Fruit thinning rate and characteristics were investigated for three years on seven promising pear (Pyrus pyrifolia Nakai) cultivars, which were treated by lime-sulfur as an eco-chemical thinning substance. Lime-sulfur was treated twice at the second and third days after full bloom by cultivar. Most of pear cultivars were significantly thinned by lime-sulfur compounds. Especially 'Whangkeumbae', 'Supergold' and 'Hanareum' exhibited high flower thinning rates, 41.5%, 40.1% and 39.9%, respectively. As weather conditions at the lime-sulfur treatment, insolation and cloud amount were correlated with flower thinning rate but not significant (r = 0.49 and r = -0.45, respectively). These results suggest that lime-sulfur is suitable for reducing labor force for flower thinning of Korean pears but flower thinning effects of lime-sulfur can vary depending on other factors such as environmental conditions. This information will provide useful data for low labor force cultivation of Korean pear cultivars.
Tricholoma matsutake is a traditional favorite food in East Asia, cultivated in fairy rings called "shiro," which are found near Pinus densiflora. For effective artificial cultivation of Tri. matsutake, microorganisms from symbiotic fairy rings are co-cultivated. In this study, one bacterial isolate (Y22_B35) and two fungal isolates (Y22_F64 and Y22_F68) displayed growth-promoting effects on Tri. matsutake mycelium (158.47, 125.00, and 122.26% enhanced growth, respectively). For identification, 16S rRNA or ITS regions from the microorganisms¡¯ genomes were sequenced. Other sequences, including BenA, CaM, and RPB2 were sequenced in the fungal isolates. The bacterial isolate Y22_B35 was identified as Bacillus cereus. Y22_F64 and Y22_F68 were identified as Umbelopsis nana and Aspergillus parvulus, respectively. To identify the effects of the dominant microorganisms on Tri. Matsutake cultivation, metagenomic analyses were performed. Discovery of these Tri. matsutake mycelium growth-promoting microorganisms and metagenomics analyses are expected to contribute to our understanding of Tri. matsutake fruiting body growth and construction of biomimicry.

This study examined a technique for cooling root zone aimed at lowering substrate temperature for sweet pepper (Capsicum annum L. 'Orange glory') cultivation in coir substrate hydroponics during hot season, from the $16^{th}$ of July to $15^{th}$ of October in 2012. The root zone cooling technique was applied by using an air duct (${\varnothing}12$ cm, hole size 0.1 mm) to blow cool air between two slabs during night (5p.m. to 3a.m.). Between the $23^{rd}$ of July and $31^{st}$ of August (hot temperature period), average daily substrate temperature was $24.7^{\circ}C$ under the root zone cooling, whereas it was $28.2^{\circ}C$ under condition of no cooling (control). In sunny day (600~700 W $m^{-2}{\cdot}s^{-1}$), average substrate temperatures during the day (6a.m. to 8p.m.) and night (8p.m. to 6a.m.) were lower about $1.7^{\circ}C$ and $3.3^{\circ}C$, respectively, under the cooling treatment, compared to that of control. The degree of temperature reduction in the substrate was averagely $0.5^{\circ}C$ per hour under the cooling treatment during 6p.m. to 8p.m.; however, there was no decrease in the temperature under the control. The temperature difference between the cooling and control treatments was $1.3^{\circ}C$ and $0.6^{\circ}C$ in the upper and lower part of the slab, respectively. During the hot temperature period, about 32.5% reduction in the substrate temperature was observed under the cooling treatment, compared to the control. Photosynthesis, transpiration rate, and leaf water potential of plants grown under the cooling treatment were significantly higher than those under the control. The first flowering date in the cooling was faster about 4 days than in the control. Also, the number of fruits was significantly higher than that in the control. No differences in plant height, stem thickness, number of internode, and leaf width were found between the plants grown under the cooling and control, except for the leaf length with a shorter length under the cooling treatment. However, root zone cooling influenced negligibly on eliminating delay in fruiting caused by excessively higher air temperature (> $28^{\circ}C$), although the substrate temperature was reduced by $3^{\circ}C$ to $5.6^{\circ}C$. These results suggest that the technique of lowering substrate temperature by using air-duct blow needs to be incorporated into the lowering growing temperature system for growth and fruit set of health paprika.
Intraspecific clonal interactions have important influences on a population structure of the cellular slime mold (CSM). This study was to investigate whether or not evolutionary change in a population could be induced by clonal competition, and to elucidate how various clones in a population evolve in a homogeneous environment of laboratory culture. The characteristic clones of Polysphondylium pallidum which had different resource consumption rates (RCR) and mating types I and II were selected for study. Investigation was conducted for 4 experimental time interval $(T_0-T_4)$; one experimental time interval took almost 10-14 days from inoculation to havest of fruiting bodies. Two sets of 50 clones were cultured from 50 clones at To, and RCR variations of the population were compared between $(T_0\;and\;T_4)$ for each set of clones. Each clone of the CSM had a diverse resource consumption rate, or growth rate, in a homogeneous and limited Cerophyl agar plate despite the passage of 48-56 generations from the beginning of the experiment. Diverse clones with different growth rate could coexist in one site of the homogeneous agar plate as well as heterogeneous soil microenvironment. When there was high clonal diversity of RCR, a clone in a population had high chances to encounter other clones with resultant increased clonal competition. In one set, 26 of 37 clones of mating type I were changed to mating type Il for the 4 experimental time intervals, which indicated that the rate of competitive exclusion among clones during total experiment from $(T_0\;to\;T_4)$ was 0.703. In another set, 31 of 37 clones of mating type I were changed to mating type II , having the rate of competitive exclusion 0.838. The frequency of each of mat~ng types changed by 0.93-1.29% in each successive generation. The competitive exclusion among clones occurred by 1.26-1.75% when approximately $2.6{\times}10^8$ bacterial cells were provided as food and thereafter one generation of myxamoebae of CSM elapsed at room temperature. This finding implicated that in the vegetative state of P, pallidurn there was 1.26-1.75% probabil~ty of evolutionary change per generation changing from one clone to another clone.
These studies were investigated for effect of concentration of sodium chloride on occurrence and growth of fruitbody in oyster mushroom, Pleurotus ostreatus. When the mycelia were inoculated and cultured on the PDA plate added with the different concentrations of sodium chloride, the growth of them were not affected at the concentration of 0.5%, started to decrease at 1.0% as compared with the PDA plate without sodium chloride, and they did not grow at 5.0%. When tested at the column filled with sawdust, the tendency was similar, but the decreasing rate of mycelial growth was gentle. In sawdust bottle cultivation sprinkled with the different concentration of sodium chloride solution at the different stage, the productivity and quality of fruitbody started to decrease at 3.0% of the solution and the yield at the treatment of 10.0% solution was only 47% of that in non-treated bottles. Treated at the different stages of mushroom development, treatment just after mushroom sprout did not show any different from that just after scraping spawn from the complete grown bottles. The sprinkle treatment at this stage looked better than that of the soaking with the solution before mushroom sprout. The yield at this treatment with 10% solution showed little damage with that of 90% of non-treated bottle. Sprinkle treatment during mushroom growth stage did not affect and showed almost same to non-treated bottle except the treatment with 10.0% solution. The treatment of this stage showed better results of growing characters than the treatment after and before mushroom sprouting even 10% solution. Morphological characteristics of oyster mushroom do not distinguish to treatment of different stage and concentration of sodium chloride solution. The analyzing data of quality on underwater usually used for oyster mushroom in Inchon, Yeongjong and Kanghwa area showed over-concentration to the standard for edible water and agricultural water. But it was not confirmed whether this water quality could affect to mushroom cultivation.
This experiment was conducted to evaluate on agronomic traits of Korean local adlay (Coix lachryma L.) germplasm, 281 cultivars (lines). The largest collected cultivars was from Kyungsangbukdo (56 cultivars), and the next was in the order of Cheonlabukdo (48 cultivars), Kyungsangnamdo (44 cultivars), Kyunggido (39 cultivars) Chungcheongnamdo (23 cultivars), Kangwondo (23 cultivars), Cheonlanamdo (17 cultivars), and Chungcheongbukdo (16 cultivars) in distribution of collected region for 281 cultivars. In the growth stages, days to emergence from seeding were required $16.4{\pm}2.03$ days, days to heading from emergence was required $73.6{\pm}5.87$ days, days to ripening from heading was required $52.3{\pm}4.91$ days and cultivation period was required $142.5{\pm}5.87$ days. Culm length was $210.3{\pm}16.39$ cm, no. of tiller per plant was $10.4{\pm}2.13$, culm diameter was $11.7{\pm}1.09$ mm, fruiting position was $4.6{\pm}0.82$ node, no. of main culm node was $11.1{\pm}0.78$, degree of lodging was $5.3{\pm}3.42$ and severity of leaf blight was $56.1{\pm}0.78$ in the growth characteristics of the germplasm. In the yield of component, no. of grains per $m^2$ was $5,938{\pm}2,152$, percentage of ripening was $67.7{\pm}33.12$, 1,000 grains weight was $123.8{\pm}33.76$ g and grain weight was $473.0{\pm}207.90$ g. Grain weight was correlates significantly positively with no. of main culm node, percentage of ripening and no. of grains per $m^2$ among agronomic traits. Correlation coefficient between grain weight and 1,000 grain weight was positive, while grain weight and severity of leaf blight was significantly negative. Correlation coefficient between grain weight and days to ripening from heading was significant negatively.
Studies were processed to confirm the differences of productivity according to sawdust size and effect of additives for sawdust cultivation of shiitake. In results of investigation until shiitake mycelia fully spread on surface of sawdust medium, mycelial growth in treatment of sawdust size $3{\sim}5\;mm$ with $CaCO_3$ and $KNO_3$, and treatment of sawdust sizw $3{\sim}5\;mm$ without $CaCO_3$ and $KNO_3$ was the fastest. And, in investigation of rate of weight reduction, treatment of sawdust size below 1 mm without $CaCO_3$ and $KNO_3$ was the highest. The tendency of fruiting was somewhat different according to treatments. In case of sawdust size below 1 mm with $CaCO_3$ and $KNO_3$, yield produced from 1st to 2nd flushing period was over 80%. However, in case of sawdust size below 1 mm plus $5{\sim}7\;mm$ without $CaCO_3$ and $KNO_3$, yield produced from 3rd to 4th flushing period was more than it from 1st to 2nd flushing period and rate of yield produced from 1st to 2nd flushing period to total yield was 34%. Results of investigation on productivity, yield and the number of fruit-bodies were remarkably different according to treatments. Sawdust size $3{\sim}5\;mm$ with $CaCO_3$ and $KNO_3$, whose total yield and the number of fruit-bodies were 568 g and 67, respectively, was the highest. And, total yield of medium of sawdust size below 1 mm plus $2{\sim}3\;mm$ without $CaCO_3$ and $KNO_3$ was lowest as 227 g and the number of fruit-bodies of medium of sawdust size below 1 mm plus $5{\sim}7\;mm$ without $CaCO_3$ and $KNO_3$ was lowest as 24. In investigation on amount of fruit-bodies over 10 g, medium of sawdust size $3{\sim}5\;mm$ with $CaCO_3$ and $KNO_3$ was highest as 397 g.
We analyzed saccharide by dividing and comparing Monosaccharide, Disaccharide and sugar Alcohol. At first, Glucose had outstanding contained quantity of ASI 7114 with 81.11 g/l even comparing with other mushrooms for medical use and edibility. And 119.98 g/l of Fructose was observed at Hericium erinaceum that was more contained quantity than Flammulina velutipes and Lentinus edodes. But, the most contained quantity observed in Ganoderma lucidum was ASI 7015 with 15.70 g/l that was the level of 1/8 approximately against Hericium erinaceum. Ribose was found at low level generally that was hardly contained. Xylose was also observed low level. ASI 7004 was detected at 0.96g/l that was the most content with imperceptible difference by comparing with other mushrooms for medical use and edibility. Next, 35.21 g/l of Trehalose, disaccharide was observed at Agaricus bisporus that was around 11 times of content than ASI 3.09 g/l that was the most content of Ganoderma lucidum. For ${\alpha}$-Lactose, Sparassis crispa has the most amount of 3.38 g/l that was around 12.5 times of ASI 7060 0.27 g/l that was the most content of Ganoderma lucidum. For Glycerol, sugar alcohol, 64.74 g/l was observed at Pleurotus eryngii. We knew it was around 8 times of ASI 7004 8.61 g/l that was the most content of Ganoderma lucidum. 0.72 g/l of Solbitol was observed at Flammulina velutipes. We knew it was around 2times of ASI 7003 0.31 g/l that was the most content of Ganoderma lucidum. Moreover most of Ganoderma lucidum didn't contain Solbitol. 2.96 g/l of Mannitol was observed at Agaricus bisporus. that was the most content among other mushrooms. Also Mannitol was contained in Lentinus edodes and leurotus cornucopiae only. Even Ganoderma lucidum didn't have Mannitol. At last, as a result of myo-Inosito content analysis, it was seemed not to be involved in any of mushrooms.
Oyster mushrooms are widely cultivated and consumed in Korea. P. ostreatus 'Suhan(ASI 2504)' is an ideal cultivar for mushroom farmers due to its dark pileus and thick stipe; however, as it is very sensitive to environmental conditions, an alternative cultivar is required. To develop a new cultivar, parental strains 'Suhan(ASI 2504)' and 'ASI 0665 (Heuktari)' were selected from various collected strains according to morphological characteristics. P. ostreatus 'Soltari' was developed by DiMon crossing between the dikaryotic strain 'Suhan' and the monokaryotic strain derived from 'Heuktari'. Thirty-eight of the 100 crossed strains were selected following analysis of mitochondrial genetic characteristics, and 'Soltari' was ultimately selected by continuous cultivation tests. The mitochondrial DNA profile of 'Soltari' was found to be the same as that of 'Heuktari, and a nuclear DNA profile of 'Soltari' was similar as those of the parental strains, 'Suhan' and 'Heuktari.' 'Soltari' mycelium grows adequately in moderate to high temperatures of $12-20^{\circ}C$, although its optimum temperature was found to be $30^{\circ}C$. Fruiting body production per 1.1-L cultivation bottle was approximately 158.8 g. Its stipe length and thickness were comparable to those of diameter and thickness were somewhat lower (42.72 vs. 51.33 mm and 18.18 vs. 22.46 mm, respectively). 'Soltari' was found to be more resistant to high $CO_2$ atmosphere than 'Suhan', and the color of the pileus of 'Soltari' was dark gray at high temperature. Therefore, it is suggested that this new cultivar 'Soltari' is a good alternative cultivar and will contribute to energy saving in oyster mushroom farms.

This study was performed to investigate the anti-diabetic mechanism of crude polysaccharides isolated from the fruiting bodies of Grifola frondosa. We treated 3T3-L1 adipocyte cells to observe whether the crude polysaccharides isolated from Grifola frondosa would stimulate insulin sensitivity. Significant insulin sensitizing activity was observed in the 3T3-L1 adipocytes, and giving the crude polysaccharide of Grifola frondosa with 1 nM of insulin caused glucose uptake to increase to a similar level as giving 50 nM of insulin alone. To confirm the mechanism for the anti-diabetic effect of the crude polysaccharides, we performed further examinations within $KK-A^{y}$ mice, an animal model of type 2 diabetes. The crude polysaccharides reduced blood glucose levels in the $KK-A^{y}$ mice for 2 weeks after feeding, and also significantly lowered plasma insulin levels. These results suggest that the anti-diabetic mechanism of the crude polysaccharide of Grifola frondosa is related to the enhancement of insulin sensitivity.
