Melanin-dispersing Effect of Vasopressin on Frog Skin Melanocyte (Vasopressin의 멜라닌 확산작용에 대하여)

  • It has been reported that vasopressin disperse the melanophore granule of frog skin. The author used hypophysectomized and adrenergic receptor blockaded animals in order to define the mechanism of vasopressin on the melanopore pigment of frog skin. The Rana niglomaculata which could be found in the Seoul area were used on this experiment. The amount of the following drugs were injected into the lymphatic sac of the frog; vaospressin $(0.05\;{\mu}g/g\;B.W.)$, dibenzylin $(0.05\;{\mu}g/g\;B.W.)$, and propranolol $(0.01\;{\mu}g/g\;B.W.)$. The following results were observed; 1. Vasopressin dispersed the melanin granules of melanocyte of frog skin. 2. The melanin granule dispersion activity of vasopressin was observed on the hypophysectomized frog. 3. The melanin granule dispersion was observed on the adrenergic receptor blockaded frog with dibenzylin or propranolol respectively, especially the later one was found to be more obvious. 4. The melanin granule dispersion was observed on the frog which was injected with vasopressin after alpha-receptor or beta-receptor blockade and the later one was found to be more obvious. 5. The melanin granule dispersion was more effective with the blockade of beta-receptor after the treatment with vasopressin on hypophysectomized frog.

  • The robust development of frog farming offered high economic benefits but created a large waste residue of frog bones and skin that received little attention. Over the years, inedible by-products have often been processed into biomolecules of potential value and environmental benefits, such as collagen, gelatin, and bioactive peptides. An overview of bioactive compounds on frog skins from various countries indicated that brevinin was the most abundant biological peptide found in frog skin. Other remaining compounds also possessed their highlighted activities, including antibacterial, stimulating insulin release and gastric hormone release, anti-cancer, and neuroregulatory. Notably, various components have been analyzed in the structure and sequence to give meaningful insight into clustering components related to their biological activity. This review may create a source of raw materials for the developmental research of by-products from frog skin and concomitantly reduce environmental pollution.

  • This study was accomplished to investigate the ultrastructure of mucous glands in dorsal skin of frog (Rana catesbeiana) by means of scanning and transmission electron microscopes. The dorsal skin of Rana catesbeiana is composed of epidermis and dermis. The cutaneous mucous glands consist of inner glandular epithelial cells and outer myoepithelial cells. Glandular epithelial cells are divided into four types by the microscopic ultrastructure; ER-rich cell, round secretory granule-containing cell, foam-like granule mass-containing cell, mitochondria-rich cell. Myoepithelial cell has a long elliptical nucleus and filled with fibrous materials in the cytoplasm. As a result of scanning microscopic observation, the surface of dorsal skin is covered with cutaneous protrusions. The opening sites of the mucous glands are irregularly distributed in dorsal skin.

Frog-inspired programmable nano-architectures for skin patches and medical applications

  • Nanoscale observation of attachment systems of animals has revealed various exquisite multiscale architectures for essential functions such as gecko's locomotion, beetles' wing fixation, octopuses' sucking and crawling. In particular, the hierarchical 3-dimensional hexanonal nano-architectures in the tree frog's adhesion is known to have the capability of the enhancement of adhesion forces on the wet or rough surfaces due to the conformal contacts against rough surfaces and water-drainable micro channels. Here, we report that tree frog-inspired patches using unique artificial 3-dimensional hexagonal structures can be exploited to form reversibly enhanced adhesion against various highly curved and rough surfaces in dry and wet condition. To investigate the adhesion effect of micro-channels, we changed the arrangement of microstructure and spacing gaps between micro-channels. In addition, we introduced the 3-dimensional hexagonal hierarchical architectures to artificial patches to enhance to conformal contacts on the various rough surfaces such as skin and organs. Using the robust adhesion properties, we demonstrated the self-drainable and comfortable skin-attachable devices which can measure EKG (electrokardiogramme) for in-vitro diagnostics. As a result, bio-inspired programmable nano-architectures can be applied in versatile devices such as, medical patches, skin-attachable electronics etc., which would shed light on future smart, directional and reversible adhesion systems.

$Na^{+}$ Metabolism in the Frog in the Winter Time (겨울철 개구리 (Rana temporaria)의 $Na^{+}$이온 대사)

  • $Na^{+}$ balance was studied in Rana temporaria, which hibenates in fresh water in the winter time. $Na^{+}$ uptake rate, skin $Na^{+}$ loss rate, urinary $Na^{+}$ loss rate and $Na^{+}-K^{+}$ adenosine triphosphatase(ATPase) were measured at two different temperatures $1{\sim}2^{\circ}C\;and\;20{\sim}24^{\circ}C$ respectively. The results obtained were as follows: 1. $Na^{+}$ uptake rates in the frog in an artificial Pond water (APW) were found to be $8.28{\pm}0.73\;and\;2.19{\pm}0.37\;{\mu}Eq/g/day\;at\;20{\sim}24^{\circ}C\;and\;1.0{\sim}2.5^{\circ}$ respectively. 2. $Na^{+}$ loss rate through the frog skin to APW were found to be $4.26{\pm}0.72\;and\;0.93{\pm}0.21\;{\mu}Eq/g/day$ at the same temperatures. 3. Mean rates of urinary $Na^{+}$ loss at $20{\sim}24^{\circ}C\;and\;3{\sim}4^{\circ}C$ were found to be $3.02{\pm}0.73\;and\;0.78{\pm}0.13\;{\mu}Eq/g/day$ respectively. 4. The activities of $Na^{+}-K^{+}$ activated ATPase of frog skin fragments were found to be $258{\pm}39.4\;and\;49.6{\pm}7.1\;{\mu}M\;Pi/g$ protein/hr at $24^{\circ}C\;and\;2^{\circ}C$ respectively. From the above results, it may be concluded that frogs can take up enough $Na^{+}$ through the skin from APW exceeding skin loss Plus urinary loss at $1{\sim}2^{\circ}C$. It is suggested that $Na^{+}$ transport across frog skin is closely related with $Na^+-K^+$ ATPase since $Q_{10}\;of\;Na^{+}$ uptake is much similar to that of the activities of $Na^{+}-K^{+}$ ATPase.

Effect of Ginseng Alcohol Extract on Short-Circuit Current Across the Frog Skin (인삼 알콜 추출물이 개구리 피부를 통한 short circuit current에 미치는 영향)

  • In an attempt to examine the effect of ginseng on sodium transport across the biological membrane, we have studied effects of ginseng alcohol extract on the short-circuit current(SCC) and the $Na^+-K^+$-activated ATPase activity in isolated frog skin preparations. 1. Ginseng alcohol extract applied to the mucosal surface of the frog skin significantly increased SCC at low concentration($1{\sim}10mg%$) but decreased SCC at higher concentration($50{\sim}250mg%$). 2. Similarly, when the drug was added to the serosal bathing medium, the SCC was stimulated at low doses($5{\sim}25mg%$) and inhibibited at high doses($50{\sim}250mg%$). 3. $Na^+-K^+$-activated ATPase activity of the frog skin epidermal homogenate was significantly inhibited by ginseng alcohol extract, the effect being proportional to the concentration of the drug in the incubation mixture. These results may suggest that a low dose of ginseng alcohol extrat enhances the transepithelial sodium transport probably by increasing the permeability of outer membrane of the transporting cell to sodium ion, whereas a high dose of drug reduces the sodium transport primarly by inhibiting $Na^+-K^+$ ATPase mediated active transport step.

Effect of N-ethylmaleimide(NEM) on $Na^+$ Transport Across the Frog Skin (N-ethylmaleimide(NEM)가 개구리 피부의 $Na^+$ 이동에 미치는 영향)

  • Studies have been conducted using isolated surviving skin of Rana temporalia in an attempt to evaluate the effect of N-ethylmaleimide (NEM) on the epithelial $Na^+$ transport. Active transport of $Na^+$ across the skin was estimated by measuring short circuit current (SCC). NEM administered to the outside surface of the skin in concentration of $0.5{\times}10^{-4}-2.5{\times}10^{-4}M$ induced $20{\sim}40%$ increase during the first 30 mintues, followed by a gradual reduction in SCC. With NEM above $4{\times}10^{-4}M$, SCC was inhibited from the beginning. Qualitatively similar results were obtained when NEM was added to the inside bathing medium. However, the concentration of NEM for a similar effect was much higher with the drug in the inside bathing medium than in the outside bathing medium. The oxygen consumption of the skin was inhibited by NEM of above $10^{-4}M$, the effect being of approximately the same magnitude as that on SCC. The activity of $Na^+-K^+$ ATPase of the skin was not inhibited by NEM below $10^{-3}M$, but it was dramatically reduced with $1.2{\times}M$ NEM. The effects of NEM $(10^{-4}M)$ on the SCC and oxygen consumption could be eliminated by adding cysteine $(10^{-4}-10^{-3}M)$ in the medium, indicating that the SH group is involved in the action of NEM in the frog skin. On the basis of these results, the mode of action of NEM on the $Na^+$ transport across the frog skin was discussed.

Histochemical Study on the Mucous Gland of the Frog (Rana nigromaculata) Skin under Dry Conditions (건조상태가 개구리(Rana nigromaculata) 피부 점액선에 미치는 영향에 대한 조직화학적 연구)

  • This study was performed in order to know the histochemical changes of mucosubstances in mucous glands under normal and dry conditions. The frogs, Rana nigromaculata Hallowell, were collected near Seoul area in mid-August. The experimental group was retained under dry conditions for 12, 24, 30 and 36 hrs to make a comparison to normal group. The results obtained were as follows: 1. Mucosubstances in mucous glands of the frog skin contain neutral mucin and sulfated and non-sulfated mucins. Mucosubstances in the cells of the gland neck are neutral mucin, while the gland body is composed of neutral, sulfated and non-sulfated mucins. 2. Mucosubstances in mucous gland of the frog skin decreased distinctively under dry conditions, as is evident from the hitochemical properties, te neutral mucins were influenced more clearly than acid mucins. 3. Histochemical compositions of the mucosubstances in the cell of the mucous gland (the gland neck and the gland body) were not changed. 4. The phenomena were considered as a general adaptation syndrome to keep homeostasis.

Histochemical Study on the Serous Gland of Frog(Rana nigromaculata) Skin under Dry Conditions (건조상태가 개구리(Rana nigromaculata) 피부 장액선에 미치는 영향에 대한 조직화학적 연구)

  • This study was performed in order to know the histochemical changes of mucosubstances in granular secretions of the serous gland under normal and dry conditions. The frogs were collected near Jinju and the experimental groups were retained under dry conditions for 24, 48, 72 and 96 hours to make a comparison with normal group. The results obtained were as follows: 1. The secreting granules in serous gland of the frog skin contained neutral and weakly acidic mucosubstances. 2. the secreting granules in serous gland of the frog skin were significantly decreased under dry conditions, as is evident from the histochemical properties. The neutral mucosubstances were decreased more than acidic mucosubstances. 3. Histochemical compositions of the mucosubstances of secreting granules in the lumen of the serous gland were not changed according to dry condition.

Studies on Adrenoceptors Involved in Regulation of Sodium Transport in Frog Skin (개구리 피부에 있어서 Na 수송을 조절하는 Adrenoceptors에 관한 연구)

  • To ascertain the existence of various adrenoceptors involved in active transport of sodium in the frog skin and to delineate their physiological roles, the influence of various adrenergic agonists and antagonists on the potential difference (PD), short-circuit current (SCC) and total skin conductance (TSC) of the isolated frog skin of Rana nigromaculata were investigated. PD and SCC were determined with Ussing's technique. Drugs were administered to the serosal side of the skin. Experimental results were summarized as follows: 1. The responses to norepinephrine (NE, $6{\times}10^{-8}-6{\times}10^{-5})M$), phenylephrine (PE, $5{\times}10^{-6}-5{\times}10^{-4}M$) and epinephrine (Epi, $5.5{\times}10^{-7}-5.5{\times}10^{-5}M$) were characterized by marked elevation of PD & SCC in dose-related fashion, but the maximal effect attained by Epi was less than those of NE and PE. 2. These increments of PD & SCC were significantly inhibited by prazosin $(2{\times}10^{-6}M)$, a speciflc ${\alpha}_1$-adrenoceptor blocker. The stimulatory effect on PD & SCC were completely abolished by phenoxybenzamine (PBZ, $3.3{\times}10^{-5}M$), an irreversible ${\alpha}$-adrenoceptor blocking agent. Furthermore, with a larger doses of Epi produced marked decline of PD & SCC after the PBZ pretreatment. 3. Isoproterenol (ISP), a ${\beta}$-adrenoceptor agonist, in concentrations ranging from $5{\times}10^{-7}$ to $5{\times}10^{-6}M$ produced dose-related decrease in PD & SCC, which could be abolished by pretreatment with propranolol $(4{\times}10^{-6}M)$, a specific ${\beta}$-adrenoceptor blocker. It was further noted that the effects of Epi on PD & SCC were markedly potentiated by Propranolol pretreatment. 4. Clonidine as well as guanabenz produced increases in PD & SCC and these effects were inhibited more specifically by prazosin pretreatment than by yohimbine. These results indicated that there exist in the frog skin two distinctive types of adrenoceptors, ${\alpha}$ and ${\beta}$, which roughly corresponds to those in mammals, and that the ${\alpha}$ type of adrenoceptors mediate the stimulation of PD & SCC, whereas ${\beta}$-adrenoceptors mediate the inhibition. However, based on evidence at hand, no conclusion could be drawn on the subtype of ${\alpha}$-adrenoceptors which is involved in the stimulation of sodium transport in the frog skin.
