We have demonstrated previously on a replication incompetent recombinant adenoviral vector, AdLPCD, in which the expression of cytosine deaminase (CD) gene is driven by the tumor-specific L-plastin promoter. The object of this study was to evaluate the efficacy of AdLPCD together with 5-fluorocytosine (5-FC) in suppression of the growth of established human tumor cells of ovary, Consistent with the knowledge that infection of OVCAR-3 cells with AdLPCD resulted in expression of a functional intracellular CD enzyme capable of converting 5-FC to 5-fluorouracil (5-FU) (Chung and Deisseroth, 2004), statistically significant differences in cytotoxicity were observed when AdLPCD infected cells were also exposed to 5-FC for 6 days (p=0.05), 9 days (p<0.0005) and 12 days (p<0.005), compared to 5-FC exposure alone, These results indicate that the CD gene delivered by adenoviral vector could efficiently sensitize OVCAR-3, otherwise non-toxic 5-FC. On the other hand, SKOV-3 cells, an ovarian carcinoma cell line, were more resistant to the CD/5-FC strategy compared with OVCAR-3 cells under the same condition. The results of present study suggest that the replacement of 5-FU with CD/5-FC in combination chemotherapy would be less toxic and much greater cytotoxicity than the conventional combination chemotherapy in some patients.
Although several artificial disc designs have been developed for the treatment of discogenic low back pain, biomechanical change with its implantation was rarely studied. To evaluate the effect of artificial disc implantation on the biomechanics of functional spinal unit, nonlinear three-dimensional finite element model of L4-L5 was developed with 1-mm CT scan data. Two models implanted with artificial discs, SB $Charit\acute{e}$ or Prodisc, via anterior approach were also developed. The implanted model predictions were compared with that of intact model. Angular motion of vertebral body, force on spinal ligaments and facet joint, and the stress distribution of vertebral endplate for flexion-extension, lateral bending, and axial rotation with a compressive preload of 400 N were compared. The implanted model showed increased flexion-extension range of motion and increased force in the vertically oriented ligaments, such as ligamentum flavum, supraspinous ligament and interspinous ligament. The increase of facet contact force on extension were greater in implanted models. The incresed stress distribution on vertebral endplate for implanted cases indicated that additinal bone growth around vertebral body and this is matched well with clinical observation. With axial rotation moment, relatively less axial rotation were observed in SB $Charit\acute{e}$ model than in ProDisc model.
무독성의 반응형 난연 도료를 제조할 목적으로 모체수지 1개의 구조단위 속에 2개의 인산기를 보유한 파이로포스포릭 락톤 변성 폴리에스터 (PATT)를 합성하고, PATT에 이소시아네이트인 toluene diisocyanate-isocyanurate를 상온경화시켜 2성분계 폴리우레탄 난연 도료(PIPUC)를 제조하였다. PIPUC로서 도막제작 후 비난연 도료와의 도막물성을 비교한 결과 난연 성분 도입에 따른 난연 도료의 물성이 저하되지 않음을 알았다. 난연성 시험 중 45$^{\circ}$ Meckel burner 법에서는 탄화길이가 3.1~4.4 cm를, LOI법에서는 LOI 27~30%를 각각 나타냄으로써 양호한 난연 효과를 보여주었고, SEM에 의한 표면 도막을 관찰한 결과 도막의 결함이나 상분리 현상이 생기지 않음을 관찰하였다.
Monitoring the carbon dioxide concentration in arterial blood is vital for the evaluation and prevention of pulmonary disease. Yet, domestic pure arterial blood carbon dioxide sensor technologies are not being developed, instead all sensors are imported. In this paper, we develop a real time monitoring system for arterial blood partial pressure of carbon dioxide($pCO_2$) gas from the wrist by using a carbon micro-heater. The micro-heater was fabricated with a thickness of 0.3 ${\mu}m$ in order to collect the carbon dioxide under the skin. The micro-heater has been designed to perform temperature compensation in order to prevent damage to the skin. Two clinical trials of the system were undertaken. As a result, we demonstrated that a portable, transcutaneous carbon dioxide analysis($TcpCO_2$) device produced domestically is possible. In addition, this system reduced the analysis time significantly. Carbon films could reduce the unit price of these sensors by replacing the gold film used in foreign models. Also, we developed a real time monitoring system which can be used with optical biosensors for medical diagnostics as well as gas sensors for environmental monitoring.
It has been well established that leaf longevity is linked to the carbon economy of plants. We used this relationship to predict leaf lifetime carbon gains from leaf lifespan, and estimated the gross primary production (GPP) of a young deciduous forest of Japanese beech (Fagus crenata) located in central Japan. The light-saturated photosynthetic rates of the leaves were measured repeatedly during the growing season. We used the leaf lifespan to calculate the conversion coefficient from the light-saturated photosynthetic rate into the realized leaf lifetime carbon gain under field conditions. The leaf turnover rate was estimated using litter traps. GPP was estimated as the product of lifetime carbon gain per unit of leaf mass, and the annual leaf turnover rate. The GPP of the forest in 2007 was estimated to be $1.2{\times}10^3gCm^{-2}y^{-1}$, which was within the range of previously reported GPP values of beech forests in Japan, and was close to the GPP of a European beech forest, as estimated by eddy flux measurements.
In manufacturing of flatfish skin collagen peptide (FSCP) and flatfish protein hydrolysate (FPH) by reuse of dead flatfish from fish farm in Jeju island, the industrial process was optimized with the laboratory scale research and the on-field process. Segmented unit processes from raw material incoming to shipment were established to produce commercial product of FSCP and FPH. Total plate counts of FSCP were twenty five times of FPH, but food poisoning bacteria were not detected in two samples. FSCP and FPH were safe from heavy metal such as Pb(II), Cd(II) and Hg(II). The residual contents of antibiotics and disinfection matter in FSCP and FPH were not detected. The optimized process for mass production made the one-third of the running time and two times of the yield. From economic analysis, the production cost was estimated to 22,000 and 12,000 won/kg for FSCP and FPH, respectively. Therefore the product from the reuse of dead flatfish was expected to have a considerable competitive price and high added-value functional food material compared with other commercially available fish products.
The sliding door and plug door are the main types of door system in the EMU(Electric Multiple Unit). The sliding door is widely used in Korea but has weak point in the noise problem. In the low operation speed, the noise coming from outer side of the EMU is not an important factor. As the speed is higher than before, noise is increased and make an issue. The main cause of noise is the imperfect air tightness in the EMU. The plug door system has advantages for the noise reduction characteristic in the high speed area. Actually the noise level is an important factor for the passenger comfort. In this paper, we will describe the characteristic of electric plug door and functions of sub component.
As a scaffolding subunit of the PIK3C3/VPS34 complex, Beclin 1 recruits a variety of proteins to class III phosphatidylinositol-3-kinase (VPS34), resulting in the formation of a distinct PIK3C3/VPS34 complex with a specific function. Therefore, the investigation of a number of Beclin 1 domains required for the protein-protein interactions will provide important clues to understand the PIK3C3/VPS34 complex, of which Beclin1-VPS34 interaction is the core unit. In the present study, we have designed a bacterial overexpression system for the Beclin 1 domain corresponding to VPS34 binding (Vps34-BD) and set up the denaturing purification protocol due to the massive aggregation of Vps34-BD in Escherichia coli. The expression and purification conditions determined in this study successfully provided soluble and functional Vps34-BD.

Purpose: In order to improve road-friendliness of heavy vehicles, a fuzzy hybrid control strategy consisting of a hybrid control strategy and a fuzzy logic control module is proposed. The performance of the proposed strategy should be effectively evaluated using a hardware-in-the-loop (HIL) simulation model of a semi-active suspension system based on the fuzzy hybrid control strategy prior to real vehicle implementations. Methods: A hardware-in-the-loop (HIL) simulation system was synthesized by utilizing a self-developed electronic control unit (ECU), a PCI-1711 multi-functional data acquisition board as well as the previously developed quarter-car simulation model. Road-friendliness of a semi-active suspension system controlled by the proposed control strategy was simulated via the HIL system using Dynamic Load Coefficient (DLC) and Dynamic Load Stress Factor (DLSF) criteria. Results: Compared to a passive suspension, a semi-active suspension system based on the fuzzy hybrid control strategy reduced the DLC and DLSF values. Conclusions: The proposed control strategy of semi-active suspension systems can be employed to improve road-friendliness of road vehicles.
Pyrophosphoric lactone modified polyester(PATT) that contains two phosphorous functional groups in one unit base resin structure was synthesized to prepare a non-toxic reactive flame retardant coatings. Then the PATT was cured at room temperature with isocyanate, Desmodur IL, to get a two-component polyurethane flame retardant coatings(PIPUC). Comparing the physical properties of the films of PIPUC with the film of non-flame retardant coatings, there was no degradation observed in physical properties by the introduction of a flame-retarding component into the resin. We found that the char lengths measured by $45^{\circ}$Meckel burner method were $3.1{\sim}4.4cm$ and LOI values recorded $27{\sim}30%$. These results indicate that the coatings prepared in this study is good flame retardant one. The surface structure of coatings investigated with SEM does not show any defects and phase separation.
