짚신나물(Agrimonia pilosa) 추출물의 생리활성 (Physiological activities of Agrimonia pilosa extract)

  • 본 연구에서 짚신나물(A. pilosa)의 뿌리부분에 대한 다양한 생리활성을 조사하여 기능성소재 응용가능성을 검토하였다. 짚신나물 뿌리 추출물은 멜라노마 세포에 대하여 낮은 세포독성을 나타냈다. 세포독성이 거의 없는 농도에서 짚신나물 뿌리 추출물 처리 시 항산화($ID_{50}$, 20.70 mg/L) 및 항균활성이 매우 우수하게 나타났으며 특히 그람양성세균에 대한 항균활성이 높았다. 또한, 짚신나물 뿌리 추출물 처리시 높은 tyrosinase 활성저해($ID_{50}$, 90.18 mg/L) 및 멜라닌 함량 저하를 보여주었다. 이는 짚신나물 추출물이 세포내 tyrosinase 발현을 억제시킴으로서 멜라닌 합성을 저해하는 것으로 판단된다. 이와 같은 결과로 미루어 볼 때 짚신나무 뿌리 추출물은 피부미백 소재 등 피부개선 효과를 비롯한 기능성 화장품에 활용하기 위한 매우 효과적인 재료가 될 수 있다고 판단된다.

혈당상승억제 기능성 소재인 달맞이꽃종자 추출물의 지표물질 분석법의 통계적 검증에 대한 연구 (A Study on the Statistical Method for the Target Component of a Glucose-lowering Functional Material in Extracts of Evening Primerose Seeds)

  • Background: The use of the extracts from evening primrose seeds as a health functional food has been gradually increased. Therefore, the monitoring and screening process has been considerably required for its quality control. Objective: This study aimed to estimate the measurement uncertainty associated with determination of penta-o-galloyl ${\beta}$-D-glucose (PGG) in extracts from evening primrose seeds by high-performance liquid chromatography. Methods: The sources of measurement uncertainty was expressed in accordance with mathematical/statistical theories of GUM & EURACHEM Guide. The expanded uncertainty was calculated by using the relative standard uncertainty between analytical result and sources of uncertainty in measurement (sample weight, final volume, extraction volume, standard solution, matrix and instrument etc). Results: In the results of 95% confidence interval, the uncertainty in measurement was $10,253.34{\pm}1,844.50{\mu}g/kg$ (k = 2.0). Conclusion: In this study, it showed that the value of uncertainty in measurement for determination of PGG in extracts from evening primrose seeds by HPLC has about 18.0% influence on PGG contents of the analytical results. The results would be very useful for the monitoring and screening of evening primrose seeds marketed in Korea for its quality control as dietary supplement.

루꼴라(Eruca sativa) 추출물의 멜라닌 생성 저해 효과 (Anti-melanogenic effect of Eruca sativa extract)

  • 본 연구에서 루꼴라(E. sativa)에 대한 다양한 생리활성을 조사하여 기능성소재 응용가능성을 검토하였다. 루꼴라 추출물은 멜라노마 세포에 대하여 낮은 세포독성을 나타냈다. 세포독성이 거의 없는 농도에서 루꼴라 추출물 처리 시 항산화 활성($ID_{50}$, 17.60 mg/L)이 매우 우수하게 나타났으며 루꼴라 추출물 처리시 높은 tyrosinase 활성저해($ID_{50}$, 132.54 mg/L) 및 멜라닌 함량 저하($ID_{50}$, 158.90 mg/L)를 보여주었다. 또한, 루꼴라 추출물 농도 의존적으로 tyrosinase 발현이 억제되었으며, 이는 루꼴라 추출물이 직접적인 tyrosinase 활성저해 및 세포 내 tyrosinase 발현을 억제시킴으로서 멜라닌 합성을 저해하는 것으로 판단된다. 이와 같은 결과로 미루어 볼 때 루꼴라 추출물은 피부미백 소재 등 피부개선 효과를 지닌 기능성 화장품에 활용하기 위한 매우 효과적인 재료가 될 수 있다고 판단되며 향후 주름개선 효과 등 추가적인 실험을 통하여 화장품 소재 개발 응용 가능성을 타진할 것이다.

돼지감자 분말 첨가 묵의 생리활성성분 분석 및 품질평가 (Physicochemical Analysis and Quality Characteristics of Jerusalem Artichoke and Mook Prepared with Jerusalem Artichoke Powder)

  • This study was performed to investigate the physiological functionality of jerusalem artichokes and to define its antioxidant potential. The study also evaluated mook prepared with jerusalem artichokes powder as a new functional food for ameliorating Diabetes Mellitus. Regarding the chromaticity of mook prepared with jerusalem artichoke, lightness decreased as the amount of jerusalem artichoke increased (p < 0.05), while the redness and yellowness values increased significantly (p < 0.05). Regarding the mechanical properties, hardness, springiness, chewiness, gumminess, and cohesiveness decreased significantly as the amount of jerusalem artichoke increased (p < 0.05). The total polyphenol and flavonoid contents of JA30 were 7.36 mg TAE/g and 2.15 mg RE/g, respectively, which were higher than those of the control group(3.50 mg TAE/g and 0.76 mg RE/g, respectively). The DPPH and ABTS scavenging abilities ($IC_{50}$) for JA30 were 4.40 and 5.70 mg/mL, respectively, indicating a higher radical scavenging ability than the control group (7.59 and 7.24 mg/mL, respectively). The ${\alpha}-glucosidase$ inhibitory effect ($IC_{50}$) of JA30 and the control group was 18.91 and 24.01 mg/mL, respectively. Based on the above-described results, mook containing jerusalem artichoke powder is expected to ameliorate diabetes with antioxidant activity, ${\alpha}-glucosidase$ inhibitory effect, and sensory factor. Also, it is believed that this functional material has the potential to improve health.

Antioxidant and QR Inductive Activities of Novel Functional Soybean 'Agakong3'

  • In order to evaluate the bioactivity of 'Agakong3', which was newly bred, quinone reductase (QR) inductive activity and antioxidant activity were both assessed. The methanol extract of 'Agakong3' showed a significantly stronger QR inductive activity than other soybeans. The methanol extract of 'Agakong3' also showed a significantly stronger cytotoxicity on hepa1c1c7 than other soybeans. 'Agakong3' exhibited the most potent antioxidant activity in the Trolox equivalent antioxidant capacity (TEAC) assay whereas it showed significantly weak antioxidant in the DPPH assay. In total phenol and flavonoid contents, 'Agakong3' showed the highest contents regarding phenol and flavonoid compounds. Major isoflavones such as daidzein and genistein were quantitified by high performance liquid chromatography. 'Agakong3' also showed the highest total isoflavone contents. Results of correlation analysis showed that there were high correlation coefficients between the contents of isoflavone and TEAC and the contents of isoflavone and QR inductive activity, respectively. These results suggest that 'Agakong3' will be a promising and functional food material.

홍화(Carthamus tinctorius L.) 순의 이화학적 특성 (Properties of the Chemical Composition of Safflower (Carthamus tinctorius L.) sprout)

  • In order to develop new materials for the functional food, the components of safflower(Carthamus tinctorius L.) sprout was studied. Chemical composition, minerals, amino acids, and fatty acids of the leaf and the stem parts of 2 weeks grown safflower sprout were analyzed. The chemical composition of safflower spout was 12.7% (w/w) moisture, 28.5% crude protein, 10.1% crude fat, and 5.3% crude ash. Mineral contents of the leaf part were P 49%, K 22%, Mg 15%, Ca 10% whereas these of the stem part were K 51%, P 27%, S 10%, Ca 7%, Mg 4%. Other mineral contents were less than 2% in both parts. Especially, Pt was 0.18 ppb and 0.17 ppb in the leaf and the stem parts, respectively. The composition of the amino acid were approximately as follow, the major amino acid in the leaf and the stem were aspartic acid and glutamic acid, the contents of these were 10.7mg/g, 10mg/g in the leaf, 11.3 mg/g, 8.4 mg/g in the stem, respectively. The major fatty acids in the leaf and the stem parts were linoleic acid and $\alpha$-linolenic acid. The linoleic acid (C18:2) in the leaf and the stem parts were 67% (w/w) and 47% whereas the $\alpha$-linolenic acid. The linoleic acid (C18:2) in those parts were 14% (w/w) and 11%, respectively. On the basis of chemical analysis, the safflower sprout showed to have relatively high contents of crude protein and crude fat, minerals including small amount of Pt, polyunsaturated fatty acid as linoleic acid and $\alpha$ -linolenic acid. These results suggested that safflower sprout was found to be a useful material of natural health food for the functional food development.

비쭈기 나무(Cleyera japonica) 추출물의 생리활성에 대한 연구 (Study on the Physiological Activities of Cleyera japonica Extract)

  • 본 연구에서 비쭈기 나무의 잎 부분에 대한 다양한 생리활성을 조사하여 기능성소재 응용가능성을 검토하였다. 비쭈기 나무 잎 추출물은 멜라노마 세포에 대하여 낮은 세포독성을 나타냈다. 비쭈기 나무 추출물 처리 시 항산화(IC50, 22.90 ㎎/L) 및 항균활성이 매우 우수하게 나타났으며 특히 그람양성세균에 대한 항균활성이 높았다. 또한, 비쭈기 나무 추출물 처리시 tyrosinase 활성저해 및 멜라닌 함량 저하(IC50, 101.90 ㎎/L)를 보여주었다. 이와 같은 결과로 미루어 볼 때 비쭈기 나무 추출물은 피부미백 소재 등 피부개선 효과를 비롯한 기능성 화장품에 활용하기 위한 매우 효과적인 재료가 될 수 있다고 판단된다.

나노리포좀을 이용한 astaxanthin의 안정화 (Stabilization of Astaxanthin Using Nanoliposome)

  • 본 연구에서는 이중결합을 가진 불포화화합물로 제조나 저장 시 열과 산화 (빛)에 의해 쉽게 파괴되어 활성이 감소하여 응용범위에 한계가 있는 아스타잔틴의 안정성 향상을 위하여 나노리포좀 제형기술을 이용하였다. 제형안정성 평가를 통하여 리포좀 제조조건 및 조성비를 확립하였고, 포접나노리포좀을 제조하여 포접전의 아스타잔틴과 안정성을 비교 검토하였다. 아스타잔틴을 포접하여 포접나노리포좀 제조 시포접전의 아스타잔틴에 비해 열에 대한 안정성이 2배 정도 향상되고, UV 안정성 또한 3배 향상된 것을 확인하였다. 이상의 결과로부터 아스타잔틴을 포접한 포접나노리포좀은 안정성이 우수한 기능성 원료로서 산업적으로 용도를 넓히고 부가가치를 더욱 향상시킬 수 있을 것으로 기대된다.

옻나무 (Rhus verniciflua Stokes) 추출물의 알코올대사 효소활성에 미치는 여향 (Effect of Rhus verniciflua Stokes Extract on the Alcohol-Metabolizing Enzyme Activities)

  • Alcohol oxidation activities and optimization of extraction conditions of Rrhus verniciflua Stokes (RVS) extract were evaluated for the development of a functional biomaterial for improving liver function. When alcohol oxidation activities of RVS was analyzed, the Rrhus verniciflua Stokes bark (RVSB) were higher than the Rrhus verniciflua Stokes heartwood (RVSH). Alcohol oxidation activity value of RVSB increased in a concentration-dependent manner. In the comparative analysis between Hovenia dulcis Thunb (HOT) and Alnus japonica Steud (AJS) which was reported as a alcohol oxidation material, alcohol oxidation activity is much higher than the others. The experimental conditions were optimized for alcohol oxidation-active components production from RVSB. The extraction conditions such as temperature, time, pH and particle size were performed. It was recommended to extract the alcohol oxidation-active components from RVSB by hot water (pH 7.0) at $85^{\circ}C$ for 8 hours.

Enzymatic Synthesis of Puerarin Glucosides Using Leuconostoc Dextransucrase

  • Puerarin (P), an isoflavone derived from kudzu roots, has strong biological activities, but its bioavailability is often limited by its low water solubility. To increase its solubility, P was glucosylated by three dextransucrases from Leuconostoc or Streptococcus species. Leuconostoc lactis EG001 dextransucrase exhibited the highest productivity of puerarin glucosides (P-Gs) among the three tested enzymes, and it primarily produced two P-Gs with a 53% yield. Their structures were identified as ${\alpha}$-$_D$-glucosyl-($1{\rightarrow}6$)-P (P-G) by using LC-MS or $^1H$- or $^{13}C$-NMR spectroscopies and ${\alpha}$-$_D$-isomaltosyl-($1{\rightarrow}6$)-P (P-IG2) by using specific enzymatic hydrolysis, and their solubilities were 15- and 202-fold higher than that of P, respectively. P-G and P-IG2 are easily applicable in the food and pharmaceutical industries as alternative functional materials.
