금은화(金銀花)가 Cationic Bovine Serum Albumin 투여로 유발된 Membranous Nephropathy Mouse Model에 미치는 영향 (Effects of the Lonicerae Flos Extract on the Membranous Nephropathy Induced by Cationic Bovine Serum Albumin in Mice)

  • Membranous nephropathy(MN) is the most common cause of adult nephrotic syndrome worldwide. But treatment of MN is not defined. This study was to evaluate the effects of Lonicerae Flos Extract(LFE) on the MN induced by cBSA in mice. Mice were divided into 4 groups. The first group named for 'Normal' was injected with a saline solution. The second group named 'Control' treated with cBSA(10 mg/kg i.p) only. The third group named 'LFE-250', treated with cBSA(10 mg/kg i.p) and LFE(250 mg/kg, p.o). The fourth group named 'LFE-500'treated with cBSA(10 mg/kg i.p) and LFE(500 mg/kg, p.o). After cBSA and LFE treatment for 4 weeks, we measured change of body weight, 24hrs proteinuria, serum albumin, total cholesterol, triglyceride, BUN, creatinine, TNF-$\alpha$, IL-6, IL-$1{\beta}$, IL-10, IFN-$\gamma$, IgA, IgM and IgG levels. The morphologic changes of renal glomeruli were also observed with a light microscope. The levels of 24 hrs proteinuria, total cholesterol, IgG , IgM, IgA, IL-6 were significantly decreased in both LFE groups. The level of triglyceride, IL-$1{\beta}$ was significantly decreased in LFE-500 group. The level of Albumin was significantly increased in LFE-250 group. The level of TNF-$\alpha$, IFN-$\gamma$ were significantly decreased in LFE-250 group. The mRNA expression of IL-$1{\beta}$ in splenocytes was consideraly decreased in LFE-500 group. In histological findings of kidney tissue, thickening of GBM decreased in both LFE groups. This study shows that the LFE might be effective for treatment of MN. More clinical data and studies are to be done for efficient application.

Interaction of Bilobalide and Ginkgolides B with Bovine Serum Albumin: A Fluorescence Quenching Study

  • The interaction of bilobalide (BB) and ginkgolides B (GB) with bovine serum albumin (BSA) was investigated by fluorescent technique and UV/vis absorption spectroscopy. The results showed that BB and GB could intensively quench the fluorescence of BSA through a static quenching procedure. The binding constants (Ka) and the average binding distance between the donor (BSA) and the acceptor (ginkgolides) were obtained ($r_{BB}$ = 5.33 nm and $r_{GB}$ = 4.20 nm) by the theory of non-radiation energy transfer, and then the thermodynamic parameters such as ${\Delta}S^0$ (0.17-0.32 kJ/mol), ${\Delta}G^0$ (-20.76 ~ -17.79 kJ/mol) and ${\Delta}H^0$ (32.47-76.52 kJ/mol) could be calculated, respectively. All these results revealed that the interaction of BB and GB with BSA were driven mainly by hydrophobie force. The synchronous fluorescence spectroscopy was applied to examine the effect of two ginkgolides on the configuration of BSA. The configuration alteration of BSA could be induced by the hydrophobicitv environment of tyrosine with the increase of the drug concentration.

알부민을 함유한 메톡시 폴리(에틸렌 글리콜)- 폴리(L-락타이드-co-글리콜라이드) 웨이퍼의 제조 및 방출거동 (Preparation and Release Behavior of Methoxy poly(ethylene glycol)- poly(L-lactide-co-glycolide) Wafer Containing Albumin)

  • 단백질 전달체로 이용하기 위한 폴리(에틸렌 글리콜)-폴리(락타이드-co-글리콜라이드) 공중합체는 카비톨 (134 g/mole)과 550, 2000 및 5000g/mole의 각기 다른 분자량을 가진 메톡시 폴리(에틸렌 글리콜)을 개시제로 하여 촉매인 Sn(Oct)$_2$를 사용하여 L-락타이드와 글리콜라이드의 개환중합에 의해 합성되었다. $^1$H-NMR, GPC, 그리고 XRD를 이용하여 합성된 블록 공증합체의 특성을 결정하였다. 이식형 웨이퍼를 준비하기 위하여 동결분쇄기를 이용하여 형광이 결합된 1% 소 혈청 알부민과 고분자를 균일 혼합 후에 3mm${\times}$1mm의 크기로 웨이퍼를 제조하였다. 형광 알부민의 방출 거동과 pH 변화는 pH 7.4의 인산염완충용액을 이용하여 37$^{\circ}C$, 100rpm의 항온조에서 30일 동안 관찰하였다. 알부민의 방출양은 형광분광기를 통하여 FITC의 강도에 의해 결정되었다. 알부민의 방출 거동은 블록 공중합체내에서 MPEG의 분자량이 증가할수록 빠른 초기방출을 보였고, PLGA의 분자량을 조절함으로서 약물의 방출기간을 결정할 수 있었다. 이러한 결과로 생분해성 PLGA에 친수성인 MPEG의 도입을 통해 약물의 방출거동을 조절할 수 있었다.

  • This paper describes of VME-BSA which is a tool for the development, mainternance and repair. In micro/mini computer system using VMEbus as a backplane bus, VME-BSA has some good facilities such as acquiring, storing and analyzering the information (address, data, etc.) on VMEbus according to the various condition which is set by users, and therefore it is easy to isolate and find many complete errors on bus.

  • 단백질 수용액 중의 초음파흡수 mechanism을 규명하기 위하여 pH 7의 소혈청 albumin(BSA)수용액의 초음파흡수 측정을 주파수 100 kHz에서 1600 MHz에 걸쳐 행하였다. 측정방법은 plano-concave 공명법(0.1${\sim}$10 MHz)과 고분해능 Bragg 반사법(120${\sim}$1600 MHz)을 사용하였다. pH7에서의 초음파흡수스펙트럼은 Davidson-Cole 분포식의 거울상인 분포함수를 사용한 완화곡선과 일치하였다. 이 완화현상을 BSA 분자의 수화평행에 의한 것으로 해석하였다.

The interaction of serum albumin with ginsenoside Rh2 resulted in the downregulation of ginsenoside Rh2 cytotoxicity

  • Background: Ginsenoside Rh2 (G-Rh2) is a ginseng saponin that is widely investigated because of its remarkable antitumor activity. However, the molecular mechanism by which (20S) G-Rh2 triggers its functions and how target animals avoid its cytotoxic action remains largely unknown. Methods: Phage display was used to screen the human targets of (20S) G-Rh2. Fluorescence spectroscopy and UV-visible absorption spectroscopy were used to confirm the interaction of candidate target proteins and (20S) G-Rh2. Molecular docking was utilized to calculate the estimated free energy of binding and to structurally visualize their interactions. MTT assay and immunoblotting were used to assess whether human serum albumin (HSA), bovine serum albumin (BSA), and bovine serum can reduce the cytotoxic activity of (20S) G-Rh2 in HepG2 cells. Results: In phage display, (20S) G-Rh2-beads and (20R) G-Rh2-beads were combined with numerous kinds of phages, and a total of 111 different human complementary DNAs (cDNA) were identified, including HSA which had the highest rate. The binding constant and number of binding site in the interaction between (20S)-Rh2 and HSA were $3.5{\times}10^5M^{-1}$ and 1, and those in the interaction between (20S) G-Rh2 and BSA were $1.4{\times}10^5M^{-1}$ and 1. The quenching mechanism is static quenching. HSA, BSA and bovine serum significantly reduced the proapoptotic effect of (20S) G-Rh2. Conclusion: HSA and BSA interact with (20S) G-Rh2. Serum inhibited the activity of (20S) G-Rh2 mainly due to the interaction between (20S) G-Rh2 and serum albumin (SA). This study proposes that HSA may enhance (20S) G-Rh2 water solubility, and thus might be used as nanoparticles in the (20S) G-Rh2 delivery process.

Fibrin glue로부터 Ampicillin-Na의 용출 및 Tissue adhesive로서의 접착력 (Release of Ampicillin-Na from fibrin glue and its adhesive strength as a tissue adhesive)

  • 광범위 항생물질인 Ampicillin sodium(AMP-Na)을 두가지 방법 즉, 단순섞임과 bovine serum albumin(BSA)으로 microsphere화한 후 loading 하는 방법으로 fibrin glue(FG)를 제조하였고 이 FG로부터 AMP-Na의 서방성 시험을 시도하였다. 단순섞임의 경우 fibrinogen(FBNG)의 농도를 조적함으로서 FG로부터 AMP-Na의 지속적 방출을 달성할 수 있었으며 특히 이 microsphere를 glutaraldehyde로 가교화시킴으로서 용출속도를 더욱 늦출 수 있었다.(tO.9 : 33hr). FG의 rat perironeum에 대한 접착력은 FBNG과 thrombin의 농도가 각각 5.0%, 25~50 NIHU/ml에서 최대로 나타났다. Factor XIII의 농도는 0~500 U/1g of FBNG의 범위내에서 접착력에 거의 영향을 미치지 않았으며 incubation time은 60분일 때 최대로 나타났다. FG에 AMP-Na 및 BSA micorsphere를 loading하여도 접착력에는 큰 영향을 미치지 않았다.

Results: The levels of 24 hrs proteinuria and serum triglyceride, BUN, IgG, TNF-${\alpha}$, IL-1${\beta}$ significantly decreased in both HHE groups, while the level of serum albumin significantly increased in both HHE groups. The mRNA expression of IFN-${\gamma}$ and IL-6 in splenocytes considerably increased in both HHE groups. The mRNA expression of IL-10 in splenocytes considerably decreased in both HHE groups. In histological findings of kidney tissue, thickening of GBM decreased in both HHE groups. Conclusions: This study shows that HHE might be effective for treatment of acute stage MN. More clinical data and studies are to be done for efficient application.

황금(黃芩)이 Cationic Bovine Serum ALbumin투여로 유발된 Membranous Nephropathy Mouse Model에 미치는 영향 (Effects of Scutellaria Radix Extract on the Membranous Nephropathy Induced by Cationic Bovine Serum Albumin in Mouse Model)

  • Background: Membranous nephropathy (MN) is the most common cause of adult nephrotic syndrome worldwide and has been defined as granular subepithelial deposition of immune complexes along the glomerular basement membrane (GBM). MN has few known treatments and gives rise to side effects under treatment with steroids and immunosuppressives. Objective: The purpose of this experimental study was to demonstrate the effects of Scutellariae Radix extract (SRE) treatment on MN mouse model induced by cBSA. Methods: We divided mice into 4 groups. The Normal group had no treatment. We induced MN mouse model to the other 3 groups by injecting cBSA into the abdominal cavity. The control group was treated with cBSA (10 mg/kg, i.p.) only. The second group, 'SRE-250', was treated with cBSA (10 mg/kg, i.p.) and SRE (250 mg/kg, p.o.). The third group, 'SRE-500', was treated with cBSA (10 mg/kg, i.p.) and SRE (500 mg/kg, p.o.). After cBSA and SRE treatment for 4 weeks, gain in body weight, 24hrs proteinuria, serum albumin, total cholesterol, triglyceride, BUN and creatinine of all groups were measured. TNF-$\alpha$, IL-6, IL-1$\beta$, IL-10, IFN-$\gamma$, IgA, IgM and IgG levels of all groups were gauged. H&E staining and electron microscopy of the kidney were observed. Results: SRE showed significant decrease in the 24hrs proteinuria, serum triglyceride, BUN, TNF-$\alpha$, IL-6, serum IgA, IgM and IgG levels compared with the control group. SRE showed increase in the serum IL-10 and IFN-$\gamma$ levels compared with control on RT-PCR. SRE considerably decreased in the thickening of the GBM on H&E staining and deposition of electron-density on electron microscopy of the kidney compared with the control. Conclusions: According to the above results, it is suggested that SRE decreases the symptoms of MN induced by cBSA in mouse model. Therefore, SRE seems to be applicable to MN in clinical practice.

Effects of α-Linolenic Acid and Bovine Serum Albumin on Frozen-thawed Boar Sperm Quality during Cryopreservation

  • This study was conducted to evaluate effect of ${\alpha}$-linolenic acid (ALA) and bovine serum albumin (BSA) on viability, acrosome reaction and mitochondrial intact in frozen-thawed boar sperm. The boar semen was collected by gloved-hand method and cryopreserved using freezing extender containing 3 ng/mL ALA and/or $20\;{\mu}g/mL$ BSA. Cryo-preserved boar sperms were thawed in $37^{\circ}C$ water-bath for 45 sec to analysis. Viability, acrosome reaction, and mitochondrial intact were analyzed using flow cytometry. In results, viability of frozen-thawed boar sperm was significantly higher in only ALA+BSA supplement group than control group (p<0.05), whereas there was no difference either in ALA or BSA supplement. However, acrosome reacted sperm in both of live and all sperm population were significantly decreased in all treatment groups than control (p<0.05). Interestingly, mitochondrial intact of boar sperm was enhanced in ALA and ALA+BSA groups compared with control (p<0.05). In this study, we showed that supplementation of ALA and BSA in freezing extender enhanced the sperm viability, mitochondrial intact and decrease acrosomal membrane damage. In conclusion, our findings suggest that quality of frozen-thawed sperm in mammalians could improve by using of ALA and BSA.
