Ergosterol Contents and Enzymatic Characteristics of Lentinula edodes During Culture and Fruiting Periods (표고 균주의 배양 기간과 자실체 발생 기간에 따른 에르고스테롤 변화와 효소적 특성)

  • Three different strains of Lentinula edodes, Sanlim 5-Ho, Sanlim 6-Ho and Nongki 3-Ho, were cultured in the sawdust media of Mongolian oak(Quercu mongolica Fisch) for 90 days under dark and light conditions(each 30 days) and fruiting period(30 days). Weight loss of sawdust media was determined after fungal cultures and the contents of ergosterol in fungal mycelia were quantified by HPLC analysis followed by solvent extraction. Compared with the two other fungal strains$(8\%)$, weight loss of Sanlim 5-Ho was slightly lowered to $7\%$. The level of ergosterol content, a parameter for fungal growth, was continuously enhanced in Sanlim 5-Ho for dark and light incubation periods. However, Sanlim 6-Ho and Nongki 3-Ho recorded the maximized fungal growth under light condition. In fruiting periods the ergosterol contents were lowered in the three strains. Intra- and extracellular enzymes during cultural and fruiting periods were also characterized. The activity of Mn-peroxidase and laccase, which are characteristics enzymes for white rot fungi as lignin degrading enzymes, were determined as a high level overall the periods. As cellulose degrading indicators, the activity of CMCase, avicelase, xylanase and glucanase were detectable in initial incubation period.

Characteristics of Flowering and Bolling in Cotton (목화의 개화 및 결삭 특성)

  • This study was carried out to ascertain the flowering and boiling habits in conditions of vinyl mulched cultivation. The effects of vinyl mulching cultivation in cotton was early flowering. The flowering period was no more than 26 days and shedding of flower bud was lower. The total boll setting rate to flower number was about 38%. The number of flowering plants was rapidly increased until Aug. 8, then later was decreased, the period of the highest flowering occured was for 5 days from Aug. 6 to Aug. 10. Boll setting was increased that the flowers opened before July 30, then the later flowers was tended to decrease belling in spite of the increased flowering. However flower bud shedding had a tendency to decrease after the highest boiling period. The effect of weather, max. temperature and hours of sunshine on flowering days were highly positive significant correlation. Minimum temperatures, high air humidity and amount of rainfall had a negative significant correlation. The demanded days to flowering on the same internode in each fruiting branches was taken average 1.2 days, these days among the fruiting branches of the same internode was 3.6 days.

Artificial Cultivation and Antioxidant Activity of Korean Native Mushroom Pycnoporus coccineus (한국산 간버섯의 인공재배 및 항산화 활성 평가)

  • The mycelial growth of P. coccineus strain was good in PDA and YMA, but mycelial growth was low in MEA. Light irradiation during the incubation period affected the pigment formation and density of mycelia. Mushroom of P. coccineus strain was able to produce fruiting bodies in both bottle and bag cultivation, and oak sawdust was found to be the most suitable substrate for spawn culture and cultivation. In artificial cultivation using sawdust medium, fruiting body was grown to the extent that visual observation was possible from the 15th day, and it formed about 5 days fast in the treatment group with low relative humidity. From 40 to 45 days of mushroom development, mature fruiting bodies could be harvested, and the lower relative humidity of the growing room favored mushroom development and growth. Antioxidant activity of fruiting bodies harvested from artificial cultivation showed that ABTS radical scavenging activity of bottle-cultivated and wild fruit bodies were shown at 505㎍/㎖ and 515㎍/㎖, respectively. However, fruiting bodies harvested in bag cultivation were high at 910㎍/㎖. As a result of DPPH radical scavenging activity, all extracts were found to be inactive, exhibiting IC50 value of more than 2,000㎍/㎖ concentration. The ethyl acetate extract of mushrooms obtained from bottle cultivation showed the highest activity with 1,550㎍/㎖ IC50 value. Methanol extract of wild fruit bodies had the highest ABTS radical scavenging activity at the same concentration (10mg/㎖).

Cultivation characteristics and yield of Sparassis crispa according to medium pH, medium moisture content, and inoculum volume of liquid spawn (배지 pH, 배지함수율 및 종균접종량에 따른 꽃송이버섯의 재배 특성 및 수량)

  • The cultivation of Sparassis crispa in the beginning of the 2000s in South Korea, and the cultivar 'Neowul' bred in the Chonbuk Agricultural Research and Extension Service were registered first in 2016. However, there is no manual for the cultivation of Sparassis crispa, and therefore, there remains a big difference in its harvest rate across farms. Herein, we aimed to study the primordium formation conditions of Sparassis crispa 'Neowul' according to the medium pH, medium moisture content, and inoculum volume of liquid spawn and develop a stable production technology. We found the annual yield per bottle relating to the cultivation period, harvest rate, and the weight of fruiting body to be the highest at 363.6 g in the area cultivated at pH 3.8. However, it is thought that cultivation by adjusting the pH to 3.9±1 would be necessary for stable production, considering that at pH 3.6, the yield sharply reduced to 189.5 g. Moreover, the culture period was shorter at pH 4.0 compared with that at pH3.8, and the cultivation period at pH 4.0 was the same as that at pH 3.8. No significant difference in the weight of the fruiting body at different conditions was recognized. Additionally, it is difficult to regulate the pH precisely in practical applications in the farms. It is thought that 341.8 g Sparassis crispa will be produced per bottle annually if the medium moisture content is adjusted to 65%, liquid spawn inoculum volume is equivalent to 4% of the medium volume, and the humidity in the culture room is set to below 50%.

Cultural and Morphological characteristics of fruit body of genus Agrocybe on Artificial Cultivation in Summer (볏짚버섯속(Agrocybe) 보존균주의 여름철 인공재배시 재배 및 형태적 특성)

  • Thirty strains of genus Agrocybe which had been kept in National Institute of Horticultural and Herbal Science, were tested for the formation of fruiting body. Nineteen strains of collected genus Agrocybe were formed fruiting body at bottle culture of sawdust medium. Five strains (including ASI 19003) and thirteen strains (including ASI 19007) could be grouped as A. cylindracea and A. chaxingu. Although ASI 19008 showed the formation of fruiting body, morphological characteristics were significantly different from the two groups. Cultural period of ASI 19003 strain at sawdust substrates was 29 days and ASI 19007 train was 30 days in liquid spawn inoculation. The yield and quality of ASI 19003 strain was excellent in the spring, fall, and winter, whereas ASI 19007 strain was excellent in the summer. Accordingly, these cultivar might be contribute to farmers' income by stable year-round production if using the season-oriented strain.

Relationship between Ectomycorrhizal Fruiting Bodies and Climatic and Environmental Factors in Naejangsan National Park

  • We collected and identified 5,721 ectomycorrhizal fruiting bodies (EcM) from Naejangsan National Park from June 2004 to 2013, belonging to 1 phylum, 1 class, 6 orders, 19 families, 40 genera, and 196 species. Of these, 2,249 individuals were identified as 89 species belonging to 11 genera in 7 families in the Agaricales; 1,511 were identified as 43 species belonging to 2 genera in 1 family in the Russulales; 1,132 were identified as 50 species belonging to 21 genera in 6 families in the Boletales; 793 were identified as 8 species belonging to 3 genera in 2 families in the Cantharellales; 29 were identified as 3 species belonging to 2 genera in 2 families in the Thelephorales; and 7 were identified as 3 species belonging to 1 genus in 1 family in the Gomphales. Thus, most of the EcMs identified belonged to the following 3 orders: Agaricales, Russulales, and Boletales. Russulaceae were most common (43 species), followed by Boletaceae (39 species), and Amanitaceae (27 species); most individuals were Russulaceae (1,511), followed by Hydnagiaceae (1,071) and Boletaceae (804). The monthly distribution showed that the greatest number of individuals and species of EcM, including the dominant ones, occur around July~September at an elevation of 200~299 m, diminishing markedly above 600 m. The greatest number of individuals and species, including the dominant ones, were collected in the period with average temperatures $25.0{\sim}26.9^{\circ}C$, lows of $21.0{\sim}22.9^{\circ}C$, and highs of $30.0{\sim}31.9^{\circ}C$, relative humidity > 76%, and rainfall > 400 mm.

  • Lentinula edodes is known by oak mushroom. It has been favored as delicious and nutritious food and the low-calorie food with a high nutritional value. It is also functional food since it contains a material well-known for its medicinal benefits. Since the growth and quality of oak mushrooms are sensitively affected by environmental conditions, an adequate environmental control is very essential to improve the yield and quality under protected cultivation. The main objectives of the study were to investigate cultural characteristics of mycelial growth and in vitro fruiting of Lentinula edodes. The optimum culture media for mycelial growth of L. edodes were PDA and MYA. Similarly, optimum temperature was $25^{\circ}C$. Malt extract(2%) and yeast extract(0.2%) were optimum carbon and nitrogen sources. Optimal culture period was 110~120 days in sawdust medium. Mycelial growth in medium(61 mm/7 days) Quercus mongolica extract the most good. Among different five log types, highest mycelial growth and fruiting productivity were observed in Quercus variabilis sawdust(20.9%).

Cultural Characteristics of A Medicinal Mushroom, Phellinus linteus

  • For comparison of mycelial colonization of Phellinus linteus on logs, several techniques of inoculation were tested; sterilized short log inoculation, drilling inoculation and log-end sandwich inoculation. The mycelial colonization of P. linteus on logs was good in the treatment of sterilized short log inoculation, but poor in the traditional methods such as drilling inoculation and log-end sandwich inoculation. The initial mycelial growth and the full mycelial colonization of P. linteus in logs were the best in case of 20 cm logs under the condition of 42% moisture content. Also, the initial mycelial growth of P. linteus was accelerated over 12 hours of sterilization. Burying method of logs after $5{\sim}6$ months of incubation was the best for formation of basidiocarps of P. linteus. The formation of fruiting body of P. linteus was quite good in the cultivation house at $31{\sim}35^{\circ}C$ and over 96% of relative humidity.

Biochemical Characterization of Protease Produced by Cordyceps nutans

  • The fruiting body of Cordyceps is derived from the pupa or larva of insects infected by the entomopathogenic fungi Cordyceps. The fruiting body has been used as an anti-cancer and anti-inflammatory ingredient in traditional Chinese medicine. The biochemical characteristics of protease isolated from Cordyceps nutans were investigated in this study. The culturing period for production of protease by C. nutans was 10days. The acidity was pH 7.0, and the temperature was $25^{\circ}C$. The carbon and nitrogen sources for the production of the protease were glucose and yeast extract, respectively. The ratio of C/N was 2% glucose and 0.6% yeast extract. 0.06% $CuSO_4$ was used as the inorganic salt. The investigation into the acidity of the protease produced by C. nutans revealed that the optimal pH and temperature were pH 7.0 and $30^{\circ}C$. The stability of the protease was shown as pH 6.0~9.0 and $30{\sim}50^{\circ}C$. The investigation into the influence of the metal ions on the enzyme activation of C. nutans revealed that it was inhibited in $ZnSO_4$ and activated in $FeSO_4$.

Physiological and Genetic Characteristics of Cultivated Mushroom, Hypsizygus marmoreus

  • A edible mushroom, Hypsizygus marmoreus is commercially cultivated in Northeast Asia. Japan's annual production is 110,000ton or more. Since 2002, cultivation is expanded in Korea. To investigate the morphological, cultural and microscopic characteristics of Hypsizygus marmoreus, 109 isolates were collected from Korea and other countries. Clamp connection, chlamydospore and arthrospore were present in all tested isolates of H. marmoreus except HYM-002 and HYM-004. Also pilealtrama, gilltrama, basidia, basidiospore and cystidia of fruiting body were no difference among the isolates in the present investigation. Morphological characteristics of fruiting body was that color of pileus was brown and white, irregular as marble, the average size 12~22mm and stipes was $46{\sim}91{\times}6{\sim}10mm$. Isolates HYM-031, HYM-047 and HYM-109 formed grayish-brown pileus with a faint pattern. Molecular analysis with RAPD and ITS rDNA sequence analysis were also performed to check the genetic relationships among H. marmoreus isolates. Based on the RAPD analysis using the URP-PCR, all isolates of H. marmoreus were clustered into large 3 groups but more than 90% showed high similarity. In addition, morphological and geographical differences have been classified as an independent cluster. The brown and white strains enclosed in same cluster. So genetically no significance difference was observed between these two strains. ITS gene sequences of 16 selected isolates which were 640 bp long, were aligned and compared. The similarity in ITS sequence was 94.8 to 99.1% among tested isolates and the H. marmoreus isolates in GeneBank. In conclusion the tested isolates were H. marmoreus. Morphological and molecular observations proved that all tested isolates were belonging to H. marmoreus. For the stable artificial cultivation, composition of optimum media, mature period and light condition were established. Optimal formula of artificial cultivation medium was Douglas sawdust: corn cob: soybean meal: wheat bran = 40:30:15:15. In addition, 7% rice bran and 3% yellow sucrose was the most effective composition for spawn's liquid medium. For the maturation of the isolates was favorable for growing for 20 to 30 days at $25^{\circ}C$ and the LED lights in mixture of white and blue was good for growth period. For effective growth, the temperature, humidity and aeration control in every step was important.
