Production of Polysaccharide by the Edible Mushroom, Grifola frondosa

  • The production of polysaccharide according to various developmental stages(mycelium growth, primordium appearance, and fruiting-body formation) in the edible mushroom Grifola frondosa was studied. The cap of the mature mushroom showed the highest amount of polysacchride. Mycelial growth and polysaccharide synthesis were optimal at pH 5 and $20^{\circ}C$. Polysaccharide synthesis was maximal after 12 days of cultivation, whereas maximum mycelial growth was shown after 18 days. Mannose, cellobiose and starch increased the level of polysaccharide as well as growth in submerged culture. Glucose and sucrose appeared to be good substrates for fruiting of Grifola frondosa.

  • Thirty-seven single spore isolates were obtained from specimens of ascomycetous fruiting bodies collected from Mt. Suri, Anyang in Korea. The fungal specimens and isolates were identified as Dumontinia tuberosa based on their morphological and cultural characteristics. This is the first record of this fungus occurring in Korea.

  • The caterpillar-shaped Chinese medicinal mushroom (DongChongXiaCao) looks like a worm in the winter and like a grass in the summer. The fruiting body has been regarded as popular folk or effective medicines used to treat human diseases such as asthma, bronchial and lung inflammation, and kidney disease. The fruiting bodies of Cordyceps militaris, C. pruinosa and Paecilomyces tenuipes that formed on the living silkworm (Bombyx mori) host were used in this examination. This study was carried out to investigate the soluble sugar, amino acid and fatty acid profiles in the fruiting-bodies. Soluble sugars such as glycerol, glucose, mannitol and sucrose were mainly found in the fruiting bodies of C. militaris, C. pruinosa and P. tenuipes. Total soluble sugar content was 29.23 mg/g in C. militaris, 8.61 mg/g in C. pruinosa and 24.00 mg/g 1in P. tenuipes on dry weight basis. Total free amino acid content was 14.09 mg/g 1in C. militaris, 34.60 mg/g in C. pruinosa and 17.09 mg/g in P. tenuipes. The content of oleic acid in fatty acids was above high more than 30% regardless of species.

Log Cultivation of the Birch Fungus Piptoporus betulinus using Betula davurica (물박달나무를 이용한 자작나무버섯의 원목재배)

  • A birch fungus, Piptoporus betulinus, is judged the mushroom with economical value that produces the fruiting body on Betula trees, but the mushroom rarely occurs in Korean birch forest. So, the fungus was applied to log cultivation for the production of fruiting body on Betula davurica. The fungus produce the fruiting bodies in the early August of the next year after inoculation in April and its mushroom matured in October. The mushroom produced 212 g to 1,298 g fresh weight in one or two mushrooms per a log. Thus, it is considered that log cultivation of the fungus has a potential to mass production of fruiting body and is expected to introduce to more researches to develop the use of this mushroom.

  • The study was conducted to investigate the characteristics of growth and fruiting in chestnut trees by SCB (Slurry Compostion and Biofilteration) liquid fertilizer instead of Chemical fertilizer. The experiment was done between April, 2008 and September, 2009, using seven years old grafting trees 'Tsukuba'. Fertilizer treatment was designed as follows; SCB liquid fertilizer, chemical fertilizer, organic fertilizers and no fertilized. Results were as follows; In the 2nd year, height and basal diameter gain of 8.04% and 25.90% over than the 1st year by SCB liquid fertilizer. The number of burrs per bearing mother branch showed no significant increment by SCB liquid fertilizer but fruiting rate was higher than other treatments. These results indicated that the SCB liquid fertilizer has a value of new eco-friendly fertilizer.

The fruiting bodies of Paecilomyces tenuipes that formed on the living silkworm (Bombyx mon) host were used in this examination. This study was carried out to investigate the proximate composition, soluble sugar, amino acid and fatty acid profiles, and contents of the bioactive ingredient including adenosine and D-mannitol in the fruiting-bodies. The moisture content was 57.56%. Soluble sugars found were glycerol, glucose, mannitol and sucrose, and the contents exceeded $24\;mgg^{-1}dry$ weight. Total free amino acid content was $17.09\;mgg^{-1}dry$ weight. Arginine, glycine, proline and tyrosine were main amino acids. The content of oleic acid in fatty acids was high. Adenosine was more abundant in fruiting bodies than corpus.

Ergosterol and Water Changes in Tricholoma matsutake Soil Colony during the Mushroom Fruiting Season

  • The purpose of this study is to understand spatio-temporal changes of active fungal biomass and water in Tricholoma matsutake soil colonies during the mushroom fruiting season. The active fungal biomass was estimated by analyzing ergosterol content at four different points within four replicated locations in a single circular T. matsutake colony at Ssanggok valley in the Sogri Mt. National Park in Korea during 2003 to 2005. The four points were the ahead of the colony, the front edge of the colony and 20 cm and 40 cm back from the front edge of the colony. Ergosterol content was 0.0 to 0.7 ${\mu}g$ per gram dried soil at the ahead, 2.5 to 4.8 ${\mu}g$ at the front edge, 0.5 to 1.8 ${\mu}g$ at the 20 cm back and 0.3 to 0.8 ${\mu}g$ at the 40 cm back. The ergosterol content was very high at the front edge where the T. matsutake hyphae were most active. However, ergosterol content did not significantly change during the fruiting season, September to October. Soil water contents were lower at the front edge and 20 cm back from the front edge of the colony than at the ahead and 40 cm back during the fruiting season. Soil water content ranged from 12 to 19% at the ahead, 10 to 11% at the edge, 9 to 11% at the 20 cm back and 11 to 15% at the 40 cm back. Our results suggest that the active front edge of the T. matsutake soil colony could be managed in terms of water relation and T. matsutake ectomycorrhizal root development.

Cultivation characteristics and genetic diversity of wild-type collections of Hericium erinaceus in Korea (국내 노루궁뎅이버섯 야생수집균주의 자실체 특성 및 유전적 다양성)

  • Wild-type isolates of H. erinaceus were collected from different geographical areas of Korea. Nineteen isolates were cultured on mushroom substrate for producing fruiting bodies. Of these, 14 isolates formed pinheads and fruiting bodies on the substrate. The morphological and cultivation characteristics of fruiting bodies were categorized by pinheading, fruit body formation, spine types, fresh weights, and colors. Microstructures, including spines, spores, and basidia on the fruiting bodies were observed using scanning electron microscopy. The H. erinaceus isolates demonstrated different PCR polymorphisms produced by universal fungal primers (UFPs) and were classified into four groups based on their high genetic diversity.

Enhanced GABA content from sodium alginate-induced Sparassis latifolia influences dendrite development in primary cortical neurons (해조류 기반 엘리시터 처리에 의한 꽃송이버섯의 GABA 함량 증가 및 흥분성 신경세포의 수상돌기 발달 억제)

  • Sparassis latifolia is a fungus abundant in β-glucan and amino acids and is highly valued as a medicinal mushroom. Among amino acids, γ-aminobutyric acid (GABA) is a free amino acid and has biological effects, such as increase/decrease of hypertension, improvement of cerebral blood flow, and prevention of dementia. In this study, biological elicitors were used to increase bioactive substances as a biofortification method. Sodium alginate extracted from seaweed (Sargassum horneri, Sargassum fulvellum, Sargassum fusiforme) were used as the elicitor. The levels of β-glucan and GABA in the mycelium and fruiting body grown by adding the elicitor to the medium were investigated. Addition of sodium alginate positively affected GABA production and negatively affected the β-glucan production in these fungi. Sodium alginates extracted from S. fulvellum induced the highest increase in GABA in the mycelium and fruiting bodies. Moreover, we investigated the effects of the extracts from mycelium and fruiting bodies on dendrite development in primary cortical neurons. We found that the extract from the fruiting bodies of sodium alginate treated fungi with increased levels of GABA inhibited the dendrite outgrowth of excitatory neurons, but not inhibitory neurons.

Identification and Characterization of Myxobacteria from Korean Soil (국내토양에서 분리한 점액세균의 동정및 특성)

  • We isolated a Myxobacteria strain from a soil sample obtained from Mt. Daedoon located in Choongnam, Korea. This strain, ARJ, secreted slime while swarmed on the surface of CT medium. It produced greenish yellow pigment in liquid or solid media, and the swarming edge showed green florescence under U. V. at 366 nm. It formed fruiting bodies when nutrient was exhausted, which is one of the most imkportant characteristics of Myxobacteria. The fruiting bodies did not have a stalk and consisted of naked myxospores when examined under the scanning electron microscope. These traits lead us to believe that this strain is very close to Myxococcus virescens. It showed antimicrobial activity, especially against Gram positive bacteria. Culture filtrate showed the activity but this was not due to protein. The culture filtrate also had proteolytic activity in which at least two enzymes are involved.
