Candida albicans에 대한 생약의 항진균성에 관한 연구(I) (Antifungal Actions of Crude Drug Water Extracts on Candida albicans(I))

  • Some crude drugs in ancient literatures have been used as traditional therapeutic agent of leucorrhea mainly caused by Trichomonas vaginalis and Candida albicans. Sixty six kinds of crude drugs in ancient literatures and ten constituents were selected as sample drugs. Trichomycin standard was tested to compare with the above drugs. To determine the anti-fungal effect of these drugs on Candida albicans Yu 1200, a test organism, screening test was conducted. Antifungal activities of crude drug water extracts were observed by means of two test methods : firstly through the agar slant method and secondly the counting chamber method which was used for acknowledged drug agents upon the result of the agar slant method. And in order to improve the fungicidal effect, the organisms were stained with 0.02% methylene blue solution. The results of the above test indicated that Fritillariae Rhizoma has antifungal action in the concentration of 310mcg/ml, Coptidis Rhizoma in 620mcg/ml, Meliae Cortex, Scutellariae Radix both in 5,000mcg/ml. Baicalin, catechol among the pure isolated constituents inhibited in the range of 50mcg/ml. This score was based on 50% inhibition in comparison with amounts of control organisms. Rhei Rhizoma, Mori Radicis Cortex, Linderae Radix, and Amomi globosi Fructus showed the antifungal effect moderately in 5,000mcg/ml, and baicalein and pectolinarin in 50mcg/ml in the limit of between 35% and 50% antifungal activity. Staining with 0.02% methylene blue showed that any of the crude drug extracts was unable to stain the cells, but trichomycin in 0.86unit/ml able to stain 12% of the cells. This result means that crude drugs probably do not have fungicidal but fungistatic action.

Gold Nanoparticles Conjugation Enhances Antiacanthamoebic Properties of Nystatin, Fluconazole and Amphotericin B

  • Parasitic infections have remained a significant burden on human and animal health. In part, this is due to lack of clinically-approved, novel antimicrobials and a lack of interest by the pharmaceutical industry. An alternative approach is to modify existing clinically-approved drugs for efficient delivery formulations to ensure minimum inhibitory concentration is achieved at the target site. Nanotechnology offers the potential to enhance the therapeutic efficacy of drugs through modification of nanoparticles with ligands. Amphotericin B, nystatin, and fluconazole are clinically available drugs in the treatment of amoebal and fungal infections. These drugs were conjugated with gold nanoparticles. To characterize these gold-conjugated drug, atomic force microscopy, ultraviolet-visible spectrophotometry and Fourier transform infrared spectroscopy were performed. These drugs and their gold nanoconjugates were examined for antimicrobial activity against the protist pathogen, Acanthamoeba castellanii of the T4 genotype. Moreover, host cell cytotoxicity assays were accomplished. Cytotoxicity of these drugs and drug-conjugated gold nanoparticles was also determined by lactate dehydrogenase assay. Gold nanoparticles conjugation resulted in enhanced bioactivity of all three drugs with amphotericin B producing the most significant effects against Acanthamoeba castellanii (p < 0.05). In contrast, bare gold nanoparticles did not exhibit antimicrobial potency. Furthermore, amoebae treated with drugs-conjugated gold nanoparticles showed reduced cytotoxicity against HeLa cells. In this report, we demonstrated the use of nanotechnology to modify existing clinically-approved drugs and enhance their efficacy against pathogenic amoebae. Given the lack of development of novel drugs, this is a viable approach in the treatment of neglected diseases.

  • Plant essential oils are used in products such as fragrances and cosmetics due to their individual aromatic characteristics. Currently, essential oils are not only used in cosmetics but also in pharmaceutical products with anti-bacterial, anti-viral, anti-fungal, anti-parasitic, insecticidal, anti-cancer, neuroprotective, psychophysiological, or anti-aging effects. Despite their pharmaceutical properties, some studies reported cytotoxic effects in high doses. Therefore, for pharmaceutical purposes, the margin of safety of essential oils needs to be examined. Herein, we evaluated the IC50 of 10 essential oil from Korean native plants: Juniperus chinensis L. var. sargentii Henry, Citrus natsudaidai Hayata, Citrus reticulata Blanco, Citrus unshiu (Yu. Tanaka ex Swingle) Marcow, Artemisia capillaris Thunb, Aster glehnii F. Schmidt, Juniperus chinensis L, Zanthoxylum schinifolium Siebold & Zucc, Zanthoxylum piperitum (L.) D, and Cinnamomum loureirii. In addition, gene regulation of the cell-cycle gene and apoptosis marker CASP3 was examined at the IC50 level. The purpose of this study was to describe the toxic concentrations of essential oils extracted from Korean native plants, thereby providing toxic concentration guidelines for inclusion in a toxicity database and in the application of plant essential oils in various fields.

Experimental design approach for ultra-fast nickel removal by novel bio-nanocomposite material

  • In the present study, novel chitosan coated magnetic magnetite (Fe3O4) nanoparticles were successfully biosynthesized from mushroom, Agaricus campestris, extract. The obtained bio-nanocomposite material was used to investigate ultra-fast and highly efficient for removal of Ni2+ ions in a fixed-bed column. Chitosan was treated as polyelectrolyte complex with Fe3O4 nanoparticles and a Fungal Bio-Nanocomposite Material (FBNM) was derived. The FBNM was characterized by using X-Ray Diffractometer (XRD), Scanning Electron Microscopy-Energy Dispersive X-Ray Spectroscopy (SEM-EDS), Fourier Transform Infrared spectra (FTIR) and Thermogravimetric Analysis (TGA) techniques and under varied experimental conditions. The influence of some important operating conditions including pH, flow rate and initial Ni2+ concentration on the uptake of Ni2+ solution was also optimized using a synthetic water sample. A Central Composite Design (CCD) combined with Response Surface Modeling (RSM) was carried out to maximize Ni2+ removal using FBNM for adsorption process. A regression model was derived using CCD to predict the responses and analysis of variance (ANOVA) and lack of fit test was used to check model adequacy. It was observed that the quadratic model, which was controlled and proposed, was originated from experimental design data. The FBNM maximum adsorption capacity was determined as 59.8 mg g-1. Finally, developed method was applied to soft drinks to determine Ni2+ levels. Reusability of FBNM was tested, and the adsorption and desorption capacities were not affected after eight cycles. The paper suggests that the FBNM is a promising recyclable nanoadsorbent for the removal of Ni2+ from various soft drinks.

Preparation of Nanomaterial Wettable Powder Formulations of Antagonistic Bacteria from Phellodendron chinense and the Biological Control of Brown Leaf Spot Disease

  • Brown leaf spot disease caused by Nigrospora guilinensis on Phellodendron chinense occurs in a large area in Dayi County, Chengdu City, Sichuan Province, China each year. This outbreak has severely reduced the production of Chinese medicinal plants P. chinense and caused substantial economic losses. The bacterial isolate JKB05 was isolated from the healthy leaves of P. chinense, exhibited antagonistic effects against N. guilinensis and was identified as Bacillus megaterium. The following fermentation medium and conditions improved the inhibitory effect of B. megaterium JKB05 on N. guilinensis: 2% glucose, 0.1% soybean powder, 0.1% KCl, and 0.05% MgSO4; initial concentration 6 × 106 cfu/ml, and a 42-h optimal fermentation time. A composite of 0.1% nano-SiO2 JKB05 improved the thermal stability, acid-base stability and ultraviolet resistance by 16%, 12%, and 38.9%, respectively, and nano-SiO2 was added to the fermentation process. The best formula for the wettable powder was 35% kaolin, 4% polyethylene glycol, 8% Tween, and 2% humic acid. The following quality test results for the wettable powder were obtained: wetting time 87.0 s, suspension rate 80.33%, frequency of microbial contamination 0.08%, pH 7.2, fineness 95.8%, drying loss 1.47%, and storage stability ≥83.5%. A pot experiment revealed that the ability of JKB05 to prevent fungal infections on P. chinense increased considerably and achieved levels of control as high as 94%. The use of nanomaterials significantly improved the ability of biocontrol bacteria to control this disease.

Biological Control Potential of Penicillium brasilianum against Fire Blight Disease

  • Erwinia amylovora is a causative pathogen of fire blight disease, affecting apple, pear, and other rosaceous plants. Currently, management of fire blight relies on cultural and chemical practices, whereas it has been known that few biological resources exhibit disease control efficacy against the fire blight. In the current study, we found that an SFC20201208-M01 fungal isolate exhibits antibacterial activity against E. amylovora TS3128, and the isolate was identified as a Penicillium brasilianum based on the 𝛽-tubulin (BenA) gene sequence. To identify active compounds from the P. brasilianum culture, the culture filtrate was partitioned with ethyl acetate and n-butanol sequentially. From the ethyl acetate layer, we identified two new compounds (compounds 3-4) and two known compounds (compounds 1-2) based on spectroscopic analyses and comparison with literature data. Of these active compounds, penicillic acid (1) exhibited promising antibacterial activity against E. amylovora TS3128 with a minimal inhibitory concentration value of 25 ㎍/ml. When culture filtrate and penicillic acid (125 ㎍/ml) were applied onto Chinese pearleaf crab apple seedlings prior to inoculation of E. amylovora TS3128, the development of fire blight disease was effectively suppressed in the treated plants. Our results provide new insight into the biocontrol potential of P. brasilianum SFC20201208-M01 with an active ingredient to control fire blight.

Antifungal Activity of Green and Chemically Synthesized ZnO Nanoparticles against Alternaria citri, the Causal Agent Citrus Black Rot

  • Citrus black rot is a serious disease of citrus plants caused by Alternaria citri. The current study aimed to synthesize zinc oxide nanoparticles (ZnO-NPs) by chemically or green method and investigate their antifungal activity against A. citri. The sizes of synthesized as measured by transmission electron microscope of ZnO-NPs were 88 and 65 nm for chemical and green methods, respectively. The studied prepared ZnO-NPs were applied, in vitro and in situ, at different concentrations (500, 1,000, and 2,000 ㎍/ml) in post-harvest treatment on navel orange fruits to verify the possible control effect against A. citri. Results of in vitro assay demonstrated that, at concentration 2,000 ㎍/ml, the green ZnO-NPs was able to inhibit about 61% of the fungal growth followed by 52% of chemical ZnO-NPs. In addition, scanning electron microscopy of A. citri treated in vitro with green ZnO-NPs showed swelling and deformation of conidia. Results showed also that, using a chemically and green ZnO-NPs at 2,000 ㎍/ml in situ in post-harvest treatment of orange, artificially-infected with A. citri, has reduced the disease severity to 6.92% and 9.23%, respectively, compared to 23.84% of positive control (non-treated fruits) after 20 days of storage. The out findings of this study may contribute to the development of a natural, effective, and eco-friendly strategy for eradicating harmful phytopathogenic fungi.

시폰케이크 제조 시 첨가한 대잎 분말이 케이크의 품질 및 보존성에 미치는 영향 연구 (Quality Characteristics and Storage Properties of Chiffon-cake Containing added Bamboo Leaf Powder)

  • Bamboo leaf powder was added to Chiffon cake to increase its neutraceutical effects and storage periods. Specifically, 0, 7, 10 and 13% leaf powder was added to the cakes. The content of total dietary fiber in the bamboo leaf powder was 65.57%. The moisture content did not differ significantly among groups. The cake containing 10% bamboo leaf powder had the greatest volume, whereas the control group (0% bamboo leaf powder) had the lowest volume (p<0.05). The Hunter's L and a values decreased significantly as the amount of bamboo leaf powder increased. The b value of the control was lowest among the groups (p<0.05). Evaluation of the consumer acceptance of flavor revealed that the cakes containing the added bamboo leaf had greater consumer acceptance than the control. However, when the color was evaluated, the 13% group showed the lowest acceptance (p<0.05). Other factors such as texture, taste and overall acceptance did not differ significantly among groups. Additionally, the elasticity did not differ among groups, while the air cells were most uniform in the control. The strength of bamboo leaf aroma, bitter taste and aftertaste increased as the amount of bamboo leaf powder added increased. Cakes containing 10% and 13% added powder had the greatest moisture content, while the control had the lowest content (p<0.05). As in previous studies, the results of this study indicated that 10% bamboo leaf powder was the optimal level for the preparation of Chiffon cake. To evaluate the storage of cakes, the 10% group and the control were inoculated with Aspergillus oryzae and then incubated at $30^{\circ}C$ for 6 days. The microbial colony counts in the control group were dramatically increased after 48 hrs; however, the fungal concentration of the 10% group did not increase for 4 days. In conclusion, the addition of 10% bamboo leaf powder to Chiffon cake increased the storage time while maintaining adequate consumer acceptance.

Coated cysteamine, a potential feed additive for ruminants - An updated review

  • For sustainable development, better performance, and less gas pollution during rumen fermentation, there is a need to find a green and safe feed additive for ruminants. Cysteamine (CS) is a biological compound naturally produced in mammalian cells. It is widely used as a growth promoter in ruminants because of its ability to control hormone secretions. It mainly controls the circulating concentration of somatostatin and enhances growth hormone production, leading to improved growth performance. CS modulates the rumen fermentation process in a way beneficial for the animals and environment, leading to less methane production and nutrients loss. Another beneficial effect of using CS is that it improves the availability of nutrients to the animals and enhances their absorption. CS also works as an antioxidant and protects the cells from oxidative damage. In addition, CS has no adverse effects on bacterial and fungal alpha diversity in ruminants. Dietary supplementation of CS enhances the population of beneficial microorganisms. Still, no data is available on the use of CS on reproductive performance in ruminants, so there is a need to evaluate the effects of using CS in breeding animals for an extended period. In this review, the action mode of CS was updated according to recently published data to highlight the beneficial effects of using CS in ruminants.

MAP 포장재인 숨쉬는 필름이 장거리 수출 조건에서 토마토의 품질과 저장성 향상에 미치는 영향 (Effect of Breathable Film for Modified Atmosphere Packaging Material on the Quality and Storability of Tomato in Long Distance Export Condition)

  • 본 연구는 토마토 (cv 'adison')의 장거리 수출을 위한 MAP에 적합한 숨쉬는 필름을 구명하기 위해 실시하였다. 저장중 생체중 감소는 5,000cc, 10,000cc, $20,000cc/m^2{\cdot}day{\cdot}atm$의 산소투과도를 가지는 숨쉬는 필름 포장 처리구에서 0.6% 이하의 낮은 수준을 보인 반면, 기존에 원예작물 포장용으로 사용되던 천공필름은 1.4%의 감소를 보였다. $5^{\circ}C$ 저장중 포장재내 이산화탄소와 산소 농도는 20,000cc 필름처리에서 토마토 MA저장에 적합한 수준으로 유지되었다. $20^{\circ}C$ 저장에서는 이산화탄소 농도는 크게 증가하였고, 반면 산소농도는 급격히 감소하였는데, 역시 20,000cc 필름 처리에서 가장 변화폭이 적었다 포장재내 에틸렌 농도변화도 $5^{\circ}C$ 저장 후 $20^{\circ}C$에서 저장한 21일부터 25일까지 크게 증가하였는데, 숨쉬는 필름처리 중에서는 20,000cc 처리에서 가장 낮았다. 토마토의 상품성은 $5^{\circ}C$ 저장 20일까지 모든 처리구에서 유지되었으나, $20^{\circ}C$ 저장 5일째부터 곰팡이가 발생하면서 품질이 급격히 열화되었다. 저장 최종일에 분석한 경도, 비타민 C, 당도는 20,000cc 처리구에서 각각 9.56N, 16.31mg/100 gFW, 그리고 $7^{\circ}Brix$로 가장 높게 유지되었다. 이상의 결과로 볼 때, $20,000cc/m^2{\cdot}day{\cdot}atm$ 산소 투과도를 가진 숨쉬는 필름이 20일 이상의 장거리 수출에서 적합한 MAP용 포장재라고 생각되었다.
