Seven Previously Unrecorded Fungal Species Isolated from Freshwater Ecosystems in Korea

  • Various freshwater ecosystems, such as streams, lakes and rivers, provide a dynamic habitat for fungi. In this study, we isolated several fungal strains from freshwater sediment and plant litter. The strains were identified using molecular phylogenetic analyses of rDNA and beta tubulin (TUB) sequences. Morphological characteristics of the fungi were also investigated using microscopy and culture characteristics of the fungi grown on several media. We identified seven species previously unrecorded in Korea, Nothophoma spiraeae, Westerdykella aquatica, W. aurantiaca, W. dispersa, Chrysosporium sanyaense, C.pseudomerdarium and Taeniolella phialosperma.

Isolation and Characterization of Three Zygomycetous Fungi in Korea: Backusella circina, Circinella muscae, and Mucor ramosissimus

  • While surveying undiscovered fungal taxa in Korea, three rare zygomycetous fungal strains, CNUFC-PTF2-1, CNUFC-TF3-1, and CNUFC-ESAF3-1, were isolated from soil, leaf, and freshwater samples, respectively. The strains were analyzed morphologically as well as phylogenetically based on the internal transcribed spacer region and 28S rDNA sequences. Sequence analysis of the two loci revealed that the isolates, CNUFC-PTF2-1, CNUFC-TF3-1, and CNUFC-ESAF3-1, were identified as Backusella circina, Circinella muscae, and Mucor ramosissimus, respectively. These species have not yet been previously described in Korea.

Clonostachys divergens and Chrysosporium merdarium: Two New Records from Soil in Korea

  • During an investigation of micro-fungi in soil, two fungal isolates belonging to the phylum Ascomycota, namely KNUF-20-NI011 and KNUF-20-NI006, were collected from Gyeongbuk Province and Dokdo Island in Korea and identified as Clonostachys divergens and Chrysosporium merdarium, respectively. The fungal isolates were confirmed through molecular phylogenetic analyses of the internal transcribed spacer regions, 28S rDNA large subunit, and β-tubulin sequences. Cultural and morphological characteristics were observed and determined using different media. These species were identified based on phylogenetic relationships along with their cultural and morphological characteristics. To our knowledge, this is the first report on Clonostachys divergens and Chrysosporium merdarium in Korea.

  • Endophytic fungi were isolated from needle leaves of two conifer species in Korea. The fungal isolates were identified based on their morphological characteristics and phylogenetic analyses of their internal transcribed spacer and large-subunit rDNA regions. Three species of endophytic fungi, namely Celosporium laricicola, Neocatenulostroma germanicum, and Phaeophleospora eucalypticola were the first records in Korea. In this study, we reported the morphological and phylogenetic characteristics of these fungi.

First Report of Leptosphaerulina australis Isolated from Soil in Korea

  • The fungal strain KNU16-004 was isolated from a field soil sample collected in Seoul. The isolate was identified as Leptosphaerulina australis based on morphological characterization and phylogenetic analysis using the internal transcribed spacer (ITS), large subunit (LSU) rDNA regions, and ${\beta}-tubulin$ (Tub2). This is the first report of Leptosphaerulina australis in Korea.

Diversity of Fungi Isolated from Soil of Jeollabuk-do and Chungcheongbuk-do, Korea

  • This study was conducted aiming with the assessment of fungal diversity in soil samples collected from different locations of Jeollabuk-do and Chungcheongbuk-do, Korea. Forty soil samples were collected in 2015 and fungi were isolated through serial dilution technique. Isolated fungi were purified and differentiated according to their morphological and microscopic characteristics. In total, 150 different representative isolates were recovered and the genomic DNA of each isolate was extracted by using QIAGEN$^{(R)}$ Plasmid Mini Kit (QIAGEN Sciences, USA) and the identification of fungi was carried out by sequence analysis of internal transcribed spacer (ITS) region of the 18S ribosomal DNA (18S rDNA). Recovered isolates belonged to 37 family, 67 genera and 108 species. Aspergillus spp., Penicillium spp., Trichoderma spp., Chaetomium spp. And Fusarium spp. were the most dominant taxa in this study. Out of total species, 20 species were identified as new records for Korea.

제주도에 서식하는 식물 잎에서 분리된 6종의 국내 미기록 내생균 보고 (Characterization of Six Novel Endophytic Fungi Isolated from Leaves of Plants Inhabiting Jeju Island)

  • 제주도에 서식하는 다양한 식물의 잎에서 내생균을 분리하였다. 분리된 균주들은 형태적 특성 및 internal transcribed spacer, large subunit rDNA 영역 및 beta-tubulin rDNA 유전자의 염기서열의 계통 분석을 통해 종을 동정하였다. 그 결과 6종의 국내 미기록 내생균을 확인하였고, 확인된 종은 각각 Diaporthe goulteri, Diaporthe vaccini, Rhizosphaera pini, Valsa friesii, Xylaria primorskensis, Zalerion arboricola 이다. 확인된 6종의 미기록 내생균 균주의 형태적 특성 및 염기서열 계통분석의 결과에 대해 기술하였다.

First Report of Phaeosphaeria chengduensis Isolated from Gametis jucunda in Korea

  • A fungal strain, KNUF-4H-A belonging to the genus Phaeosphaeria was isolated from the citrus flower chafer (Gametis jucunda) in Chungcheongbuk-do, Korea. This strain was further identified as Phaeosphaeria chengduensis through phylogenetic analyses based on a concatenated dataset of DNA sequences of internal transcribed spacer (ITS) regions, as well as the small subunit rDNA (SSU), large subunit rDNA (LSU), RNA polymerase II second largest subunit (RPB2), and translation elongation factor 1-α (TEF1-α) genes. The isolate KNUF-4H-A exhibited typical cultural characteristics of P. chengduensis, producing colonies that were flattened, greenish-grey at the edges, grey-white at the center, and dark brown on the reverse side. Phaeosphaeria species have been reported in various ecosystems, including terrestrial and freshwater environments. The isolation of KNUF-4H-A from the citrus flower chafer provides valuable insights into the habitat diversity of Phaeosphaeria. This is the first record of Phaeosphaeria chengduensis in Korea.

동충하초의 계통분류 및 시판동충하초의 분류학적 위치 (Phylogenetic Analysis of the Entomopathogenic Fungal Species and Taxonomical Positions of Their Commercial Products)

  • 5.8S rDNA를 포함한 ITS부위에 대한 염기서열 분석결과, 종에 따라 다양한 염기서열을 가지고 있어 분류에 이용될 수 있었으며, 특히 ITS2부위보다 ITS1부위에서 종에 대한 변이율이 높은 것으로 나타났다. 아울러 균종에 따라 정도의 차이는 있으나 사용된 모든 종들이 서로 계통분류학적 거리가 멀어서 종간의 구분이 명확하게 나타났다. P. tenuipes, I. japonica, P. japonicus는 multiple alignment분석에서 매우 유사한 염기서열을 가지고 있어, 이들 세종은 같은 종이지만 다른 이름으로 불리고 있는 것으로 나타났으며, 아울러 Paecilomyces sp. KACC 40220과 KACC 40656도 동일한 염기서열을 가지고 있어 p. tenuipes로 판단된다. 국내에서 유통되는 동충하초제품 35건과 중국산 1건에 대해 실험한 결과 23건은 P. tenuipes / japonica로, 11건은 C. militaris로, 1건은 B. bassiana로 분류되었으며, 중국산 제품 1건은 C. multiaxialis로 분류되었다.

  • 본 연구에서는 소나무(Pinus densiflora)와 아로니아(Aronia melanocarpa)의 잎을 채취하여 표면살균한 후 내생균을 분리하였다. 분리한 균주는 형태적 특징과 internal transcribed spacer (ITS) rDNA 지역과 28S rDNA 지역 그리고 ${\beta}-tubulin$ 유전자의 염기서열을 이용하여 계통분석을 통해 동정하였다. 소나무에서 분리한 두 종의 균주인 Pestalotia lawsoniae와 Zasmidium fructicola, 그리고 아로니아에서 분리한 세 종의 균주인 Pestalotiopsis chamaeropis, Pestalotiopsis jesteri, Stagonosporopsis cucurbitacearum는 국내 미기록 진균으로 보고하고자 한다.
