MoJMJD6, a Nuclear Protein, Regulates Conidial Germination and Appressorium Formation at the Early Stage of Pathogenesis in Magnaporthe oryzae

  • In plant-pathogen interactions, Magnaporthe oryzae causes blast disease on more than 50 species of 14 monocot plants, including important crops such as rice, millet, and most 15 recently wheat. M. oryzae is a model fungus for studying plant-microbe interaction, and the main source for fungal pathogenesis in the field. Here we report that MoJMJD6 is required for conidium germination and appressorium formation in M. oryzae. We obtained MoJMJD6 mutants (ΔMojmjd6) using a target gene replacement strategy. The MoJMD6 deletion mutants were delayed for conidium germination, glycogen, and lipid droplets utilization and consequently had decreased virulence. In the ΔMojmjd6 null mutants, global histone methyltransferase modifications (H3K4me3, H3K9me3, H3K27me3, and H3K36me2/3) of the genome were unaffected. Taken together, our results indicated that MoJMJD6 function as a nuclear protein which plays an important role in conidium germination and appressorium formation in the M. oryzae. Our work provides insights into MoJMJD6-mediated regulation in the early stage of pathogenesis in plant fungi.

국제기능장애 건강분류: 아동 청소년 버전을 이용한 개별화교육지원팀 중재목표 분석 및 개별화교육계획 구성원으로서 작업치료사의 필요성: 체계적 고찰 (Analysis of Individualized Education Support Team Intervention Objectives Using International Classification of Functioning, Disability and Health-Children and Youth Version and the Necessity of Occupational Therapists as IEP Members: A Systematic Review)

  • 목적 : 본 연구에서는 국제기능장애 건강분류: 아동 청소년 버전(International Classification of Functioning, Disability and Health-Children and Youth version, ICF-CY) 프레임워크를 활용하여 개별화교육계획(individualized education plan, IEP) 협력적 팀 중재에 대해 체계적으로 고찰하고자 한다. 이를 통해 국내에서 IEP 협력적 팀 중재 속 작업치료사의 전문적인 영역을 마련하는 데 근거를 만들고, 개별화교육계획에서 협력적 팀 접근 중재의 목표를 통해 전문가로서 작업치료사의 역할에 대한 기반을 마련하고자 한다. 연구방법 : EBSCOhost, ProQuest, Web of Science를 통하여 2013년 1월부터 2023년 2월까지의 국외 논문을 검색하였다. 국외 검색어에는 "Special education", "Individualized education plan (IEP)", "IEP process", "IEP implementation", "Occupational therapy"를 사용하였다. 2차 분류를 통해 최종 10편의 연구를 분석하였다. 결과 : 분석 대상 연구의 근거 수준은 무작위 실험설계 연구가 가장 많았고, 중재 대상은 자폐성 장애가 가장 많았으며, 중재 방법은 환경 개선이 가장 많이 적용되었다. ICF-CY를 이용한 IEP 협력적 팀 중재의 목표 분석을 통해 활동에 관하여 5편, 참여와 관련하여 4편, 신체 구조 및 신체 기능과 관련하여 1편인 것으로 확인되었다. 결론 : IEP에서 협력적 팀 접근 중재 속 작업치료사의 역할은 중재의 목표에서 중요한 역할임을 확인할 수 있었다. 이를 기반으로 국내의 IEP에서 협력적 팀 접근의 전문가 중 하나로 작업치료사의 전문성을 설명할 수 있는 근거가 될 것으로 보인다.

음선 기반 중주파수 대역 잔향음 모델 개발 및 실측 데이터 비교 (Mid Frequency Band Reverberation Model Development Using Ray Theory and Comparison with Experimental Data)

  • 실제 해양에서 음파는 해수면/해저면의 거친 경계면이나 기포층/어군과 같이 불규칙적으로 분포된 체적에 의해 산란 되며, 잔향음 신호는 이러한 산란 신호의 합으로 형성된다. 실측된 잔향음 신호를 정확하게 모의하기 위해서는 각 산란 메커니즘에 적합한 산란 모델을 음파 전달 손실 모델과 결합시켜야 한다. 본 논문에서는 기존의 산란 모델과 결합이 용이한 음선 이론을 기반으로 잔향음 모델을 개발하였다. 개발된 잔향음 모델은 (1) 해수면에 대한 산란 신호로 실험 기반의 Chapman-Harris 식과 이론 기반의 APL-UW/SSA 모델, (2) 해저면에 대해서는 실험 기반의 Lambert 법칙과 이론 기반의 APL-UW/SSA 모델을 선택적으로 사용하도록 한다. 개발된 잔향음 모델의 타당성을 검증하기 위해서 정상 모드법 기반으로 개발된 Ellis 모델 결과와 2006 잔향음 공동웍크�乍【� 발표된 여러 잔향음 모델 결과와 비교하였다. 모델간의 비교를 통해 검증된 잔향음 모델을 이용하여 한국 근해의 중주파수 대역 잔향음 신호를 모의하고, 이를 실측 데이터와 시간 영역에서 직접 비교하였다. 이러한 비교를 통해 각 해역의 해양 환경의 특성에 따라 상호 다른 잔향음 신호 경향을 고찰 할 수 있으며, 나아가 각 해역 특성을 반영하는 산란 강도 함수를 본 잔향음 모델을 통해 선정할 수 있다.

풍하중을 고려한 확률론적 운동특성 평가기법 개발에 관한 연구 (Development of a Probabilistic Approach to Predict Motion Characteristics of a Ship under Wind Loads)

  • 지난 10년간, 복원력 상실로 인한 어선의 해양 사고는 지속해 증가하고 있다. 특히, 소형선박 사고의 대부분은 갑작스러운 바람이 주요 원인으로 지목되었다. 바람에 의한 소형선박의 갑작스러운 사고를 예방하기 위해서는 체계적인 분석기법 개발이 필요한 실정이다. 본 연구는 확률론적 극값 추정법을 기반으로 선박의 운동성능에 바람이 미치는 영향을 평가하는 데 그 목적이 있다. 이를 위해 운동 해석, 극값 추출, 운동 특성 분석 등의 연구를 수행하였다. 운동 해석은 Sea State 5의 파랑에서 파도, 파도와 균일 바람, 파도와 NPD풍속 모델 바람이 작용하는 3가지 조건을 적용하였다. 극값 추출은 Hysteresis 필터링 및 Peak-Valley 필터링 기법을 적용하였다. 추출된 극값을 이용하여 적합도 시험(Goodness of Fit Test)을 4가지 분포함수에 대해 수행하여 극값을 가장 잘 표현하는 최적의 분포함수를 선정하였다. 어선의 운동 특성은 3가지 주기 운동에 대하여 (Heave, Roll, Pitch)에 평가 후, 결과를 비교하였다. 선박의 운동성능 해석은 상용 솔버인 ANSYS-AQWA를 이용하였다.

지도학습 오토인코더를 이용한 전문어의 범용어 공간 매핑 방법론 (Domain-Specific Terminology Mapping Methodology Using Supervised Autoencoders)

  • 최근 비정형 자료인 텍스트를 벡터로 변환하고 이를 통해 다양한 목적으로 방대한 양의 자연어를 분석하는 시도가 이루어지고 있다. 특히 코퍼스 규모가 제한적일 수밖에 없는 전문적인 도메인의 텍스트에 대해서도 분석 수요가 급증하면서, 해당 전문 분야의 문서를 범용 문서와 함께 분석하기 위한 연구가 활발하게 이루어지고 있다. 특정 전문어를 해당 전문어 코퍼스 외부의 일반적인 범용어와 함께 분석하기 위해서는, 전문어 임베딩 공간을 범용어 임베딩 공간과 일치시키는 것이 필요하다. 기존에는 변환 행렬 또는 매핑 함수 등을 통해 전문어 코퍼스로부터 얻은 전문어 임베딩 값을 범용어 임베딩 공간으로 변환, 일치시키려는 시도가 있었지만, 변환 행렬을 기반으로 하는 선형 변환은 국지적인 범위에서만 근사적인 변환 효과가 있다는 일반적인 선형 변환의 한계를 극복하지 못했다. 이러한 선형 변환의 한계를 극복하기 위해 최근에는 다양한 형태의 비선형적인 변환 방법이 제안되고 있으며, 본 연구에서는 오토인코더(Autoencoder)와 회귀 모델을 동시에 학습하는 종단형 학습을 통해 전문어 임베딩 공간을 범용어 임베딩 공간으로 변환하여 임베딩 공간을 일치시키는 모델을 제안한다. 실제 "보건의료" 분야의 R&D 문서에 대해 임베딩 변환 실험을 진행한 결과, 제안 방법론이 기존의 오토인코더를 활용한 방법 대비 변환 정확도 측면에서 우수한 성능을 보임을 확인하였다.

Forced vibrations of an elastic rectangular plate supported by a unilateral two-parameter foundation via the Chebyshev polynomials expansion

  • The present study deals with static and dynamic behaviors including forced vibrations of an elastic rectangular nano plate on the two-parameter foundation. Firstly, the rectangular plate is assumed to be subjected to uniformly distributed and eccentrically applied concentrated loads. The governing equations of the problem are derived by considering the dynamic response of the plate, employing a series of the Chebyshev polynomials for the displacement function and applying the Galerkin method. Then, effects of the non-essential boundary conditions of the plate, i.e., the boundary conditions related to the shearing forces, the bending moments and the corner forces, are included in the governing equation of motion to compensate for the non-satisfied boundary conditions and increase the accuracy of the Galerkin method. The approximate numerical solution is accomplished using an iterative process due to the non-linearity of the unilateral property of the two-parameter foundation. The plate under static concentrated load is investigated in detail numerically by considering a wide range of parameters of the plate and the foundation stiffnesses. Numerical treatment of the problem in the time domain is carried out by assuming a stepwise variation of the concentrated load and the linear acceleration procedure is employed in the solution of the system of governing differential equations derived from the equation of motion. Time variations of the contact region and those of the displacements of the plate are presented in the figures for various numbers of the two-parameter of the foundation, as well as the classical and nano parameters of the plate particularly focusing on the non-linearity of the problem due to the plate lift-off from the unilateral foundation. The effects of classical and nonlocal parameters and loading are investigated in detail. Definition of the separation between the plate and the two-parameter foundation is presented and applied to the given problem. The effect of the lift-off on the static and dynamic behavior of the rectangular plate is studied in detail by considering various loading conditions. The numerical study shows that the effect of nonlocal parameters on the behavior of the plate becomes significant, when nonlinearity becomes more profound, due to the lift-off of the plate. It is seen that the size effects are significant in static and dynamic analysis of nano-scaled rectangular plates and need to be included in the mechanical analyses. Furthermore, the corner displacement of the plate is affected more significantly from the lift-off, whereas it is less marked in the time variation of the middle displacement of the plate. Several numerical examples are presented to examine the sensibility of various parameters associated with nonlocal parameters of the plate and foundation. Both stiffening and softening nonlocal parameters behavior of the plate are identified in the numerical solutions which show that increasing the foundation stiffness decreases the extent of the contact region, whereas the stiffness of the shear layer increases the contact region and reduces the foundation settlement considerably.

중심성 교뇌수초용해 환자의 치료 경과에 따른 보행 기능의 정량적 평가 (Case Report: Quantitative Evaluation of Gait Function Following Treatment Progression in a Patient with Central Pontine Myelinolysis)

  • Introduction: Central pontine myelinolysis (CPM) is a rare neurological disorder marked by symmetric nerve fiber damage, commonly following rapid hyponatremia correction, with no established treatment and a poor prognosis. The condition, often linked to alcoholism, malnutrition, and various diseases, lacks comprehensive studies on its impact on gait. This research aims to quantitatively analyze gait changes in CPM patients receiving both traditional Korean and conventional rehabilitation treatments, addressing a gap in current understanding of CPM management. Case presentation: A 56-year-old male diagnosed with CPM following an initial misdiagnosis underwent combined electoracupuncture and rehabilitative treatment at ○○ university Korean medical center, resulting in significant gait improvements. A treadmill gait analysis system was used to measure changes in key gait parameters at 2-week intervals, and the patient's progress was documented. Conclusions: The quantitative analysis revealed significant gait improvements. Foot rotation decreased from 8.9° to 6.4° (right) and from 11.1° to 7.2° (left); lateral symmetry improved from -7.8 to 0.8; step length increased from 21 cm to 44 cm (right) and from 19 cm to 44 cm (left); and velocity increased from 1.2 m/s to 2.7 m/s. These findings highlight decreased foot rotation and lateral symmetry, along with increased step length and velocity, suggesting a positive outcome of the treatment regimen. Notably, the patient experienced no adverse effects related to the treatments. Despite limitations, including the singe case focus and lack of prior gait-focused CPM research, this case report provides valuable insights into effective CPM management strategies, paving the way for future research in this domain.

Biological Functions of N- and O-linked Oligosaccharides of Equine Chorionic Gonadotropin and Lutropin/Chorionicgonadotropin Receptor

  • Members of the glycoprotein family, which includes CG, LH, FSH and TSH, comprise two noncovalently linked $\alpha$- and $\beta$-subunits. Equine chorionic gonadotropin (eCG), known as PMSG, has a number of interesting and unique characteristics since it appears to be a single molecule that possesses both LH- and FSH-like activities in other species than the horse. This dual activity of eCG in heterologous species is of fundamental interest to the study of the structure-function relationships of gonadotropins and their receptors. CG and LH $\beta$ genes are different in primates. In horse, however, a single gene encodes both eCG and eLH $\beta$-subunits. The subunit mRNA levels seem to be independently regulated and their imbalance may account for differences in the quantities of $\alpha$ - and $\beta$ -subunits in the placenta and pituitary. The dual activities of eCG could be separated by removal of the N-linked oligosaccharide on the $\alpha$-subunit Asn 56 or CTP-associated O-linked oligosaccharides. The tethered-eCG was. efficiently secreted and showed similar LH-like activity to the dimeric eCG. Interestingly, the FSH-like activity of the tethered-eCG was increased markedly in comparison with the native and wild type eCG. These results also suggest that this molecular can implay particular models of FSH-like activity not LH-like activity in the eCG/indicate that the constructs of tethered molecule will be useful in the study of mutants that affect subunit association and/or secretion. A single-chain analog can also be constructed to include additional hormone-specific bioactive generating potentially efficacious compounds that have only FSH-like activity. The LH/CG receptor (LH/CGR), a membrane glycoprotein that is present on testicular Leydig cells and ovarian theca, granulosa, luteal, and interstitial cells, plays a pivotal role in the regulation of gonadal development and function in males as well as in nonpregnant and pregnant females. The LH/CGR is a member of the family of G protein-coupled receptors and its structure is predicted to consist of a large extracellular domain connected to a bundle of seven membrane-spanning a-helices. The LH/CGR phosphorylation can be induced with a phorbol ester, but not with a calcium ionophore. The truncated form of LHR also was down-regulated normally in response to hCG stimulation. In contrast, the cell lines expressing LHR-t63I or LHR-628, the two phosphorylation-negative receptor mutant, showed a delay in the early phase of hCG-induced desensitization, a complete loss of PMA-induced desensitization, and an increase in the rate of hCG-induced receptor down-regulation. These results clearly show that residues 632-653 in the C-terminal tail of the LHR are involved in PMA-induced desensitization, hCG-induced desensitization, and hCG-induced down-regulation. Recently, constitutively activating mutations of the receptor have been identified that are associated with familial male-precocious puberty. Cells expressing LHR-D556Y bind hCG with normal affinity, exhibit a 25-fold increase in basal cAMP and respond to hCG with a normal increase in cAMP accumulation. This mutation enhances the internalization of the free and agonist-occupied receptors ~2- and ~17-fold, respectively. We conclude that the state of activation of the LHR can modulate its basal and/or agonist-stimulated internalization. Since the internalization of hCG is involved in the termination of hCG actions, we suggest that the lack of responsiveness detected in cells expressing LHR-L435R is due to the fast rate of internalization of the bound hCG. This statement is supported by the finding that hCG responsiveness is restored when the cells are lysed and signal transduction is measured in a subcellular fraction (membranes) that cannot internalize the bound hormone.

구조토양에서의 침출수와 잔존수농도의 파과곡선에 관한 비교연구 (COMPARISON OF FLUX AND RESIDENT CONCENTRATION BREAKTHROUGH CURVES IN STRUCTURED SOIL COLUMNS)

  • 오염물질 이동현상 연구에서는 침출수 혹은 잔존수농도 형태가 사용되는데 이의 선택은 모니터링 방법에 의존하게 된다. 파과곡선 실험에서 모니터링 농도 형태에 관한 선택은 임의적이며, 각 농도 형태에서 얻어진 운송 파라미터들은 동등하며 다공성매질의 수리적 특성을 각각 대표하는 것으로 알려져왔다. 그러나, 현장상태의 구조적 발달을 보이는 토양에서는 농도 형태별 운송계수의 동등성이 의문시 된다. 본 연 구에서는 불교란 현장시료(직경 20cm, 높이 20cm)에 대하여 두가지 농도 형태에 의한 파과곡선 실험을 시행하므로써 모니터링 방법에 따른 농도 형태와 그에 따른 운송 파라미터들을 비교분석 하였다. 침출수 농도와 잔존수 농도는 토양상부에서 20cm와 loom 떨어진 지점에서 EC-meter와 TDR 을 이용하여 각각 측정하였다. 연구결과, 침출수 농도는 잔존수 농도보다 첨두농도가 훨씬 높게 그리고 첨두농도의 운송시간이 짧게 나타났음을 알 수 있었다. 따라서 침출수농도곡선으로부터 추정된 운송파 라미터들은 잔존수농도곡선으로부터 추정된 수치들과 상당한 차이를 보였으며 그 차이는 CLT 모델보다 CDE 모델에서 더 크게 나타났다. 특히 CDE 모델에서는 침출수곡선으로부터 도출된 계수값들이 잔존수곡선으로부터 도출된 계수값들보다 훨씬 크게 나타났다. 이는 구조토양내에 존재하고 있는 대공극을 통한 오염물질 우회통과와 평형조건에서의 CDE 모델이 연구대상토양에서의 오염물질 이동현상을 표현하는데 부적합하였기 때문인 것으로 사료된다. 분자 분산에 대한 동수리학적 확산의 비와 Peclet number와의 상관관계를 나타내는 도표영역에서 두가지 농도는 모두 역학적 확산이 오염물질 운송을 좌우하는 영역에 속하였다. 그러나 분자분산은 토양내 대공극부분보다 matrix 부분에서의 오염물질 확산에 더 많은 기여를 하는 것으로 나타났으며 이는 공극유속과 확산계수사이에 존재하는 비선형성에 기인하기 때문인 것으로 사료된다.
