시판 유통중인 한약재에서의 위해성 진균 분포에 관한 연구 (A Survey on the population of Toxigenic Fungi in Crude Drugs in Seoul)

  • We just carried out this study in order to obtain an appropriate information of the population of toxigenic fungi in crude drugs in seoul. Results of fungal examination on twenty-three kinds crude drugs were described in this report. In 21 crude drugs, colonies of fungi were possible to identify into 7 genera. Predominant genera of fungi in crude drugs were Aspergillus spp. $(14,\;46.6\%)$, Scopulariospsis spp. $(7,\;23.3\%)$, Penicillium spp., Rhizopus spp., Fusarium spp $(2,\;6.6\%,\;respectively)$ and Phoma sp., Chaetomium sp. $(1,\; 3.3\%,\;respectively)$. Mycotoxin producing fungi like Aspergillus spp., Penicillum spp., and Fusarium spp. were 18 colonies$(60\%)$, totally.

Fungal Biodiversity in Cardamom Protected Forests and Seima Biodiversity Conservation Area of Cambodia

  • Mushroom surveys and collections were conducted in the western and eastern forest areas in Cambodia, and then fungal biodiversity was analyzed by identifying mushrooms. One thousand and three hundreds eighty three specimens were identified by morphological and genetical characteristics, and were classified into 238 species, 160 genera, 52 families, 15 orders, and 3 phylums. The collected mushrooms were immersed in 70% ethyl alcohol for DNA extraction, and the rest of them were dried in the portable mushroom dryer for 12 hrs. Among these mushrooms, the genera Mycena (8.7%), Ganoderma (5.6%), Microporus (5.3%), Marasmius (4.2%), Marasmiellus (3.0%), Phellinus (2.5%), Trametes (2.5%), Hygrocybe (1.9%) and Pycnoporus (1.5%) were dominant. In the western Cambodia, 1,061 specimens were collected from Koh Kong forests, while 263 specimens were collected from the eastern Cambodia, Seima and Mondulkiri forests. Elevations of surveyed sites were ranged from 0 to 750 m above sea level. The number of species observed in the elevation of 251-500 m was the highest as compared to the other ranges of elevation. Daldinia concentrica, Microporus vernicipes, Microporus xanthopus, Pycnoporus coccineus, Stereum hirsutum, and Stereum ostrea were commonly distributed in all ranges of elevation, while the distribution of Ceratomyxa fruticulosa, Panus fulvus, Schizophyllum, Trametes versicolor, and Tyromyces chioneus were limited under 500 m. One hundred and forty one species including Amauroderma sp., Bjerkandera adusta, Trichaptum abietinum, and Tyromyces chioneus were collected only in Cardamom, while 20 species including Auricularia auricula-judae, Coriolopsis sanguinaria, Rigidoporus microporus, and Xylaria polymorpha were collected only in Seima. Ganoderma sp., Mycena sp., Marasmius sp., Microporus xanthopus, Phellinus sp., and Russula sp. were dominant species in both the western and eastern Cambodia. Species diversity indices in the eastern and western survey sites were 1.83 and 1.77, respectively, while evenness indices were 0.92 and 0.90. The species similarity index between two survey sites was 0.42.

순천만에 자생하는 염생식물의 뿌리로부터 분리된 내생진균의 유전적 다양성 (Genetic Diversity of Endophytic Fungi Isolated from the Roots of Halophytes Naturally Growing in Suncheon Bay)

  • 순천만에 자생하는 2종의 염생식물의 뿌리로부터 내생 진균을 분리하였다. 사용 되어진 염생식물은 칠면초와 천일사초이며, 스트렙토마이신이 포함된 최소배지를 사용하여 내생진균을 배양한 후 순수 분리하였다. 분리된 15 내 생진균 균주 rDNA의 ITS 영역의 염기서열을 분석한 결과 칠면초에서 5속, 천일사초에서 7속의 내생진균이 확인되었다. 그리고 Shannon's diversity index(H')를 이용하여 칠면초와 천일사초로부터 분리된 내생진균들의 종 다양성은 각각 1.561과 1.889로 확인되었으며 유연관계를 확인하기 위하여 계통분석을 수행하였다. 순천만 자생식물인 칠면초와 천일사초의 내생진균에서 자낭균문 Pezizomycotina 아문이 많이 분포하고 있음을 알 수 있었으며, Pezizomycotina 아문 중에서도 Sordariomycetes 강, Dothideomycetes 강, Eurotiomycetes 강에 속하는 내생진균이 칠면초와 천일사초에 모두 분포는 것을 확인하였다.

Fungal Diversity of Rice Straw for Meju Fermentation

  • Rice straw is closely associated with meju fermentation and it is generally known that the rice straw provides meju with many kinds of microorganisms. In order to elucidate the origin of meju fungi, the fungal diversity of rice straw was examined. Rice straw was collected from 12 Jang factories where meju are produced, and were incubated under nine different conditions by altering the media (MEA, DRBC, and DG18), and temperature ($15^{\circ}C$, $25^{\circ}C$, and $35^{\circ}C$). In total, 937 strains were isolated and identified as belonging to 39 genera and 103 species. Among these, Aspergillus, Cladosporium, Eurotium, Fusarium, and Penicillium were the dominant genera. Fusarium asiaticum (56.3%), Cladosporium cladosporioides (48.6%), Aspergillus tubingensis (37.5%), A. oryzae (31.9%), Eurotium repens (27.1%), and E. chevalieri (25.0%) were frequently isolated from the rice straw obtained from many factories. Twelve genera and 40 species of fungi that were isolated in the rice straw in this study were also isolated from meju. Specifically, A. oryzae, C. cladosporioides, E. chevalieri, E. repens, F. asiaticum, and Penicillium polonicum (11.8%), which are abundant species in meju, were also isolated frequently from rice straw. C. cladosporioides, F. asiaticum, and P. polonicum, which are abundant in the low temperature fermentation process of meju fermentation, were frequently isolated from rice straw incubated at $15^{\circ}C$ and $25^{\circ}C$, whereas A. oryzae, E. repens, and E. chevalieri, which are abundant in the high temperature fermentation process of meju fermentation, were frequently isolated from rice straw incubated at $25^{\circ}C$ and $35^{\circ}C$. This suggests that the mycobiota of rice straw has a large influence in the mycobiota of meju. The influence of fungi on the rice straw as feed and silage for livestock, and as plant pathogens for rice, are discussed as well.

적송 (Pinus densiflora) 림내 송이(Tricholoma matsutake) 발생지와 미발생지의 토양 균류의 수직 분포 (Microfungal flora of Tricholoma matsutake producing and nonproducing sites in the forest of Pinus densiflora)

  • 송이 (Tricholoma matsutake) 생산의 대표적인 지역으로 알려진 강원도 양양 지역과 명주 지역을 선택하여 송이 발생지와 미발생지의 미세균류를 분리 동정하였으며 수직분포를 조사하였다. 진균의 선별방법으로 희석법, $42^{\circ}C$에서 48시간 배양 후 $37^{\circ}C$로 배양하는 방법, $70^{\circ}C$에서 15분간 열처리 방법, 그리고 ethanol처리 방법으로 토양의 수직적 분포를 파악하였다. $42^{\circ}C$배양법에서는 두 장소에서 분리된 토양균은 7속 18종이 분리되었으며, 송이 발생지에서는 Aspergillus fumigatus, A. ochraceus, A. terreus, Acremonium sp., Penicillium frequentans, Talarom-yces stipitatus가 분리되었으며, 송이 미발생지에서는 Aspergillus fumigatus, Penicillium lilacinum, P. oxalicum, Westerdykella multispora가 우점종으로 나타났다. $70^{\circ}C$열처리 방법으로 분리한 경우 7속20종이 분리되었으며, 송이발생지에서는 Aspergillus fumigatus, Mucor sp. 가 우점적으로 나타났으며, 송이 미발생지에서는 A. iumigatus, Atternaria alter-nate, Mucor sp., Neurospora sitophita가 우점종으로 나타났다. Ethanol처리 방법으로 분리한 경우는 1속 1종이 분리되었으며, 송이 발생지와 미발생지에 관계없이 우점종은 Mortierella sp.로 분류되었다. 이들 분리된 토양균은 전체균의 수와 우점종균수 그리고 나타나는 종의 빈도수는 그 수직적 분포에 있어서 상층에서 하층으로 내려갈수록 감소하였다.

태안반도에 자생하는 해안식물 뿌리에서 분리한 내생진균의 다양성 분석 (Diversity Analysis of Endophytic Fungi Isolated from the Roots of Coastal Plants in Taean Peninsula)

  • 태안반도에 자생하는 해안식물인 해국(Aster sphathulifolius Maxim.), 갯개미취(Aster tripolium L.), 갈대(Phragmites australis), 갯꾸러미풀(Puccinellia nipponica Ohwi) 의 뿌리로부터 총 42주의 내생진균이 분리되었다. 그리고 분리된 내생진균의 ITS지역의 염기서열을 이용하여 동정한 결과 모두 자낭균문에 속하며, 8종류의 목(Botryosphaeriales, Capnodiales, Diaporthales, Dothideales, Eurotiales, Helotiales, Hypocreales, and Pleosporales)과 13종류의 속(Alternaria, Aureobasidium, Cadophora, Cladosporium, Davidiella, Diaporthe, Fusarium, Gibberella, Macrophomina, Metarhizium, Neosartorya, Penicillium, and Phoma)으로 분류되었으며, Eurotiales목의 Penicillium속의 분포비율이 가장 높게 존재하였다. Shannon 다양성지수를 비교한 결과 4종의 해안식물 중 갯개미취에서 분리한 내생진균의 다양성이 가장 높게 확인되었다.

국내산 미곡에 발생하는 곰팡이와 곰팡이독소 (Natural Occurrence of Mycotoxin and Fungi in Korean Rice)

  • 국내산 변질미와 저장 중 미곡에 발생하는 곰팡이로는 Aspergillus 속이 가장 많이 관찰되었으며, 분리한 곰팡이들의 일부는 아플라톡신 생성능을 보유하고 있었다. 대부분의 미곡시료에서는 Fusarium, Aspergillus, Penicillium, Alternaria 속 등의 다양한 곰팡이가 발생했으나 시료 내 아플라톡신 발생이나 오염수준은 허용기준치 이하로 미미한 반면 Fusarium 독소인 데옥시니발레놀, 니발레놀, 제랄레논, 푸모니신 등의 발생은 빈번하였다. 미곡종합처리장의 벼와 가공부산물의 경우, 곰팡이 및 곰팡이독소의 발생수준이 가공단계를 거칠수록 감소하여 백미에서 가장 낮았다. 현재, 벼의 가공부산물을 포함한 국내산 미곡의 경우, 아플라톡신 보다 Fusarium 곰팡이독소에 대한 안전관리가 더욱 필요한 것으로 판단된다.

곰팡이 노출에 따른 건강영향 및 민감 시설 내 곰팡이 분포 현황 (Health Effects of Exposure to Indoor Mold and the Levels of Mold in Facilities with Susceptible Populations in Korea)

  • Objectives: Exposure to mold is strongly associated with adverse health effects (development or exacerbation of allergic diseases). We reviewed the health effects of mold exposure and explored to determine the annual distribution of indoor mold in facilities with susceptible populations. Methods: The health effects of mold exposure were mainly summarized by reviewing related papers and WHO research reports. We selected 10 facilities, including daycare centers, postpartum care centers, medical institutions, and elderly care facilities within the Seoul Metropolitan. Mold sampling was performed once every week or once every quarter from February 2016 to 2017. In addition, fungal species analyses was performed, and distribution status by month and facility was analyzed in the same manner as concentration. Results: Adverse health effects attributed to fungal exposure are largely divided into allergic symptoms, toxic effects, and infectious effects. Monthly mean concentrations of mold indoors and outdoors was 368.8 CFU/㎥ (geometric mean 213.4 CFU/㎥) and 496.0 CFU/㎥ (327.9 CFU/㎥), respectively. The indoor concentration has begun to increase in February, peaked in July, declined in August, increased again until October, and then decreased in November. About 36 genera of indoor fungal species were found in each facility. Cladosporium sp., Penicillium sp., Fusarium sp., Aspergillus sp., Alternaria sp., and Arthrinium sp. were observed as the dominant species. Conclusions: Our findings showed that the overall level of indoor mold was below the 500 CFU/㎥ level recommended by the Ministry of Environment. The development of DNA-based assessment and expanding facilities to be monitored for mold would be necessary for preventive aspects.

At Death's Door: Alternaria Pathogenicity Mechanisms

  • The fungal genus Alternaria is comprised of many saprophytic and endophytic species, but is most well known as containing many notoriously destructive plant pathogens. There are over 4,000 Alternaria/host associations recorded in the USDA Fungal Host Index ranking the genus 10th among nearly 2,000 fungal genera based on the total number of host records. While few Alternaria species appear to have a sexual stage to their life cycles, the majority lack sexuality altogether. Many pathogenic species of Alternaria are prolific toxin producers, which facilitates their necrotrophic lifestyle. Necrotrophs must kill host cells prior to colonization, and thus these toxins are secreted to facilitate host cell death often by triggering genetically programmed apoptotic pathways or by directly causing cell damage resulting in necrosis. While many species of Alternaria produce toxins with rather broad host ranges, a closely-related group of agronomically important Alternaria species produce selective toxins with a very narrow range often to the cultivar level. Genes that code for and direct the biosynthesis of these host-specific toxins for the Alternaria alternata sensu lato lineages are often contained on small, mostly conditionally dispensable, chromosomes. Besides the role of toxins in Alternaria pathogenesis, relatively few genes and/or gene products have been identified that contribute to or are required for pathogenicity. Recently, the completion of the A. brassicicola genome sequencing project has facilitated the examination of a substantial subset of genes for their role in pathogenicity. In this review, we will highlight the role of toxins in Alternaria pathogenesis and the use of A. brassicicola as a model representative for basic virulence studies for the genus as a whole. The current status of these research efforts will be discussed.

Rice Straw-Decomposing Fungi and Their Cellulolytic and Xylanolytic Enzymes

  • Filamentous fungi colonizing rice straw were collected from 11 different sites in Korea and were identified based on characterization of their morphology and molecular properties. The fungi were divided into 25 species belonging to 16 genera, including 14 ascomycetes, one zygomycete, and one basidiomycete. Fungal cellulolytic and xylanolytic enzymes were assessed through a two-step process, wherein highly active cellulase- and/or hemicellulase-producing fungi were selected in a first screening step followed by a second step to isolate the best enzyme-producer. Twenty-five fungal species were first screened for the production of total cellulase (TC), endo-${\beta}$-1,4 glucanase (EG), and endo-${\beta}$-1,4 xylanase (XYL) using solid-state fermentation with rice straw as substrate. From this screening, six species, namely, Aspergillus niger KUC5183, A. ochraceus KUC5204, A. versicolor KUC5201, Mucor circinelloides KUC6014, Trichoderma harzianum 1 KUC5182, and an unknown basidiomycete species, KUC8721, were selected. These six species were then incubated in liquid Mandels' media containing cellulose, glucose, rice straw, or xylan as the sole carbon source and the activities of six different enzymes were measured. Enzyme production was highly influenced by media conditions and in some cases significantly increased. Through this screening process, Trichoderma harzianum 1 KUC5182 was selected as the best enzyme producer. Rice straw and xylan were good carbon sources for the screening of cellulolytic and xylanolytic enzymes.
