Skin Moisturizing Activity of Unripe Apple(Immature Fruit of Malus pumila) in Mice (풋사과 추출물의 피부 보습 효과)

  • Objectives : Skin aging is generally characterized by wrinkles, sagging, loss of elasticity roughness, pigmentation and dryness. This changes is caused by reducing the elements constituting the extracellular matrix contributing to the physiological properties of the skin, such as collagen fiber, elastic fiber, and hyaluronic acid. Adequate skin hydration is important to maintain normal skin function and reduce skin aging. The present study is objective to observe skin moisturizing effects of Unripe apple(UA, Immature fruit of Malus pumila Mill) in vivo and its underlying molecular mechanisms. Methods : ICR mice were orally administerd UA(100, 200 and 400mg/kg/day) for 8 weeks, and skin water contents and the expression of transforming growth factor (TGF)-𝛽1, ceramide, hyaluronan and collagen type I(COL1) were measured in dorsal back skin of the mice. Gene expression of hyaluronan synthase(HAS1, HAS2, HAS3), collagen synthase(COL1A1, COL1A2) and TGF-𝛽1 were also determined by realtime RT-PCR. Results : Skin water contents and the expression of TGF-𝛽1, ceramide, COL1 and hyaluronan were significantly increased in UA group(100, 200 and 400mg/kg/day) compared to vehicle control. The mRNA expression of HAS isoform(HAS1, HAS2, HAS3), COL1A1, COL1A2, and TGF-𝛽1 were also significantly increased by UA. Conclusions : UA has skin moisturizing effects and enhancement activities in skin function related components(COL1, hyaluronan, ceramide and TGF-𝛽1). These results suggested that UA can be a developing candidate for developing alternative skin protective agent or functional food ingredient.

Study on Skin Permeation of Tocopheryl Acetate Using Swollen Micelle (Swollen Micelle을 이용한 Tocopheryl Acetate 의 피부흡수 연구)

  • A study was performed to increase skin permeation of tocopheryl acetate, an oil-soluble component among tocopheryl acetate, salicylic acid, and niacinamide, which are the active ingredients of the anti-hair loss toner. As a method of transparently solubilizing tocopheryl acetate while containing salicyl acid and niacinamide, we used a swollen micelle structure that increased the size of the micelle formed by the aggregation of surfactants. The prepared swollen micelle solution contains three kinds of active ingredients, and poloxamer 407 and octyldodeceth-16 were used as surfactants to increase transparent properties and stability. In addition, isostearic acid was used as a co-surfactant to increase the size of micelles. To evaluate the physical properties of the prepared swollen micelles, turbidity at room temperature (25 ℃) was measured. The Franz diffusion cell method was used to evaluate the skin permeation rate of tocopheryl acetate among the hair-loss prevention components contained in swollen micelles. After 24 h, tocopheryl acetate showed a 6-fold improvement in skin permeation compared to the control group. Therefore, it can be seen that the swollen micelles developed in this study can be applied to hair-loss prevention products or solubilized formulations of various functional cosmetics.

Assessing Carotenoid Levels and Antioxidant Properties in Korean Sweet Corn Inbred Lines to Develop High-Quality Sweet Corn Varieties through Breeding (기능성 단옥수수 품종 육성을 위한 자식계통의 카로티노이드 함량 및 항산화 활성 평가)

  • Sweet corn is widely consumed due to its high nutritional content and diverse phytochemical composition, including carotenoids and phenolic compounds, which have several benefits for human health. This study aims to identify breeding materials for developing high-functional sweet corn varieties by evaluating the phytochemical and antioxidant activities of 37 Korean sweet corn inbred lines. The results revealed genetic variation in various components, such as carotenoid content (range of 120.7~1239.3 mg 100 g-1), polyphenol content (490.5~740.6 mg gallic acid equivalent 100 g-1), and flavonoid content (7.3~68.6 mg catechin equivalent 100 g-1). In addition, the free radical scavenging capacity, measured using 1,1-diphenyl-2-picrylhydrazyl and 2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid), also varied among the inbred lines. Therefore, in this study, we identified Korean sweet corn inbred lines with high phytochemical content and excellent antioxidant activity. The development of sweet corn varieties with improved functionality is expected to further expand the role of sweet corn as a source of antioxidants in the Korean diet.

A Study on the Architecture for Avionics System of Jet Fighters (제트 전투기의 항공전자 시스템 아키텍처에 관한 연구)

  • The development trend of jet fighter's avionics system architecture is the digitization of subsystem component functions, increased RF sensor sharing, fiber optic channel networks, and modularized integrated structures. The avionics system architecture of the fifth generation jet fighters (F-22, F-35) has evolved into an integrated modular avionics system based on computing function integration and RF integrated sensor systems. The integrated modular avionics system of jet fighters should provide improved combat power, fault tolerance, and ease of jet fighter control. To this aim, this paper presents the direction and requirements of the next-generation jet fighter's avionics system architecture through analysis of the fifth generation jet fighter's avionics system architecture. The core challenge of the integrated modularized avionic system architecture requirements for next-generation fighters is to build a platform that integrates major components and sensors into aircraft. In other words, the architecture of the next-generation fighters is standardization of systems, sensor integration of each subsystem through open interfaces, integration of functional elements, network integration, and integration of pilots and fighters to improve their ability to respond and control.

The Empirical Research on Relationship of Consumption Value, Satisfaction, Trust, Loyalty of Green Product in Elderly Consumer (실버 소비자의 친환경 제품에 대한 소비 가치가 만족도, 신뢰, 충성도에 미치는 영향 - 하이브리드 카를 중심으로 -)

  • The purpose of this study is to analyse how consumption value affects the loyalty of green product through satisfaction and trust among elderly consumers. Data were collected from a cross-sectional survey of 314 older adults (age≥60) in the U. S., who bought and possessed a hybrid car, a representative green product. The statistical methodology is employed a structural equation model. The results demonstrated several important findings. First, perceived social value among elderly population had significant effects on green product satisfaction, while hedonic value did not. Second, both perceived functional value and environment friendly value had a significant positive effect on trust in green products. Third, satisfaction with green products also led to trust in green products. Finally, trust in green products showed their significant effects on loyalty in green product. These results provide practical implications to improve the trust and loyalty in green products among the elderly consumers. Furthermore, by deriving major components of consumption values in green products among the elderly, and analyzing the mechanism of satisfaction, trust, and loyalty, the study emphasizes relationship marketing in implementing "green" marketing strategies.

A Study on the Prediction Model for Bioactive Components of Cnidium officinale Makino according to Climate Change using Machine Learning (머신러닝을 이용한 기후변화에 따른 천궁 생리 활성 성분 예측 모델 연구)

  • Climate change has emerged as a global problem, with frequent temperature increases, droughts, and floods, and it is predicted that it will have a great impact on the characteristics and productivity of crops. Cnidium officinale is used not only as traditionally used herbal medicines, but also as various industrial raw materials such as health functional foods, natural medicines, and living materials, but productivity is decreasing due to threats such as continuous crop damage and climate change. Therefore, this paper proposes a model that can predict the physiologically active ingredient index according to the climate change scenario of Cnidium officinale, a representative medicinal crop vulnerable to climate change. In this paper, data was first augmented using the CTGAN algorithm to solve the problem of data imbalance in the collection of environment information, physiological reactions, and physiological active ingredient information. Column Shape and Column Pair Trends were used to measure augmented data quality, and overall quality of 88% was achieved on average. In addition, five models RF, SVR, XGBoost, AdaBoost, and LightBGM were used to predict phenol and flavonoid content by dividing them into ground and underground using augmented data. As a result of model evaluation, the XGBoost model showed the best performance in predicting the physiological active ingredients of the sacrum, and it was confirmed to be about twice as accurate as the SVR model.

Study on the optimization of additive manufacturing process parameters to fabricate high density STS316L alloy and its tensile properties (고밀도 STS316L 합금 적층 성형체의 제조공정 최적화 및 인장 특성 연구)

  • To optimize the process parameters of laser powder bed fusion process to fabricate the high density STS316L alloy, the effect of laser power, scanning speed and hatching distance on the relative density was studied. Tensile properties of additively manufactured STS316L alloy using optimized parameters was also evaluated according to the build direction. As a result of additive manufacturing process under the energy density of 55.6 J/mm3, 83.3 J/mm3 and 111.1 J/mm3, high density STS316L specimens was suitably fabricated when the energy density, power and scan speed were 83.3 J/mm3, 225 W and 1000 mm/s, respectively. The yield strength, ultimate tensile strength, and elongation of STS316L specimens in direction perpendicular to the build direction, show the most competitive values. Anisotropic shape of the pores and the lack of fusion defects probably caused strain localization which result in deterioration of tensile properties.

Metabolic Regulation of Longevity and Immune Response in Caenorhabditis elegans by Ingestion of Lacticaseibacillus rhamnosus IDCC 3201 Using Multi-Omics Analysis

  • Probiotics, specifically Lacticaseibacillus rhamnosus, have garnered attention for their potential health benefits. This study focuses on evaluating the probiotic properties of candidate probiotics L. rhamnosus IDCC 3201 (3201) using the Caenorhabditis elegans surrogate animal model, a well-established in vivo system for studying host-bacteria interactions. The adhesive ability to the host's gastrointestinal tract is a crucial criterion for selecting potential probiotic bacteria. Our findings demonstrated that 3201 exhibits significantly higher adhesive capabilities compared with Escherichia coli OP50 (OP50), a standard laboratory food source for C. elegans and is comparable with the widely recognized probiotic L. rhamnosus GG (LGG). In lifespan assay, 3201 significantly increased the longevity of C. elegans compared with OP50. In addition, preconditioning with 3201 enhanced C. elegans immune response against four different foodborne pathogenic bacteria. To uncover the molecular basis of these effects, transcriptome analysis elucidated that 3201 modulates specific gene expression related to the innate immune response in C. elegans. C-type lectin-related genes and lysozyme-related genes, crucial components of the immune system, showed significant upregulation after feeding 3201 compared with OP50. These results suggested that preconditioning with 3201 may enhance the immune response against pathogens. Metabolome analysis revealed increased levels of fumaric acid and succinic acid, metabolites of the citric acid cycle, in C. elegans fed with 3201 compared with OP50. Furthermore, there was an increase in the levels of lactic acid, a well-known antimicrobial compound. This rise in lactic acid levels may have contributed to the robust defense mechanisms against pathogens. In conclusion, this study demonstrated the probiotic properties of the candidate probiotic L. rhamnosus IDCC 3201 by using multi-omics analysis.

A Genotoxicity Study of Transgenic Tomatoes using CRISPR/Cas9 (CRISPR/Cas9에 의한 유전자교정 토마토의 유전독성평가)

  • Tomato is a widely distributed, cultivated, and commercialized vegetable crop. Recently, an increasing trend has been observed in the consumption of transgenic crops with enhanced functional components. However, consumer concerns regarding genotoxicity have been increasing. This study examined the genotoxicity of transgenic tomato (LTT) using the CRISPR/Cas9 system through a bacterial reverse mutation assay, chromosomal aberration assay, and mammalian micronucleus test. In the bacterial reverse mutation assay, LTT did not induce mutagenicity in Salmonella typhimurium strains TA98, TA100, TA1535, and TA1537, or Escherichia coli WP2uvrA, irrespective of the presence or absence of S9. LTT did not cause clastogenic or aneugenic chromosomal abnormalities during metaphase in CHL cells. Moreover, LTT did not increase the frequency of micronucleated polychromatic erythrocytes in the polychromatic erythrocytes. These findings can be used as a foundation to assess the genotoxicity of transgenic crops using the CRISPR/Cas9 system in the future.

Comparison of whitening effect of Rubus coreanus fruit according to maturity (성숙도에 따른 복분자 열매의 미백 활성 비교)

  • Purpose: The Rubus coreanus fruit (RF) is an important traditional medicinal herb having antioxidant, anti-inflammatory, and immunoregulatory properties. These activities are known to change dramatically, depending on maturity of the RF. It is presumed that change of functional components, such as flavonoids, tannins, phenolic acids, triterpenoids and organic acids in RF, affect the various bioactivities. This study aimed to confirm changes in the anti-melanogenic effects of RF based on maturity, and to identify the bioactive compounds responsible. Methods: The cell viability of mature RF (MRF) and immature RF (IRF) extracts was investigated using B16F10 cells. To compare the anti-melanogenic effect of MRF and IRF extracts, we first assessed the melanin content. High-performance liquid chromatography analysis was performed to evaluate changes in the level of ellagic acid according to maturity of the RF. In addition, tyrosinase inhibitory activity of both extracts was examined. Results: MRF and IRF extracts (50-200 ㎍/mL) do not affect the cell viability of B16F10 melanoma cells. IRF extract more effectively inhibited melanin synthesis than MRF extract. The content of ellagic acid in IRF extract was higher than that obtained in MRF extract. Furthermore, greater inhibition of tyrosinase activity was observed after exposure to IRF extract than MRF extract. A positive correlation was determined between ellagic acid content and tyrosinase inhibitory activity, and a negative correlation was obtained between ellagic acid content and melanin content. Taken together, our results indicate that ellagic acid is one of the major bioactive compounds of RF that imparts a whitening effect. Conclusion: Our results indicate that ellagic acid in MRF and IRF extracts affect the anti-melanogenesis effect through inhibition of tyrosinase activity. Therefore, the ellagic acid rich IRF has greater potential for application as a natural and functional cosmetic material.
