A small cationic peptide (JH8944) was tested for activity against a number of pathogens of agricultural crops. JH8944 inhibited conidium growth in most of the tested plant pathogens with a dose of $50{\mu}g/ml$, although one isolate of Fusarium oxysporum was inhibited at $5{\mu}g/ml$ of JH8944. Most conidia of Fusarium graminearum were killed within 6 hours of treatment with $50{\mu}g/ml$ of JH8944. Germinating F. graminearum conidia required $238{\mu}g/ml$ of JH8944 for 90% growth inhibition. The peptide did not cause any damage to tissues surrounding maize leaf punctures when tested at a higher concentration of $250{\mu}g/ml$ even after 3 days. Liposomes consisting of phosphatidylglycerol were susceptible to leakage after treatment with 25 and $50{\mu}g/ml$ of JH8944. These experiments suggest this peptide destroys fungal membrane integrity and could be utilized for control of crop fungal pathogens.
White rot, which is caused by Sclerotium cepivorum, is a lethal disease affecting green onions. Three different types of nanosilver liquid (WA-CV-WA13B, WA-AT-WB13R, and WA-PR-WB13R) were tested in several different concentrations on three types of media to assess their antifungal activities. Results from in vitro experiments showed that all three of the nano-silver liquids had more than 90% inhibition rates at a concentration of 7 ppm. Greenhouse experiments revealed that all of the nano-silver liquids increased biomass and dry weights, and there were minimal changes in the population of various bacteria and fungi from the soil of greenhouse-cultivated green onions. In addition, a soil chemical analysis showed that there were minimal changes in soil composition.
In the present study, we analyzed the defensin protein deduced from Korean radish (Raphanus sativus L.) seeds.To express the genes in E. coli, we constructed a recombinant expression vector with a defensin gene, named rKRs-AFP gene isolated from Korean radish seeds. Over expressed rKRs-AFP proteins was separated by SDS-PAGE to determine the purity, and protein concentration was determined by the Bradford method. Antifungal activity was assessed by disk assay method against the tested fungi. As a result, when 500 mL of cell culture were disrupted by sonicator, 32.5 mg total proteins were obtained. The purified protein showed a single band on SDS-PAGE with estimated molecular weight about 6 KDa, consistent with the molecular mass calculated from the deduced amino acid sequence. The purified rKRs-AFP protein showed remarkable antifungal activities against several fungi including Aspergillus niger, Botrytis cinerea causing the gray mold disease, and Candida albicans. In field tests using the purified rKRs-AFP protein, the protein showed the reducing activity of disease spot and the mitigating effect of spreading of disease like agrichemicals. The immuno-assay of rKRs-AFP protein showed that the purified protein entirely accumulated at B. cinerea cytoplasm through the hyphal septa shown by fluorescence imaging. There was no fluorescence inside the cell, when the hypha was incubated without the protein. These all results indicate that the recombinant rKRs-AFP proteins can be utilized as a potential antifungal drug to control harmful plant fungal pathogens.
Turkey has a very rich fungal flora due to its phytogeographical position. The screening of chemical content and active substances of mushrooms becomes an important subject not only for Turkey but also for all over the world. In the last decade, Analyses on phytochemical and biological activity of fungi have gradually increased as a result of improvement in the number and quality of facilities. In the scope of the present research, four medicinal mushrooms; Morchella elata, Lactarius volemus, Cantharellus cibarius and Tricholoma terreum were analyzed for their fatty acid compositions and antioxidant capacities. The fungal species have been found with unsaturated fatty acid/saturated fatty acid ratio of 6.73 for Morchella elata, 4.12 for Lactarius volemus, 5.21 for Cantharellus cibarius, 3.73 for Tricholama terrum. In addition, the concentration of malondialdehyde which was an indicator of lipid peroxidation was also determined in these species. According to the results, free radical scavenging activity of Morchella elata and Lactarius volemus were found higher than the other species. Any of the mushroom species investigated were found having very high metal chelating activity. The results showed that the extract of Morchella elata and Lactarius volemus exhibited significant antioxidant activities. Hence, the mushrooms have a potential to be a natural antioxidant in food industries as antioxidant agent.
Essential oil of Chamaecyparis obtusa showed antimicrobial on relatively broad spectrum of bacterial and fungal species. Staphylococcus epidermidis was highly sensitive to the essential oil but Streptococcus aureus and Streptococcus mutans were not. Vibro parahemolyticus, Pseudomoas aeruginosa and Pseudomoas putida showed sensitivity at the concentration higher that 400 ppm, Thegrowth of a pathogenic yeast Candida albicans was inhibited by the essential oil above 200ppm. The radialgrowth of several filamentous fungi was also inhibited The antifungal activity of the essential oil was effective on two plant pathogens Fusarium oxysporum and Altenaria mali. These results sug-gest that essential oil of Chamaecyparis obtusa has an antimicrobial activity by inhibiting bacterial and fungal species.
Objectives: The purpose of this study was to investigate the antifungal effect of bee venom (BV) and sweet bee venom (SBV) against Candida albicans (C. albicans) clinical isolates. Methods: In this study, BV and SBV were examined for antifungal activities against the Korean Collection for Type Cultures (KCTC) strain and 10 clinical isolates of C. albicans. The disk diffusion method was used to measure the antifungal activity and minimum inhibitory concentration (MIC) assays were performed by using a broth microdilution method. Also, a killing curve assay was conducted to investigate the kinetics of the anti-fungal action. Results: BV and SBV showed antifungal activity against 10 clinical isolates of C. albicans that were cultured from blood and the vagina by using disk diffusion method. The MIC values obtained for clinical isolates by using the broth microdilution method varied from $62.5{\mu}g/mL$ to $125{\mu}g/mL$ for BV and from $15.63{\mu}g/mL$ to $62.5{\mu}g/mL$ for SBV. In the killing-curve assay, SBV behaved as amphotericin B, which was used as positive control, did. The antifungal efficacy of SBV was much higher than that of BV. Conclusion: BV and SBV showed antifungal activity against C. albicans clinical strains that were isolated from blood and the vagina. Especially, SBV might be a candidate for a new antifungal agent against C. albicans clinical isolates.
Although siderophores are induced primarily in response to iron deficiency, soil and other ecological factors can affect on this process. This study was to evaluate the production of siderophores by different fungal species isolated from heavy metal contaminated and uncontaminated soils. More than thirty fungal strains were isolated from heavy metal contaminated and rhizosphere uncontaminated soils. Chrome azurol sulfonate (CAS) was used for both quantitative and qualitative evaluation of siderophores production. No significant correlations were observed between the tested variables such as ultraviolet (UV) irradiation method and CAS-agar plate and heavy metal concentration in both soils. The production of siderophores in rhizosphere fungi was higher than those isolated from the contaminated soil; however, the difference was not significant. The siderophore production (%) by fungi isolated from heavy metal contaminated soil using UV irradiation method was positively correlated with the qualitative values using CAS-plate method (P<0.05). Pearson correlation test indicated a positive correlation between the quantitative and qualitative methods of detection for fungi isolated from rhizosphere and also those isolated from heavy metal contaminated soil.
A fungal disease of the cultivated mushroom, Ganoderma lucidum, caused by Xylogone sphaerospora was epidemic throughout all cultivation areas in Korea which caused a lot of yield losses in the mushroom production. For controlling the disease, the screening of effective fungicides against the pathogenic fungus were conducted. Thirty seven commercially available fungicides were tested for their inhibitory activities on potato dextrose agar media supplemented with these fungicides at various concentrations. Twenty one fungicides significantly inhibited mycelial growth of the pathogen, Xylogone sphaerospora, but 16 fungicides had no inhibitory effect. Among these 21 fungicides, 17 fungicides also inhibited mycelial growth of Ganoderma lucidum as well, but imazalil, procymidone, triforine, and vinclozolin had no inhibitory effects. However, vinclozolin showed no inhibitory effect on mycelial growth of the mushroom even at the concentration of 50 $\mu\textrm{g}$/ml vinclozolin solution for 2 hours, and then the pathogen was inoculated. After two month-cultivation of the mushroom, over 90% of logs treated with vinclozolin without pathogen inoculation produced fruiting bodies. However, fruiting bodies were not produced form the logs inoculated with the pathogen, but not treated with vinclozolin. Fifty seven percent of logs. which were pre-treated with vinclozolin and then inoculated with the pathogen produced fruiting bodies. Based on the results, vinclozolin is effective for the control of yellow disease of the Ganoderma lucidum caused by Xylogone sphaerospora.

The biotransformation of monoterpenic agro-industrial wastes (turpentine oil and essential orange oil) was studied. More than 40 fungal strains were isolated from Brazilian tropical fruits and eucalyptus trees and screened for biotransformation of the waste substrates. Solid phase microextraction was used to monitor the presence of volatile compounds in the headspaces of sporulated surface cultures. The selected strains were submitted to submerged liquid culture. The biotransformation of R-(+)-limonene and ${\alpha},\;{\beta}-$ pinenes from the oils resulted in ${\alpha}-terpineol$ and perillyl alcohol, and verbenol and verbenone, respectively, as the main products. The selected strains were also placed in contact with ${\alpha}-$ and ${\beta}-$ pinenes standards. It was confirmed that verbenol, verbenone, and ${\alpha}-terpineol$ were biotransformation products from the terpenes. A concentration of 90 mg/L of verbenone was achieved by Penicillium sp. 2360 after 3 days of biotransformation.
In this study, the decolorization of Evercion Blue P-GR (EBP) and Ostazin Black H-GRN (OBH) was investigated using white-rot fungi named as Trametes versicolor (T. versicolor) by Membrane Bioreactor (MBR) system. This study involved experiments employing synthetic textile wastewater in Membrane Bioreactor (MBR) system (170 ml), initially inoculated with a pure culture of fungi, but operated, other than controlling pH (4.5±0.2) and temperature (25±1℃), under non-sterile conditions. The effect of dye concentrations on fungal biodegradation was also investigated. The decolorization efficiencies were 98%, 90%, and 87% respectively, for EBP when the initial dye concentration of 50, 100, and 200 mg L-1 were used. However, the decolorization percentages for OBH dye were obtained 95% for 50 mg L-1 dye solution in 2 days and 66% for 100 mg L-1 dye solution in 5 days. Possible interactions between dye molecules and the fungal surface were confirmed by SEM, EDX, and FTIR analyses.
