In this study, the antioxidant and tyrosinase inhibitory activities of aqueous ethanolic extract from the immature fruits of Malus pumila cv. Fuji were evaluated. The antioxidant capacities of the extract was investigated employing radical scavenging assays using DPPH and $ABTS^+$ radicals. The skin-whitening effect of M. pumila cv. Fuji extract was tested using mushroom tyrosinase assay. In addition, the total phenolic content was measured by a spectrophotometric analysis. All tested samples showed a dose-dependent radical scavenging and tyrosinase inhibitory activities. Among the tested samples, the ethyl acetate (EtOAc)-soluble portion from the immature fruits of M. pumila cv. Fuji was showed the significant DPPH and $ABTS^+$ radicals scavenging activities. Also, the tyrosinase inhibitory activity of the ethyl acetate (EtOAc)-soluble portion from immature apples was higher than other solvent-soluble portion. These results suggest that unripe fruits of M. pumila cv. Fuji could be considered as a new valuable source of natural antioxidant and skin-whitening agents. Systematic investigation of immature fruits of Malus pumila cv. Fuji will be performed for the further development of its biological properties.
The purpose of this study was to evaluate for the cytotoxicity of glass-ionomer cement liners(GC liningcement, Ketac-bond, Vitrebond and Fuji lining LC) on the fibroblasts cultured from human pulp. The fibroblasts were cultured in DMEM-10% FBS medium. The measurement of pH, succinate dehydrogenase (SDH) activity test and $^{51}Chromium$ release test were performed. Viable cell count and $^{14}C$-leucine incorporation rate were evaluated following culture time of 2, 4 and 6 days. The results of this study were as follows : 1. The pH in all cements was to be neutralized as time elapsed, and Fuji lining LC was the lowest pH value among them. 2. SDH activity was more inhibited in GC lining cement and Vitrebond than Ketac-bond and Fuji lining LC with the setting process, and GC lining cement and Ketac-bond were reduced after 5 minute's setting and then elevated as time elapsed. 3. In SDH activity test following exposure time, the activity in Vitrebond, GC lining cement and Fuji lining LC was inhibited with increased exposure time, but it was fairly constant in Ketac-bond. 4. Overall the liquid component was more inhibited than the powder component of glass-ionomer cement in SDH activity test. 5. In $^{51}Cr$-release test, Fuji lining LC was the most released of all the cements tested and followed by : Vitrebond, Ketac-bond, GC lining cement. 6. In viable cell count, the number of cells increased as the culture day proceeded in Ketac-bond, but they decreased in GC lining cement. Fuji lining LC was only observed after 2 days culture and there was not observed the whole culture days in Vitrebond. 7. In $^{14}C$-leucine incorporation rate test, protein synthesis was decreased with the number of culture days in GC lining cement, Vitrebond and Fuji lining LC, but it was followed that of control in Ketacbond.
In this study, we investigated the limit temperature range which is effective for dormancy release of 'Hongro' and 'Fuji' apples during the endodormancy period. The germination rate was 50% or more in all treatments except of 'Hongro' $-5^{\circ}C$ treatment. The germination rate of 'Fuji' was 86.3% at the temperature of $-5^{\circ}C$. The concentrations of carbohydrate and mineral components were significantly different between treatments but did not show any tendency or specific change. However, the sorbitol contents of 'Hongro' $-5^{\circ}C$ treatment were higher at 29.62 mg/g than those of 'Fuji', which was not affected for dormancy release. The results of hormone analysis showed that ABA(abscisic acid) and JA(jasmonic acid) in 'Hongro' $-5^{\circ}C$ treatment were respectively 176.48 and 15.72 ng/g. ABA, JA and SA(salicylic acid) contents in 'Fuji' apple tree were significantly lower than those of 'Hongro'. As a result of this study, the limit temperature range effective for dormancy release was different according to the varieties, and 'Hongro' did not affect with 30.3% of germination rate at $-5^{\circ}C$. However, 'Fuji' is 86.3% even at $-5^{\circ}C$ it was suggested that 'Fuji' had a chilling accumulation for dormancy release.
In order to use Osmia cornifrons more effectively in apples, we investigated the pollination effect and nesting activity of O. cornifrons on 'Hongro', the middle-season cultivar, and 'Fuji', the late-season cultivar. There was a significant difference in the nesting activity and pollination effect of O. cornifrons depending on the apple cultivar. The nesting activity, rate of trap nesting, and reproduction in 'Fuji' were 2.5, 1.5, and 3.8 times greater than in 'Hongro', respectively. The pollination effect according to cultivar 'Fuji' was 1.6 times greater than that of 'Hongro' in the central fruit set. In terms of the quality of fruit, the asymmetry index of 'Fuji' was 2.5 times lower than that of 'Hongro', and the number of apple seeds of 'Fuji' was 1.9 times greater than that of 'Hongro'. The main reason for this result was the air temperature. The activity of O. cornifrons was most affected by air temperature (R2 = 0.578). It is expected that the nesting activity and pollination effect are great in 'Fuji' (17.4-24.1℃) when the temperature during the blooming period is higher than that of 'Hongro' (12.5-20.2℃). Our study provides important information on stable apple production for apple cultivators, and can be used as an evidence for changes in flora and insect fauna caused by climate change.
The internal breakdown of Fuji apple during CA storage classified as watercore breakdown, low temperature breakdown and CO2 injury. This study was undertaken to investigate the watercore breakdown injury factors of Korean Fuji apple during CA storage. The development of internal breakdown was more increased with the larger size, the later harvest time and the hither CO2 gas level. But in internal breakdown fruit of the titratable acidity and soluble solid decreased significantly, the pH of fruit juice and the production of carbon dioxide was greatly increased. The best gas levels of CA storage was 2% oxygen and 3% carbon dioxide. Thus, the predictable parameters of internal breakdown of fruit were increase in pH on decrease titratable acidity within 2 months of CA storage, increase carbon dioxide. So, it was found that the best CA sotrage for internal breakdown control of fruit during CA storage was delayed CA storage methods after low temperature storage immediate harvest of apple and than took a step. The delayed CA storage after low temperature storage for 2 months was more effective in the prevention of development of internal breakdown than immediate CA storage after harvest.
Cytotoxicity of dental restorative materials using cell culture technique has been extensively studied by various quantitative assays. The aim of this study was to investigate the microstructural change of damaged L292 cells which could not observed with light microscope. Cytotoxic effect of ZOE, Prisma APH (Densply International Inc., U.S.A.), Clearfil FII(Kuraray Co., Japan), Fuji II(GC Co., Japan) and Fuji II LC(GC Co., Japan) on cultured L292 cells were observed. Irreversible cell damage and cytolysis were found in ZOE and Fuji II groups. In Clearfil FII, mild to moderate cell damage was observed. APH group showed variable cytotoxicity. Moderate cell damage was found in Fuji II LC group. Cytotoxic effect were as follows : A condensation of the chromatin occureds along or adjacent to the inner membrane of the nuclear envelops. The nuclear envelope remained resonably intact but the contents were partially or completely lost. The cell nucleus contains clusters of markedly electron-dense interchromatin granules. The rough endoplasmic reticulum were dilated. In some mitochondira, matrix was disoriented and fused cristae were discernible. Mitochondiral swelling and woolly appearance were recognized. Large vacuoles and autolysosmes were found in cytoplasm. Some breaks of the cytoplasmic membrane and even cytolysis could be seen in dying cells.

Near-infrared spectroscopy (NIRS) was used to investigate the possibility for application in identification of apple cultivars. Three apple cultivars ‘Kamhong, Hwahong, and Fuji’ produced in Korea were scanned over the range of 1100-2500nm using NIRS (Infra Alzer 500). Two types of samples were used for scanning; one was apple with skin and the other was apple without skin. For cultivar identification, the NIR absorbance spectrums were analyzed by qualitative calibration in “Sesame” analysis program, and the various influence properties such as sugar contents, acidity, color, firmness, and micro-structure were compared in scanned samples. The ‘Kamhong’ cultivar could be identified from ‘Hwahong’ and ‘Fuji’ cultivars using the cluster model analysis. The test samples in calibration between ‘Kamhong’ and ‘Fuji’ cultivars could be completely identified. The test samples in calibration between ‘Kamhong’ and ‘Hwahong’ cultivars could be identified most of all. But, ‘Hwahong’ and ‘Fuji’ cultivars could not be quite classified each other. The apple skin influenced the identification process of apple cultivars. The samples without skin were more difficult to classify in calibration than the samples with skin. The physicochemical properties of apple cultivars showed like the result of identification in calibration using NIRS. Some physicochemical properties of ‘Kamhong’ cultivar were different from those of the other cultivars. Those of ‘Hwahong’ and ‘Fuji’ cultivars showed. similar to each other. The sucrose contents of ‘Kamhong’ cultivar were higher and the fructose contents and firmness of skin and flesh were lower than those of the others. The hypodermis layer of skin in ‘Kamhong’ cultivar was thinner than those of the others. In this studies, the identification of all apple cultivars by NIRS was not quite accurate because of the physicochemical properties which were different in the same cultivar, and inconsistent patterns by culivars in some properties. To solve these problems in NIRS application for apple cultivar identification, further study should be focused on the use of peculiar properties among the apple cultivars.
Cell culture methods have been used to assess the cytotoxicity of dental materials. Different paramaters are used to monitor cytotoxic effects. But it is difficult to compare each investigator's results with different methods. The objective of this study was to investigate cytotoxic effect of several retrograde filling materials according to cell lines and assay methods. Cytotoxicity of Bestalloy (Dogmyung, Korea), Prisma APH(Densply International Inc., U.S.A.), Clearfil FII (Kuraray Co., Japan), Fuji II (GC Co., Japan), Fuji II LC (GC Co., Japan) and IRM (Densply Co., U.S.A.) on L929, 3T3 and KB permanent cell lines was measured. Radiochromium, Lactate dehydrogenase (LDH) release method and colorimetric assays, namely neutral red (NR) and MTT were used. Each material was mixed according to the manufacturer's instruction. They were tested as solid and extracted state. Cell culture media were added to each mixed or solid materials then the solution was collected and used as extract solutions. Solid Fuji II showed mild cytotoxicity on three cell lines using radiochromium release method. There was no difference in cytotoxicity of extract solution group using radiochromium release method. In colorimetric assay immediate Fuji II group and all the IRM groups showed severe cytotoxic effect. Difference in cyctotoxicity was due to rather kinds of cell lines than assay methods. Solid Fuji II and IRM showed mild cytotoxicity on three cell lines. But extract solutions had different cytotoxic effect according to cell lines using LDH release assay. Light-cured glass ionomer had mild to moderate degree of cytotoxicity on three cell lines. Cytotoxicity was affected by specimen prepaton. Susceptibility of each cell ines were also affected by assay emthods. It was suggested that cytotoxicity study using only one cell line and/or assay method might not accurately reflect the real toxic nature of dental biomaterials.
Fuji apple variety introduced in Japan has excellent storage quality and good taste so it is most commonly cultivated in the Korean Peninsula. Accurate prediction of harvest maturity allows farmers to more efficiently manage their farm, such as working time, fruit storage, market shipment and labor distribution so it is very important. This study was carried out to predict the harvest maturity of 'Fuji' apple using DTS (Days Transformed to Standard temperature) model based on the Arrhenius law in the Gunwi province of the South Korea. Input data are daily average temperature and apple harvest maturity. Predicted the harvest maturity of Fuji apple after estimating the optimal parameters by using the Nelder-Mead method. The differences of observed and predicted harvest maturity day are approximately 1 to 4 days and the RMSE is 2.9.
The purpose of this study was to examine the effect of surface treatment on the flexural strength of glass ionomer cement with time elapsed. Glass ionomer cement (Fuji ionomer type II, (GC Co.) was used as experimental materials. Glass ionomer cement was placed in a beam-shaped teflon mold (3mm ${\times}$ 3mm ${\times}$ 25mm) that was rest on a glass plate. Another flat glass was placed on the top of the mold with pressure. After the cement was set, the specimens were divided into three groups and thirty two specimens in each group were surface-treated as follows: No treatment group: Specimens were no surface-treated and stored at $35.6^{\circ}C$ in distilled water. Fuji varnish application (FA) group: Specimens were surface-treated with Fuji varnish (GC Co.) and stored at $35.6^{\circ}C$ in distilled water. Vaseline storage (VS) group: Specimens were no surface-treated and stored at $35.6^{\circ}C$ in vaseline. The flexural strength was measured after I day, 1 week, 2 weeks, 4 weeks from the start of mixing using Instron Universal Testing Instruments. Results were as follows: 1. After 4 weeks, vaseline storage group exhibited the maximum flexural strength (p <0.0005). 2. The flexural strengths in no treatment group were slightly increased with time elapsed, but its difference was not significant, statistically. 3. The flexural strengths in Fuji varnish application group were increased with time elapsed (p <0.05). 4. The flexural strengths in Fuji varnish application group and vaseline storage group were greater than that in no treatment group (p <0.0005).
