양식넙치 멜라닌 농축 호르몬의 특성 (Characterization of Melanin-concentrating Hormone from Olive Flounder (Paralichthys olivaceus))

  • 멜라닌 농축 호르몬(melanin-concentrating hormone, MCH)은 17개의 아미노산으로 구성된 환형의 시상하부 펩티드로 색소 침착의 조절인자로서 연어에서 처음 분리되었다. 포유동물의 MCH는 19개의 아미노산으로 구성되어 있으며 섭식 및 에너지 항상성을 조절하는데 관여한다. 본 연구에서는 양식넙치의 다양한 조직에서 MCH 유전자의 발현 분포, 멜라닌 함유 세포의 집적, 포유동물 MCH 수용체와 양식넙치 MCH의 상호작용을 조사하였다. Real-time qPCR을 이용하여 뇌, 정소, 난소에서 MCH 유전자의 발현이 나타나는 것을 확인하였고, 수정 후 발달 단계에서도 MCH 유전자의 발현을 확인할 수 있었다. 합성된 연어 sMCH, 포유류 hMCH, 양식넙치 fMCH, dN-fMCH, dC-fMCH를 양식 넙치의 표피에 처리했을 때 다양한 농도에 따라 멜라닌 함유 세포의 집적이 다양하게 나타났다. 연어 sMCH, 포유류 hMCH에 비해 양식넙치 fMCH의 멜라닌 함유세포의 집적도가 36~99.85%로 비역가를 나타났으나 양식넙치 dN-fMCH, dC-fMCH를 처리한 경우 양식넙치 fMCH에 비해 높은 농도에서 집적이 나타나고 짧은 시간에 분산되었다. 또한, 인간 MCH 수용체와 쥐 MCH 수용체가 발현된 포유동물의 세포주에 양식넙치 fMCH를 처리하여 각 수용체와 결합하는 것을 확인하였다. 이러한 결과는 어류에서 발현되는 MCH가 포유동물의 MCH와 유사한 구조를 가지고 있어 MCH 수용체에 대한 새로운 리간드로서 제공될 수 있으며, 향후 어류의 MCH 수용체에 확대 적용할 수 있을 것이다.

삼백초 추출물의 자궁경부암세포 억제 효능 (The Efficacy of Saururus chinensis on Cervical Cancer Cells : The Inhibitory Effect on the Function of E6 and E7 Oncogenes of HPV Type 16)

  • Cervical cancer is one of the leading causes of female death from cancer worldwide with about 500,000 deaths per year. A strong association between certain human papilloma viruses (HPV type 16 and 18) and cervical cancer has been well known. An extract of Saururus chinensis, named as PE-46, has been used to investigate whether this agent has the ability of inhibiting the oncogenes E6 and E7 of HPV type 16. PE-46 inhibited the proliferation of human cervical cancer cell lines in a dose response manner. PE-46 also inhibited the in vitro binding of E6 and E6AP which are essential for the binding and degradation of the tumor suppressor p53. In addition, PE-46 inhibited the in vitro binding of E7 and Rb which is essential tumor suppressor for the control of cell cycle. The levels of mRNA for E6 and E7 were also decreased by PE-46. SiHa cells treated with PE-46 induced G0/G1 arrest, resulting in inhibition of growth. Our study showed that the PE-46 can inhibit the cervical carcinomas via both inhibition of bindings between oncogenes and tumor suppressors, and inhibition of G1longrightarrowS transition. PE-46 inhibited the oncogenecity of E6 and E7 of HPV 16 type, thus could be used as a putative modulating agent for the treatment of cervical carcinomas caused by HPV.

우리나라 지방자치단체의 외국인직접투자 증감요인 분해 분석 (Decomposition Analysis of Regional Governments in Foreign Direct Investment Increase or Decrease)

  • 본 연구에서는 우리나라 각 지방자치단체 등지에서 추진하고 있는 외국인직접투자 유치 정책에 기초 정보를 제공한다. 변이-비중 모형(Shift-Share Model)을 이용해 외국인직접투자 유치 실적의 증감 요인은 지역별 각 산업의 분해 요인에 따라 구조적 특징을 파악함으로써 찾을 수 있다. 분석은 2009년을 기준으로 2010년, 2011년 전국(수도권과 비수도권으로 구분)의 외국인직접투자 실적을 비교, 전산업(한국표준산업분류 중분류)에 대해 실시했다. 그 결과는 국가성장효과(NS), 산업구조효과(IM), 지역할당효과(RS)로 구분하여 외국인직접투자 증감요인을 살펴보면 2010년과 2011년 수도권, 비수도권 모두에서 국가성장효과(NS)는 음(-)의 값을 갖고 있었다. 이는 세계경기침체의 여파가 국내 경제에 미치는 영향이 큰 것으로 나타났기 때문인 것으로 분석된다. 산업구조효과(IM)의 경우 수도권은 교육, 문화, 비즈니스 및 교통 등의 발달로 비수도권에 비해 서비스 산업의 발달에 유효한 환경을 가지고 있다. 특히 서비스(음식숙박, 비즈니스서비스업, 문화오락) 부문에서 그 특징을 찾을 수 있었다. 비수도권은 제조업(비금속광물, 전기전자) 등의 부문에서 산업발달의 전후방 연계효과 및 지역적 특징이 큰 역할을 한 것으로 분석된다. 그러나 지역할당효과(RS)에서는 수도권에서 제조업 분야의 금속 및 화공분야에서 경쟁력이 있는 것으로 도출되었다.

한국인에서 씹기 불편감과 우울증의 연관성: 2016 국민건강영양조사를 이용한 단면 연구 (Association of depression with chewing problems in Koreans : A cross-sectional study using the Korea National Health and Nutrition Examination Survey 2016)

  • 목적: 본 연구는 일반 인구의 대표 표본에서 우울증의 발현과 심각도, 씹기 불편감 사이의 관계를 분석하였다. 연구 재료 및 방법: 전국 대표 표본(n = 8150)을 대상으로 하였으며, 씹기 불편감은 해당 문제에 관한 설문 조사에 응답한 사람들로 정의하였다. 우울증은 PHQ-9 설문 조사에서 총 점수가 10점 이상인 대상자로 정의하였다. 인구 통계, 사회적 경제적 특성 및 동반 질환에 관한 데이터를 포함하여 다중 로지스틱 회귀 분석을 수행하고 분석하였다. 결과: 한국인의 경우 씹기 불편감이 없는 사람(10.2%)보다 씹기 불편감이 있는 사람(17.2%)에서 우울증의 유병률이 유의하게 더 높았다. 다변량 로지스틱 회귀 분석에서 우울증의 존재는 CP와 유의하게 관련 있었다(adjusted odd ratio [aOR]: 1.90, P < 0.001). 씹기 불편감의 위험은 우울증의 심각도가 증가함에 따라 증가하였다: 중증 우울증(OR: 2.62, P < 0.001), 중등도 우울증(OR: 2.19, P < 0.001). 결론: 중증 우울증을 보이는 개인에서 우울증의 공존여부가 씹기 불편감과 상당히 연관되었다. 씹기 불편감 환자 치료에 있어 우울증의 선별검사가 고려되어야 한다.

야생 검은댕기해오라기에서 대퇴관절 탈골의 수술적 교정 (Surgical Repair of Coxofemoral Joint Luxation in a Wild Black-Crowned Night Heron (Nycticorax nycticorax))

  • 왼쪽 뒷다리의 파행을 보이는 야생 검은댕기해오라기가 내원하였다. 신체검사에서 왼쪽 뒷다리가 외측으로 $90^{\circ}$ 돌아가 있었으며 대퇴관절부위의 부종이 관찰되었다. 혈액검사와 더불어 영상학적 검사가 진행되었다. 골반 방사선 사진상 왼쪽 대퇴관절의 앞쪽 등쪽 방향 탈골이 확인되었다. 비흡수성 봉합사를 이용하여 탈골을 고정한 후 관절낭 성형술을 이용한 수술적 방법으로 교정하였다. 수술 10일 후 거의 정상에 가까운 보행을 보였으며, 수술 20일 후 자연으로 복귀되었다. 본 보고는 야생조류에서 대퇴골 탈골의 영상 진단 과정과 수술적 교정방법에 대해 소개하였다.

The Near-Infrared Imaging Spectroscopy to Visualize the Distribution of Sugar Content in the Flesh of a Melon

  • To improve the accuracy of sweetness sensor in automated sorting operations, it is necessary to clarify unevenness of the sugar content distribution within fruits. And it is expected that the technique to evaluate the content distribution in fruits contribute to the development of the near-infrared (NIR) imaging spectroscopy. Sugiyama (1999) had succeeded to visualize the distribution of the sugar content on the surface of a half-cut green fresh melon. However, this method cannot be applied to red flesh melons because it depends on information of the absorption band of chlorophyll (676 nm), which is affected by the color of the fresh. The objective of this study was to develop the universal visualization method depends on the absorption band of sugar, which can be applied to various kinds of melons and other fruits. The relationship between the sugar contents and absorption spectra of both green and red fresh melons were investigated by using a NIR spectrometer to determine the absorption band of sugar. The combination of 2$\^$nd/ derivative absorbances at 902 nm and 874 nm was highly correlated with the sugar contents. The wavelength of 902 nm is attributed to the absorption band of sugar. A cooled charge-coupled device (CCD) imaging camera which has 16 bit (65536 steps) A/D resolution was equipped with rotating band-pass filter wheel and used to capture the spectral absorption images of the flesh of a vertically half-cut red fresh melon. The advantage of the high A/D resolution in this research is that each pixel of the CCD is expected to function as a detector of the NIR spectrometer for quantitative analysis. Images at 846 nm, 874 nm, 902 nm and 930 nm were acquired using this CCD camera. Then the 2$\^$nd/ derivative absorbances at 902 nm and 874 nm at each pixel were calculated using these four images. On the other hand, parts of the same melon were extracted for capturing the images and squeezed for the measurement of sugar content. Then the calibration curve between the combination of 2$\^$nd/ derivative absorbances at 902 nm and 874 nm and sugar content was developed. The calibration method based on NIR spectroscopy techniques was applied to each pixel of the images to convert the 2$\^$nd/ derivative absorbances into the Brix sugar content. Mapping the sugar content value of each pixel with linear color scale, the distribution of the sugar content was visualized. As a result of the visualization, it was quantitatively confirmed that the Brix sugar contents are low at the near of the skin and become higher towards the seeds. This result suggests that the visualization technique by the NIR imaging spectroscopy could become a new useful method fer quality evaluation of melons.

  • [ $TiO_{2},\;Pd,\;Pt$ ]와 In을 사용하여 trimethylamine gas에 대한 감도를 향상시키는 $SnO_{2}$ 가스센서에 대하여 연구하였다. 금속-$SnO_{2}$ 후막은 백금전극이 내장된 알루미나 지지체에 스크린 법으로 제작하였다. 센서의 특성은 검출가스 농도의 함수로서 반응기내 각 센서의 전기적 저항을 측정하여 조사하였으며, $100{\sim}1000ppm$범위의 trimethylamine, dimethylamine과 암모니아가스에 대하여 측정하였다. 그 결과, 금속-$SnO_{2}$의 가스 검출특성은 금속의 종류와 양에 따라 결정되었으며, $250^{\circ}C$에서 trimethylamine에 대한 센서의 감도와 선택성은 1 wt%, Pd와 10 wt%, $TiO_{2}$를 첨가하였을 때 감도가 가장 우수한 것으로 나타났다.

농가소득 창출을 위한 식용식물의 항산화, 항염, 항혈전 및 항비만 효과 탐색 (Evaluation of Antioxidant, Anti-Inflammatory, Antithrombotic, and Antiobesity Activities in Cultured Edible Plants to Increase Farm Income)

  • We studied the antioxidant, the anti-inflammatory, antithrombotic, and antiobesity activities of seven different kinds of edible plants. 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ABTS radical scavenging activities were determined as a measurement of antioxidant activity. NO production inhibition by the macrophage cell line (Raw 264.7) treated with lipopolysaccharide (LPS) was carried out to assess anti-inflammatory activity. Thrombin inhibitory activity was measured for its antithrombotic function and inhibition of 3T3-L1 cell differentiation was evaluated as a measurement of antiobesity activity. Total phenolic components and total flavonoid contents were measured to determine functional materials in medicinal plants. Common sage, Japanese lady bell, and hairy agrimony showed high antioxidant activity ($IC_{50}$) of less than $100{\mu}g/mL$. All samples used in this study showed anti-inflammatory activity. Common sage, hairy agrimony, and hooker chives showed antithrombotic effects. Hairy agrimony showed the highest antithrombotic effect (98.1%). Common sage, Japanese lady belly, hairy agrimony, and hooker chives showed reduced 3T3-L1 cell differentiation and hooker chives strongly inhibited lipid accumulation in the cells compared to other medicinal plants. Common sage and hairy agrimony contained more than 1 mg GAE/g of phenolic compounds and more than 1 mg CE/g of flavonoids. Functional activities were different by plant part and extraction method from each sample. These results suggest that common sage, Japanese lady belly, hairy agrimony, and hooker chives may be used as healthy food sources with antioxidant, anti-inflammatory, antithrombotic, and antiobesity activities, and appropriate extracting methods from each plant need to be developed.

Extremely Low Frequency Magnetic Field is an Environmental Stress Factor by Exerting Oxidative Stress

  • The previous study reported the biological effect of magnetic field exerted by acting on endocrine and anti-oxidant system. The present study aims to study whether ELF-MF (extremely low frequency magnetic field) affects the physiological endocrine systems such as thyroid and whether ELF-MF affects the defense system against oxidative stress when it alters the function of thyroid. Finally, we correlate the effects of MF on oxidative stress, and adrenal and thyroid with an environmental stress factor. We exposed sham or MF to rats for 5 or 25 days. After the exposure, we determined pain sensitivity, level of TSH, $T_3$ and free $T_4$ in plasma. We also assayed in whole brain, lipid peroxidation, the activity of enzymatic anti-oxidant defense including superoxide dismutase(SOD) and glutathione peroxidase (GPx), and non enzymatic defense such as reduced or oxidized glutathione contents. MF induced the hypersensitivity to thermal stimuli with the reduction of latency. $T_3$ and $T_4$ levels were also increased by the exposure of MF. In addition, we observed the rise of MDA level in rat brain by MF although the MF did not change superoxide dismutase and glutathione peroxidase activity. The effect of MF on both reduced and oxidized glutathione results in decrease in reduced or oxidized glutathione in whole brain. In every experiment, there was no significant difference in MF influence between short term (5 days) and long term (25 days) exposure. Taken together, MF exposure affects the thyroid hormonal control in brain. The elevated thyroid hormone acts on brain, leading to hyper-utilization of oxygen. This phenomenon may be correlated with oxidative stress resulting from MF exposure. In conclusion, we suggest that MF exposure may be an environmental stress by exerting oxidative stress.

미취학 아동의 치아우식증과 식습관 및 간식섭취와의 관계 (A study on the correlation between the dental caries and dietary habits and snack intake of pre-school children)

  • Objectives : The purpose of this study was to correlation between the index of dental caries in primary teeth and the habits of diet and snack which are expected to influence on the index. The subjects of this study were 165 parents of the children who visited a pediatric dental hospital located in Gyeong-gi do from Jun. 15th to Sep. 26th, 2009, and the self-report questionnaire survey was performed. Methods : A questionnaire used in a previous study was revised and completed for this study and it consisted of 15 items on general characteristic, nine items on dietary habit, and five items on snack intake habit. The collected data was analyzed using SPSS program and findings are as follows Results : 1. The index of dental caries in primary teeth by ages was that children of four years old has 4~5 with 27.5%, children of five years old has 2~3 and 4~5 with 33.3% and children of six years old has 1 and 8~9 with 25.6%, showing a statistically significant(${\chi}^2$=37.72, p<.01). 2. The frequency of brushing teeth was found to have statistically significant negative correlation with the number of primary teeth with dental caries (${\chi}^2$=61.20, p<.001). 3. The frequency of taking snack was found to have statistically significant positive correlation with the number of primary teeth with dental caries (${\chi}^2$=51.42, p<.001). 4. The desirability of dietary habit was found to have statistically significant negative correlation with the number of primary teeth with dental caries (${\chi}^2$=36.51, p<.001). 5. The frequency of taking biscuit was found to have statistically significant positive correlation with the number of primary teeth with dental caries (${\chi}^2$=67.74, p<.001). 6. The frequency of taking bread was found to have statistically significant positive correlation with the number of primary teeth with dental caries (${\chi}^2$=29.63, p<.01). 7. The frequency of taking caramel and candy was found to have statistically significant positive correlation with the number of primary teeth with dental caries (${\chi}^2$=38.85, p<.001). 8. The frequency of taking soft drinks was found to have statistically significant positive correlation with the number of primary teeth with dental caries (${\chi}^2$=52.92, p<.001). 9. The frequency of taking ice-creams was found to have statistically significant positive correlation with the number of primary teeth with dental caries (${\chi}^2$=75.07, p<.001). Conclusions : Those findings show that the children with undesirable dietary habit and higher frequency of taking snack have higher index of dental caries in primary teeth, therefore, it is considered that the regular brushing teeth and establishment of desirable dietary habit are very important for keeping health oral cavity. It is recommended that the continuous concern and repeated learning are needed in house and group of rearing infants so that the children take foods of fruits which have function of self-purification than the food of carbohydrates which has high adhesion and level of sweetness.
