensory systems are described. The competition is comprised of two events called the Autonomous Challenge and the Navigation Challenge: For the autonomous challenge, line following, obstacle avoidance, and detection are required. Line following is accomplished with a camera system. Obstacle avoidance and detection are accomplished with a laser scanner. For the navigation challenge, waypoint following and obstacle detection are required. The waypoint navigation is implemented with a global positioning system. The TailGator has provided an educational test bed for not only the contest requirements but also other studies in developing artificial intelligence algorithms such as adaptive control, creative control, automatic calibration, and internet-base control. The significance of this effort is in helping engineering and technology students understand the transition from theory to practice.

과실(果實)의 점탄성(粘彈性) 특성(特性)(I) - 응력이완거동(應力弛緩擧動) - (Viscoelastic Properties of Fruit Flesh(I) - Stress Relaxation Behavior -)

  • Fruits are generally subjected to mechanical forces during harvesting, handling, and transportation that may cause damage in the form of bruises, punctures, and cracks. In order to prevent damage, and insure better quality fruits for consumers, it is very essential to study physical properties of these materials. The studies were conducted to examine the effect of storage period, storage condition, and other factors, such as loading rate and initial strain, on the stress relaxation behavior of the fruit flesh, and develop nonlinear viscoelastic models to represent its stress relaxation behavior. The following results were obtained from the study : 1. Since the viscoelastic behavior of the fruits flesh was nonlinear, the behavior was satisfactorily modelled as follows ; $${\delta}({\varepsilon},\;t)={\varepsilon}^A[B\;{\exp}(-Ct)+D\;{\exp}(-Ft)+G(-Ht)]$$ But, for the every strain applied, the stress relaxation behavior of the fruit flesh, such as apple and pear, could be well described by the Generalized Maxwell model, respectively. 2. The effect of loading rate on the stress relaxation behavior was remarkable. The higher loading rate resulted in the higher initial stress, and the faster stress relaxation. 3. The higher initial strain resulted in the higher initial stress, and stress relaxed at the large initial strain was also much higher than at the small initial strain. 4. Stress relaxation rate and quantity stored in the fruits at the low temperature storage were much higher than those at the normal temperature storage in the same storage period. Also, in all fruits tested, the longer storage period was the more relaxation rate and quantity were shown. These trends in the normal temperature condition was the more significant than in the low temperature condition.

약물-용출 생분해성 고분자 스텐트를 위한 EGCG와 디자인 파라미터의 영향에 대한 연구 (A Study on Effects of EGCG and Design Parameter for Drug-Eluting Biodegradable Polymer Stents)

  • Finite element analysis(FEA) has been extensively applied in the analyses of biomechanical properties of stents. Geometrically, a closed-cell stent is an assembly of a number of repeated unit cells and exhibits periodicity in both longitudinal and circumferential directions. This study concentrates on various parameters of the FEA models for the analysis of drug-eluting biodegradable polymeric stents for application to the treatment of coronary artery disease. In order to determine the mechanical characteristics of biodegradable polymeric stents, FEA was used to model two different types of stents: tubular stents(TS) and helicoidal stents(HS). For this modeling, epigallocatechin-3-O-gallate (EGCG)-eluting poly[(L-lactide-co-${\varepsilon}$-caprolactone), PLCL] (E-PLCL) was chosen as drug-eluting stent materials. E-PLCL was prepared by blending PLCL with 5% EGCG as previously described. In addition, the effects of EGCG blending on the mechanical properties of PLCL were investigated for both types of stent models. EGCG did not affect tensile strength at break, but significantly increased elastic modulus of PLCL. It is suggested that FEA is a cost-effective method to improve the design of drug-eluting biodegradable polymeric stents.
