Studies on antimicrobial agent produced by lactobacillus acidophilus (Lactobacillus acidophilus가 생산한 항균물질(抗菌物質)에 관한 연구(硏究))

  • The research was conducted(1) to confirm the agent(s) responsible for the antimicrobial activity contained in the fermented tomato juice with L. acidophilus(2) to extract and purify the antimicrobial agent(s)(3) to find the biological, physical and chemical properties of the agent(s). The following results were obtained and summarized as followings; 1. The agent responsible for the inhibitory activity was confirmed by both well assay method using fermented tomato juice with L. acidophilus and turbidimetric technique using the cell-free filtrate or neutralized filtrate of tomato acidohilus culture and found exerted antimicrobial agent other than lactic acid. 2. The procedures of purification : The isolation and purification of antimicrobial agent from the lyophilized acidophilus tomato culture were carried out by (1) methanol extraction (2) acetone extraction, (3) Sephadex G-50 gel filtration (4) paper chromatography and (5) thin layer chromatography. 3. The biological, physical and chemical properties of antimicrobial agent: The biological, physical, chemical properties of the purified antimicrobial agent were: (1) The antimicrobial activity was strong against test organisms; Bacillus subtilis(ATCC 6633), Escheichia coli(ATCC 25922), Staphylococcus aureus(ATCC 167), Pseudomonas fluorescens(KFCC 32394), Proteus vulgaris and Shigella dysenteriae. (2) The pH value of the agent was 2.0 and the inhibitory activity was lost when it was neutralized at 7.0 of pH and the agent was heat stable at $121^{\circ}C$ for 60 minutes. (3) The ultraviolet light absorption spectra of methanol-acetone extract and TLC fraction exhibited a maximum absorption at 260nm and 224nm respectively. (4) The most purified agent from TLC plate increased about 130-fold in activity. (5) The agent isolated from TLC plate was free from $H_2O_2$ or lactic acid. 4. Bioautographic assy: By means of bioautography of the agent on silica gel of TLC plate a strong inhibitory activity against B. subtilis was demonstrated. 5. Mass spectrometry: The agent obtained from TLC plate was analyzed by mass spectrometry which show the parent peak at m/e 264 suggesting the molecular weight of the compound and molecular group such as [$C_2H{^+}_4$], [CO], [CH=NH], [$C_3{H^}4_7$], [$\begin{array}{rcl}O\\{\parallel}\\CH_3-C\\\end{array}$], [$C_6-H{^+}_{11}$], [$C_5H{^+}_{11}$], [$\begin{array}{rcl}O\\{\parallel}\\C_5H_7-C^+\\\end{array}$] were suggested.

  • This study was conducted to investigate the physiological activities and nutrition contents of Gonji No.10 and Solhyang, the strains of Neolentinus lepideus. In nutritional contents of two strains of N. lepideus, the content of free sugars and sugar alcohols and total polyphenol were more in Gonji No.10 than Solhyang. The total contents of amino acids and ${\beta}$-glucan in Solhyang are more than those in Gonji No.10. Solhyang also showed higher DPPH radical scavenging activity than Gonji No.10. In particular, hot water extract of Solhyang has higher inhibitory activity of ${\alpha}$-amylase and ${\alpha}$-amyloglucosidase while Gonji No.10 has higher anti-inflammatiory effect than Solhyang. But, they have similar effects on nitrite scavenging activity.

Modulation of Nrf2/ARE and Inflammatory Signaling Pathways by Hericium erinaceus Mycelia Extract

  • Hericium erinaceus is an edible mushroom used as a medicinal food in Asian countries. In this study, the chemopreventive effects of H. erinaceus mycelia hot water extract (HEW) were evaluated. HEW remarkably induced the luciferase activity of the antioxidant response element (ARE), located in the promoter region of phase 2 and antioxidant genes and regulated by nuclear factor E2-related factor 2 (Nrf2). The up-regulation of ARE activity by HEW corresponded with the induction of Nrf2 and the antioxidant enzyme, hemeoxygenase-1. The inhibition of cyclooxygenase-2 (COX-2) activity is a promising effective approach in cancer chemoprevention, and HEW prominently suppressed COX-2 protein expression in HepG2 cells. Furthermore, HEW showed anti-inflammatory activity by modulating inflammatory mediators such as nitric oxide (NO), inducible NO synthase, tumor necrosis factor-${\alpha}$, interleukin-$1{\beta}$, and the transcription factor, nuclear factor-${\kappa}B$, in lipopolysaccharide-stimulated RAW 264.7 cells. These results suggest that H. erinaceus possessed anti-tumor and anti-inflammatory effects via the modulation of Nrf2/ARE and inflammatory signaling pathways, and may therefore have potential use as a natural chemopreventive agent.

Development of Functional Beverage (SanYa) from Fermented Medical Plants and Evaluation of Its Physiological Activities (산야초를 이용한 기능성 발효음료개발 및 생리활성 연구)

  • This study was investigated to analyze the contents of flavonoid compounds and the effects of fermentation on the physiological activities of medical plants, also known as SanYa (SY). Antioxidative activity of the fermented SanYa (FSY) was measured by using DPPH radical scavenging and SOD-like activity. DPPH radical scavenging and SOD-like activity of FSY were 94.3% and 45.0%, respectively. Nitric oxide (NO) synthesis was increased 11 times through the addition of FSY. However, NO production of the macrophages RAW264.7 cells stimulated with lipopolysaccharide (LPS) was reduced to 56% through the addition of FSY. FSY showed fibrinolytic activity and indicated about 69.8% and 73.7% of xanthine oxidase and angiotensin converting enzyme inhibitory activities, respectively. These results suggested that FSY plays a significant role in fibrinolytic activity and have strong xanthine oxidase and angiotensin converting enzyme inhibitory activities.

Reproductive outcomes of retransferring retained embryos in blastocyst transfer cycles

  • Objective: To determine the incidence of embryo retention (ER) in the transfer catheter following embryo transfer (ET) in blastocyst transfer and investigate whether retransferring retained embryos has an impact on reproductive outcomes in patients undergoing in vitro fertilization-ET. Methods: We retrospectively analyzed the records of 1,131 blastocyst transfers, which comprised 223 single blastocyst transfer (SBT) and 908 double blastocyst transfer (DBT) cycles. Each SBT and DBT group was classified depending on whether ET was performed without retained embryos in the catheter during the first attempt (without-ER group) or whether any retained embryos were found following ET (ER group) for the purpose of comparing reproductive outcomes in a homogenous population. Results: The overall incidence of finding retained embryos was 2.8% (32/1,131). There were no retained embryos in SBT cycles. In DBT cycles, implantation rates (30.0% vs. 26.6%), positive ${\beta}-hCG$ rates (57.2% vs. 56.2%), clinical pregnancy rates (45.3% vs. 46.9%), and live birth rates (38.9% vs. 43.8%) were not significantly different between the without-ER and ER groups. There were no significant differences in the mean birth weight (g) $2,928.4{\pm}631.8$ vs. $2,948.7{\pm}497.8$ and the mean gestational age at birth ($269.3{\pm}17.2days$ vs. $264.2{\pm}25.7days$). A total of nine cases of congenital birth defects were found in this study population. Eight were observed in the without-ER group and one in the ER group. Conclusion: Our results suggest that retransfer of retained embryos does not have any adverse impact on reproductive outcomes in blastocyst transfer cycles. Furthermore, our results support finding that SBT might be advantageous for decreasing the incidence of retained embryos in catheters.

Antiinflammatory and Antiallergic Activity of Fermented Turmeric by Lactobacillus johnsonii IDCC 9203 (Lactobacillus johnsonii IDCC 9203으로 발효한 울금의 소염 및 항알레르기 효과)

  • Although turmeric has numerous pharmacological effects, the poor water-solubility of curcuminoids, active components of turmeric, restricts their systemic availability in orally administered formulations and limits their therapeutic potential. In this study we attempted turmeric fermentation using several probiotic bacteria to improve its solubility, and also investigated the effects of turmeric and fermented turmeric on anti-inflammatory activity. Fermented turmeric, by L. johnsonii IDCC 9203, more strongly inhibited LPS-induced expression of the pro-inflammatory cytokines than non-fermented turmeric and fermented turmeric by other probiotic strains. We used an NC/Nga mouse model for mite antigen-induced atopic dermatitis to examine the efficacy of the fermented turmeric. Fermented turmeric-fed mice exhibited a significantly reduced serum IgE level and mitigated acute inflammation. When the fermented turmeric was pre-treated by oral administration, it had more preventive activity against acute anaphylactic reaction than the non-fermented group. In addition, we observed that fermentation of turmeric leads to increased water-solubility of curcumin and a change in the active components ratios for bisdemethoxycurcumin, demethoxycrucumin and curcumin. Taken together, these results strongly suggest that fermented turmeric by L. johnsonii IDCC 9203 could be used as a functional food ingredient for improving treatments for atopic dermatitis.

Effect of Ascidian Tunic Extracts on Cholesterol Accumulation and Pigmentation of Kuruma Prawn, Penaeus japonicus (우렁쉥이 껍질 추출물이 보리새우 Cholesterol 축척 및 착색에 미치는 영향)

  • The effect of various levels of ascidian tunic extracts and carophyll pink on the growth rate, pigmentation, lipid and total cholesterol accumulation, and fatty acid compositions were studied in kuruma prawn, Penaeus japonicus. The kuruma prawn was fed the purified diets with or without ascidian tunic extract and carophyll pink at the levels of 100, 200, and 400 ppm for 8 weeks. In the experiment diet with ascidian tunic extracts or carophyll pink, the values of daily growth rate were ranged between $1.065\;to\;1.292%$, compared with control group. The content of astaxanthin in kuruma prawn was not significantly affected by the feeding levels of tunic extracts. Feeding of the tunic extracts, on the other hand, increased the kuruma prawn lipid and total cholesterol content, and pigment deposition in concentration-dependent manners without influencing the free astaxanthin concentration of prawn flesh and heads between two feeding groups(200 and 400 ppm). And it was also demonstrated that the dietary astaxanthin was deposited in kuruma prawn body tissue mainly as astaxanthin esters. The results suggest that the best feeding strategy for pigmentation in kuruma prawns is the diets with ascidian tunic extracts at the level of 4g/kg feed (200 ppm) for 8 weeks.

  • For centuries, Panax ginseng Meyer (Korean ginseng) has been widely used as a medicinal herb in Korea, China, and Japan. Ginsenosides are a class of triterpene saponins and recognized as the bioactive components in Korean ginseng. Ginsenosides, which can be classified broadly as protopanaxadiols (PPD), protopanaxatriols (PPT), and oleanolic acids, have been shown to flaunt a vast array of pharmacological activities such as immune-modulatory, anti-inflammatory, anti-tumor, anti-diabetic, and antioxidant effects. In recent years, a number of ginseng and ginsenoside researches have increasingly gained wide attention owing to its unique pharmacological properties. Although good efficacies of ginsenosides have been reported, lack of target specific delivery into tumor sites, low solubility, and low bioavailability due to modifications in gastro-intestinal environments limit their biomedical application in clinical trials. As a result to this major challenge, nanotechnology and drug delivery techniques play a significant role to solve this problematic issue. Thus, we reported the preparation of poly-ethylene glycol (PEG) and glycol chitosan (GC) functionalized to ginsenoside (Compound K and PPD) conjugates via hydrolysable ester bonds with improved aqueous solubility and pH-dependent drug release. In vitro cytotoxicity assays revealed that PEG-CK, and PPD-CK conjugates exhibited lower cytotoxicity compared to bare CK and PPD in HT29 cells. However, GC-CK conjugates exhibited higher and similar cytotoxicity in HT29 and HepG2 cells. Furthermore, GC-CK-treated RAW264.7 cells did not exhibit significant cell death at higher concentration of treatment which supports the biocompatibility of the polymer conjugates. They also inhibited nitric oxide production in lipopolysaccharide (LPS)-induced RAW64.7 cells. In addition to polymer-ginsenoside conjugates, silver (AgNps) and gold nanoparticles (AuNps) have been successfully synthesized by green chemistry using different m. The biosynthesized nanoparticles demonstrated antimicrobial efficacy, anticancer, anti-inflammatory, antioxidant activity, biofilm inhibition, and anticoagulant effect. Special interest on the effective delivery methods of ginsenoside to treatment sites is the focus of metal nanoparticle research.In short, nano-sizing of ginsenoside results in an increased water solubility and bioavailability. The use of nano-sized ginsenoside and P. ginseng mediated metallic nanoparticles is expected to be effective on medical platform against various diseases in the future.

Protective Effects of Cellular Membrane and Component Analysis of Polygonum aviculare Extracts (마디풀 추출물의 세포 보호 효과 및 주성분 분석)

  • In this study, the antioxidative effects and component analysis of Polygonum aviculare (P. aviculare) extracts were investigated. The ethyl acetate and the aglycone fraction from P. aviculare extracts were more active than (+)-${\alpha}$-tocopherol and $\small{L}$-ascorbic acid, which are known as strong antioxidants for their antioxidative activity by the DPPH method and chemiluminescence assay. The cellular protective effects of fractions of P. aviculare on the rose-bengal sensitized photohemolysis of human erythrocytes, increased in a concentration dependent manner ($1-10{\mu}l$). In particular, the ethyl acetate fraction at a concentration of $10{\mu}l$ showed the most prominent protective effect among all the extracts (${\tau}_{50}$, 314.70 min). TLC and HPLC chromatogram of the ethyl acetate fraction of P. aviculare extracts revealed 3 main bands (PA8, PA5, PA6) and peaks (peak 1, peak 2, peak 3), which were identified as myricetin-3-O-rhamnoside (myricitrin, PA8, peak 1), quercetin-3-${\alpha}$-rhamnoside (quercitrin, PA6, peak 3) by LC/ESI-MS/MS and $^1H$-NMR respectively. These results indicate that fractions from P. aviculare could be applicable to new functional cosmetics as antioxidants.

중력이상을 이용한 한반도 모호면 추출에 관한 연구

  • 중력이상 및 수치고도모델을 이용하여 한반도 모호면 심도를 추출하였다 중력이상값은 인공위성고도레이더 관폭값을 주로 이용한 전지구 모델을 이용하여 데이터영역 뿐 만 아니라 주파수영역에서도 자료의 균질성을 확보하였다. 모호면 추출은 Kim et al. [2000a]에 의해 제안된 스펙트럼 대비법 및 후리에급수를 이용한 파워스펙트럼분석법을 이용하였다. 전자는 지각근형을 전제로, 지형에 의한 중력효과와 후리에어 중력이상을 파동수영역에서 대비하여 모호면의 심도를 계산하는 방법이고, 후자는 완전부우게 중력이상으로부터 푸리에변환을 이용하여 지하 밀도 변화층의 심도를 계산하는 방법이다. 이 두 모호면은 서로 0.53의 상관관계를 갖고 있으며, 이는 모호면 산출의 방법론적인 차이 및 계산상의 오차인 것으로 사료된다. 이렇게 두 가지 독립적인 방법으로 추출된 모호면을 하나로 통합하기 위한 한 방법으로, 두 모호면의 차이를 계산한 후, 이를 최소자승법을 이용, 두 모호면을 보정하였다. 결과적으로 한반도의 최종 모호면의 평균심도는 32.0km, 표준편차는 2.5km 이며, 최소, 최대 심도는 20.3, 36.6km으로 나타났다. 이 경우 지형에 의한 중력효과는 스펙트럼대비법에 의해 제거된 결과이나, 한반도의 지각이 완전한 지각판 내에 놓여 있어서 Airy-Heiskanin 지각균형설의 가정이 타당성이 있는가, 혹은 국부적인 응력장에 의해 한반도의 지각이 과연 얼마나 지지되고 있는가 하는 것에 대한 추가적인 연구가 필요하며, 이에 앞서, 일정한 밀도차를 갖는 연속적인 밀도변화층이 존재한다는 가정이 반드시 필요하다.에는 관련성을 갖고 있으며, 이는 유류 분해정도를 파악하는 지시자로써 특정 무기 오염물질을 이용할 수 있을 가능성이 있으므로 좀더 이들 관계성에 대한 연구가 진행될 필요성이 있다고 판단된다.고 과학적으로 분석할 수 있는 방법이 될 수 있을 것으로 기대된다. 의미를 되새기는 것으로 짧은 연구를 시작하겠다. 등은 활성 값이 70% 이상으로 퇴적물 독성이 상대적으로 낮았다. 이중나선 DNA 함량은 28.4 % - 49%로 대조군에 비해서 감소가 크다. 대부분의 정점이 대조군의 30% 내외로 정점 간의 차이는 크지는 않다. 그러나 다른 측정자료와 같이 정점 22에서 18%로 최소치를 나타내고, 정점 2, 12에서 20% 내외의 값을 보인다. 종합적으로 볼 때 오염물질의 유입이 크고, 광양제철 인근 정점 들이 모두 다른 정점에 비해서 낮아서, 퇴적물 독성이 높은 정점으로 조사되었다.hiwo의 광합성 능력은 낮은 농도들에서는 대조구와 유사하였으나, 5 $\mu\textrm{g}$/l의 높은 농도에서는 초기에 매우 낮은 광합성 능력을 보이다가 시간이 경과하면서 대조군보다 더 높은 경향을 나타냈다. 이러한 결과는 식물플랑크톤이 benso[a]pyrene의 낮은 농도에서 노출될 때는 이 물질을 탄소원으로 사용할 가능성이 있음을 시사한다. 본 연구의 결과들은 연안해역에 benso[a]pyrene과 같은 지속성 유기오염물질이 유입되었을 때 내정여부에 따라 식물플랑크톤 군집내 종 천이와 일차생산력에 크게 영향을 미칠 수 있음을 시사한다.TEX>5.2개)였으며, 등급별 회수율은 각각 GI(8.5%), GII
