Cheese Manufacturing and Bioactive Substance Separation: Separation and Preliminary Purification of cAMP from Whey

  • Cheese consumption has been gradually increased in China. However, both the manufacturing process of cheese and the utilization of its main by-product were not well developed. Based on the sensory evaluation, Box-Behnken Design (BBD) was performed in the present study to optimize the cheese processing, which was proved more suitable for Chinese. The optimal parameters were: rennet 0.052 g/L, start culture 0.025 g/L and $CaCl_2$ 0.1 g/L. The composition analysis of fresh bovine milk and whey showed that whey contained most of the soluble nutrients of milk, which indicated that whey was a potential resource of cyclic adenosine-3', 5'-monophosphate (cAMP). Thus, the cAMP was isolated from whey, the results of high-performance liquid chromatography (HPLC) analysis showed that the macroporous adsorption resins (MAR) D290 could increase the concentration of cAMP from $0.058{\mu}mol/mL$ to $0.095{\mu}mol/mL$. We firstly purified the cAMP from the whey, which could become a new source of cAMP.

A Computational Investigation of the Stability of Cyclopropyl Carbenes

  • The conformations of dicyclopropyl, isopropyl cyclopropyl, and diisopropylcarbenes were optimized using density functional theory (B3LYP/6-31G(d)). We showed that the optimized geometries of carbenes with cyclopropyl groups are fully in accord with those expected for bisected W-shaped conformations, in which the effective hyperconjugation of a cyclopropyl group with singlet carbene can occur. The stabilization energies were evaluated at the B3LYP/6-311+G(3df, 2p)//B3LYP/6-31G(d) + ZPE level using an isodesmic equation. The relative stability of carbenes is in the order $(c-Pr)_2$C: > (i-Pr)(c-Pr)C: > $(i-Pr)_2$C:, and a cyclopropyl group stabilizes carbene more than an isopropyl group by nearly 9 kcal/mol. Energies for the decomposition of diazo compounds to carbenes increase in the order $(c-Pr)_2$ < (i-Pr)(c-Pr) < $(i-Pr)_2$ by ~9 kcal/mol each. From a singlettriplet energy gap ($E_{ST}$) calculation, the singlet level is lower than the triplet level and the $E_{ST}$ shows a trend similar to the stabilization energy calculations. For comparison, the optimized geometries and stabilization energies for the corresponding carbocations were also studied at the same level of calculation. The greater changes in geometries and the higher stabilization energies for carbocations compared to carbenes can explain the greater hyperconjugation effect.

여수해만에서 분리한 유독 와편모조류, Gymnodinium catenatum (Graham)의 성장에 미치는 수온, 염분과 광 조건 (Effects of Water Temperature, Salinity and Irradiance on the Growth of the Toxic Dinoflagellate, Gymnodinium catenatum (Graham) Isolated from Yeosuhae Bay, Korea)

  • 여수해만에서 분리한 Gymnodinium catenatum의 성장에 있어서 최적 수온과 염분 조건을 파악하기 위해 수온 6단계,염분 5단계의 조합에 의한 30단계의 배치배양을 실시하여, 성장속도를 계산하였다. C. catenatum은 약 18${^{\circ}C}$ 이상의 수온과 전 염분단계에서 0.3day$^{-1}$ 이상의 성장속도를 보였으며 수온의 감소와 함께 성장속도도 감소하였다. 하지만 염분은 성장에 그다지 영향을 미치지 않았다. 최대성장속도(0.5day$^{-1}$)는 수온 25${^{\circ}C}$와 염분 30psu에서 얻어졌다. 따라서, 수온과 염분에 따른 성장속도를 고려할 때, C. catenatum은 이른 여름부터 가을까지 한국남해연안해역에서 서식 가능한 것으로 판단된다 이 종의 출현을 예측하기 위한 모델식은 $\mu$ = 0.005-T$^2$ - 0.0001164 T$^3$ - 0.063-S + 0.005-S$^2$ - 0.00007608-S$^3$ - 0.003-T-S + 0.00005308-T$^2$-S로 나타났다. 한편. 수온 18${^{\circ}C}$와 염분 30psu 조건하에서 6단계의 광도 실험을 실시한 결과, 광량에 따른 C. catenatum의 성장은 $\mu$ = 0.16 (I - 10.4)/(1+21.8), (r=0.96)의 관계식으로 Ks는 42.6$\mu$mol m$^{-2}$s$^{-1}$, I$_0$는 10.4$\mu$mol m$^{-2}$s$^{-1}$이었다. 따라서 수온, 염분과 광 조건만을 고려할 경우 C. catenatum은 고수온기 한국 남해 내만과 연안해역에서 우점종으로 출현할 가능성이 높을 것으로 판단되었다.

  • Microstructure and electrical properties of ZnO-doped (0-5 mol%) 0.05 Pb(Mn1/3Sb2/3)O3-0.95 PZT ceramics were investigated. Sintering temperature was decreased to 100$0^{\circ}C$ due to eutetic reaction between PbO and ZnO. Grain-size increased up to adding 1mol% ZnO and then decreased. Compositions of grain and grain-boundary were investigated by WDS. Lattice parameter was decreased with ZnO addition. Density increased with ZnO addition and reached to the maximum of 7.84(g/cm2) at 2 mol% ZnO. The effect of ZnO on electrical properties of PMS-PZT was investigated. At 3mol% ZnO addition electromechanical coupling factor(kp) was about 50% and relative dielectric constant($\varepsilon$33/$\varepsilon$0) was 997 Mechanical quality factor(Qm) decreased with ZnO addition. Lattice parameters and tetragonality(c/a) were measured to investigate relationship between the electric properties and substitution of Zn2+. At 3 mol% ZnO tetragonality was maximiged at c/a=1.0035 Curie temperature (Tc) decreased slightly with ZnO addition.

  • In recent year, Ferroelectric $BLT(Bi_{4-x}La_{x}Ti_{3}O_{12})$ is a promising candidate materials. This study was practiced to make good conditions of BL T targets. In this study, calcination and sintering temperature were kept at $750^{\circ}C$, $1100^{\circ}C$ for 2 hour respectively. the density obtained 7.612, 7.98, $7.877g/cm^{3}$ as $La_{2}O_{3}$ contents were 0.0mol%, 0.25mol%, 0.5mol%. Especially, the lanthanum content of 0.5 mol% measured C-axis (117) preferred orientation more than the others targets in the XRD. In $\varepsilon_{r}-f$ relationship using by HP 4194 A impedance analyzer, the 0.5 mol% observed above 200 relative dielectric constant. but the dissipation factor was higher than others targets at 100Hz~13MHz range. SEM photograph with the content of $La_{2}O_{3}$ was observed like rod and plate types.

전동싸리 근류의 질소고정에 대한 온도의 영향 (Effect of Temperature on the Nitrogen Fixation Activity of Root Nodules of Melilotus suaveolens)

  • Effects of wintering and temperature on nitrogen fixation activity of nodules of Melilotus suaveolens Ledeb. grown in the field and growth chamber conditions were investigated. The biennial plants transfered to the growth chamber from winter field recovered the activity in 3 weeks of incubation and attained the maximum rate of $153{\mu}mol\;C_2H_4{\cdot}g$ fr wt $nodule^{-1}{\cdot}h^{-1}$ in 5 weeks. When root nodules which adapted to different temperatures, were pretreated with 10, 20 and $30^{\circ}C$ for 1 hour, and then transfered to $30^{\circ}C$, nitrogen fixation activity was promoted in the nodules exposed to lower field temperature ($12^{\circ}C$) with 1$0^{\circ}C$ pretreatment. M. suaveolens maintained nitrogen fixation activity in the wide range of temperatures, and was more tolerant to lower temperature than those of other woody leguminous plants, Diurnal changes of nodule activity showed increase with sunrise and decrease with sunset during spring and autumn, but the activity was inhibited during July and August because of high temperature with stron irradiation. Nitrogen fixation activity of annual plant appeared in mid-April, and showed two peaks (104 and 43 mol $C_2H_4{\cdot}g$ fr wt $nodule^{-1}{\cdot}h^{-1}$) in July and September, and then disappeared after October. Nitrogen fixation activity of biennial plant reappeared in mid-March after wintering and attained two peaks (102 and 82 ${\mu}mol\;C_2H_4{\cdot}g$ fr wt $nodule^{-1}{\cdot}h^{-1}$) in April and June of flowering period, and then disappeared after July due to plant withering by severe drought.

국산 장석으로부터의 Analcime합성 (Synthesis of Analcime by Treating of Domestic Feldspar)

  • Domestic Anyang feldspar was treated in autoclave with 1N sodium hydroxide solution for 1-6 hrs at 100-20$0^{\circ}C$ By X-ray diffraction patterns and cation exchange capacity, the products were examined. The following concecutive reaction was valid. AlbitelongrightarrowAmorphous aluminosilicatelongrightarrowAnalcime The reaction rate constants, k1 and k2, were 0.20 and 0.15(1/hr) at 20$0^{\circ}C$ respectively. The activiation energy from albite to amorphous aluminosilicate was 10 kcal/mol, and from amorphous aluminosilicate to analcine 13 kcal/mol. The analcime obtained was excellent in cation exchange capacity, amounting to 210meq./100g.

  • Characteristics of cell growth and poly(3-hydroxybutyrate-co-4-hydroxybutyrate) [P(3HB-co-4HB)] synthesis was investigated through flask and batch cultures of Alcaligenes latus and Comamonas acidovorans. The specific growth rate of C. acidovorans increased with yeast extract concentration and decreased with 1,4-butanediol concentration. Optimum glucose concentration for growth of C. acidovorans was 20 g/L. In one-step flask cultures of C. acidovorans, final dry cell weight and PHA content decreased with the ratio of 1,4-butanediol to glucose, while the 4HB fraction in copolymers gradually increased to 100 $mol\%$ with an initial 1,4-butanediol concentration of 20 g/L as single carbon source. The specific growth rate of A. latus decreased with v-butyrolactone concentration and optimum sucrose concentration for growth was 10 g/L. In batch cultures of A. latus, 4HB fraction increased with initial v-butyrolactone concentration. P(3HB-co-4HB) with 19 $mol\%$ 4HB was obtained when the initial ratio of v-butyloractone (g/L) to sucrose (g/L) was 10 : 10.

녹조 납작파래 (Enteromorpha compressa)의 질산염 흡수, 배아 생장 및 지방산 조성에 대한 광 및 온도의 영향 (Effects of Light and Temperature on Nitrate Uptake, Germling Growth and Fatty Acid Composition of Enteromorpha compressa (Chlorophyta))

  • Effects of light and temperature on the nitrate uptake and germling growth of Enteromorpha compressa (L.) Greville (Chlorophyta) were studied based on samples from Cheongsapo near Busan, Korea. In addition, their effects on fatty acids composition in thallus were examined. Nitrate uptake showed saturation kinetics. $V_{max}$ (maximal uptake rate) and its $K_s$ (half-saturation constant) at $20^{\circ}C,\;80\;{\mu}mol\;m^{-2}s^{-1},$ white light were $1.571\;{\mu}mol{\cdot}g\;fr\;wt^{-1}{\cdot}h^{-1}$ and 3.56 ${\mu}M$, respectively. In nitrate uptake with irradiance, wavelength and temperature, its rate represented respectively the highest value as $1.405\pm0.020,\;0.623\pm0.040,\;1.422\pm0.022\;{\mu}mol{\cdot}g\;fr\;wt^{-1}{\cdot}h^{-1}\;at\;100\;{\mu}mol\;m^{-2}s^{-1},$ red light, $20^{\circ}C$ and exhibited significant difference among the examined conditions (p<0.001). Germling growth of E. compressa also showed saturation kinetics, and $V_{max}$ and its $K_s$ value at $20^{\circ}C,\;100\;{\mu}mol\;m^{-2}s^{-1},$ 12:12 h were $56.18\%\;day^{-1}$ and 0.33 ${\mu}M$, respectively. SGR (specific growth rate) recorded a maximal value as 49.33-54.80, 39.07-50.72, $47.20-54.53\%\;dat^{-1}$ at $120\;{\mu}mol\;m^{-2}s^{-1},$ blue light and $18^{\circ}C$ respectively, and showed significant difference (p<0.001). Red light made the effective nitrate uptake, but germling growth was largely limited by the light. In fatty acids analysis, PUFAs (polyunsaturated fatty acids) were high at blue light, $18^{\circ}C,\;100\;{\mu}M\;NO_3^-.$ However, irradiance did not affect the production of PUFAs. In conclusion, nitrate uptake and germling growth of E. compressa showed saturation kinetics to external nitrate concentration, and were significantly affected by irradiance, wave length and temperature. Fatty acid composition was also influenced by the factors except for irradiance. Their maximal values, together with the highest production of PUFAs, were found at blue light band, $20^{\circ}C,\;100\;{\mu}mol\;m^{-2}s^{-1},\;and\;100\;{\mu}M\;NO_3^-.$

카본블랙의 농도 및 단량체 구성비에 따른 스티렌-부틸메타크릴레이트 공중합체 입자의 유동성 (Effect of Carbon Black Concentration and Monomer Compositional Ratio on the Flow Behavior of Copoly(styrene/butyl methacrylate) Particles)

  • 소수성실리카를 안정제로 하는 현탁중합반응법을 이용하여 합성한 스티렌(St)/부틸메타크릴레이트(BMA) 공중합체 (co-PSB) 입자의 전단점도를 모세관 레오미터 (capillary rheometer)를 이용하여 $170^{\circ}C$에서 측정하였다. co-PSB 입자의 전단점도는 중량평균분자량이 74,800 g/mol 이하인 경우 낮은 전단속도에서는 뉴톤거동을 보였다. 중량평균분자량이 136,800 g/mol을 초과하면서 전단점도는 전단속도의 전 영역에 걸쳐 감소하였고 전단속도에 대한 기울기의 절대값은 분자량의 증가와 함께 증가하였다. St/BMA의 구성비가 7/3, 5/5 및 3/7의 co-PSB 입자는 유사한 분자량을 나타내었지만 BMA의 구성비가 증가하면서 유리전이온도와 전단점도는 감소하였다. St/BMA의 구성비가 1/9 인 co-PSB의 경우 유리전이온도는 더욱 감소하였으나 초기 전단점도는 크게 증가하였다. 카본블랙을 함유하는 co-PSB 복합체 입자의 전단점도는 카본블랙의 증가에 따라 증가하였으나, 카본블랙의 농도에 따른 전단점도의 변화는 분자량 및/혹은 구성비의 변화 효과에 비교하여 미약하였다.
